Abstract
본 발명은 파이코시아노빌린 (PCB)를 포함하는 펩타이드, 또한 상기 펩타이드 및 PCB의 확인된 신경보호 및/또는 신경재생 효과를 이용하여 이를 의약으로 사용하는 용도에 관한 발명이다. 부가적으로, 본 발명은 상기 펩타이드 및 PCB와 단백질들 또는 다른 펩타이드들의 약학적 조합이 상승 효과를 갖는다는 점을 이용하여, 신경 퇴행성 및 허혈성 손상을 포함하는 중추 신경계 질병들의 치료에 사용하기에 적합한 약학적 조성물을 제공하는 것에 관한 것이다.
Description
본 발명은 생명과학, 약물학, 신경생물학, 생명공학 및 의과학에 관한 것으로서, 특히 신경학 및 내과학에 관한 것이다. 본 발명은, 일반적으로, 허혈성, 염증성 및/또는 신경퇴행성 손상으로부터 진전되거나 또는 유래하는 중추신경계 질환들에 대한 치료법들을 개발하는 것에 관한 것이다.
본 발명은, 조직 저산소증을 나타내고 염증성 및 신경퇴행성 특성들을 나타내는 뇌혈관 질환 치료를 위해서, 그 구조 내에 피롤 고리들을 갖는 C-파이코시아닌 (C-Phycocyanin: C-파이코) 유도체 또는 그 부분을 사용하는 사항에 기초한다. 본 발명은 또한 다른 생분자들과 함께 상기 화합물이 조합된 약제학적 조성물을 포함하며, 상기 조성물은 예방적 및 치료적으로 투여된다.
뇌혈관성, 탈수초성 (demyelinating) 및 신경퇴행성 질환들은 신경과학계에서 새로운 개척 분야로 떠오르고 있다. 뇌혈관성 사고들 (cerebrovascular accidents: CVA)은 65세 이상의 전세계 인구 중 대략 5%에 영향을 미치며, 영향을 받은 개인들에게는 심각한 물리적 장애를 야기할 수 있다 (World Health Organization: Statistical Information System. World Health Organization, 2004). 90% 이상의 사망이 50세 이상의 환자들에게 발생되며, 생존 환자들 중 대략 15% 내지 30%는 몇몇 유형의 후유증을 겪는다 (Buergo-Zuaznabar M A, et al. Revista Electronica de las Ciencias Medicas en Cienfuegos: 2-22; Miranda Q J A. Cerebrovascular Diseases, 2004; 1: 17-21; Rosamond W, et al. Circulation. 2007:169-171).
세계 보건 기구 (World Health Organization: WHO) 데이터에 따르면, 대략 80% 정도의 CVA는 허혈성 유형으로서, 혈전 (thrombus) 또는 색전 (embolus)에 의해서 주뇌 동맥들 중 하나가 급성 폐색됨으로써 발생하고 (World Health Organization: Statistical Information System. World Health Organization, 2004), 해당 동맥에 의해서 관개되는 (irrigated) 영역의 관류 (perfusion)를 감소시킨다.
신경보호는 예방적이고 치유적인 치료 전략으로서, 허혈과 같은 CNS 질환들 중에 발생되는 병리적 뉴런 손실을 예방하기 위한 근본적 목표를 갖는다. 신경재생은 다발성 경화증 (multiple sclerosis: MS)과 같은 신경염증성 및 신경퇴행성 질환들에서 발생되는 손상을 회복시키기 위한 근본적 목표를 갖는다.
뇌 허혈의 치료의 주된 목표는: (1) 허혈 구역의 크기를 감소시켜서, 인접 영역으로의 확장 가능성을 제한하고; (2) 적절한 치료적 윈도우 내의 회복가능한 반음영 구역 (recoverable zone of penumbra)으로 세포 사멸이 점진적으로 진행하는 것을 제한하는 것이다 (Muhammad S H A S. Eur Neurol. 2008; 59:4-14).
신규 신경보호 및/또는 신경재생 치료제 후보물질은 허혈 과정 도중 및 이후에 점진적 뇌 손상으로 이어지는 세포 대사 생화학적 과정들을 차단 및/또는 약화시킬 수 있어야 한다. 더 나아가, 이러한 물질들은 뇌 손상 병리 메카니즘 내의 가능한 약물학적 표적들에 대한 광범위한 스펙트럼을 커버할 수 있어야 한다 (Ovbiagele B, et al. Curr Treat Options Cardiovasc Med. 2003; 5:441-449).
허혈성 유형의 질환들뿐만 아니라 신경퇴행성 질환들에서 신경보호제 및/또는 신경재생제로서의 효과를 갖는 천연 물질들로부터 추출된 화합물들이 보고된 바 있다. 몇몇 연구들은 천연 및 합성 카나비노이드류 (cannabinoids)가 뇌 허혈증 (Mauler F, el at. J Pharmacol Exp Ther. 2002; 302(1): 359-68; Iadecola C. Curr Opin Neurol. 2001; 14: 89-94; Sinor A D, et al. Neurosci Lett. 2000; 278(3): 157-60), 다발성 경화증 (Baker D, Pryce G, Croxford J L, Brown P, Pertwee RG, et al. Nature. 2000; 404(6773): 84-87), 헌팅턴병 (Lastres-Becker I, et al. Brain Res. 2002; 929(2): 236-42) 및 파킨슨병 (Lange J M, et al. DDT. 2005; 10(10): 693-702)에서, 아미노산 및 염증 매개체들의 방출을 감소 및 차단함으로써, 다른 항산화 메카니즘 및 흥분세포독성 (excitotoxicity) 억제에 의해서 신경보호 효과를 나타낼 수 있다는 점을 보고한 바 있다.
이러한 사실에도 불구하고, 대부분의 경우, 각 투여에 있어서 카나비노이드류 화합물들의 농도에 의존하는 투여량 의존성에 따른 카나비노이드류와 관련된 중요한 부작용들이, 소비자의 경험 및 소비 시점에 따라서 존재한다.
급성 효과들이 보고된 바 있는데, 이는 상당 부분 카나비노이드류의 항콜린성 (anticholinergic) 효과에 기인한 것으로서: 구강 건조, 안구 충혈, 시각 흐림, 혈압 감소, 심장 박동 증가, 반응 능력 감소, 인지 감각 증가, 조정능력 상실 및 정신운동 속도의 저하를 포함한다. 만성 효과 또한 보고된 바 있으며, 예를 들어: 암 발생 증가를 수반할 수도 있으며, 폐암의 경우에는 흡연자들의 발병률을 초과하는, 면역 체계 기능장애를 들 수 있다. 급성 심근경색의 위험성 증가, 불임, 간염 환자들에서 간 섬유증 및 간질에 대한 잠재적 위험성 증가는 카나비노이드 소비자들 사이에서 현재까지도 논쟁의 대상이 되고 있으며; 카나비스 소비가 환각, 시공간 지각의 왜곡, 또는 이인증 (depersonalization) 및/또는 현실감 소실 (derealization) 현상과 같은 인지 변화를 유발할 수 있다는 사실에 비추어 볼 때, 일부 정신적 효과들도 논쟁의 대상이 되고 있다. 이외에도, 단기 기억 상실 또는 사고 속도 저하와 같은 인지력 장애, 비가역적 인지력 변화, 기존 정신적 장애의 악화, 정신분열증 위험의 증가뿐만 아니라, 불안증, 정신적 유형의 급성 위기감, 도취감, 과도한 수다증도 논쟁의 대상이 되고 있다. 경우에 따라서는 다른 약물들에 대한 소비를 유발하는 카나비스 의존증뿐만 아니라, 기분 변화 (우울 및/또는 광증), 사회적 주변성도 언급된 바 있다. 특히, 임신 중 카나비스 소비는 태어난 후손의 청소년기에 인지 및 정신병리학적 변화를 유도할 수도 있다.
C-파이코는 스피룰리나 플란테시스 (Spirulina platensis)와 같은 일부 청녹 조류에서 발견되는 빌리단백질 (biliprotein)이며, 입증된 영양 및 세포보호 특성들을 지니는 바, 세계 각국에서 식이보충제로서 종종 사용된다 (Bockow B I. United States Patent. No. 05709855 (1998); Kay, R.A. Crit. Rev. Food Sci. Nutr.1991; 30: 555-573; Gonzalez De R C, et al. Life Sci.1993; 53: 57-61).
C-파이코에 대한 많은 연구들이 그 항산화적 특성들을 입증하기 위한 것이었다. C-파이코 자유 라디칼 격리 작용 (C-Phyco free radical sequestrating action)이: (1) 화학-발광, 및 (2) 2-데옥시리보오스 손상의 저해에 대한 분석에 의해서 입증된 바 있다 (Romay C, et al. Inflamm. Res. 1998; 47:36-41; Bhat V B, et al. Biochem. Biophys. Res. Commun. 2000; 275:20-25).
아스코르빅산 + Fe2 + 처리 이후 (Romay C, et al. Inflamm. Res. 1998; 47:36-41) 또는 2,2' 아조비스 2-아미노프로판 하이드로클로라이드 (HAAP)로 처리한 이후에, C-파이코가 쥐 간 마이크로좀의 지방 과산화물의 증가를 현저하게 저해하는 것으로 증명된 바 있다. HAAP는 자유 라디칼 형성의 저해제이며 (Bhat V B, et al. Biochem. Biophys. Res. Commun. 2000; 275:20-25), 따라서 이러한 화합물이 지방 과산화 반응에 대한 훌륭한 저해제라는 점을 알 수 있다.
반면에, C-파이코는 ONOO-에 의해서 매개되는 DNA에 대한 산화적 손상을 저해함으로써 항종양제로서 기능하며, 종양 번식을 억제한다 (Li B, et al. Biomed Pharmacother. 2005; 59: 551-60). 더욱이, 이러한 천연 화합물이 혈소판 응집을 저해하는 것으로 알려진 바 있다 (Hui-Fen Ch, et al. British Journal of Nutrition. 2006; 95: 435-440).
1-3 mg/mL 농도의 C-파이코는, 뇌 입상 세포들 (brain granulous cells)의 배양에 있어서, 칼륨 및 혈청 부존재로 인한 뉴런 사멸을 24시간 동안 방지할 수 있다 (Rimbau V, et al. Naunyn Schmiedebergs Arch Pharmacol. 2001; 364:96-104).
C-파이코 활성은 카이닌산 (kainic acid)에 의해서 유도된 쥐의 뇌 손상 모델에서도 확인할 수 있다 (Rimbau V, et al. Neuroscience Letters.1999; 276: 75-78). C-파이코를 100 mg/kg의 용량으로 투여함으로써 징후 및 변이를 감소시켰는데, 이는 미처리된 대조군 동물들 대비 처리된 동물들에서의 감소를 통해서 입증된다.
이에 더해서, C-파이코는 COX-2를 저해하는 것으로 알려져 있는데: 이는 1) 분리된 효소의 분석 및 (2) 완전 혈액 분석 (Reddy C M, et al. Biochem. Biophys. Res. Commun. 2000; 277:599-603)을 통해서 입증되었다.
C-파이코를, 마우스 및 쥐에서 카라지닌 (carragenin) 및 글루코오스 산화제에 의해서 유도된 부어오른 발 모델과 (Romay Ch, et al. Pharm. Pharmacol. 2000; 52: 367-368; Madhyastha HK, et al. J Cell Mol Med. 2008), 아라퀴돈산 (araquidonic acid)에 의해서 유도된 마우스-귀 염증 모델 (Romay Ch, et al. Pharm. Pharmacol. 2000; 52: 367-368)에 있어서, 100-200 mg/kg의 투여량으로 구강 경로에 의해서 투여하는 경우, 부종을 감소시킬 수 있는 것으로 알려져 있다.
C-파이코를 사용하여 다른 동물 모델들에 대해서 수행된 주된 연구들을 요약하는 검토 내용도 있다 (Curr Protein Pept Sci. 2003 Jun; 4(3):207-16). 대부분의 실험들에서 치료적 효과를 관찰하기 위해서는 고용량의 C-파이코를 투여하여야만 했다 (100 mg/kg 내지 300 mg/kg).
파이코시아노빌린 (phycocyanobilin: PCB)은 C-파이코의 발색단 (chromophore)으로서; 화학적 관점에서 보면, 단백질 단편 없이 피롤 고리들이 존재한다는 특징을 갖는다.
인터페론 (IFNs)은 초기에는 항바이러스 활성을 갖는 가용성 단백질로서 발견되었으며: IFNs 타입 I (IFN 알파 및 베타) 및 타입 II (IFN 감마)로 분류될 수 있다. 비록, IFNs 알파 및 베타가 공통된 복합 수용체를 사용하는 것으로 일반적으로 알려져 있지만, 몇몇 연구들에 따르면 특정 생물학적 효과들을 유도하는데 있어서 IFN 알파 및 베타의 용량 면에서 차이점이 있는 것으로 보고된 바 있다. 이러한 생물학적 효과들에는, IFN-특이 유전자의 차별적 유도 (Rani MRS, et al. J Biol Chem 1996, 271: 22878- 22884; Platanias LC, et al. J Biol Chem 1994, 269:17761-17764), 다양한 성장 인자들의 저해 효과 (Rosenblum MG, et al. J Interferon Res 1990, 10: 141-151) 및 적혈구생성 효과 (Means RT, et al. 알파 및 베타 IFNs 사이의 서로 다른 신호전달 현상에 대한 개연성 있는 설명은, 베타 IFN에 특이적인 수용체와 결합하는 인산화단백질의 존재이며, 이는 인산화 타이로신인 것으로 보이고, 알파 IFN (IFNAR1)의 수용체 1과 결합한다 (Croze E, et al. J Biol Chem. 1996: 271: 33165-33168; Platanias LC, et al. J Biol Chem. 1996, 271: 23630-23633).
알파 및 베타 IFN의 작용 메카니즘은 매우 복잡하다. 이러한 두 가지 사이토카인들은 서로 다른 신호전달 경로를 통해서 작용한다. 후자는 실험적으로 입증되었다. JAK 키나아제 TYK2가 존재하지 않는 UIA 세포들에서 수행된 연구들로부터 (Velazquez L, et al. 1992. Cell 70: 313-322), 이러한 세포들이 알파 IFN에 결합할 수 없고, 또한 반응할 수 없지만, 베타 IFN과는 결합 및 반응이 가능하다는 점이 밝혀졌다 (Pellegrini S, et al. 1989. MS에서 알파 IFN으로 수행된 임상 실험들 (CT)의 경우 낮은 효율을 나타내었다 (Gilhus EN, World Neurology 1995, 5: 10-12; Sheridan P (ed), Multiple Sclerosis Research in progress 1993-1994. Clinical Trials. International Federation of Multiple Sclerosis Societies, London, 1995, pp. 3-35; Trials with Alferon, human leukocyte interferon alpha. Clinical Trials Monitor, 1997; 4 (12): 4).
베타 IFN은 미국 식약청 (FDA)에 의해서 MS에 대해 승인된 약물들 중의 하나이지만, 그 효과가 좋지 않고, 약효를 위해서 고용량을 투여하여야 하는 의존성이 있다는 점이 보고된 바 있다 (Zaragoza Garcia F et al. Farm Hospit. 2002; 26:294-301).
이에 더해서, IFN 치료법의 경우, 투여량 및 투여 경로에 따라서 부작용이 보고된 바 있다. 환자들이 각 주사 후 24-48 시간 사이에서 발열, 근육통, 떨림 및 일반적 불편함을 겪을 수도 있다. 환자들 중 5%에서는 주사 부위의 괴사가 발생된다.
한편, 인터루킨 2 (IL-2)의 주된 인 비보 작용들 중 하나는, 흉선 발달 및 조절 T 세포 (Treg) 말초 확장 (cTreg)을 촉진하는 것이다. IL-2 또는 IL-2R 베타 결핍 마우스에서 Treg 활성의 손실은 심각한 항원 의존성 임파선염에 이은 치명적 자가면역성을 야기한다. IL-2 의존성 Treg의 존재는 몇몇 양자 전이 (adoptive transfer) 및 유전적 실험들에 기초한다. 최근에, Treg가 후-허혈성 염증 뇌 손상 (post-ishcemic inflammatory brain damage)의 뇌 보호 조절자로서 필수적인 기능을 수행한다는 점이 밝혀진 바 있다 (Liesz A et al. Nat Med 200915, 192-199).
(MS와 같은) 자가면역 질환 및 뇌 허혈증은 Treg의 상대적 결핍이라는 특성을 갖는다. 따라서, Treg 확장은 이러한 질병들을 개선할 수 있다. IL-2는 인 비보에서 Treg 확장을 야기함으로써, 이러한 치료 양상의 잠재적 임상 적용가능성을 열어준다 (Liu R. Eur J Immunol. 2010; 40:1577-89).
IL-2는 생물학적 반응 변형자로 간주되며, HIV 뿐만 아니라, 흑색종 및 신장 세포 종양과 같은 암 치료법에 사용되어 왔다. IL-2는 고용량으로 투여되는 경우들이 테스트된 바 있는데, 이는 저용량의 경우 원하는 치료적 효과를 달성하지 못하기 때문이다.
고용량 투여법은 IL-2를 정맥 내 경로를 통해서 매 8시간 간격으로 투여하는 것을 의미하는데, 환자가 견뎌낼 수 있는 경우, 15회까지 투여된다. 이러한 투여 스케줄은 현저한 부작용을 나타내는데, 이러한 부작용은 대부분의 경우 치료를 중단하면 멈추는 가역적인 것들이지만, 일부 부작용들의 경우는 매우 심각해서, 환자들의 입원 치료를 요하고, 때로는 약물을 투여하는 동안 집중적인 간호를 필요로 한다.
특히 관심을 끄는 또 다른 물질은 GHRP-6로 알려진 펩타이드이다 ("Growth Hormone Releasing Peptide-6"). 이러한 펩타이드는, 원래는 창자 메타-엔케팔린 (bowel meta-encephalin)으로부터 유래되는 것으로 알려져 있는 바, 이는 이후에 인간을 포함하는 다양한 포유류 종들에서 예기치 못한 성장 호르몬 (GH) 분비자극성 효과를 나타내는 것으로 보고되었다 (Bowers CY, et al. Endocrinology. 1984, 114: 1537-45; Pandya N, et al. J Clin Endocrinol Metab. 1998, 83:1186-9). 상기 분자는 다양한 형태의 왜소증을 차별적으로 임상 진단하기 위한 분비자극성 시약으로서 인간에 정맥 내 투여되어 왔다 (Popovic V, et al. Lancet. 2000; 356:1137-42).
GHRP-6는 CNS에서 인슐린형 성장 인자 1 (IGF-1) 발현을 증가시킨다 (Frago L.M, et al. Endocrinology 2002, 143:4113-4122). IGF-1는: (1) 희돌기교세포 (oligodendrocyte) 성숙과 관련된 과정들의 증가 (Wilson H.C, et al. GH/IGF-1 축의 노화는 GH 생산 및 분비를 자극하는 치료법들을 통해서 회복되어야만 한다. GHRP-6를 사용하여 성체 쥐들을 장기간에 걸쳐 전신적으로 치료하게 되면 시상하부 및 소뇌와 같은 몇몇 뇌 영역들에서 IGF-1 수준이 증가한다. 또한, 일반적으로 항-세포사멸 작용과 관련된 세포 내 신호전달 경로는 이러한 영역들에서 활성화된다. 비정상적으로 높은 소듐 글루타메이트 농도는 뉴런 과흥분을 일으킴으로써 세포 손상 및/또는 사멸을 야기할 수 있다. 본 발명은 전술한 문제점들을 해결하기 위한 것으로서, 첨부된 서열목록에 나타난 서열을 가지며, 테트라-피롤 구조를 갖는 발색성 (chromogenic) 펩타이드 (PCB-aa)를 제공한다. 본 발명에서는, 최초로, 이러한 화합물들이 조직 허혈 또는 퇴행의 예방 또는 치료에 사용되기에 적합한 특성들을 갖는다는 점을 입증하였다. 특정 구현예에서, 상기 화합물들은 허혈 및 신경퇴행성 질환 치료에 사용될 수 있다.

PCB-aa 및 PCB 펩타이드들은 PCB-aa의 경우 25배 더 낮은 몰농도, 또한 PCB의 경우 10배 더 낮은 농도에서 보호 효과를 나타낸 반면에 (2 μM의 PCB-aa 및 5 μM의 PCB가 손상된 세포들 중 100%를 보호), 대략 75% 정도의 세포들을 보호하는데 50 μM의 C-파이코가 필요하였다.
부가적으로, 특정 실시예를 통해서, 동물들을 PCB-aa 및 PCB로 처리한 경우에, 몽고 게르빌루스쥐 (Mongolian Gerbils)에서 뇌 허혈성 재관류 모델 I/R (brain ischemia reperfusion model I/R) 중 경색 부피의 감소가 관찰되었다. 결과는 PCB (43,1%)에 비해서 PCB-aa (49,2%)의 경우 더 높은 효율성을 보였다.

비록 알파 및 베타 IFN들이 타입 I IFN들이며, 공통의 복합체 수용체를 사용하지만, 몇몇 연구들에 의하면, 알파 및 베타 IFN들이 특정 생물학적 효과들을 야기함에 있어서 특성 차이를 나타낸다는 점이 알려져 있다.
특정 실시예에서, PCB-aa/IFN-a 및 PCB/IFN-a 조합들에 대한 평가를, 실험적 자가면역 뇌척수염 (experimental autoinmune encephalomyelitis; EAE) 모델 중 예방적 스케줄에서 수행하였으며, 질병 진전의 예방 면에서, 각각 개별적인 활성 성분들에 비해서 상기 언급한 조합들에서 상승 효과가 나타났다.
또 다른 특정 실시예에서, PCB-aa 및 PCB와 IFN-a 및 IFN-b 조합을 그들 각각의 개별적인 활성 성분들과 비교하여 평가하였는 바, 3,375 mg/Kg 투여량의 PCB-aa, 750 ㎍/Kg 투여량의 PCB 및 500 ng/Kg 투여량의 2가지 IFNs에 대해서 평가한 결과, IFN-a와 PCB 및 PCB-aa의 조합에 대해서 각각 83,3% 및 89,3%; IFN-b와 PCB 및 PCB-aa의 조합에 대해서 각각 87,0% 및 93,6%의 뇌경색 부피의 감소 면에서의 상대적 효율성이 나타났으며, 이는 몽고 게르빌루스쥐에서 뇌 I/R 모델 중에서 각각 개별적인 활성 성분들보다 더 높은 것이다.
부가적으로, 또 다른 특정 실시예에서, PCB-aa/IFN-b 및 PCB/IFN-b 조합들을 평가하였으며, 일일 투여량으로 3,375 mg/Kg의 PCB-aa/Kg, 750 ㎍/Kg의 PCB 및 6회 복용량의 500 ng/Kg의 IFN-b를 투여하였다. 결과들은 EAE 모델 중, 임상적 징후들의 감소에 있어서 각각 개별적인 활성 성분들의 수치보다 더 큰 수치를 나타내었다. 따라서, EAE 동물 모델에서의 치료적 효과 역시 검증되었다.
다른 실시예들은 EAE 모델 중에서, 다른 경로를 통해서 투여된 (복강, 비강, 구강 및 직장) 약학적 조성물로서 PCB-aa/IFN-b 및 PCB/IFN-b의 조합들에 대한 평가를 포함하며, 임상적 징후들 면에서 유사한 약학적 효과가 나타났다. 전술한 약학 조성물의 일부를 구성하는 화합물들은 동일한 환자 내에서 단일 치료 과정 동안 동시에 또는 개별적으로 투여될 수 있다. 본 발명에서 언급된 약학 조성물들은 비구강, 비강 또는 직장 투여용일 수 있으며; 이러한 투여 경로를 위해서 적당한 부형제들을 포함할 수 있다.
또 다른 특정 실시예에서, PCB-aa, PCB 및 IL-2의 뇌 허혈증에 대한 효과를 평가하였다. PCB-aa/IL-2 및 PCB/IL-2의 조합은 활성 성분들 독립적일 때에 비해서, 몽고 게르빌루스쥐의 뇌 허혈증 모델에서 뇌 경색 부피의 감소 면에서 상승 효과를 나타내었다 (PCB-aa에 대해서 49,2% 효율성, PCB에 대해서 43,1% 효율성, IL-2에 대해서 25,8% 효율성, PCB-aa/IL-2 조합에 대해서 84,3% 효율성 및 PCB/IL-2에 대해서 74,5% 효율성).
다른 한편으로, PCB/GHRP-6 조합 및 독립적인 활성 성분들에 대한 평가를 수행하였는 바, 몽고 게르빌루스쥐 뇌 I/R 동물 모델 중에서 이들을 복강내 투여함으로써 수행하였다. 형태측정 (morphometric) 관점에서 볼 때, 조합의 경우 상승 효과가 관찰되었는 바, PCB에 대해서 35.8% 및 GHRP-6에 대해서 36.1%인 경우에 비해서 85%의 효율성이 관찰되었다.

실시예 1. 메탄올 처리에 의해서 얻어진 파이코시아노빌린 (PCB) 및 발색성 펩타이드들 (PCB-aa)의 질량 스펙트럼
도 1A는 C-파이토 메탄올 추출물의 분별한외여과 (differential ultrafiltration)에 의해서 얻어진, 발색단 PCB에 해당하는 m/z 신호 587.26을 도시한 것이다.
도 1B, C 및 D는 C-파이코의 트립신 소화에 의해서 얻어진 PCB-aa의 질량 스펙트럼 패턴을 도시한 것이다.
실시예 2. PC12 세포주에서 소듐 글루타메이트에 의해서 유도된 손상에 대해서 C-파이코, PCB 및 PCB-aa가 갖는 신경보호 효과
PC12 세포들 (1,5x104 세포들/웰)을 24시간 동안 C-Phyco (25, 50 μM), PCB (0.5; 1; 5 μM) 또는 PCB-aa (0.25; 1; 2 μM)로 사전처리한 다음, 대응되는 산물 (다른 투여량)과 함께 50 μM 소듐 글루타메이트로 4시간 동안 공동배양하였다. 세포 생존도는 (3-(4,5-디메틸티아졸-2-일)-2,5-디페닐테트라졸륨 브로마이드 방법 (MTT)에 의해서 측정하였으며, 대조군에 대한 백분율을 도 2에 도시된 바와 같이 측정하였다. C-파이코의 세포 생존도와 유사한 세포 생존도를 얻기 위해서는 더 낮은 농도의 PCB 및 PCB-aa가 필요함을 알 수 있다. PCB 및 C-파이코와 유사한 효과를 달성한 PCB-aa (펩타이드 1) 농도들은 상기 화합물들에 비해서 더 낮았다.
실시예 3. 몽고 게르빌루스 쥐 I/R 모델 중에서, 경색 부피 감소에 의해서 측정한 PCB-aa 펩타이드들의 신경보호 및/또는 신경재생 특성
허혈성 사건 후 30분, 3시간, 6시간 및 12시간 경과한 시점에, 동물들을 식염수 용액 또는 PCB-aa 펩타이드들의 축적량 (매회 3.375 mg/kg)으로 복강내 투여에 의해서 처리하였다.
매 처리 당 유효 백분율은 하기 식에 따라서 계산하였다: 유효 % = (1 - Vi / VI/R) x 100. Vi: 대응되는 산물로 처리한 허혈군의 경색 부피; VI/R: 식염수 용액으로 처리한 허혈군의 경색 부피.
도 3에 도시된 바와 같이, 서열번호 1 내지 서열번호 5로 구성되는 발색성 펩타이드들 (PCB-aa)로 처리된 동물들은 식염수 용액으로 처리된 허혈군에 비해서 경색 부피 면에서 현저한 감소를 나타내었다.
실시예 4. 게르빌루스 쥐의 양방향 허혈-재관류 모델에서 PCB/ IFN-a, PCB-aa (펩타이드 1)/ IFN-a, PCB/IFN-b 및 PCB-aa (펩타이드 1)/IFN-b 조합의 효과
허혈성 사건 후 30분, 3시간, 6시간 및 12시간 경과한 시점에, 도 4에 도시된 투여량에 따라서, 동물들을 식염수 (복강내 투여) 또는 개별적인 화합물들로서 소정 투여량의 PCB (750 ㎍/Kg, 복강내 투여), 또는 PCB-aa (펩타이드 1) (3.375 mg/kg), IFN-a 및 IFN-b (500 ng/Kg, 피하 투여) 또는 PCB/IFN-a, PCB/IFN-b, PCB-aa (펩타이드 1)/IFN-a 및 PCB-aa (펩타이드 1)/IFN-b의 조합으로 처리하였다. 유효 백분율은 실시예 3에 서술된 바에 따라서 계산하였다.
각 군별로 뇌 경색 부피 감소는, 평가된 처리들이 효과적이라는 것을 입증하며 (도 4), PCB로 처리된 군에서의 경색 지수 감소는 43.1%; PCB-aa (펩타이드 1)의 경우 49.2%; IFN-a의 경우 35,4%; IFN-b의 경우 37.0%; PCB/IFN-a의 경우 83.3%; PCB-aa (펩타이드 1)/IFN-a의 경우 89.3%; PCB/IFN-b의 경우 87.0%; PCB-aa (펩타이드 1)/IFN-b의 경우 93.6%의 효율로 나타났고; 이는 조합으로 처리된 동물들에서 두 가지 활성 성분들의 상승 효과가 나타남을 반증하는 것이다.
실시예 5. EAE 모델에서의 임상적 징후들을 참조한, 개별적인 활성 성분들과 비교하였을 때 PCB/ IFN-a 및 PCB-aa (펩타이드 1)/IFN-a 조합들의 약학적 효과
반면에, PCB/ IFN-a 및 PCB-aa (펩타이드 1)/IFN-a 조합들의 평가를, EAE 모델 (표 1) 중에서 예방적 스케줄로 수행하였으며 (표 1), 그 결과, 상기 조성물들이 전술한 투여량에서, 질병들의 진전 예방 면에서 상승적 효과를 나타냄을 알 수 있었다.

실시예 6. EAE 모델에서의 임상적 징후들을 참조한, 개별적인 활성 성분들과 비교하였을 때 PCB/IFN-b 및 PCB-aa (펩타이드 1)/IFN-b 조합들의 약학적 효과
C57BL6 마우스에서의 임상적 징후들의 감소와 관련해서, 예방적 (표 2) 및 치료적 스케줄 (도 5) 하에서, PCB/IFN-b 및 PCB-aa (펩타이드 1)/IFN-b 조합들의 약학적 효과를 입증하기 위해서, 하기 개별적 화합물들을 사용하였다: PCB (750 ㎍/kg, 복강내 경로), PCB-aa (3.375 mg/kg, 복강내 경로)는 15일 동안 매일 투여하였고, IFN-b (500 ng/Kg, 피하 경로)는 일주일에 세 번 6회 투여량으로 투여하였으며, 이들의 조합들 역시 표 2에 나타낸 투여량에 따라서 사용하였다.
각 처리의 유효 백분율은 하기 식에 따라서 계산하였다: 유효 백분율 (%) = (1 - ACi / ACEAE) x 100. ACi: 대응되는 산물로 처리된 군의 곡선 하 영역; ACEAE: EAE 군의 곡선 하 영역.
예방적 스케줄에서는, EAE가 유도되기 15일 전에 처리를 수행하였으며, 치료적 스케줄에서는 임상적 징후가 개시된 때로부터 수행하였다. 대조군은 어떠한 치료도 받지 않은 건강한 동물들에 해당된다.
표 2는 예방적 스케줄에서 얻어진 결과들을 나타낸 것으로서, PCB/IFN-b 및 PCB-aa (펩타이드 1)/IFN-b 조합들은 각각 90% 내지 100%의 동물들을 보호하였다. 그러므로, 개별적인 활성 성분들에 비해서 상승 효과가 관찰됨을 알 수 있다.

도 5는 PCB (750 ㎍/kg, 복강내 경로), PCB-aa (3.375 mg/kg, 복강내 경로), IFN-b (500 ng/Kg, 피하 경로)의 개별적인 활성 성분들로부터 얻어진 감소치보다 더 큰 임상적 징후들의 감소를 도시한 것이며, 치료적 스케줄에서도, EAE 모델에서 PCB (750 ㎍/kg) / IFN-b (500 ng/Kg) 또는 PCB-aa (3.375 mg/kg) / IFN-b (500 ng/Kg) 조합들로 처리된 군에서 상승 효과가 나타남을 보여주는 것으로서, PCB/IFN-b 조합에 대해서 87.7%; PCB-aa/IFN-b 조합에 대해서 94.5%; PCB에 대해서 46.1%; PCB-aa에 대해서 53.9% 및 IFN 베타에 대해서 34.8%의 효율로 나타났다.
실시예 7. EAE 모델에서 다른 경로들에 의한 PCB-aa (펩타이드 1)/ IFNb 조합의 치료적 효과
EAE 군의 동물들에 대해서 복강내 경로에 의해서 식염수 용액을 매일 투여하였다. PCB-aa (펩타이드 1)/IFN-b (PCB-aa 3.375 mg/kg + 500 ng의 IFN-b/Kg) 조합으로 처리된 마우스들을 투여 경로에 따라서 다른 군들로 나누었다: 복강내, 구강, 비강 및 직장. 치료적 스케줄을 면역화 후 9일부터 24일까지, 15일 연속 준수하였다. 대조군은 어떠한 치료도 받지 않은 건강한 마우스에 해당된다. 임상적 평가는 면역화 후 27일째 수행하였다.
도 6에 도시된 바와 같이, 평가된 다른 투여 경로들 (복강내, 비강, 구강 및 직장) 사이에서 통계적으로 유의성 있는 차이점들이 발견되지는 않았으며, 이러한 사실은 상기 경로들이 동일한 효율로 사용될 수 있다는 것을 의미한다.
실시예 8. 게르빌루스 쥐의 양방향 I/R 모델에서 PCB/IL-2 및 PCB-aa (펩타이드 2)/IL-2 조합의 신경보호 및/또는 신경재생 효과
동물들을 식염수 (복강내 경로) 또는, 허혈성 사건 이후 30분, 3, 6 및 12시간 동안 PCB (750 ㎍/Kg, 복강내 경로), PCB-aa (펩타이드 2) (3.375 mg/kg), IL-2 (100 ng/Kg, 피하 경로) 또는 PCB/IL-2 및 PCB-aa (펩타이드 2)/IL-2로 축적 투여량으로 처리하였다 (대응되는 투여량 및 투여 경로 유지). 도 7을 참조하면, 군 별로 경색 부피의 감소를 관찰할 수 있다.
각 처리의 유효 백분율을 실시예 3에 서술된 바와 같이 계산하였으며, 이는 PCB에 대해서 43.1%; PCB-aa (peptide 2)에 대해서 49.2%; IL-2에 대해서 25.8% for IL-2; PCB/IL-2 조합에 대해서 74.5%; 및 PCB-aa (peptide 2)/IL-2)에 대해서 84.3%였으며, 이는 상기 조합으로 처리된 동물들에서 양 활성 성분들 사이에 상승 효과가 존재한다는 것을 입증하는 것이다.
실시예 9. 게르빌루스 쥐의 양방향 I/R 모델에서 PCB/GHRP-6 조합 및 그 개별적 활성 성분들의 치료적 효과
PCB-aa (펩타이드 3), GHRP-6 펩타이드 또는 그 조합들의 치료적 처리에 대한 형태측정 평가를, 일시적 (10 분)으로 총경동맥이 폐색된 몽고 게르빌루스 쥐에서 수행하였다. 허혈성 사건 이후 30분, 3, 6 및 12시간 동안 PCB-aa (펩타이드 3) (750 ㎍/Kg, 복강내 경로), GHRP-6 (6,25 ㎍/kg, 복강내 경로), 또는 PCB-aa (펩타이드 3)/GHRP-6로 축적 투여량으로 처리한 다음 평가하였다 (대응되는 투여량 및 투여 경로 유지). 양방향 세포 계수를 각 실험군에 대해서 C2, CA3 및 CA4 영역들에서 수행하였으며, 가짜 (sham) (음성 또는 가짜 수술)에 대한 백분율로 표시하였다. 결과로부터, I/R 군의 동물들에서, 상기 세포주의 거의 완전한 소멸이 있었으며, 이는 실제적으로 모든 해마 영역들을 포함한다 (CA2, CA3, CA4).

Claims (16)

- 허혈성, 염증성 또는 신경퇴행성 손상에 따라서 진전되는 중추신경계 질환들의 예방 또는 치료를 위한 약물을 제조하기 위한, 제4항에 따른 화합물의 용도.
- 제4항에 따른 화합물 중 적어도 하나를 포함하는 약학 조성물을 투여하는 것을 특징으로 하는 허혈증 또는 조직 퇴행증을 예방 또는 치료하기 위한 방법.
- 제6항에 있어서, 상기 허혈증 또는 조직 퇴행증은 허혈성, 염증성 또는 신경퇴행성 손상에 따라서 진전되는 중추신경계 질환들을 야기하는 것을 특징으로 하는 방법.
- 상승 효과를 갖는 성분들로서, 제1 성분으로서, 서열번호 1 내지 서열번호 5로 표시되는 펩타이드들 및 파이코시아노빌린으로 이루어진 군으로부터 선택된 제4항에 따른 화합물을 포함하고, 제2 성분으로서, 타입 I 인터페론, 인터루킨-2 (IL-2), 에리스로포에틴 (EPO), 무시알로EPO 및 인간 성장 호르몬의 분비자극성 펩타이드 (GHRP-6)로 이루어진 군으로부터 선택된 화합물을 포함하는 약학 조성물.
- 제8항에 있어서, 상기 제2 성분이 인터페론 알파 또는 인터페론 베타인 것을 특징으로 하는 약학 조성물.
- 제8항에 있어서, 상기 서열번호 1 내지 서열번호 5로 표시되는 펩타이드들 중 어느 하나를 0.9 내지 3.375 mg/Kg 중량으로, 상기 파이코시아노빌린을 300 내지 750 ㎍/Kg 중량으로, 또한 상기 알파 또는 베타 인터페론을 500 내지 5000 ng/Kg 중량으로 포함하는 것을 특징으로 하는 약학 조성물.
- 제1 성분으로서, 서열번호 1 내지 서열번호 5로 표시되는 펩타이드들 및 파이코시아노빌린으로 이루어진 군으로부터 선택된 화합물을 포함하고, 제2 성분으로서, 알파 인터페론 (IFN-a) 및 베타 인터페론 (IFN-b)을 포함하는 타입 I 인터페론; 인터루킨-2 (IL-2); 에리스로포에틴 (EPO), 무시알로EPO 및 인간 성장 호르몬의 분비자극성 펩타이드 (GHRP-6)로 이루어진 군으로부터 선택된 화합물을 포함하는 약학 조성물의, 허혈성, 염증성 또는 신경퇴행성 기원의 CNS 질환들을 예방 또는 치료하기 위한 의약을 제조하기 위한 용도.
- 제11항에 있어서, 상기 의약은 급성 또는 만성 질환의 결과로서 손상된 뇌 유조직 (brain parenchyma)을 보호하는 것을 특징으로 하는 용도.
- 제11항에 있어서, 상기 CNS 질환들은 다양한 기원의 뇌 허혈증, 다발성 경화증 (multiple sclerosis), 알츠하이머 질환 및 파킨슨병 (Parkinson's disease)을 포함하는 것을 특징으로 하는 용도.
- 허혈성, 염증성 또는 신경퇴행성 기원의 CNS 질환들을 예방 또는 치료하기 위한 방법으로서, 제1 성분으로서, 서열번호 1 내지 서열번호 5로 표시되는 펩타이드들 및 파이코시아노빌린으로 이루어진 군으로부터 선택된 화합물을 포함하고, 제2 성분으로서, 인터페론 알파 (타입 IFN-a) 및 인터페론 베타 (IFN-b)를 포함하는 타입 I 인터페론; 인터루킨-2 (IL-2); 에리스로포에틴 (EPO), 무시알로EPO 및 인간 성장 호르몬의 분비자극성 펩타이드 (GHRP-6)로 이루어진 군으로부터 선택된 화합물을 포함하는 약학 조성물을 환자에게 투여하는 단계를 포함하는 것을 특징으로 하는 방법.

- 제14항에 있어서, 상기 조성물을 구성하는 성분들을, 다양한 기원들을 갖는 뇌 허혈증, 다발성 경화증, 알츠하이머 질환, 루게릭병 (lateral amyotrophic sclerosis), 척수-소뇌성 운동 실조증 (spinal-cerebellar ataxia), 헌팅턴병 (Huntington's disease) 및 파킨슨병 (Parkinson's disease)을 포함하는 CNS 질환의 의료 치료 과정에서 동일한 환자에게 동시에 또는 연속적으로 투여하는 것을 특징으로 하는 방법.
