Abstract
본 발명은 (ⅰ) 공지된 방법으로 비만세포 및/또는 호염기구에 결합하며, 그에 의해서 흡수되는 단백질과, (ⅱ) 비만세포 및/또는 호염기구 분비장치의 하나 이상의 단백질을 분해하는 프로테아제로 구성되는 혼성 단백질에 관한 것이다.
Description
직접형 알레르기 반응(allergic reactions of the immediate type)은 해당 환자가 알레르겐(예를 들면, 꽃가루, 집안의 먼지, 진드기, 동물의 털)에 대하여 IgE형의 항체를 형성하는 특성이 있다. 이 항체들은 혈액내에서 순환할 뿐만 아니라, 원형질막에 나타나는 세포조직의 세포들에 IgE분자의 일부에 대한 특정한 수용체인 Fc 단편(Fc fragment)을 결합한다(Fishman Lorberboum-Galski 1997; Hamawy 1997). IgE 수용체를 가진 세포들은 오로지 비만세포(mastocytes)와 호염기구(basophils)이다. 이 세포들은 직접형 알레르기 반응을 일으키는 세포들이다. 이 세포들은 혈관활성아민과 프로스타글란딘(prostaglandins)과 류코트리엔(leukotrienes)(아라키돈산의 유도체)을 포함하는 소포(vesicle)를 저장한다. 비만세포의 과립소실을 초래하는 이 물질들의 방출은, 특정하거나 특정하지 않은 메카니즘을 거쳐서 일어난다. 일단, 예를 들어, 피부의 긁힘과 같이 세포가 기계적으로 파괴되면, 히스타민이 불특정하게 방출된다. 상처난 피부는 붉어진다. 부종이 생기고, 피부가 가려워진다(3중 반응). 특히, 히스타민을 방출하는 물질들은 상대적으로 낮은 농도에서 효과적이며, 이하의 연속단계 반응(signal cascade: 신호 연속단계)을 일으킨다: 포스포리파아제 C의 활성화 - 제 2메신저“디아실글리세롤”과“IP3”의 형성 - 세포 저장소로부터의 칼슘의 유동 - 세포막과 과립의 융합 - 세포융해없이 과립의 세포외 배출작용 - 헤파린과 염기성 단백질의 복합체인 양으로 하전된 히스타민에 대한 나트륨의 교환 - 과립기질로부터의 히스타민의 방출. 세포 표면의 IgE분자는 알레르기성 체질인 사람의 비만세포와 알레르겐 사이에 접촉하여 제공되어, 이 알레르겐과 결합하게 된다. 일단 알레르겐 분자는 원형질막에서 충분한 양의 수용체 집합체와 결합된다. 그 집합체는 세포 내부에서 상술한 신호 연속단계를 유도하기 위한 특정한 자극제이다. 방출된 물질들은 알레르기 증상(결막염, 비염, 천식, 후두 부종, 두드러기, 혈압의 저하로 인한 과민성 쇼크)을 유발시킨다. 비만세포 과립소실 펩타이드(MCD)와 같이 벌의 독소에 포함된 펩타이드도 비만세포의 과립소실을 초래한다. 게다가, 일부 약품들은 바람직하지 않은 효과로서 히스타민의 특정한 방출을 일으킨다. 인간에게서 히스타민의 방출은 근육이완제, 덱스트런, 아세틸살리실산(아스피린), 모르핀, 항생제, 방사선 사진에서의 조영제, 이질 혈청등에 대하여 기술된다.
원형질막과 소포의 융합이 성공적으로 억제되면, 아민과 아라키돈산 유도체의 방출이 일어나지 않는다. 결과적으로, 알레르기 반응이 일어나지 않는다. 몇몇 단백질(융합 단백질)은 단백질이 분비성 소포막 및/또는 원형질막에 결합될 수 있는 분비과정과 방출에 각각 연관된다. 마찬가지로, 그것들은 세포기질에서 나타난다. 대표적인 단백질로 SNAP 25, 시납토브레빈(synaptobrevin)(VAMP), 신택신(syntaxine)과 그 이소형이 있다. 이 단백질들은 분비성 소포를 원형질막의 내측에 고정시키는 복합체(fusion complex: 융합 복합체)를 형성한다. 고정은 원형질막과 소포의 융합을 예견하며, 상기 융합은 IgE에 의해서 야기되는 Ca++의 유입에 의해서 일어난다. 이 단백질 중 하나의 비활성화 예를 들면, 단백질 분해에 의해서, 복합체의 형성이 억제된다.
언급된 융합 단백질은 신경세포에서 보툴리누스균(bacillus Clostridium botulinum)에 의해서 생성된 신경독소의 L쇄의 타겟분자(기질)인 것으로 알려져 있다(Ahnert-Hilger & Bigalke, 1995; Bigalke 1999 in press). 현재 보툴리누스 독소의 7가지 다른 형태가 알려져 있다(A, B, C1, D, E, F, G). 언급된 시납토브레빈은 추가적으로 파상풍균(Clostridium botulinum)에 의해서 생성된 TeNT(Link et al., 1993)과 임균(Neisseria gonorrhoeae)으로부터의 프로테아제(Binscheck et al., 1995)의 타겟분자이다. 프로테아제로부터 분리된 독소는 적어도 두 개의 기능영역으로 구성된다. 단백질의 C말단부(heavy chain: H쇄)는 신경세포로의 결합을 맡는 반면에, N말단부(light chain: L쇄)는 상술한 매우 특별한 단백질분해 활동을 하는 특징이 있다. 독소는 H쇄를 통하여 신경세포에 결합하고, 수용체를 중재하는 세포내흡입작용과 잇따른 이동을 거쳐서 세포기질에 도달하며, 거기서 번갈아 융합 복합체를 구성하는 언급된 하나 이상의 융합 단백질을 분해시킨다. 각각의 단백질을 분해한 후에 신경 세포로부터 아세틸콜린과 다른 신경전달물질의 분비가 억제된다(Binscheck and Wellhoner, 1997).
신경전달물질의 분비억제는 과거에 이상긴장의 운동신경 장애를 치료하고 과도한 부교감 신경의 활동을 억제하기 위한 치료법으로 사용되어 왔다.(Benecke and Kessler, 1995) 방추형 신경독소에 대하여 융합 단백질 이외의 생물학적 기질이 알려져 있지 않다. H쇄는 말초신경세포에 대하여 높은 친화력을 가짐으로써, 그것에 연결된 L쇄가 이 세포들에만 도달하고 이 세포들에서만 효력을 갖게 된다 - 그러나 비록 분비과정이 일어나는 비만세포 및 호염기구와 같은 다른 형태의 세포들이 상술한 기질을 가지고 있다 할지라도, 프로테아제의 흡수를 위한 메카니즘을 갖지는 않는다(Marxen et al., 1989).
본 발명의 일실시예는 혼성 단백질과 관련이 있으며,
(ⅰ) 비만세포 및/또는 호염기구에 결합하고, 통상적으로 공지된 방식으로 이 세포들에 의해서 흡수되는 통상적으로 알려진 단백질과,
(ⅱ) 비만세포 및/또는 호염기구 분비장치의 하나 이상의 단백질을 분해하는 통상적으로 알려진 프로테아제
를 포함하여 구성된다.
본 발명의 또 다른 실시예는
(ⅰ) 비만세포 및/또는 호염기구에 결합하고, 이 세포들에 의해서 흡수되는 단백질에 있어서,
IgE와,
IgE 단편, 특히 IgE Fc 단편과,
비만세포 및/또는 호염기구의 IgE수용체에 대한 항체와,
비만세포 및/또는 호염기구의 IgE수용체에 대한 항체의 단편, 특히 Fab 단편과,
비만세포-특정 칼륨채널(channel)에 대한 항체와,
비활성이지만 결합을 하는 MCD 펩타이드로 구성된 그룹에서 채택된 단백질과,
(ⅱ) 특히 비만세포 및/또는 호염기구 분비장치의 하나 이상의 단백질을 분해하는 통상적으로 알려진 프로테아제
를 포함하여 구성되는 혼성 단백질과 관련이 있다.
본 발명의 또 다른 실시예는
(ⅰ) 특히 비만세포 및/또는 호염기구에 결합하고, 통상적으로 공지된 방식으로 이 세포들에 의해서 흡수되는 통상적으로 알려진 단백질과,
(ⅱ) 비만세포 및/또는 호염기구 분비장치의 하나 이상의 단백질을 분해하는 프로테아제에 있어서,
보툴리누스균독소, 특히 A, B, C1, D, E, F, G형 독소의 L쇄와,
보툴리누스균독소, 특히 A, B, C1, D, E, F, G형 독소 L쇄의 촉매적으로 활성화된 단편과,
파상풍균독소(TeNT)의 L쇄와,
파상풍균독소 L쇄의 촉매적으로 활성화된 단편과,
임균의 IgA 프로테아제와,
임균의 IgA 프로테아제의 촉매작용 영역으로 구성된 그룹에서 채택된 프로테아제를 포함하여 구성되는 혼성 단백질과 관련이 있다.
본 발명의 혼성 단백질은 단백질(ⅰ)과 프로테아제(ⅱ)가 이전의 단백질 및 프로테아제의 그룹에서 각각 선택되는 것을 특징으로 한다.
또한 본 발명의 혼성 단백질은 각각의 독소(HN단편)나 그 단편의 H쇄의 N말단부가 보툴리누스균독소나 파상풍균독소의 L쇄에 더하여 혼성 단백질의 일부가 될 수 있는 특징을 갖는다.
결국, 본 발명의 일실시예는 비만세포의 과립형성을 억제하기 위한 혼성 단백질의 사용에 관한 것이다.
비만세포가 죽으면, 일단 죽은 비만세포가 저장된 내인성 아민을 방출시켜 알레르기 쇼크가 발생하는 위험이 존재한다. 게다가, 비만세포 수의 감소는 번갈아 알레르기 반응에 다시 이용할 수 있는 세포들의 새로운 합성을 자극할 것이다. 본 발명의 혼성 단백질은 ADP 리보실화 활동에 의해서 단백질 합성을 억제하여 세포를 죽이는 IgE-Fc/녹농균 균체외독소 접합체(conjugate)와 근본적으로 다르다(Fishman & Lorberboum-Galski, 1997). 완전히 반대로, 본 발명의 혼성 단백질은 비만세포를 죽이는 데 도움이 되지 않는다. 오히려, 세포는 본 발명의 혼성 단백질의 처리 후에도 살아남게 되며, 오직 혈관수축 아민을 방출할 정도까지만 그 수용력을 잃는다.
새로운 합성의 자극이 일어나지 않는다. 치료법으로 사용될 때, 완벽한 세포독성의 녹농균독소나 비교할 만한 세포독소에 기초한 접합체의 예상 가능한 독성부작용을 피할 수 있다.
따라서 본 발명의 주제는 (ⅰ) 비만세포/호염기구에 대한 높은 친화력을 나타내는 단백질이나 펩타이드(수송 단백질/펩타이드)와 (ⅱ) 접합체가 세포의 과립소실과 분비 메카니즘을 각각 차단하는 특정 프로테아제로 구성되는 접합체(혼성 단백질)일 수 있다. 그 접합체는 직접형 알레르기 반응의 치료와 예방에 유용하다.
(ⅰ) 접합체의 고친화성 비만세포 결합 구성성분은 E형의 면역 글로블린(IgE)과 그것의 단편(예를 들면, Fc-단편)이다. 게다가, 비만세포/호염기구의 특정한 표면분자에 대한 항체가 이용되며, 항체는 이 세포들의 원형질막에 선택적으로 결합한다. 특히, IgE 수용체에 대한 항체들이 이 목적을 달성한다. 또한, 비만세포 과립소실 펩타이드의 비활성이지만 결합하는 돌연변이는 혼성 단백질에서 수송 펩타이드/단백질로서 사용된다. 이 수송 펩타이드/단백질은 프로테아제를 세포로 보내기 위해 사용된다. 이 프로테아제는 단백질이 세포의 과립소실 메카니즘을 개시하는 매우 특별한 방식으로 비만세포의 융합 복합체에서 단백질을 분해한다.
(ⅱ) 금속프로테아제(metalloprotease), 예를 들면 A, B, C1, D, E, F, G형의 보툴리누스균독소(BoNT/X)와 파상풍균독소(TeNT)의 L쇄나 임균의 IgA 프로테아제는 매우 특별한 프로테아제로서 사용된다. 이 프로테아제는 시납토솜결합 단백질(MR25,000)(SNAP 25)이나 시납토브레빈 혹은 신텍신(syntaxin)을 분해시킨다. 이 단백질/펩타이드 중 하나만이 분해되어도, 비만세포의 과립소실이 억제된다. 결과적으로, 히스타민, 프로스타글랜딘, 루커트리엔이 분비되지 않으며, 알레르기 증상이 더 이상 일어나지 않게 된다.
본 발명에서, 독소의 무독성 L쇄는 비만세포와 호염기구에 각각 결합하는 단백질들을 운송하도록 부착되어서 오직 이 세포들에 의해서만 흡수될 수 있으며, 마치 승객을 실어나르는 것과 같이 세포에 도달하게 된다. L쇄는 유기체의 신경세포와 다른 형태의 세포에 침입할 수 없어서 그 영향이 비만세포와 호염기구로 제한된다. 기질 중의 하나가 단백질 분해적으로 파괴되면, IgE가 부착된 세포가 알레르겐이나 상술한 약품 중의 하나와 접촉한 후에 알레르기 증상이 나타나지 않는다.

L쇄는 거기서 그것의 기질을 분해하며, 그 분해가 히스타민과 다른 물질들의 분비를 억제한다. 접합체를 준비하기 위한 두번째 방법은 L쇄의 유전자와 네 개의 비만세포-결합 단백질 중 하나의 유전자를 융합함으로써 혼성 단백질이 적당한 숙주세포(host cell)에서 발현되는 것이다. 이렇게 생물공학적으로 생성된 혼성 단백질은 두 개의 단백질 구성성분으로부터 준비된 접합체와 유사하게 비만세포로부터 분비과정을 차단한다. 혼성 단백질의 준비는 특히 종양치료 분야에서 통상적으로 공지된 절차이다(Vogel, 1987; Magerstadt, 1991). 이러한 치료개념에서, 종양세포의 표면 단백질에 대한 항체는 세포독소 단백질, 예를 들어, 리신, 디프테리아 독소에 부착되어 암세포를 죽인다. 본 발명의 방법에 있어서의 새로운 양상은 비만세포의 과립소실을 억제하는 항알레르기 치료법을 위한 혼성 단백질에서 특정 프로테아제와 단백질분해 영역을 각각 사용하는 것이다. 이 혼성 단백질은 건강에 해로운 알레르기 증상(발열, 천식, 신경피부염)을 피하기 위해서 사용되는 것만은 아니다. 혼성 단백질은 구명약품으로 치료하는 동안에 알레르기 반응을 피하기 위해서 예방적으로 복용될 수 있다. 게다가, 혼성 단백질은 알레르기를 없애는 과정에서 일어나는 알레르기 증상을 피할 수 있다.
예시 1: IgE와 BoNT/A의 L쇄로부터의 혼성 단백질의 합성
15mM의 사붕산나트륨과 pH 8.4의 30mM 인산염에서 평형을 이룬 후, QAE Sephadex 관(1.0×3.0cm)에 동일한 완충제로 평형을 이루면서 A형의 정제된 보툴리누스균독소(5.0 mg)를 가했다. 관은 120mM의 디티오에리트롤과 2M의 우레아와 1mM의 EDTA 10ml로 씻고, 밤새 항온처리하였다. 그 후, L쇄가 10mM의 붕산염 완충제에 의해서 관으로부터 용리되고(elute), pH7.0의 20mM 인산염에 대하여 투석되었다.
면역 글로블린 E(쥐)가 얻어졌다. 1ml의 인산염 완충제에서 50㎍의 파파인으로 10mg의 면역 글로블린이 분해되었다(밤새 4℃). Fc 단편이 겔여과관(Sephacryl S200)을 거쳐 정제되었다. 정제된 Fc 단편 3.0mg이 16시간에 걸쳐서 pH 7.0의 인산나트륨 2ml에서 10mM의 디티오비스숙신이미딜프로피오네이트(dithiobissuccinimidylpropionate)(2기 작용제)로 정제된 보툴리누스균독소의 L쇄 3.0mg과 항온처리되었다. 이런 식으로 합성된 혼성 단백질은 겔 여과기(Sephacryl S200)를 거쳐서 정제되고, SDS 겔 전기이동을 거쳐서 그것의 순도가 분석되었다.
비만세포의 과립소실을 억제하는 것이 두 가지 실험적인 접근방법에 의해서 검사된다. 제 1접근방법에서는 쥐에서 분리해낸 비만세포를 혼성 분자와 함께 항온처리한다. 그 후, 히스타민의 방출을 자극한다. 자극은 MCD 펩타이드와 콘카나발린A(콘카나발린A는 실험적으로 이용되는 물질)와 같은 특정 히스타민 유리자에 의해 일어나며, 또한 세포내 칼슘농도를 직접적으로 증가시킴으로써 일어난다. 세포내 칼슘농도의 직접적인 증가는 다른 비만세포로 칼슘을 주입함으로써 달성된다. 따라서, 칼슘 농도의 증가와 같이 상기 언급된 신호 연속단계의 짧은 회로는 소포의 융합에 의해서 일어나는 분비과정 동안의 단계이다. 히스타민의 방출을 나타내는 비만세포의 과립소실은 위상차 현미경에서 나타난다. 다음에 형광 측정에 의해서 방출된 히스타민의 양을 측정하는 것이 가능하다. 결국, 과립소실 동안에 소포막을 원형질막에 결합시킴으로써 야기되는 비만세포의 확대는 전기생리학적으로 판정될 수 있다. 혼성 단백질로 처리되는 새포에서는 대조 세포와 비교하여, (1) 형태 변화가 일어나지 않고, (2) 세포의 상층에서 형광성의 증진이 일어나지 않으며, (3) 세포의 확대도 일어나지 않을 것이다. 따라서, 히스타민의 방출이 혼성 단백질에 의해서 차단되는 것을 증명하는 것이 가능하다.
제 2접근방법에서는 살아있는 쥐에 혼성 단백질을 주입한다. 쥐는 며칠 후에 죽고, 비만세포는 통상적으로 분리된다. 과립소실과 히스타민의 방출은 각각 상술한 바와 같이 판정된다. 이 접근법에서는 접합체가, 살아있는 동물에서도 비만세포가 위치하는 구역에 도달할 수 있는지와, 접합체가 살아있는 동물의 비만세포를 비활성화시키는지를 검사한다.
예시 2: A형의 보툴리누스균의 L쇄를 암호화한 유전자를 면역 글로블린 E와 그것의 단편(Fc 단편)중의 하나를 암호화한 유전자에 각각 조작가능하게 결합함으로써 재조합형 혼성 단백질을 생산
A형 보툴리누스균독소의 L쇄를 암호화한 유전자는 PCR(polymerase chain reaction: 중합효소 연쇄반응)을 거쳐서 적절한 시발체에 의해서 분리된다. A형의 보툴리누스균의 배양이 준비되며, 그로부터 DNA가 준비된다. 공지된 독소 유전자의 배열로부터(Binz et al.) 한 쌍의 시발체가 얻어지며, 라이트 소단위(light sub-unit)에 대한 유전자는 PCR을 거쳐서 증폭된다. 그 후, 이 유전자는 생산자의 처방에 따르는 상업용 발현벡터 pQE로 복제된다.
인간의 면역 글로블린 E의 Fc 단편을 암호화한 유전자(Helman L.)는 PCR을 거쳐서 상업용 cDNA 자료로부터 분리되고, A형 보툴리누스균독소의 L쇄 유전자와 벡터구조(vector construct)로 용융되었다.
이러한 구조로 콤피덴트균 M15(competent M15 cells)(E. coli)이 형질전환된다. 이러한 발현체계에서 삽입된 유전자가“표지(his tag)”와 함께 갖추어질 때, 재조합 단백질이 니켈 친화성 관을 거쳐서 정제된다. Sephacryl S300을 통한 겔여과에 단백질을 고도로 정제하는 과정이 잇따른다.
생물학적 활성의 측정이 생체외 분리된 비만세포에서 다시 실행된다.
예시 3: 파상풍균독소의 라이트 소단위체(light sub-unit)를 암호화한 유전자를, 비만세포 과립소실 펩타이드(MCD)를 암호화한 돌연변이 유전자와 결합함으로써 재조합 혼성 단백질을 준비
비만세포 과립소실 펩타이드에 대하여 22분자배열(22mer)이 알려져 있다(Gmachl and Kreil). 대응하는 올리고뉴클레오티드에 기초하여 합성된다.
파상풍균독소의 라이트 소단위체의 배열을 분리하기 위해서 파상풍균의 배양이 준비되며, 그것으로부터 DNA가 회수되었다. 공지된 파상풍균독소의 핵산배열로부터 PCR에 대한 시발체가 얻어지며, 이어서 독소의 라이트 소단위체에 대한 유전자가 얻어진다.
예시 1에서 나타낸 바와 같이, 양쪽의 핵산 배열은 발현벡터pQU로 융합되며, 그 후 E.coli에서 발현된다. 표지(his-tag)와 함께 차례로 갖추어진 혼성 단백질은 친화 크로마토그래피와 잇따른 겔여과를 거쳐서 정제된다. 비만세포 과립소실 펩타이드를 암호화한 정제된 유전자는 펩타이드의 활성영역에서 점돌연변이를 포함하여 화학적으로 합성된다. 그 유전자는 파상풍균독소의 L쇄를 암호화한 유전자에 결합된다. 혼성 단백질은 E.coli에서 발현되고, 정제된다. 이런 식으로 생성된 혼성 단백질은 실험관내에서 비만세포 과립소실 검정으로 테스트된다.
예시 4: IgA프로테아제를 암호화한 유전자에 IgE의 Fc단편을 암호화한 유전자를 결합함으로써 재조합 혼성 단백질을 준비
IgE의 Fc단편을 암호화한 유전자가 예시 1에 설명한 바와 같이 분리되었다.
임균으로부터 IgA프로테아제를 암호화한 유전자가 알려져 있다. 그로부터 시발체가 얻어지고, 특정 프로테아제를 암호화한 유전자가 임균에서 얻어진 핵산의 준비로부터 PCR에 의해서 회수된다.
양쪽의 핵산이 생산자에 의해서 주어진 처방에 따르는 상업용벡터로 통합되고, 혼성 단백질이 친화 크로마토그래피에 의해서 정제되었다(예시 2 참조).
억제 활성도(inhibitory activity)가 생체외 분리된 비만세포에서 다시 입증된다(상기 참조).
예시 5: IgE 수용체에 대한 항체의 Fab 단편과 B형 보툴리누스 독소의 L쇄로 구성된 혼성 단백질의 준비
비만세포에서 IgE 수용체에 대한 단일클론항체가 얻어지며, 크로마토그래피로 재정제되었다. 0.5mg의 항체가 보툴리누스 독소 F의 정제된 L쇄 0.4g에 접합되었다. 일단 예시 1의 절차에 따라서 신경독소가 준비되면, 신경독소의 분해와 뒤이은 이온교환 크로마토그래피를 거친 정제에 의해서 라이트 소단위체가 분리된다.
양쪽의 단백질(F형 독소의 라이트 소단위체와 단일클론항체)은 2기 작용제를 사용하여 서로 결합된다. 이러한 목적을 위해서 분리된 단백질이 10mM의 말레이미도벤조일-N-히드록시-숙신이미드에스테르(maleimidobenzoyl-N-hydroxy-succinimidester)와 항온처리된다. 이어서 혼성 단백질이 Sephacryl S300에 걸쳐서 겔여과에 의해서 비접합된 단백질로부터 정제된다.
다시 분리된 비만세포는 합성된 혼성 단백질이 히스타민의 분비를 억제하는 것을 증명하기 위해서 사용되었다.

Claims (6)
- 혼성 단백질에 있어서,(ⅰ) 비만세포(mastocytes) 및/또는 호염기구(basophils)에 결합하고, 통상적으로 공지된 방식으로 이 세포들에 의해서 흡수되는 통상적으로 알려진 단백질과,(ⅱ) 비만세포 및/또는 호염기구 분비장치의 하나 이상의 단백질을 분해하는 통상적으로 알려진 프로테아제를 포함하여 구성되는 것을 특징으로 하는 혼성 단백질.
- (ⅰ) 비만세포 및/또는 호염기구에 결합하고, 이 세포들에 의해서 흡수되는 단백질로서,IgE와,IgE 단편(fragment), 특히 IgE Fc 단편과,비만세포 및/또는 호염기구의 IgE수용체에 대한 항체와,비만세포 및/또는 호염기구의 IgE수용체에 대한 항체의 단편, 특히 Fab 단편과,비만세포-특정 칼륨채널(channel)에 대한 항체와,비활성이지만 결합을 하는 MCD 펩타이드로 구성된 그룹에서 채택된 단백질과,(ⅱ) 특히 비만세포 및/또는 호염기구 분비장치의 하나 이상의 단백질을 분해하는 통상적으로 알려진 프로테아제를 포함하여 구성되는 혼성 단백질.
- (ⅰ) 특히 비만세포 및/또는 호염기구에 결합하고, 통상적으로 공지된 방식으로 이 세포들에 의해서 흡수되는 통상적으로 알려진 단백질과,(ⅱ) 비만세포 및/또는 호염기구 분비장치의 하나 이상의 단백질을 분해하는 프로테아제로서,보툴리누스균독소, 특히 A, B, C1, D, E, F, G형 독소의 L쇄(light chain)와,보툴리누스균독소, 특히 A, B, C1, D, E, F, G형 독소의 L쇄의 촉매적으로 활성화된 단편과,파상풍균독소(TeNT)의 L쇄와,파상풍균독소의 L쇄의 촉매적으로 활성화된 단편과,임균의 IgA 프로테아제와,임균의 IgA 프로테아제의 촉매작용 영역으로 구성된 그룹에서 채택된 프로테아제를 포함하여 구성되는 혼성 단백질.
- 제 2항 또는 제 3항에 있어서,(ⅰ) 제 2항에 따른 그룹에서 채택된 단백질과,(ⅱ) 제 3항에 따른 그룹에서 채택된 프로테아제인 것을 특징으로 하는 혼성 단백질.
- 제 4항에 있어서,각각의 독소(HN단편)나 그 단편의 H쇄(heavy chain)의 N-말단부는 보툴리누스균독소나 파상풍균독소의 L쇄에 더하여 혼성 단백질의 일부인 것을 특징으로 하는 혼성 단백질.
- 비만세포의 과립소실을 억제하기 위한 제 1항 내지 제 5항에 기재된 혼성 단백질의 사용방법.
