제약학적 조성물

FMdC를 캡슐화 하는 가장 바람직한 방법은 방법이 당업계에서 잘 알려진 FMdC를 함유하는 장 코팅 정제, 캡슐, 과립 또는 비드에 의한다. 장 코팅을 위한 바람직한 물질은, 예로써, 셀룰로오스 아세테이트 프탈레이트, 히드록시프로필 메틸셀룰로오스 프탈레이트, 폴리(비닐 아세테이트 프탈레이트), 히드록시프로필 메틸셀룰로오스 아세테이트 숙시네이트, 셀룰로오스 아세테이트 프탈레이트/디에틸프탈레이트, 및, 바람직하게는 폴리(메트)아크릴레이트를 포함한다. 후자는 메타크릴산 및 아크릴산 에스테르 및/또는 메타크릴산 에스테르의 코폴리머를 포함한다. 캡슐이 코팅되는 경우, (히드록시 프로필 메틸셀룰로오스 아세테이트 숙시네이트/트리에틸 시트레이트 또는 특히 셀룰로오스 아세테이트 프탈레이트/디에틸프탈레이트와 같은)가소제가 코팅에서의 부서짐을 최소화하고 코팅의 균열을 억제하기 위하여 사용되어야 한다. 또한 정제 및 과립이 사용될 수 있다.
FMdC의 흡수를 향상시키기 위한 물질 및 화합물이 정제, 캡슐, 과립 또는 비드내에 또한 포함될 수 있다.
완충제가 정제, 캡슐, 과립 또는 비드에 또한 첨가되어 소장의 직접적인 국부 환경의 산성도를 감소시키고 따라서 FMdC의 안정성을 유지시켜 그것이 소장을 통하여 혈류내로 흡수될 수 있도록 한다.
하기 실시예는 본 발명을 설명한다.
조제예 1
하기 성분을 함유하는 경질 젤라틴 캡슐을 제조한다:
상기 성분들을 혼합하여 340mg 양으로 경질 젤라틴 캡슐에 채운다. 필요하면, 경질 젤라틴 캡슐을 약 4 내지 5 또는 그이상의 pH까지 용해되지 않는 제약학적으로 허용가능한 물질로 오버코팅한다.
조제예 2
정제 제형을 아래의 성분을 사용하여 제조한다.
구성 성분들을 배합하고 압착하여 무게가 각각 100mg인 정제를 형성시킨다. 정제를 약 4 내지 5 또는 그이상의 pH까지 용해되지 않는 제약학적으로 허용가능한 물질로 오버코팅한다.
조제예 3
정제 제형을 아래의 성분을 사용하여 제조한다.
구성 성분들을 배합하고 압착하여 무게가 각각 100mg인 정제를 형성시킨다. 정제를 약 4 내지 5 또는 그이상의 pH까지 용해되지 않는 제약학적으로 허용가능한 물질로 오버코팅한다.
조제예 4
각각 FMdC의 30mg을 함유하는 정제를 하기와 같이 제조한다:
FMdC, 전분 및 셀룰로오스를 No. 20 메시 U.S.체를 통과시키고 충분히 혼합한다. 폴리비닐피롤리돈의 용액을 결과적인 분말과 혼합하고, 다음에 그것을 16 메시 U.S.체를 통과시킨다. 그렇게 생성된 과립을 50℃ 내지 60℃에서 건조시키고 16 메시 U.S.체를 통과시킨다. 다음에 미리 No. 30 메시 U.S.체를 통과시킨 나트륨 카르복시메틸 전분, 스테아르산 마그네슘 및 활석을 과립에 첨가하고, 그것을 혼합후 정제 기계에서 압착시켜 무게가 각각 150mg인 정제를 얻는다. 다음에 정제를 약 4내지 5 또는 그이상의 pH까지 용해되지 않는 제약학적으로 허용가능한 물질로 오버코팅한다.
조제예 5
각각 FMdC 40mg을 함유하는 캡슐을 하기와 같이 만든다:
활성 성분, 셀룰로오스, 전분, 스테아르산 마그네슘을 배합하고, No. 20 메시 U.S.체를 통과시키고, 150mg 양으로 경질 젤라틴 캡슐에 채운다. 필요하면, 경질 젤라틴 캡슐을 약 4 내지 5 또는 그이상의 pH까지 용해되지 않는 제약학적으로 허용가능한 물질로 오버코팅한다.
Claims (8)
- 포유 동물에서 신생물 질환 또는 바이러스 질환을 치료하기 위한 2′-디옥시-2′-(플루오로메틸렌)시티딘 유효량을 포함하는 경구적으로 송달가능한 제약학적 조성물로서, 상기 조성물이 약 4 내지 5 이하의 pH에서는 내 용해성이고 약 4 내지 5를 초과하는 pH에서는 쉽게 용해되도록 선택되는 물질에 캡슐화되어 있는 것을 특징으로 하는 제약학적 조성물.
- 제 1 항에 있어서, 제약학적으로 허용가능한 부형제 또는 부형제들을 더 포함하는 것을 특징으로 하는 경구적으로 송달가능한 제약학적 조성물.
- 제 2 항에 있어서, 제약학적으로 허용가능한 부형제 또는 부형제들이 단지 캡슐화 물질을 포함하는 것을 특징으로 하는 경구적으로 송달가능한 제약학적 조성물.기
- 제 2 항에 있어서, 별도의 제약학적으로 허용가능한 부형제 또는 부형제들이 캡슐화 물질에 포함되는 것을 특징으로 하는 경구적으로 송달가능한 제약학적 조성물.
- 제 1 항, 제 2 항, 제 3 항 또는 제 4 항에 있어서, 캡슐화 물질이 셀룰로오스 아세테이트 프탈레이트, 히드록시프로필 메틸셀룰로오스 프탈레이트, 폴리(비닐 아세테이트 프탈레이트), 히드록시프로필 메틸셀룰로오스 아세테이트 숙시네이트, 폴리(메트)아크릴레이트 및 셀룰로오스 아세테이트 프탈레이트/디에틸프탈레이트로 구성되는 군으로 부터 선택되는 것을 특징으로 하는 경구적으로 송달가능한 제약학적 조성물.
- 제 5 항에 있어서, 캡슐화 물질이 메타크릴산 및 아크릴산 에스테르의 코폴리머 그리고 메타크릴산 및 메타크릴산 에스테르의 코폴리머로 구성되는 군으로 부터 선택되는 것을 특징으로 하는 경구적으로 송달가능한 제약학적 조성물.
- 제 1 항에 있어서, 조성물이 제약학적으로 허용가능한 부형제(들) 약 50 내지 약 99.5 중량% 및 2′-디옥시-2′-(플루오로메틸렌)시티딘 약 0.5 내지 약 50 중량%를 포함하는 것을 특징으로 하는 경구적으로 송달가능한 제약학적 조성물.
- 포유 동물에 경구적으로 송달되는 경우 2′-디옥시-2′-(플루오로메틸렌)시티딘의 경구적 생체이용률을 향상시키는 방법으로서,(a) 4 내지 5 이하의 pH에서 내 용해성이고 4 내지 5를 초과하는 pH에서는 쉽게 용해되도록 선택된 제약학적으로 허용가능한 물질에 2′-디옥시-2′-(플루오로메틸렌)시티딘을 캡슐화 하는 단계; 및(b) 상기 (a)에서 제조된 제품을 상기 포유동물에 경구적 송달하는 단계를 포함하는 방법.
