Abstract
본 발명은 중간엽 줄기세포 유래 단백질을 유효성분으로 포함하는 소뇌 운동실조증의 치료 및 개선용 약학적 조성물에 관한 것으로, 소뇌성운동실조증 환자(CA-MSC)와 건강한 지원자(H-MSC)에서 파생된 중간엽 줄기 세포(MSC)를 비교하여 면역 조절 능력 및 신경 보호 효과를 평가하고, 소뇌성운동실조증 환자(CA-MSC)와 건강한 지원자(H-MSC)에서 파생된 중간엽 줄기 세포에서 수준이 유의하게 차이가 나는 단백질을 분석하여 선정된 FSTL1(follistatin-like 1)가 미세아교세포에서 항염증 활성으로 세포를 보호하는 효과를 확인함으로써, FSTL1(follistatin-like 1)을 소뇌성운동실조증의 단백질 치료제로 제공한다.
Description
본 발명은 중간엽 줄기세포 유래 단백질을 포함하는 소뇌성운동실조증의 치료 예후를 예측하기 위한 바이오마커, 및 이를 이용한 소뇌성운동실조증의 치료용 조성물에 관한 것이다.
소뇌성운동실조증(cerebellar ataxia, CA)은 신경퇴행성 질환 중 하나로, 주로 진행성 운동 실조, 구음 장애, 연하(삼킴) 장애 등을 특징으로 하는 질환이다. 소뇌성운동실조증의 주요 증상은 보행 기능장애, 불균형, 손상된 사지 협응 및 변형된 언어이다. 많은 실조증 장애에서 소뇌성운동실조증은 소뇌 피질 및 그의 구심성 또는 원심성 섬유 연결의 변성으로 인한 것으로, 환부 뇌 영역은 주로 소뇌, 후주, 추제로 및 기저핵이며, 신경세포가 감소하거나 운동 뉴런의 기능이 감소된 것이 특징이다. 소뇌성운동실조증은 유전적으로 발병되기도 하지만, 퇴행성 및 후천성으로 발병하기도 한다. 예컨대 퇴행성 실조증은 다계통 위축 실조증 및 산발성 성인-발병 실조증이고, 후천성 실조증은 알콜성/독소-유발된 소뇌 변성 또는 부신생물 소뇌 변성과 연관될 수 있다.
소뇌성운동실조증의 진단은 알콜중독, 비타민 결핍, 다발성 경화증, 혈관 질환, 종양 및 부신생물성 질환을 포함하는 실조증의 후천적인 비유전성 원인을 배제하여 진행된다. 결정적인 진단은 유전자 시험 또는 혈족 내에서의 발생을 검사하고, 신경학적 결핍과 관련 합병증으로 인해 발병하기도 한다. 소뇌성운동실조증은 척수 소뇌 변성이 주요 특징으로 나타나며, 느린 진행성, 대칭성, 측정 이상, 즉 정확성 상실을 갖는 중간선 및 사지 실조증; 변환운동곤란증, 즉 교대 운동을 수행하는데 어려움이 있는 리듬의 상실; 안구 주시에 영향을 끼치는 안구 운동 속도 감소 (안진 및 복시 포함); 언어 이상, 즉 조음장애; 연하 곤란; 손/발 비협응, 즉 사지 실조증; 비정상적 스테이션; 및 비정상적 보행이 나타난다. 소뇌성운동실조증의 징후 및 증상은 초기 성인기에 시작되지만 소아기부터 후기 성인기까지 언제든지 나타날 수 있다.
한편, 줄기세포 기반의 치료법은 다양한 신경 장애 치료에 유용하게 사용되고 있다. 다양한 줄기세포 유형 중에서 중간엽 줄기세포(MSC)는 치료 잠재력은 자가 재생 능력, 분화의 가소성, 신경 영양 특성 및 면역 조절로 인해 광범위하게 주목되고 있다. MSC는 윤리적 문제가 거의 없어서 쉽게 접근 가능하고 안정적이다. 2000 명 이상의 환자를 치료하기 위해, 자가 또는 배양된 동종 이형 MSC가 사용된다고 보고되었다. 일반 자가 MSC로 치료받는 것이 안전하지만, 노화, 악성 형질 전환, 교차 오염, 생착의 빈약함과 같은 제한이 있으며, 환자의 MSC는 연령, 유전적 특성 및 질병 중증도에 따라 배양 품질 및 능력이 낮아 치료용으로 사용되는 데에 한계가 있다. 따라서 이러한 한계를 극복하기 위해, 최근에는 MSC의 능력을 향상시키는 방향으로 치료 방법이 개발되고 있다.
상기와 같은 문제점을 해결하기 위해, 본 발명의 목적은 소뇌성운동실조증의 치료용 세포 치료제의 치료 효능을 예측하기 위한 바이오마커로서, 소뇌성운동실조증의 치료용 세포 치료제인 중간엽 줄기세포(MSC)에서 유래된 단백질들의 수준을 건강한 정상 MSC와 소뇌성운동실조증 환자의 MSC를 비교 평가하여 유의한 차이가 나타나는 단백질들을 세포 치료제의 치료 효율을 예측할 수 있는 바이오마커로 제공하는 것이다.
또한, 본 발명의 다른 목적은 건강한 정상 MSC와 소뇌성운동실조증 환자의 MSC에서 차이가 나타나는 단백질 수준을 비교 평가하여 소뇌성운동실조증의 치료용 세포 치료제의 치료 효율을 증진시킬 수 있는 중간엽 줄기세포(MSC)에서 유래된 단백질들을 포함하는 소뇌성운동실조증의 치료용 보조제를 제공하는 것이다.
또한, 본 발명의 다른 목적은 소뇌성운동실조증의 치료용 세포 치료제와 함께 병용 투여될 수 있는 중간엽 줄기세포(MSC)에서 유래된 단백질들을 포함하는 소뇌성운동실조증의 치료용 병용 투여용 조성물을 제공하는 것이다.

본 발명에 따르면, 건강한 지원자에 비해 소뇌성운동실조증 환자의 중간엽 줄기세포(MSC)에서 발현 수준이 유의미하게 증가 또는 감소하는 단백질들을 분석함으로써, 변화가 나타나는 단백질들을 소뇌성운동실조증에 대한 중간엽 줄기세포(MSC)의 치료 효율을 예측할 수 있는 새로운 바이오마커로 제공될 수 있고, 소뇌성운동실조증 환자의 중간엽 줄기세포(MSC)에서 발현 수준이 유의미하게 감소되는 단백질들을 소뇌성운동실조증에 대한 중간엽 줄기세포(MSC)의 치료 증진을 위한 보조제 및 병용 투여제로 제공될 수 있으며, 중간엽 줄기세포(MSC)에서 유래되는 단백질인 FSTL1(follistatin-like 1)이 미세아교세포에서 항염증 활성을 나타내는 것을 확인함으로써, FSTL1(follistatin-like 1)은 소뇌성운동실조증의 치료제로 제공될 수 있다.

상기 단백질들은 중간엽 줄기세포 유래 단백질이며, 상기 세포 치료제는 소뇌성운동실조증의 치료를 위해 투여되는 중간엽 줄기세포(MSC)이다. 구체적으로 치료제는 소뇌성운동실조증의 치료를 위해 중간엽 줄기세포(MSC) 치료제를 투여받은 환자에서 상기 단백질들의 발현 수준을 측정하고, 이를 소뇌성운동실조증이 발병하지 않은 정상인과 비교하여 발현 수준이 감소하거나 증가하는 것을 확인함으로써, 투여된 중간엽 줄기세포(MSC) 치료제의 치료 효율을 예측할 수 있다.
예컨대, FSTL1(follistatin-like 1), TGFB1(transforming growth factor-beta 1), IGFBP3(insulin-like growth factor-binding protein 3), GAS6(growth arrest-specific 6), FNDC1(fibronectin type III domain containing 1), COL11A1(collagen type XI alpha 1 chain), COL12A1(collagen type XII alpha 1 chain), PCOLCE(procollagen C-endopeptidase enhancer), NID2(nidogen 2), MMP2(matrix metallopeptidase 2), LRP1(LDL receptor related protein 1), SPARC(secreted protein acidic and cysteine rich), COL5A2(collagen type V alpha 2 chain), LUM(lumican), SERPINE1(serpin family E member 1), EDIL3(EGF like repeats and discoidin domains 3), COL5A1(collagen type V alpha 1 chain), THBS2(thrombospondin 2), AGRN(agrin), AEBP1(AE binding protein 1), COL6A1(collagen type VI alpha 1 chain), 및 LTBP2(latent transforming growth factor beta binding protein 2)로 구성되는 단백질들은 소뇌성운동실조증 환자의 중간엽 줄기세포(MSC)에서 낮은 수준으로 발현되며, 소뇌성운동실조증의 치료를 위해 중간엽 줄기세포(MSC) 치료제를 투여받은 후에 상기 단백질들의 수준이 질환을 갖지 않은 정상인과 비슷한 수준으로 회복되면, 소뇌성운동실조증에 대한 중간엽 줄기세포(MSC) 치료제의 치료 효과가 있음을 예측 할 수 있다.
또한, PRG1(p53-responsive gene 1) 단백질은 소뇌성운동실조증 환자의 중간엽 줄기세포(MSC)에서 높은 수준으로 발현되며, 소뇌성운동실조증의 치료를 위해 중간엽 줄기세포(MSC) 치료제를 투여받은 후에 상기 단백질의 수준이 질환을 갖지 않은 정상인과 비슷한 수준으로 회복되면, 소뇌성운동실조증에 대한 중간엽 줄기세포(MSC) 치료제의 치료 효과가 있음을 예측할 수 있다.
또한, 본 발명은 소뇌성운동실조증의 치료용 세포 치료제의 치료 효능을 증진하기 위한 보조제 조성물로서, 상기 조성물은 FSTL1(follistatin-like 1), TGFB1(transforming growth factor-beta 1), IGFBP3(insulin-like growth factor-binding protein 3), GAS6(growth arrest-specific 6), FNDC1(fibronectin type III domain containing 1), COL11A1(collagen type XI alpha 1 chain), COL12A1(collagen type XII alpha 1 chain), PCOLCE(procollagen C-endopeptidase enhancer), NID2(nidogen 2), MMP2(matrix metallopeptidase 2), LRP1(LDL receptor related protein 1), SPARC(secreted protein acidic and cysteine rich), COL5A2(collagen type V alpha 2 chain), LUM(lumican), SERPINE1(serpin family E member 1), EDIL3(EGF like repeats and discoidin domains 3), COL5A1(collagen type V alpha 1 chain), THBS2(thrombospondin 2), AGRN(agrin), AEBP1(AE binding protein 1), COL6A1(collagen type VI alpha 1 chain), 및 LTBP2(latent transforming growth factor beta binding protein 2)로 이루어진 군에서 선택된 어느 하나 이상의 단백질들을 포함하는 보조제 조성물을 제공한다.
상기 보조제 조성물은 PRG1(p53-responsive gene 1)의 억제제를 추가로 포함할 수 있고, 상기 PRG1(p53-responsive gene 1)의 억제제는 PRG1(p53-responsive gene 1)에 특이적으로 결합하는 항체, 펩타이드 또는 뉴클레오티드일 수 있다.

상기 병용 투여용 조성물은 소뇌성운동실조증의 치료를 위한 세포 치료제와 병용 투여되며, 상기 세포 치료제는 소뇌성운동실조증의 치료를 위해 투여되는 중간엽 줄기세포(MSC)이다.
또한, 본 발명은 FSTL1(follistatin-like 1)을 유효성분으로 포함하는 소뇌성운동실조증의 치료 또는 개선용 약학적 조성물을 제공한다.
상기 약학적 조성물은 TGFB1(transforming growth factor-beta 1), IGFBP3(insulin-like growth factor-binding protein 3), GAS6(growth arrest-specific 6), FNDC1(fibronectin type III domain containing 1), COL11A1(collagen type XI alpha 1 chain), COL12A1(collagen type XII alpha 1 chain), PCOLCE(procollagen C-endopeptidase enhancer), NID2(nidogen 2), MMP2(matrix metallopeptidase 2), LRP1(LDL receptor related protein 1), SPARC(secreted protein acidic and cysteine rich), COL5A2(collagen type V alpha 2 chain), LUM(lumican), SERPINE1(serpin family E member 1), EDIL3(EGF like repeats and discoidin domains 3), COL5A1(collagen type V alpha 1 chain), THBS2(thrombospondin 2), AGRN(agrin), AEBP1(AE binding protein 1), COL6A1(collagen type VI alpha 1 chain), 및 LTBP2(latent transforming growth factor beta binding protein 2)로 이루어진 군에서 선택된 어느 하나 이상을 추가로 포함한다. 또한, 상기 약학적 조성물은 PRG1(p53-responsive gene 1)의 억제제를 추가로 포함할 수 있다.
상기 FSTL1(follistatin-like 1)는 NO 생성 및 미세아교세포(microglia)의 염증 활성을 억제하는 효과가 있으며, 산화 스트레스로부터 신경세포를 보호하는 효과가 있다.
본 발명의 약학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다.
상기 약제학적으로 허용되는 담체는 제제 시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸 히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 약학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다.
본 발명에 있어서, 상기 약학적 조성물에 포함되는 첨가제의 함량은 특별히 한정되는 것은 아니며 통상의 제제화에 사용되는 함량 범위 내에서 적절하게 조절될 수 있다.
상기 약학적 조성물은 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 환약, 캡슐, 과립, 정제, 크림, 젤, 패취, 분무제, 연고제, 경고제, 로션제, 리니멘트제, 파스타제 및 카타플라스마제로 이루어진 군으로부터 선택되는 하나 이상의 외용제 형태로 제형화될 수 있다.
본 발명의 약학적 조성물은 제형화를 위해 추가로 있는 약학적으로 허용가능한 담체 및 희석제를 포함할 수 있다. 상기 약학적으로 허용가능한 담체 및 희석제는 전분, 당, 및 만니톨과 같은 부형제, 칼슘 포스페이트 등과 같은 충전제 및 증량제, 카르복시메틸셀룰로오스, 히드록시프로필셀룰로오스 등과 같은 셀룰로오스 유도체, 젤라틴, 알긴산염, 및 폴리비닐 피롤리돈 등과 같은 결합제, 활석, 스테아린산 칼슘, 수소화 피마자유 및 폴리에틸렌 글리콜과 같은 윤활제, 포비돈, 크로스포비돈과 같은 붕해제, 폴리소르베이트, 세틸알코올, 및 글리세롤 등과 같은 계면활성제를 포함하나, 이에 한정되지 않는다. 상기 약학적으로 허용가능한 담체 및 희석제는 대상체에게 생물학적 및 생리학적으로 친화적인 것일 수 있다. 희석제의 예로는 염수, 수용성 완충액, 용매 및/또는 분산제(dispersion media)를 들 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 약학적 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구 투여(예를 들어, 정맥 내, 피하, 복강 내 또는 국소에 적용)할 수 있다. 경구 투여일 경우, 정제, 트로키제 (troches), 로젠지 (lozenge), 수용성 현탁액, 유성 현탁액, 조제 분말, 과립, 에멀젼, 하드 캡슐, 소프트 캡슐, 시럽 또는 엘릭시르제 등으로 제형화될 수 있다. 비경구 투여일 경우, 주사액, 좌제, 호흡기 흡입용 분말, 스프레이용 에어로졸제, 연고, 도포용 파우더, 오일, 크림 등으로 제형화 될 수 있다.
본 발명의 약학적 조성물의 투여량은 환자의 상태 및 체중, 연령, 성별, 건강상태, 식이 체질 특이성, 제제의 성질, 질병의 정도, 조성물의 투여시간, 투여방법, 투여기간 또는 간격, 배설율, 및 약물 형태에 따라 그 범위가 다양할 수 있으며, 이 분야 통상의 기술자에 의해 적절하게 선택될 수 있다. 예컨대, 약 0.1 내지 10,000 mg/kg의 범위일 수 있으나 이제 제한되지 않으며, 하루 일회 내지 수회에 나누어 투여될 수 있다.
상기 약학적 조성물은 목적하는 방법에 따라 경구 투여되거나 비경구 투여(예를 들면, 정맥 내, 피하 내, 복강 내 또는 국소에 적용)될 수 있다. 본 발명의 약학적 조성물의 약학적 유효량, 유효 투여량은 약학적 조성물의 제제화 방법, 투여 방식, 투여 시간 및/또는 투여 경로 등에 의해 다양해질 수 있으며, 당해 기술 분야에서 통상의 지식을 가진 자는 목적하는 치료에 효과적인 투여량을 용이하게 결정하고 처방할 수 있다. 본 발명의 약학적 조성물의 투여는 하루에 1회 투여될 수 있고, 수회에 나누어 투여될 수도 있다.
또한, 본 발명은 FSTL1(follistatin-like 1)을 유효성분으로 포함하는 세포 치료제와 병용 투여되는 병용 투여용 조성물을 제공한다. 상기 세포 치료제는 소뇌성운동실조증의 치료를 위한 중간엽 줄기세포(MSC)이다.
이하, 본 발명의 이해를 돕기 위하여 실험예 및 실시예를 들어 상세하게 설명하기로 한다. 다만 하기의 실험예 및 실시예는 본 발명의 내용을 예시하는 것일 뿐 본 발명의 범위가 하기 실험예 및 실시예에 한정되는 것은 아니다. 본 발명의 실험예 및 실시예는 당업계에서 평균적인 지식을 가진 자에게 본 발명을 보다 완전하게 설명하기 위해 제공되는 것이다.
<실험예> 실험 재료 및 방법
하기의 실험예들은 본 발명에 따른 각각의 실시예에 공통적으로 적용되는 실험예를 제공하기 위한 것이다.
1. 세포 배양
MSC(중간엽 줄기세포)는 건강한 지원자 및 소뇌성운동실조증 환자의 골수로부터 수집하였다. 분리된 MSC는 국제세포치료학회(ISCT, International Society of Cellular Therapy) 지침에 따라 CORESTEM Inc. (Seoul, Korea)에서 GMP(Good Manufacturing Practice) 조건으로 배양되었다. MSC는 10%(v/v) 열-불활성 소 태아 혈청(FBS, heat-inactivated fetal bovine serum)(Invitrogen, Carlsbad, CA), 100 U/mL 페니실린(penicillin), 100 μg/mL 스트렙토마이신(streptomycin)(Invitrogen) 및 2.5 mM L-알라닐-L-글루타민(Gibco, Grand Island, NY)을 포함하는 CSBM-A06 배지 (CORESTEM Inc.)에서 배양되었다. 세포는 배양기에서 37℃, 5% CO2 조건으로 배양되었다.
BV-2 미세아교세포주(microglia)는 5% (v/v) FBS 및 50 μg/ml 젠타마이신(gentamicin)을 함유하는 DMEM(Dulbecco’s modified Eagle medium)(Lonza, Basel, Switzerland)에서 37℃, 5% CO2 조건으로 배양되었다. 미세아교세포(microglia)의 초대 배양을 위해, 3일된 C57BL/6의 뇌 전체를 잘게 자르고, 나일론 메쉬로 기계적으로 분쇄하여 혼합 신경교세포(mixed glial cells)를 얻었다. 수집된 세포는 10% FBS, 100 U/mL 페니실린, 및 100 μg/mL 스트렙토마이신을 함유하는 DMEM에서 37℃, 5% CO2 조건으로 배양되었다. 배양 배지는 배양 시작일로부터 5일 째에 교체하고, 이후로는 3일 마다 교체하였다. 세포 배양 14일 뒤, 미세아교세포(microglia)는 혼합 신경교세포(mixed glial cells)를 18시간 동안 250 rpm으로 흔들어 일차로 수집하였고 추가로 가벼운 트립신화(mild trypsinization)를 이용하여 수집하였다. 초대 배양된 미세아교세포의 순수도를 확인하기 위해, Iba-1(ionized calcium-binding adapter molecule 1), Cx3cr1, F4/80, CD11c, 및 CD11d와 같은 microglia 또는 myeloid에 대한 세포 특이 유전자를 PCR로 증폭하였다. 성상교세포(astrocyte)의 오염도는 GFAP 면역세포화학염색법에 의해 확인되었다.
소뇌 과립 뉴런(Cerebellar granule neuron)은 16.5일차에 마우스 배아로부터 수집되었다. 마우스 배아의 뇌를 분리하여 피루브산 나트륨(Gibco) 및 HEPES(Gibco)를 포함하는 차가운 HBSS(Hanks’ balanced salt solution)에 넣었다. 소뇌를 분리하고 37℃에서 30분 동안 0.25% 트립신-EDTA를 처리하였다. 소뇌 조직을 피펫팅으로 세포 단위로 분리하고, 2 mM 글루타민(Sigma-Aldrich), 페니실린-스트렙토마이신, N2 보충제(Invitrogen), 및 B27 보충제(Invitrogen)를 포유하는 신경 세포 전용 배지를 이용하여 poly-D-lysine(Sigma-Aldrich, St. Louis, MO) 코팅된 플레이트에 배양하였다. 이후 세포를 Ca2+ 및 Mg2+이 없는 HBSS 10 mL를 사용하여 5회 세척하고, 무혈청 CSBM-A06 배지에 74 시간 동안 배양하였다. MSC가 배양된 배지(MSC-CM)를 수확하였고, 4℃, 500xg에서 10 분 동안 원심분리하여 죽은 세포를 제거하고 -80℃에 분석 전까지 냉동보관 하였다.
2. 세포 증식 분석 (Cell proliferation assay)
세포를 IncuCyte® ImageLock plates (Essen Bioscience, Ann Arbor, MI)에 Well 당 1 X 103 개 세포로 배양하였다. 24 시간 후, IncuCyte ZOOM Live-Cell Imaging System (Essen Bioscience)을 사용하여 2~3 시간 마다 세포 증식을 실시간으로 분석하였다. IncuCyte ZOOM 소프트웨어(Essen Bioscience)에 의해 생성된 시간 경과 곡선에 따라 세포 증식을 정량화 하였다.
3. 세포 이동 분석 (Cell migration assay)
IncuCyte ZOOM Live-Cell Imaging System (Essen Bioscience)을 사용하여 세포 이동 분석을 수행하였다. 세포는 IncuCyte® ImageLock plates (Essen Bioscience)에서 well당 2.5 X 104 개 세포로 배양하였다. 그 후 세포 증식을 억제하기 위해 세포 이동 분석 2 시간 전 5 μg/mL 마이토마이신 C(mitomycin C)를 처리하였다. IncuCyte WoundMaker™ (Essen Bioscience)를 사용하여 well 중앙을 긁은 후, IncuCyte ZOOM Live-Cell Imaging System (Essen Bioscience)를 사용하여 긁힌 부분으로 세포가 이동하는 것을 자동으로 분석하였다. 실시간 이미지는 48 시간 동안 2~3 시간마다 획득하였다. ZOOM software (Essen Bioscience)에 의해 생성된 시간 경과 곡선을 사용하여 세포 밀도를 정량화 하였다.
4. 아질산염 정량화 및 세포 생존력 평가
아질산염의 양을 측정하여 산화 질소(NO) 생성을 평가하였다. 24 시간 배양 후, 50 μL의 세포 배양 배지를 96 웰 마이크로 플레이트에서 동일한 부피의 Griess 시약 (0.1 % 나프틸에틸렌 디아민 디하이드로클로라이드 및 5% 설파닐아마이드를 포함하는 5% 인산)과 혼합하였다. 흡광도는 마이크로 플레이트 리더에서 540 nm로 측정하였다. 아질산 나트륨은 NO농도 계산을 위해 표준 곡선을 만드는 데에 사용되었다. 세포 생존력은 MTT(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) 분석을 통해 측정되었다. 0.5 mg/mL MTT를 포함하는 인산 완충 식염수(PBS)를 세포에 첨가하고, 5% CO2 배양기에서 2 시간 동안 37℃로 배양되었다. 불용성 포르마잔 결정(formazan crystals)은 DMSO로 완전히 용해시켰고, 흡광도는 마이크로 플레이트 리더를 사용하여 570 nm에서 측정되었다.
5. Secretome 분석
용액 내 단백질을 분해하기 위해, 침전된 단백질을 40 mM NH4HCO3에 재현탁 하였다. 5 mM dithiothreitol를 처리하고 56℃에서 20분 동안 배양한 후, 단백질 침전물을 10 mM iodoacetamide로 10분 동안 상온에서 처리하였다. 그 후, 단백질 침전물을 1 : 100 trypsin-Lys C 혼합물 (Promega, Madison, WI)로 37℃에서 12 시간 동안 처리하였다. 트립신으로 분해된 펩타이드는 제조사의 지침에 따라 탈염 컬럼 (# 89873, Thermo Fisher Scientific, San Jose, CA)을 사용하여 탈염되었다.
트립신이 분해된 펩타이드는 EASY-Spray 소스와 인터페이스 되는 Q-Exactive 플러스 하이브리드 사중극 orbit-trap 질량 분석기 (Thermo Fisher Scientific)를 사용하여 분석되었다. 펩타이드의 크로마토그래피 분리는 로딩 컬럼으로 Acclaim PepMapTM 100 (75mm × 2cm, 3μm, nanoViper, Thermo Fisher Scientific), 분리 컬럼으로 EASY-Spray 컬럼 PepMapTM RSLC C18 (75 μm x 50 cm, 2 μm, Thermo Fisher Scientific) 장착한 Ultimate 3000 RSLCnano 시스템 (Thermo Fisher Scientific)에서 진행되었다. 펩타이드는 RS 자동 시료 주입기로 로딩되고, 0.1 % 포름산 함유 ACN/물의 선형 구배로 300 nL/min의 유속에서 분리되었다. LC 용리액을 분석 컬럼에서 전기 분무하고, 2.0 kV의 전압을 나노 스프레이 소스의 액체 접합을 통해 적용하였다. 펩타이드 혼합물을 40분 동안 5%에서 40%로 증가하는 CAN의 구배로 분리되었다. 분석 방법에는 350 ~ 2000 m/z 범위로 전체 질량 분석기(MS) 스캔 및 전체 MS 스캔에서 가장 강렬한 10 개의 이온에 대한 데이터 의존적 MS/MS (MS2)가 포함되었다. 질량 분석기는 데이터 종속 모드에서 데이터를 수집하도록 프로그래밍 되었다. 질량분석기의 교정은 제조사의 지침에 따라 제안된 교정 솔루션으로 수행되었다. 데이터 베이스 검색을 수행하기 위해, Proteome Discoverer software version 2.3 (Thermo Fisher Scientific)을 사용하여 직렬 질량 스펙트럼을 처리하였다. 스펙트럼 데이터는 human Uniprot database (release version 2019_07)로 검색하였다. 사용된 분석 워크플로워는 Spectrum Files (데이터 입력), Spectrum Selector (스펙트럼 및 기능 검색), Sequest HT (시퀀스 데이터베이스 검색), Percolator (펩티드 스펙트럼 일치 또는 PSM 검증 및 FDR 분석)의 4 개 노드가 포함되었다. 확인된 모든 단백질은 펩타이드 수준에서 계산된 FDR이 ≤ 1%로 나타났다. 검증은 q-값을 기반으로 하였다. 검색 매개 변수는 고정된 변형인 시스테인의 메틸티오 변형 및 동적 변형인 메티오닌의 산화와 같은 최대 2개의 누락된 분열의 트립신 특이성을 허용하였다. +1, +2 및 +3 이온에 대한 질량 검색 매개 변수에는 전구체 이온의 경우 20ppm, 조각 이온의 경우 0.6 Da의 질량 오차 허용 오차가 포함되었다.
6. 기존 RT-PCR
제조사의 지침에 따라 QIA-zol 시약 (QIAGEN, Valencia, CA)을 사용하여 세포에서 총 RNA를 추출하였다. 역전사 및 PCR 증폭은 특정 프라이머 세트와 함께 DNA Engine Tetrad Peltier Thermal Cycler (MJ Research, Waltham, MA)를 사용하였다. 프라이머 세트는 하기 표 1과 같다. Gapdh는 동량 분석 대조 용으로 사용되었다.

7. 효소 면역 분석법
BV-2 미세아교세포주 또는 초대 배양 미세아교세포에 LPS 및 MSC-CM을 24 시간 또는 48 시간 배양 후 배지를 수집하였다. 미세아교세포 배양 배지에서 포획 항체인 마우스 단일클론 항-TNF-α 항체(1:180; R & D Systems, Minneapolis, MN) 및 검출 항체인 비오틴화 다클론 항-TNF-α 항체 (1:180; R & D Systems)를 사용하여 TNF-α의 수준을 측정하였다. 비오틴화 항-TNF-α 항체는 streptavidin-horseradish peroxidase (HRP) 접합체(1:120; R&D Systems) 및 3,3 ', 5,5'-tetramethylbenzidine (TMB) 기질 (R & D Systems)과의 순차적 반응으로 검출하였다. 20분 배양 후, 2 N H2SO4를 첨가하여 반응을 중단하고, 마이크로 플레이트 리더를 사용하여 450nm와 540nm에서 흡광도를 측정하였다.
8. 통계 분석
데이터는 평균 ± SEM 또는 SD로 표시되었다. 두 그룹의 비교는 t- 검정 또는 일반적인 단방향 분산 분석 (ANOVA)에 이어 Tukey 사후 검정을 사용하여 수행되었다. 모든 통계 분석은 Prism 소프트웨어 버전 8.0 (GraphPad Software, La Jolla, CA)을 사용하였다. p- 값 <0.05는 통계적으로 유의한 것으로 간주하였다.
실시예 1. 세포 형태, 증식 및 이동
소뇌성운동실조증 환자(CA-MSC)와 건강한 지원자(H-MSC)에서 유래된 중간엽 줄기세포(MSC)의 생물학적 특성을 비교하였다. 도 1에 나타난 바와 같이, 각 MSC를 24 시간 배양하고 위상차 현미경으로 관찰한 결과, CA-MSC는 H-MSC에 비해 세포 돌출이 적고 가늘고 덜 평편한 형태를 갖는 것으로 나타났다.
배양 확장성을 확인하기 위해 MSC의 증식 정도 및 이동 정도에 대해 분석하였다. MSC의 증식 가능성은 초기 세포 밀도와 비교하여 시간 의존적 세포 밀도의 배수 변화 및 배가 시간으로 평가하였다. 세포 증식 속도는 IncuCyte Time-lapse Microscopy System을 사용하여 평가되었으며 세포 밀도의 배가 변화로 표시되었다. 도 2에 나타난 바와 같이, CA-MSC의 증식 능력은 H-MSC의 증식 능력보다 유의하게 낮은 것으로 나타났다.
또한, 세포 이동 분석을 통해 MSC의 이동성을 정량화 하였다. Well 중앙을 긁은 후 0 시간, 36 시간, 및 72 시간에 MSC의 이동 정도를 촬영하였다. 세포 이동은 2 시간 마다 평가되었다. 세포 이동은 초기에 긁혀진 영역을 기준으로 세포가 이동하여 채워진 세포 밀도를 백분율로 계산하고, 시간 당 채워지는 속도(μm/h)를 계산하였다. 도 3에 나타난 바와 같이, CA-MSC의 이동 및 속도는 H-MSC에 비해 크게 낮은 것으로 나타났으며, 상기 결과는 H-MSC보다 CA-MSC에서 세포 증식 및 이동성이 낮다는 것을 보여준다.
실시예 2. 아교 활성화에 대한 MSC의 면역 조절 능력
in vitro 및 in vivo에서 MSC의 항염증 및 면역 조절 능력이 보고된 바 있다. 따라서 미세아교세포 활성화에 대한 소뇌성운동실조증 환자 유래 중간엽 줄기세포(CA-MSC)의 면역 활성 조절 능력을 평가하였다.
LPS(100 ng/mL)가 포함 또는 비포함된 중간엽 줄기세포 배지(MSC-CM)를 BV-2 미세아교세포주 또는 초대 배양 미세아교세포에 24 시간 동안 처리하였다. 미세아교세포의 활성을 나타내는 산화 질소(NO)는 Griess 시약을 사용하여 검출하였다. TNF-α 수준은 효소결합면역법을 사용하여 측정되었다.
도 5 및 6에 나타난 바와 같이, 정상 중간엽 줄기세포 배지(H-MSC-CM)과 함께 배양된 경우 LPS에 의한 NO 생산이 현저하게 감소되었다. 그러나 환자 중간엽 줄기세포 배지(CA-MSC-CM)는 H-MSC-CM에 비해 미세아교세포 활성 억제 효과가 낮게 나타났다. 또한, 도 7에 나타난 바와 같이, H-MSC-CM은 LPS로 유도된 TNF-α 생산을 감소시켰으나, CA-MSC-CM은 H-MSC-CM보다 감소 효과가 낮게 나타났다.
실시예 3. MSC의 신경 보호 능력
소뇌 과립 뉴런(cerebellar granule neurons, CGN)을 이용하여 소뇌성운동실조증 환자에서 파생된 중간엽 줄기세포(CA-MSC)의 산화 스트레스 또는 활성화된 미세아교세포 독성에 대한 신경 보호 능력을 가지고 있는지 평가하였다. MSC가 배양된 배지와 200 μM H2O2를 소뇌 과립 뉴런에 첨가하였다. 도 9에 나타난 바와 같이, H2O2첨가는 소뇌 과립 뉴런(cerebellar granule neurons, CGN)에서 세포 생존력을 감소시켰지만, H-MSC-CM은 CGN의 세포 생존력을 완전히 회복시켰다.
또한, 활성화된 미세아교세포 독성에 대한 MSC의 신경 보호 효과를 평가하였다. BV-2 미세아교세포주를 LPS (100 ng/mL)로 24 시간 동안 자극하였다. 그 후, 신경세포 배지, MSC-CM (무 혈청 배지 또는 MSC의 CM) 및 M-CM (PBS 또는 LPS 처리 BV-2 세포의 CM)의 혼합물 (1:1:1)을 제조하였다. LPS에 의해 자극된 BV-2 세포의 CM에는 사이토 카인/케모카인, 핵산, 흥분성 아미노산, 활성 산소 종 및 프로테아제가 포함되었다. 도 9에 나타난 바와 같이, LPS로 자극된 BV-2-CM의 혼합물은 CGN의 세포 생존력을 현저하게 감소시킨 반면, H-MSC-CM을 포함하는 혼합물은 감소된 세포 생존력을 회복시켰다. CA-MSC-CM을 포함하는 혼합물은 산화 스트레스 유발 신경 독성과 미세아교세포 신경 독성을 모두 감소시켰지만, 그 효과는 H-MSC-CM보다 낮게 나타났다.
실시예 4. MSC secretome 비교
H-MSC와 CA-MSC 사이의 분비 단백질 차이를 확인하기 위해 MSC-CM에 존재하는 secretome을 LC/MS-MS로 측정하였다. 도 11에 나타난 바와 같이, 총 143개의 다른 단백질이 확인되었으며, H-MSC-CM에서 50 개의 단백질이 고유하게 검출되었고 76 개의 단백질이 CA-MSC-CM과 중첩되었다. 반면에 17 개의 단백질은 CA-MSC-CM에서만 검출되었다.
각 MSC에서 유래된 분비단백질의 기능을 확인하기 위해, DAVID 생물 정보학 데이터베이스를 이용하여 유전자 온톨로지 (GO) 분석을 수행하였다. 데이터에서 세포 이동, 세포 산화 환원 항상성 및 변형 성장 인자-베타 수용체 신호 전달 경로와 같은 기능이 CA-MSC에서 상당히 낮은 것으로 나타났다. 두 그룹에 겹친 76 개의 단백질 중 MSC-CM에서 다르게 검출된 단백질 세트를 분류하고, H-MSC와 비교하여 CA-MSC의 하향 조절된 단백질을 선별하였다. 55 개의 단백질은 CA-MSC에서 분비 단백질 발현이 낮았으며, 50 개의 단백질은 CA-MSC에서 검출되지 않았다. ClueGO 데이터베이스로 분석한 결과, 도 12에 나타난 바와 같이, 축삭 손상에 대한 반응, 엔도 펩티다아제 활성의 음성 조절, 아교 세포 발달 및 성장 인자-베타 생산 변형과 같은 기능이 CA-MSC에서 낮은 것으로 나타났다. 따라서 상기 결과는 CA-MSC의 신경 보호 잠재력이 결핍되어 있음을 입증한다.
실시예 5. MSC secretome 분석 검증
상기 실시예 3의 secretome 분석 결과에 대한 발현 검증을 수행하였다. 역전사 PCR에 의해 mRNA 발현에 따른 secretome 분석에서 확인된 단백질의 발현을 검사하였다. 건강한 지원자 유래 중간엽 줄기세포(H-MSC) 대비 소뇌성운동실조증 환자 유래 중간엽 줄기세포(CA-MSC)에서 낮은 수준으로 나타나는 단백질은 하기 표 2와 같다.

표 2에 나타난 바와 같이, 건강한 지원자 유래 중간엽 줄기세포(H-MSC) 대비 소뇌성운동실조증 환자 유래 중간엽 줄기세포(CA-MSC)에서 FNDC1(fibronectin type III domain containing 1), COL11A1(collagen type XI alpha 1 chain), COL12A1(collagen type XII alpha 1 chain), PCOLCE(procollagen C-endopeptidase enhancer), NID2(nidogen 2), MMP2(matrix metallopeptidase 2), LRP1(LDL receptor related protein 1), SPARC(secreted protein acidic and cysteine rich), COL5A2(collagen type V alpha 2 chain), LUM(lumican), SERPINE1(serpin family E member 1), EDIL3(EGF like repeats and discoidin domains 3), FSTL1(follistatin like 1), COL5A1(collagen type V alpha 1 chain), THBS2(thrombospondin 2), AGRN(agrin), AEBP1(AE binding protein 1), COL6A1(collagen type VI alpha 1 chain), 및 LTBP2(latent transforming growth factor beta binding protein 2)의 발현은 낮게 나타났고, PRG1(p53-responsive gene 1)의 발현은 높게 나타났다.
또한, 도 13에 나타난 바와 같이, 두 그룹에서 검출되었지만 CA-MSC에서 더 낮은 55 개의 단백질 중, FSTL1(follistatin-like 1), TGFB1(transforming growth factor-beta 1), IGFBP3(insulin-like growth factor-binding protein 3), 및 GAS6(growth arrest-specific 6) 등의 mRNA 발현이 H-MSC보다 CA-MSC에서 유의하게 낮게 나타났다. 또한, 건강한 지원자 유래 중간엽 줄기세포(H-MSC) (n = 9) 및 소뇌성운동실조증 환자 유래 중간엽 줄기세포(CA-MSC) (n = 12)의 배양 배지에 존재하는 FSTL1 단백질을 검증한 결과, 도 14에 나타난 바와 같이, CA-MSC-CM에서 FSTL1 단백질의 검증은 H-MSC에서보다 낮은 수준으로 나타났다.
상기 결과는 중간엽 줄기세포(MSC)에서 유래된 단백질인 FSTL1(follistatin-like 1), TGFB1(transforming growth factor-beta 1), IGFBP3(insulin-like growth factor-binding protein 3), 및 GAS6(growth arrest-specific 6) 등의 단백질이 소뇌성운동실조증 환자 유래 중간엽 줄기세포(CA-MSC)에서 유의하게 낮게 나타났다.
실시예 6. 미세아교세포의 염증 활성에 대한 FSTL1의 억제능
FSTL1는 미세아교세포 활성에 대한 억제능이 있다. FSTL1이 미세아교세포 매개 신경염증을 조절하는지 확인하기 위해, LPS (100 ng/mL) 자극하기 전에 재조합 FSTL1 단백질 (0.0001 ~ 10 μg/mL의 용량 범위)을 BV-2 미세아교세포주 및 초대 배양 미세아교세포(primary microglia)에 처리하였다. 도 15 및 16에 나타난 바와 같이, FSTL1 단백질을 처리한 경우 미세아교세포의 산화질소 생산이 억제되었다. 상기 결과는 MSC-CM의 FSTL1 단백질이 미세아교세포에 대한 항염증 역할을 나타낸다는 것을 입증한다.
상기 결과는 중간엽 줄기세포(MSC)에서 유래된 단백질인 FSTL1(follistatin-like 1)이 NO 생성을 억제하여 산화 스트레스로부터 미세아교세포를 보호하는 항염증 활성이 있음을 입증하며, FSTL1(follistatin-like 1)이 신경 세포 손상으로 발병하는 소뇌성운동실조증에 대한 치료용 성분으로 활용될 수 있음을 입증한다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 즉, 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다.
