Abstract
인간 간장을 포함하는, 복수의 인간 장기세포로 분화가능한 새로운 인간 줄기세포가 개시되어 있다. 본 발명의 인간 줄기세포는 인간 타액선에 유래하고, CD49f 양성이며, 생체외에서의 배양에 의해, (1)네스틴 양성 및 알부민 양성세포, (2)인슐린 양성세포 및 (3)글루카곤 양성세포로 분화될 수 있다.
Description
본 발명은, 인간 타액선유래 줄기세포에 관한 것이다.
재생의료에 유용한 포텐셜을 갖는 줄기세포에 대한 연구가 활발하게 이루어지고 있다. 지금까지 보고된 대표적인 줄기세포로서, 간엽계 줄기세포, 신경줄기세포, 조혈줄기세포, 그리고 췌줄기세포를 들 수 있다.
간엽계 줄기세포는 인간 성체 골수액으로부터 분리되었다(Pittenger, M. F. et al., Science 284,143(1999)). 이 세포는 지방세포, 연골세포, 골세포에의 인비트로(in vitro)에서의 분화 유도가 가능하다. 신경줄기세포(Gage, P.H., Science 287,1433-1438(2000))에 대해서는 1992년에 성체의 중추신경계로부터의 최초의 분리의 보고가 이루어졌으며, 2001년에는 성체의 피부진피로부터 신경세포로 분화가능한 줄기세포의 분리(Toma, J.G.et al., Nature Cell Biology, 3,778-784(2001))가 보고되었다.
조혈줄기세포는 이미 많은 연구가 이루어지고 있지만, 그 분화 기능에 대해서 보고된 것은 비교적 새롭다. 1999년에 골수세포가 간(肝)세포로 분화되는 것이 Petersen들에 의해 밝혀지고(Petersen B.E. et al., Science 284, 1168(1999)), 다음해에는 쥐 조혈줄기세포를 c-kittil, Thy-llow, Linneg, Sca-1+로 소팅(sorting)한 세포분획이, 줄기세포로 분화 전환되는 것이 나타내어져 있다(Lagasse, E. et al., Nature Medicine 6,1229-1234(2000)). 이 외에도 조혈줄기세포에는 분화 전환능이 있다고 생각되어지고 있으며, 심근(Orlic, D. et al., Nature 410,701-705(2001))이나, 또한 폐포상피, 장관상피, 피부(Orlic, D. et al., 상기에 기재)로의 분화도 보고되고 있다.
이상과 같이, 간엽계 또는 외배엽계의 세포에 대한 줄기세포 연구는 진행되고 있지만, 내배엽계 줄기세포의 보고는 아직 적다. 인간 간줄기세포에 대해서는 그 존재는 확실시되고 있지만, 아직 확정적인 줄기세포의 보고는 없다. 췌장에 대해서는 Cornelius들의 그룹이 성체 쥐 췌장으로부터 췌도생산 줄기세포(islet producing stem cells(IPSCs))의 분리를 행하고 있으며, 또한 IPSCs로부터 인비트로에서 제작한 췌도의 이식실험을 보고하고 있다(Ramiya, V.K. et al., Nature Medicine 6,278-282(2000)). 이 세포에 대해서도, α, β, δ세포로의 분화는 확인되고 있지만, 그 밖의 세포로의 분화능은 확인되고 있지 않다. 췌도로부터 네스틴(nestin) 양성에 의해 분리된 줄기세포가 췌장 중, 외분비 및 간장의 표현형으로 분화되었다라는 보고는 있지만(Zulewski, H. et al., Diabetes 50,521-533(2001)), 분화 마커의 면역 조직학적 검색은 나타내어져 있지 않다.
ES세포(배성 줄기세포)로부터, 간장 또는 췌장세포의 유도도 시도되고 있으며, 실제로 췌장의 α, β세포에 대해서는 분화 유도가 가능하다(Lumeisky, N, et al., Science 292,1389-1394(2001)). 그러나 인간 줄기세포로의 유도에 대해서는 아직 보고는 없다.
이상과 같이, 인간 간세포로 분화가능한 줄기세포는 아직 얻어지지 않고 있다. 따라서, 인간 간장을 포함하는, 복수의 인간 장기세포로 분화가능한 줄기세포도 얻어지지 못하고 있다. 또한 인간 줄기세포를, 재생 의료용도로 사용하는 경우, 이식에 의한 거절반응을 방지하는 관점에서, 환자자신의 세포를 이식하는 것이 가장 바람직하다. 따라서, 성체로부터도 조제하는 것이 가능한 인간 줄기세포가 얻어지면 재생의료에 있어서 특히 유리하다.
본 발명의 목적은, 인간 간세포로 분화가능한 줄기세포를 제공하는 것이다. 또한 본 발명의 목적은, 인간 간장을 포함하는, 복수의 인간 장기세포로 분화가능한 인간 줄기세포를 제공하는 것이다. 또한, 본 발명의 목적은, 인간 성체로부터도 조제할 수 있는, 인간 간세포로 분화가능한 인간 줄기세포를 제공하는 것이다.
본원 발명자들은, 예의 연구의 결과, 인간 타액선으로부터, 인간 신경세포, 인간 간세포 및 인간 췌장세포로 분화가능한 인간 줄기세포를 분리하는 것에 성공하여 본 발명을 완성했다.
즉 본 발명은, 인간 타액선에 유래하고, CD49f 양성이며, 생체외에서의 배양에 의해, (1)네스틴 양성 및 알부민 양성세포, (2)인슐린 양성세포 및 (3)글루카곤 양성세포로 분화될 수 있는 인간 줄기세포를 제공한다. 또한 본 발명은, 인간 타액선세포를 생체외에서 배양함으로써 유도되는 네스틴 양성 및 알부민 양성세포이며, 생체외에서의 배양에 의해 인슐린 양성세포 및 글루카곤 양성세포로 분화될 수 있는 인간 줄기세포를 제공한다. 또한, 본 발명은, 상기 본 발명의 인간 줄기세포를 생체외에서 배양함으로써 유도된 인슐린 양성세포 및 글루카곤 양성세포를 제공한다. 또한, 본 발명은, 인간 타액선조직 유래의 세포로부터 CD49f 양성세포를 수집하고, 이들을 상피세포 성장인자의 존재하에서 계대배양하는 것을 포함하는, 인간 타액선에 유래하고, CD49f 양성이며, 생체외에서의 배양에 의해, (1)네스틴 양성 및 알부민 양성세포, (2)인슐린 양성세포 및 (3)글루카곤 양성세포로 분화될 수 있는 인간 줄기세포를 유도하고, 유도된 상기 인간 줄기세포를 회수하는 것을 포함하는, 인간 줄기세포의 취득방법을 제공한다. 또한, 본 발명은, 인간 타액선에 유래하고, CD49f 양성이며, 생체외에서의 배양에 의해, (1)네스틴 양성 및 알부민 양성세포, (2)인슐린 양성세포 및 (3)글루카곤 양성세포로 분화될 수 있는 인간 줄기세포를, 섬유 아세포 증식인자, 상피세포 성장인자 및 백혈병 억제인자의 존재하에서, 생체외에서 배양해서 네스틴 양성 및 알부민 양성세포를 유도하고, 유도된 네스틴 양성 및 알부민 양성세포를 회수하는 것을 포함하는, 인간 줄기세포의 취득방법을 제공한다. 또한, 본 발명은, 인간 타액선세포를 생체외에서 배양함으로써 유도되는 네스틴 양성 및 알부민 양성세포이며, 생체외에서의 배양에 의해 인슐린 양성세포 및 글루카곤 양성세포로 분화될 수 있는 인간 줄기세포를, 생체외에서 배양함으로써, 인슐린 양성세포를 유도하고, 유도된 인슐린 양성세포를 회수하는 것을 포함하는, 인슐린 양성세포의 취득방법을 제공한다. 또한, 본 발명은, 인간 타액선세포를 생체외에서 배양함으로써 유도되는 네스틴 양성 및 알부민 양성세포이며, 생체외에서의 배양에 의해 인슐린 양성세포 및 글루카곤 양성세포로 분화될 수 있는 인간 줄기세포를, 생체외에서 배양함으로써, 글루카곤 양성세포를 유도하고, 유도된 글루카곤 양성세포를 회수하는 것을 포함하는, 글루카곤 양성세포의 취득방법을 제공한다.
본 발명에 의해, 인간 간장을 포함하는, 복수의 인간 장기세포로 분화가능한 인간 줄기세포가 제공되었다. 본 발명의 인간 줄기세포는, 적어도 인간 간세포 및 인간 췌세포로의 분화가 가능하므로, 인간 간장 및 인간 췌장을 재생하기 위한 재생치료에 유용하다. 특히, 본 발명의 줄기세포는, 성체로부터도 얻을 수 있으므로, 재생치료를 받는 환자 자신으로부터 채취할 수 있어, 이식시의 거절반응을 회피할 수 있으므로 매우 유리하다.
하기 실시예에 있어서 상세하세 서술하듯이, 본 발명의 인간 줄기세포는, 인간 타액선조직 유래의 세포로부터 CD49f 양성세포를 수집하고, 이들을 상피세포 성장인자(EGF)의 존재하에서 계대배양함으로써 얻어지는 것이며, 생체외에서의 배양에 의해, (1)네스틴 양성 및 알부민 양성세포, (2)인슐린 양성세포 및 (3)글루카곤 양성세포로 분화될 수 있다(이하, 이 인간 줄기세포를 편의적으로 「제1 인간 줄기세포」라고 하는 일이 있다). 계대배양시의 EGF의 농도는, 특별히 한정되지 않지만, 5ng/㎖∼80ng/㎖정도가 바람직하고, 또한 10ng/㎖∼40ng/㎖정도가 바람직하고, 특히 15∼25ng/㎖정도가 바람직하다. 또한 배지는, 인간 세포의 배양에 상용되고 있는, 예를 들면 윌리엄즈 E 배지 등을 사용할 수 있다. 또, CD49f는 α6 인테그린이라고도 불리고 있으며, CD29(β1 인테그린)과 헤테로 2량체를 형성하는 막단백이다. CD29와 2량체를 형성한 CD49f는, 세포외 매트릭스인 라미닌의 수용체(VLA-6)로서 알려져 있다. 일반적으로는 T세포, 혈소판, 단구, 상피세포, 내피세포, 태반의 트로포블라스트(trophoblast)에 있어서 발현이 알려져 있다(Knapp W. et al., eds. Leukocyte Typing IV:White Cell Differentiation Antigens, Oxford University Press, New York.(1989)). 그 밖에 태아기의 타액선 원기를 구성하는 미분화 상피세포(Lazowski KW. et al., Differentiation 56:75-82(1994))나 태아 줄기세포(Suzuki A.et al., Hepatology 32, 1230-1239(2000)) 등에서의 발현이 보고되고 있다.
이 제1 인간 줄기세포를, 섬유 아세포 증식인자(FGF), 상피세포 성장인자(EGF) 및 백혈병 억제인자(LIF)의 존재하에서, 뉴로스피어(neurosphere)법으로 배양해서 스피어를 형성시킴으로써, 네스틴 양성, 알부민 양성의 세포로 이루어지는 스피어가 형성된다. 이 때의 FGF의 농도는, 특별히 한정되지 않지만, 5ng/㎖∼80ng/㎖정도가 바람직하고, 또한 10ng/㎖∼40ng/㎖정도가 바람직하고, 특히 15∼25ng/㎖정도가 바람직하다. 또한 EGF의 농도는, 특별히 한정되지 않지만, 10ng/㎖∼160ng/㎖정도가 바람직하고, 또한, 20ng/㎖∼80ng/㎖정도가 바람직하고, 특히 30∼50ng/㎖정도가 바람직하다. 또한 LIF의 농도는, 특별히 한정되지 않지만, 2ng/㎖∼40ng/㎖정도가 바람직하고, 또한 5ng/㎖∼20ng/㎖정도가 바람직하고, 특히 7∼15ng/㎖정도가 바람직하다.
이 세포는, 상법에 따라, 스페로이드 배양하는 것에 의해 스페로이드체를 형성시킬 수 있다. 배지로서는, 인간 세포의 배양에 상용되고 있는, 윌리엄즈 E 배지와 같은 배지에, 혈청, 예를 들면 소태아혈청(FBS)을 첨가한 것이 바람직하다. FBS의 농도는, 특별히 한정되지 않지만, 5∼20중량%가 바람직하고, 7∼15중량%정도가 더욱 바람직하다. 이 스페로이드체의 외연(外緣)은 글루카곤 양성세포, 그 내측에는 인슐린 양성세포로 구성되다. 스페로이드의 중심핵의 부분은, 인슐린, 글루카곤의 어느 것에나 음성이다. 또한 스페로이드 배양시에 있어서 글루카곤형 펩티드-1(GLP-1) 10mM, 액티빈A 5ng/㎖를 분화 유도 배지에 첨가하면, 최외연부의 글루카곤 양성세포가 결여된 스페로이드체가 형성된다.
따라서, 뉴로스피어법에 의해 스피어를 형성한 세포도 또 인간 줄기세포이다(이하, 편의적으로 「제2 인간 줄기세포」라고 하는 일이 있음). 본 발명의 제2 인간 줄기세포, 나아가서는 제1 인간 줄기세포는, 이 방법에 의해 글루카곤 양성세포 및 인슐린 양성세포로 유도할 수 있다.
또, 본 발명의 줄기세포는, 인간 세포이기 때문에, 생체외에서의 배양은, 인간의 체온인 37℃에서 행하는 것이 바람직하고, 하기 실시예에서도 특별히 언급하지 않는 한, 생체외에서의 배양은 37℃에서 행했다.
주지한 대로, 알부민은 간세포의 분화 마커이기 때문에, 본 발명의 제2 인간 줄기세포, 나아가서는 제1 인간 줄기세포는, 간세포로 분화시킬 수 있다. 또한 글루카곤 및 인슐린은 췌장의 분화 마커이기 때문에, 본 발명의 제2 인간 줄기세포, 나아가서는 제1 인간 줄기세포는 췌세포로 분화시킬 수 있다.
본 발명의 제1 및 제2 인간 줄기세포는, 하기 실시예에 있어서, 성체로부터 얻어졌다. 재생 의료를 위해 세포를 이식하는 경우, 이식에 의한 거절반응을 방지하는 관점에서, 환자자신의 세포를 이식하는 것이 가장 바람직하다. 본 발명의 인간 줄기세포는, 성체로부터 얻을 수 있으므로, 이식을 받는 환자자신으로부터 조제할 수 있어, 이식하는데에 매우 유리하다. 또, 본 발명의 인간 줄기세포는, 성체 뿐만 아니라, 미성숙한 아이로부터 얻을 수도 있다.
본 발명의 인간 줄기세포는, 적어도 간장 및 췌장으로 분화될 수 있으므로, 본 발명의 인간 줄기세포를 이식해서 이들의 인간 장기의 재생을 행하는 것이 가능하다. 인간 줄기세포의 이식은, 인간 줄기세포의 부유액을 인간에게 주입함으로써 용이하게 행할 수 있다. 주입은, 췌장내나, 재생하고자 하는 장기 또는 그 근방, 또는 정맥내 등에 대해서 행할 수 있다. 또한 주입하는 줄기세포의 수는, 특별히 한정되지 않고, 증상이나 환자의 체중, 투여방법 등에 따라 적당하게 선택할 수 있지만, 통상, 102∼1O10개 정도이다.
실시예
(1)타액선세포의 초대배양
다음의 방법에 의해 타액선조직을 소화시켰다.
타액선조직의 소화법
1.수술시에 적출한 편측 악하선(2∼4g정도)을, 지름 1∼2mm 크기로 될 때까지 가늘게 절단했다.
2.가늘게 절단한 조직은 EGTA 완충액(1L중, NaCl 8g, KCl 0.4g, NaH2PO4 69mg, Na2HPO4 75mg, HEPES 2.38g, EGTA(에틸렌글리콜 4초산) 0.19g, NaHCO3 0.35g,글루코오스 0.9g, 페놀 레드 6mg)30㎖와 함께 50㎖ 원심관으로 옮기고, 37℃에서 20분간, 10회/분의 속도로 회전 진탕했다. 인큐베이션후, 조직세편액은 원심(100Xg, 5분, 실온)하고, 상청은 버렸다.
3.얻어진 펠릿은 60㎖의 소화액(digestion medium)(60㎖중, DMEM/F 12:1;1 60㎖, 콜라게나아제 100mg, 히알루로니다아제 80mg)에 분산시킨 후, 50㎖ 원심관에 옮겨 37℃에서 40분간, 10회/분의 속도로 회전 진탕했다. 인큐베이션후, 조직세편액은 원심(100Xg, 5분, 실온)하고, 상청은 버렸다.
4.얻어진 펠릿은 60㎖의 분산액(dispersion medium)(60㎖중, DMEM/F12:1;1 60㎖, 디스파아제 80mg(100OU/㎖))에 분산시킨 후, 50㎖ 원심관에 옮겨 37℃에서 60분간, 10회/분의 속도로 회전 진탕했다. 회전 진탕 도중에 수회, 1O㎖ 피펫으로 피페팅을 행하고, 조직세편액을 기계적으로 분산시켰다. 인큐베이션후, 조직세편액은 세포여과기를 통해 세포부유액을 얻었다. 세포부유액은 원심(100Xg, 5분, 4℃)하고, 이것으로부터 세포 펠릿을 얻었다.
5.세포 펠릿은 30㎖의 윌리엄즈 E 배지(Williams'E medium)에 현탁하고, 동배지로 3회 세정했다.
6.세포수 카운트와 세포생존률의 체크를 행했다. 4g의 타액선조직으로부터 얻어지는 세포의 총수는, 2.0∼3.0x107개 정도였다. 또 세포의 생존률은 90%이상이었다.
다음 방법에 의해 세포를 분리했다.
세포분리법(1.O×1O7세포의 경우)
1.상기 (1)에서 회수한 타액선유래 세포는, 0.1% BSA/PBS(500μℓ)에 재현탁후, 항-인간 CD49f 모노클로날 항체(클론:GoH3/BD 파민겐)로, 20분간의 인큐베이션(빙중)을 행한다. 사용하는 항체량은 FCM 분석시에 준해서, 1μg/1.O×1O6 세포로 라벨링을 행했다.
2.라벨링 종료후에, 5㎖의 완충액(상동, 기재하지 않는 한 이하 동일)을 첨가하여, 세정 및 원심한다.
3.세포 펠릿을 100μℓ의 완충액에 재현탁. 20㎖의 자기세포분리(MACS)용 염소항-쥐 IgG 마이크로 비즈를 첨가하고, 혼화후에 15분간의 인큐베이션(수중).
4.다시 세포를 세정 및 원심후, 세포 펠릿을 3㎖의 완충액에 재현탁.
5.샘플을 AutoMACS의 세포분리 프로그램(POSSEL)에 공급한다. CD49f(+) 분획의 세포를 회수한다.
6.회수한 세포는 윌리엄즈 E 배지에서 세정후, 플레이팅을 행했다. 배양접시는 I형 콜라겐―피복 접시, 유지 배지에는, 20ng/㎖ 재조합형 인간 (rh)EGF, 10% 소태아혈청(FBS), 1O-8mol/L 인슐린, 1O-6mol/L 덱사메타존, 100U/mL 페니실린G, 100μU/mL 스트렙토마이신 첨가 윌리엄즈 E 배지를 사용했다.
7.그 상태에서 2∼3주간 배양을 계속하면, 섬유 아세포형의 세포로 이루어지는 콜로니가 접시상에 5∼7개소 정도 형성된다. 이들 이외의 세포는 대부분이 탈락되었지만 이 후의 계대시에 재플레이팅할 수 없어 탈락된다.(이하, 이 세포는 hSGSC(human salivary gland derived stem cell)이라고 칭한다.)
8.100mm접시로부터 트립신-EDTA 처리후에 세포를 회수하고, 6웰 플레이트의 2웰(I형 콜라겐)에 재플레이팅(P2)을 행한다.
(2)단기간(∼P5)의 계대 배양법
이 세포(hSGSC)는 초대 배양에 사용하고 있는 함혈청 배지에서만의 장기 계대는 불가능하다. 분리 초기에는 섬유 아세포형의 형태를 나타내고 있으며, I형 콜라겐 플레이트상에 있어서의 세포밀도도 높지만, P6 이후에서는 대부분이 신전되어 버린다. 또 초기의 계대 직후에는 다수 관찰된 유사분열형태(mitotic figure)도 거의 보여지지 않게 되어 버린다. 또 후술하는 바와 같이, 뉴로스피어법에 의한 스피어유도가 불가능하게 된다. 이들 신전된 세포는, 비멘틴 양성, GFAP 양성이며, 미숙한 아스트로사이트(astrocyte)라고 생각된다.
형태변화를 막고, 계대 회수를 연장시키기 위해서는, (1)계대시의 세포밀도를 높게 유지하고, 및 (2)계대 배양시에, 컨디션드 배지(conditioned medium)를 사용한다는 2가지 점에 유의했다. 계대시의 세포밀도는, 1.0×1O4세포/㎠이상을 유지했다. 계대 배양시에 컨디션드 배지는, 유지 배지로 3배 희석해서 사용했다. 계대는 5∼6일 마다 행했다. 이것에 의해 현재 10대이상의 계대가 가능하다.
(3)타액선유래 세포를 사용한 인슐린생산 세포의 유도법
이 세포는 뉴로스피어법에 따라, bFGF, rhEGF, rhLIF를 함유하는 무혈청 배지에서 배양하면, 구형의 세포덩어리를 형성했다. 여기에서, 뉴로스피어법은 구체적으로 다음과 같이 해서 행했다. 배지조성(Neurobasal medium(인비트로젠), B-27첨가물(인비트로젠) 2%, N-2첨가물(인비토로젠) 1%, 염기성 섬유 아세포 증식인자(R&D) 20ng/㎖, 리콤비넌트 인간 상피세포 성장인자(시그마) 40ng/㎖, 리콤비넌트 인간 백혈병 억제인자(케미콘) 10ng/㎖)에 hSGSC를 현탁해서, 폴리-D-리딘코트 접시(팔콘:60mm직경)에 퍼트렸다. 세포수는 60mm 접시 1매당 2∼3×105 세포정도로 행했다. 배지교환은 1일마다 행하고, bFGF의 반량 보충을 배지교환의 중간일에 행했다. hSGSC는 플레이팅후의 몇일간은 접시면에 접착되어 있지만, 점차로 증식 응집된다. 디시면으로부터 유리되어 이후에는 부유상태 그대로 세포가 증식된다. 이 세포덩어리는, 대부분이 신경줄기세포의 마커인 네스틴 양성세포로 이루어지고, 또 함혈청배지에서 분화 유도를 가한 경우에 Tuj-1 양성세포, GFAP 양성세포, GaIC 양성세포 각각의 출현을 확인했다. 이상의 결과로부터, 이 세포는 신경줄기세포의 전구세포의 능력을 갖는다라고 생각되었다.
이들 타액선으로부터 조정된 hSGSC로부터 스페로이드 배양을 행했다. 플레이트에 수미토모 베이크라이트사의 스페로이드 96U 플레이트(96웰 플레이트)를 사용했다. 배지에는, 윌리엄즈 E 배지의 유지 배지에 GLP-1을 첨가해서 사용했다. 세포수는 3.0×103세포/웰의 밀도로 퍼트렸다.
스페로이드 배양 개시 직후에는, hSGSC는 개별세포의 상태이지만, 2시간이내에 응집을 개시하고, 12시간후에는 완전하게 단일의 스페로이드를 형성했다.(hSGSC 이외의 세포(공포(空胞)상의 대형 포체를 갖는 세포)가, 플레이팅시에 혼입되는 경우가 있지만, 이들 외의 세포는 스페로이드에는 받아들여지지 않는다.)
배양 개시후 7일에, 이들 스페로이드를 항인슐린, 항글루카곤 및 항GFAP 항체로 면역 형광염색을 행했다. 그 결과, 이 스페로이드의 외연은, 글루카곤 양성세포, 스페로이드 코어는 인슐린 양성세포로 이루어져 있었다.
또한 이들 스페로이드로부터 RNA를 회수하고, RT-PCR을 행했다. PDX-1, ngn-3, Glut-2, PAX-4, 인슐린에 대해서 각각 프라이머를 설계해서 PCR을 행한 결과, PDX-1, ngn-3, Glut-2, 인슐린은, 발현이 확인되었다. PAX-4에 대해서는, 발현이 보여지지 않았다. 또, RT-PCR에 사용한 프라이머세트의 염기배열은, 각각 이하와 같았다.
(4)타액선유래 세포를 사용한 무혈청 배지에 있어서의 스피어형성의 유도와 알부민 양성세포의 유도
hSGSC는 상술한 바와 같이 뉴로스피어법에 의해 네스틴 양성세포로 이루어지는 뉴로스피어형의 스피어를 형성한다. 또한 뉴로스피어법의 경우에 배지에 포함되는 FGF-2(basic-FGF) 대신에 FGF-4를 사용해서 배양을 행한 경우에도, 동일 스피어를 형성했다. 이들 스피어(F2스피어, F4스피어)에 대해서 네스틴 RT-PCR(상술)을 행하면 양성이었다. 또 HNF-3a, AFP에 대해서도 동시에 발현이 확인되었다. 또 알부민에 대해서도 발현이 확인되었다. 이들 스피어에 있어서는, PDX-1, ngn-3, Glut-2, PAX-4, 인슐린 등의 발현은 전혀 보여지지 않았다. 하기 표1에 RT-PCR의 프로파일을 나타낸다.

HNF-3a는 성체에 있어서는 내피조직에 한정되어 발현되지만, E8-E9에 있어서는 초기신경조직(early neural tissue)이나 척삭(脊索)(중배엽)에도 발현되는 것이 알려져 있다. F2스피어로부터의 신경마커 양성세포의 유도가 가능하다는 점에서도, hSGSC는 배기(胚期)의 성격을 가진 2관능성의 줄기세포이다라고 생각된다.
또, 본 실시예에 있어서, 세포가 발현되는 마커는 면역세포 염색법 또는 플로사이트 메트리에 의해 조사했다. 염색에 사용한 형광표식 항체는, 모두 시판되는 것이며, 방법의 프로토콜도 시판의 형광표식 항체 또는 시판의 플로사이트 메트리장치에 첨부된 지시서에 따라 행했다. 즉, 면역세포염색은, DAKO사의 면역조직/세포염색 가이드에 따라 행했다. 또한 플로사이트 메트리는 BD 파민젠사의 플로사이트 메트리 염색 프로토콜에 따라 행했다. 사용한 항체의 일람을 하기에 나타낸다.

본 발명의 인간 줄기세포는, 적어도 인간 간세포 및 인간 췌세포로의 분화가 가능하므로, 인간 간장 및 인간 췌장을 재생하기 위한 재생치료에 유용하다. 특히, 본 발명의 줄기세포는, 성체로부터도 얻을 수 있으므로, 재생치료를 받는 환자자신으로부터 채취할 수 있어, 이식시의 거절반응을 회피할 수 있으므로 매우 유리하다.

Claims (10)
- 인간 타액선에 유래하고, CD49f 양성이며, 생체외에서의 배양에 의해, (1)네스틴 양성 및 알부민 양성세포, (2)인슐린 양성세포 및 (3)글루카곤 양성세포로 분화될 수 있는 단리된 인간 줄기세포.
- 인간 타액선세포를 생체외에서 배양함으로써 유도되는 네스틴 양성 및 알부민 양성세포이며, 생체외에서의 배양에 의해 인슐린 양성세포 및 글루카곤 양성세포로 분화될 수 있는 인간 줄기세포.
- 제2항에 기재된 인간 줄기세포를 생체외에서 배양함으로써 유도된 인슐린 양성세포.
- 제2항에 기재된 인간 줄기세포를 생체외에서 배양함으로써 유도된 글루카곤 양성세포.
- 성체유래인 제1항 내지 제4항 중 어느 한 항에 기재된 세포.
- 인간 타액선조직 유래의 세포로부터 CD49f 양성세포를 수집하고, 이들을 상피세포 성장인자의 존재하에서 계대배양하는 것을 포함하는, 인간 타액선에 유래하고, CD49f 양성이며, 생체외에서의 배양에 의해, (1)네스틴 양성 및 알부민 양성세포, (2)인슐린 양성세포 및 (3)글루카곤 양성세포로 분화될 수 있는 인간 줄기세포를 유도하고, 유도된 상기 인간 줄기세포를 회수하는 것을 포함하는 인간 줄기세포의 취득방법.
- 인간 타액선에 유래하고, CD49f 양성이며, 생체외에서의 배양에 의해, (1)네스틴 양성 및 알부민 양성세포, (2)인슐린 양성세포 및 (3)글루카곤 양성세포로 분화될 수 있는 인간 줄기세포를, 섬유 아세포 증식인자, 상피세포 성장인자 및 백혈병 억제인자의 존재하에서, 생체외에서 배양해서 네스틴 양성 및 알부민 양성세포를 유도하고, 유도된 네스틴 양성 및 알부민 양성세포를 회수하는 것을 포함하는 인간 줄기세포의 취득방법.
- 인간 타액선세포를 생체외에서 배양함으로써 유도되는 네스틴 양성 및 알부민 양성세포이며, 생체외에서의 배양에 의해 인슐린 양성세포 및 글루카곤 양성세포로 분화될 수 있는 인간 줄기세포를, 생체외에서 배양함으로써, 인슐린 양성세포를 유도하고, 유도된 인슐린 양성세포를 회수하는 것을 포함하는 인슐린 양성세포의 취득방법.
- 인간 타액선세포를 생체외에서 배양함으로써 유도되는 네스틴 양성 및 알부민 양성세포이며, 생체외에서의 배양에 의해 인슐린 양성세포 및 글루카곤 양성세포로 분화될 수 있는 인간 줄기세포를, 생체외에서 배양함으로써, 글루카곤 양성세포를 유도하고, 유도된 글루카곤 양성세포를 회수하는 것을 포함하는 글루카곤 양성세포의 취득방법.
- 생체외에서의 배양이 스페로이드 배양에 의해 행해지는 제8항 또는 제9항에 기재된 방법.
