Abstract
Description
도 2는 본 발명에 따른 일실시예에서 발효 된장 추출물이 세포독성을 나타내는지를 확인하기 위하여, 다양한 농도의 발효 된장 추출물(10 ~ 500μg/mL)을 MG-63 세포에 24시간 동안 처리했을 때 수행한 MTT 분석 결과를 각 농도별로 비교하여 나타낸 것이다.
도 3은 본 발명에 따른 일실시예에서 발효 된장 추출물이 세포독성을 나타내는지를 확인하기 위하여 발효 된장 추출물(100μg/mL)을 MG-63 세포에 12 ~ 72시간 동안 처리했을 때 수행한 MTT 분석 결과를 처리 시간 별로 비교하여 나타낸 것이다.
도 4는 본 발명에 따른 일실시예에서 다양한 농도의 발효 된장 추출물(10 ~ 500μg/mL)을 48시간 동안 MG-63 세포주에 처리하였을 경우 처리 농도에 따른 염기성 인산분해효소(ALP)의 활성을 비교하여 나타낸 것이다.
도 5는 본 발명에 따른 일실시예에서 다양한 농도의 발효 된장 추출물(10 ~ 500μg/mL)을 72시간 동안 MG-63 세포주에 처리하였을 경우 처리 농도에 따른 오스테오칼신(OC)의 농도를 비교하여 나타낸 것이다.
도 6은 본 발명에 따른 일실시예에서 다양한 농도의 발효 된장 추출물(10 ~ 500μg/mL)을 24시간 동안 MG-63 세포주에 처리하였을 경우 처리 농도에 따른 BMP-2 단백질의 농도를 비교하여 나타낸 것이다.
도 7은 본 발명에 따른 일실시예에서 다양한 농도의 발효 된장 추출물(10 ~ 500μg/mL)을 72시간 동안 MG-63 세포주에 처리하였을 경우 처리 농도에 따른 제1형 콜라겐 단백질의 농도를 비교하여 나타낸 것이다.
도 8(a)는 본 발명에 따른 일실시예에서 미네랄화작용을 확인하기 위하여 다양한 농도의 발효 된장 추출물(10 ~ 500μg/mL)을 MG-63 세포주에 처리하고 7일 후에 Alizarine red-S로 염색하여 측정한 히드록시아파타이트 및 미네랄 형태의 칼슘 아파타이트의 양을 처리 농도에 따라 비교하여 나타낸 것이고, 도 8(b)는 발효 된장 추출물(100μg/mL)을 처리한 경우 세포 내 칼슘의 양을 처리하지 않은 대조군과 비교하여 나타낸 것이다.

Claims (9)
- 발효 된장 추출물을 유효성분으로 포함하는 골질환 예방 또는 치료용 조성물.
- 제1항에 있어서,
상기 발효 된장 추출물은 된장을 25 ~ 35℃에서 40 ~ 60일 동안 발효시킨 발효 된장을 물 또는 유기용매에 가용하여 추출한 것임을 특징으로 하는 골질환 예방 또는 치료용 조성물. - 제1항 또는 제2항에 있어서,
상기 추출물은 제니스테인(genistein), 다이드제인(daidzein) 및 글리시테인(glycitein) 중에서 선택된 어느 하나 이상의 이소플라본 아글리콘을 함유하는 것을 특징으로 하는 골질환 예방 또는 치료용 조성물. - 제3항에 있어서,
상기 이소플라본 아클리콘은 상기 발효 된장 추출물 1g 당 다이드제인 120 ~ 610㎍/g, 제니스테인 390 ~ 560㎍/g 및 글리시테인 130 ~ 450㎍/g의 농도로 함유되어 있는 것을 특징으로 하는 골질환 예방 또는 치료용 조성물. - 제1항에 있어서,
상기 골질환은 골다공증, 골연화증, 구루병, 골감소증, 저인산혈증, 섬유성 골염, 무형성 골질환, 골형성부전증, 골위축, 골이영양증, 파제트병(변형성골염), 암세포의 골전이에 의해 유발되는 뼈의 손상 및 고칼슘혈증으로 이루어진 군에서 선택되는 것을 특징으로 하는 골질환 예방 또는 치료용 조성물. - 제1항에 있어서,
상기 조성물은 조골세포(osteoblast)의 분화를 촉진하는 것을 특징으로 하는 골질환 예방 또는 치료용 조성물. - 제1항에 있어서,
상기 조성물은 조골세포(osteoblast)에 의한 염기성 인산분해효소(alkaline phosphatase; ALP)의 활성을 증가시키고, 오스테오칼신(osteocalcin; OC)의 발현을 증가시켜 골 형성을 촉진하는 것을 특징으로 하는 골질환 예방 또는 치료용 조성물. - 제1항에 있어서,
상기 조성물은 제1형 콜라겐의 합성을 유도함으로써 골 형성을 촉진하는 것을 특징으로 하는 골질환 예방 또는 치료용 조성물. - 발효 된장 추출물을 유효성분으로 포함하는 골질환 예방 또는 개선용 건강기능식품.
