Abstract
본 발명은, 억제제로서 미세입자의 형태를 띠고 있는 선형 불수용성 다당류, 바람직하게는 다당류 폴리(1,4-알파-D-글루칸)을 전체적 또는 부분적으로 포함하고 있는, 방출이 조절되는 정제(a controlled release tablet)에 관한 것이다. 상기 정제는 또한 활성제의 방출을 조절할 수 있다.
Description
현대 제약 기술에서는, 그 투여 형태가 생체분포(biodistribution), 생체내 효율(bioavailability), 생체적합성(biocompatibility) 및 흡수에 대하여 영향을 미치는 부형제(excipient)의 조제(formulation)가 중요하다. 더 나아가 부형제는 적당한 경도(hardness)와 장력(tension) 및 응력(stress)에 대한 저항과 같은 좋은 역학적 특성을 가져야만 한다. 비록 몇몇 화합물은 이미 압착(press)되어 꽉 짜인 안정화된 덩어리 (예:자당 또는 젖당)가 되었다 할 지라도, 결합제(binder), 충전제(filler), 윤활제(lubricant) 및 첨가제(additive)와 같은 성분, 즉 정제 보조제(tablet auxiliaries)도 역시 필요하다. 여기서 사용되는 안정도를 증가시키는 전형적인 건조 결합제(dry binder)는 다음과 같다.
즉, 인산칼슘(calcium phosphate), 미세결정질 셀룰로스(microcrystalline cellose)[예: Avicel(등록상표), PH102(등록상표), 특히 Celsphere], 폴리비닐피로리돈(polyvinylpyrrolidones)[예:Kollidon(등록상표), Luviskol VA64(등록상표), Plasdone(등록상표)], 옥수수, 밀가루 또는 감자 전분, 유도된 다당류, 이른바 고무(gum)(예:크산틴 고무), 셀룰로스 유도체[예:하이드록시프로필 메틸셀룰로스; Klucel(등록상표)] 또는 에틸셀룰로스[Aqualon(등록상표)]. 게다가 부형제는 체내에서 체액과 접촉하여 최적적으로 분해되어야하고, 그 분해를 조절할 수 있어야한다. 따라서 이른바 붕괴제(disintegrants)를 분해의 조절을 위해서 때때로 첨가할 수 도 있다. 이러한 목적을 위한 전형적인 화합물은 옥수수 전분, 젤라틴화된 전분 및 전분 변형물이다. 또한 수분흡수 및 이에 수반되는 팽창으로 인한 분해력(disintegrating power)을 증가시키는 물질도 상기 목적을 위해서 이용될 수 있는데, 이러한 것들로서는 교차결합된(crosslinked) 폴리비닐피로리돈[Kollidon CL(등록상표)], 카르복시 메틸셀룰로스 및 이들의 칼슘염 또는 갈락토마난(galactomannan)이 있다. 몇몇 화합물의 경우[예:Avicel(등록상표) 및 PH 102(등록상표)], 필요한 역학적 안정성을 획득하고 정제의 분해를 조절하는 것이 모두 가능하다.
아밀로오스를 포함한 특정한 전분들이 정제 조제에 적합한 부형물이라고 기술되어 있다[Journal of Pharmaceutical Sciences 55(1966), 340]. 그러나 미국의 A.E.Stanley Manufacturing Co에서 사용된 Nepol 아밀로오스는 부적합한 것으로 밝혀졌다. 왜냐하면 활성화합물이 별로 방출(release)되지 아니하고 부형제의 수분함량이 10 - 12%로 높기 때문인데, 이는 가수분해되어 불안정한 활성화합물을 조제(formulated)할 수 없는 원인이 된다. 특히 S.T.P. Pharma Science 4 (1994), 329-335 와 Journal of Controlled Release 15,(1991) 39-46 및 Journal of Controlled Release 15, (1991)3946에서는 교차결합된 아밀로오스(교차결합도 15%)를 우수한 결합제(binding agent)로 기술하고 있는 데, 이는 수분흡수력 때문에 분해촉진제(disintegration accelerator)로 작용한다. WO 94/21236에서는 교차결합된 아밀로오스(교차결합도 25%)를 결합제와 붕괴제로 사용하고 있다. 그러나 높은 교차결합도는 생물학적 적합성(biological compatibility)에 부정적인 영향을 미친다. 사용된 교차결합제는 불내성(intolerable) 에피클로로히드린(epichlorohydrin) 약 30 중량% 이다. 교차결합도가 수 % 정도로 낮다 하더라도 반응이 급격히 느려지고, 따라서 남아있는 비반응 교차결합제(crosslinker)의 잔류물이 존재할 것으로 예상된다.
현재까지 시장에 나와있는, 모든 전분과 아밀로오스를 함유한 부형제는 식물로부터 만들어진 것이다. 자연에 존재하는 물질과 같은 이러한 생중합체(biopolymer)에, 조성물과 구조의 변이(variation)가 상당량 존재하는 것은 불이익한 점이 많으며, 이로 인해 필요한 재현성(reproducibility)과 일정한 생산물의 품질을 보장할 수 없게되는 바, 특히 활성화합물의 방출을 조절하는 면에서 그러하다.
천연전분의 경우, 아밀로오스와 아밀로펙틴의 함량은 그 출처에 따라 상당히 달라진다. 예를 들면, 감자전분은 아밀로오스를 약 20 중량%, 아밀로펙틴을 약 80 중량%를 함유하고 있으나, 옥수수전분은 아밀로오스를 약 50 중량%, 아밀로펙틴을 약 50 중량%를 함유하고 있다. 식물체내의 추가적인 변동은 토양조건, 비료의 흡수, 계절적 기후변화 등에 따라 발생한다.
게다가 분자량이 약 50,000 내지 150,000 달톤(dalton)이며 1,4-결합 폴리글루칸(polyglucan)인 아밀로오스와, 분자량이 약 300,000 내지 2,000,000 달톤이며 가지가 많은 1,4- 및 1,6-결합 폴리글루칸인 아밀로펙틴은, 광범위한 분자량분포를 가지고 있다.
가지가 많은 모양에서 선형으로의 구조변이는 유동적이며 원래의 식물물질에서는 가지각색이므로, 명확하게 한계를 정하는 것은 거의 불가능하다. 특히 아밀로펙틴을 함유한 부형제는 그 가지들 때문에 비정상적인 팽창을 야기시키며, 이로 인하여 운반체의 안정성(carrier stability)은 악영향을 받는다. 따라서 효소에 의한 탈분지작용(enzymatic debranching)에 의하여, 힘들더라도 아밀로펙틴을 제거시키는 것이 일반적이다 [Journal of Controlled Release 45, (1997) 25-33 및 EP 0499 648 B1=US 5,468,286].
이러한 뚜렷한 불이익한 점 외에도, 광범위한 분자량 분포 또는 다른 공간배열을 가진 중합체의 혼합물인 천연중합체(native polymer)에는 저분자량 화합물(예:지방 및 기름)과 같은 성분이 포함되어 있는데, 이들을 분리하는 것은 쉬운 일이 아니며, 더 나아가 처리 및 가공하는데 있어서도 불리한 영향을 미친다(예:US 3,490,742) 특히 수율을 감소시키는 작업단계를 수행해야만 하는데, 어느 경우에는 불순물을 완전히 제거하는 것이 불가능하다.
식물을 유전적으로 변형시켜서 생중합체(즉 전분의 경우에도)를 최적화시키는 실험이 알려져 있다. WO 94/03049는 유전자변형 옥수수로부터 아밀로오스를 다량 함유한 전분을 제조하는 방법 및 그 용도에 대해서 기술하고 있다. 그럼에도 불구하고 불균일성(nonuniformity) 및 오염의 문제는 계속 존재한다.
재현성 및 품질은 균일성(uniformity)과 순도에 따라 크게 달라진다. 고품질의 산물을 생산하기 위해서는 이러한 개시물질(starting substance)에 대해 명확하게 정의를 내리고 그 특성을 설명할 수 있어야 한다.
본 발명은 선형 불수용성 다당류를 포함한 천천히 방출되는 정제, 그 제조 공정 및 그 사용방법에 관한 것으로서, 특히 활성 화합물의 방출을 조절하는 정제, 그 제조방법 및 그 사용방법에 관한 것이다.

본 발명은 전술한 문제점들을 회피하면서도, 활성화합물의 방출되는 정도를 조절할 수 있는 약학적 조성물을 갖는, 천천히 방출되는 정제(slow-release tablet)로서 이용 가능한, 천천히 방출되는 물질(slow-release material)을 얻고자하는 것이다. 여기서는 경구 투여(oral administration)가 바람직하다.
추가적인 첨가제 없이도 활성화합물의 방출되는 정도를 조절할 수 있는, 화학적으로 불활성이고 압력에도 안정적인 개시물질이며 또한 생체적합성을 가진(biocompatible) 선형 불수용성 다당류를, 천천히 방출되는 물질로 이용함으로써, 본 발명의 목적을 달성할 수 있다. 사용된 개시물질은 구형(spherical) 미세 입자 또는 이와 비슷한 형태를 띤 선형 불수용성 폴리(1,4-alpha-D-glucan)인 것이 바람직하다.
본 발명의 "천천히 방출되는 정제"에는 특히 정제, 당의정(coated tablet), 환제(pill), 작은 환제(pellet), 압착물(pressings), 작은 판(plate), 디스크 형태등이 포함된다. 이 때, 이들을 제조하는 데에는 압축과정이 필요하다. 마찬가지로 천천히 방출되는 물질이 함유된 캡슐도 이에 포함된다. 이하에서 천천히 방출되는 물질이라 함은, 선형 불수용성 다당류를 의미하는 것으로 간주될 것이다.
본 발명에서의 선형 불수용성 다당류는 다당류, 바람직스럽게는 폴리글루칸(polyglucan), 특히 단당류, 이당류, 이들의 올리고머(oligomer) 또는 그 유도체로 구성된 폴리(1,4-알파-D-글루칸)이 적당하다 할 것이다.
이것들은 항상 서로간에 동일한 방법으로 연결되어 있다. 이런 식으로 정의된 각 염기 단위(base unit)는 정확하게 2개의 연결고리(linkage)를 가지고 있는 데, 각 연결고리는 다른 단위체(monomer)에 연결되어 있다. 이러한 구조에서는 다당류의 첫 부분과 끝 부분을 형성하는 2개의 염기단위가 배제(excluded)된다. 이러한 염기단위는 다른 단위체에 대해서 단지 1개의 연결고리만 가지고 있다. 어느 단위체가 다른 그룹, 바람직하게는 다른 당류 단위(saccharide unit)와 3개 또는 4개의 연결고리(공유결합)로 연결된 경우에는 가지치기(branching)가 적용된다(is referred to). 각 당류단위로부터 최소한 3개의 글리코시드 결합(glycosidic bond)이 중합체 뼈대(polymer backbone)상에 존재하게 된다.
본 발명에 의하면, 가지치기는 발생하지 않거나 또는 아주 적은 비율로 존재하여 종래의 분석방법으로는 이를 분석할 수 없는 것이 일반적이다. 예를 들면, 선형 다당류의 분석에 필요하지 않은 100개의 수산기(hydroxyl groups)에 존재하는 모든 수산기를 기준으로 했을 때, 최대 5개의 수산기를 다른 당류 단위와의 결합에 의하여 분석할 수 있다.
각 당류단위에서, 유리(free) 수산기(또는 존재하는 다른 작용기)가 다른 당류와 추가적인 글리코시드(또는 다른) 결합을 한다면, 곁가지도(degree of branching)는 최대(100%)가 된다. 어느 당류에서, 중합체의 선형도(linearity)를 결정하는 수산기는 별론으로 하고, 화학반응에 의하여 수산기가 더 이상 변형되지 않는다면, 곁가지도는 최소(0%)가 된다.
선형 불수용성 다당류의 바람직한 예로서는, 선형 폴리-D-글루칸(linear poly-D-glucan)이 있는데, 여기서는 본 발명에서 의미하는 선형도가 존재하는 한 그 결합형태는 별로 중요하지 않다. 폴리(1,4-알파-D-글루칸), 폴리(1,3-베타-D-글루칸) 및 폴리(1,4-알파-D-글루칸)이 특히 바람직한 예이다.
만약 염기단위가 3개 이상의 연결고리를 가지고 있다면, 가지치기(branching)가 적용된다. 이른바 곁가지도는 선형 중합체 뼈대의 합성에 관계없으면서 가지치기를 형성하는, 100 염기단위당 수산기수로 정의된다.
본 발명에 의하면, 선형 불수용성 다당류는 곁가지도가 8%미만, 즉 100염기단위 당 8개 미만의 가지를 가지고 있다. 바람직하게는 곁가지도가 4% 미만이 적당하며, 특히 1.5%가 가장 적당하다.
선형 불수용성 다당류가 폴리글루칸(polyglucan), 예를 들면 폴리(1,4-알파-D-글루칸)이라면, 6번 위치의 곁가지도는 4%미만, 바람직하게는 최대 2%가 적당하며, 특히 최대 0.5%가 가장 적당하다. 선형결합에 관련되지 않는 다른 위치 [예:폴리(1,4-알파-D-글루칸)의 2번 또는 3번 위치] 상의 곁가지도는, 각각의 경우 최대 2%, 특히 최대 1%가 바람직하다.
특히 다당류, 구체적으로 폴리-알파-D-글루칸이 바람직한데, 이는 가지(branching)를 가지고 있지 아니하거나 또는 곁가지도가 너무 적어서 종래의 방법으로는 이를 탐지할 수 없다.
본 발명에서, 접두어 "alpha","beta" 또는 "D" 는 중합체의 뼈대를 형성하는 결합(linkage)에 관련된 것이지, 가지(branching)에 관련된 것이 아니다.
본 발명상의 "불수용성(water insolubility)"라 함은 통상의 조건, 즉 25℃ 의 상온과 101,325 Pa의 기압 또는 이 값과의 오차가 최대 20%인 조건 하에서 어떤 화합물의 용해성을 탐지할 수 없다는 것을 의미한다.
본 발명에서 사용된 다당류, 특히 폴리(1,4-알파-D-글루칸)과 같은 폴리글루칸의 경우에, 이는 사용량의 최소한 98%, 바람직하게는 99.5% 초과의 양이 물에 녹지 않는다는 것을 의미한다. 아래의 관찰을 통하여 불용해성(insolubility)이란 용어를 설명할 수도 있다. 선형 다당류 1g을 1bar 압력하의 탈이온수(deionized water) 1ℓ에서 130 ℃까지 가열한다면, 그 용액은 몇 분 동안 잠시 안정된 상태로 존재한다. 통상의 조건 하에서 냉각시킬 때, 물질은 재침전 된다. 더 냉각시킨 후 원심분리기를 사용하여 분리하면, 실험용 손실량을 포함하여 사용량의 최소한 66%정도를 이런 방법으로 회수할 수 있다.
본 발명의 경우에는, 일반적으로 정의된 생물공학적 또는 유전공학적 방법에 의하여 얻을 수 있는 선형-불수용성 다당류를 사용하는 것이 바람직하다. 여기서 기술된 특히 바람직한 실시예는 생물공학적 공정, 특히 생체촉매공정(biocatalytic process)으로 제조된 것이다.
본 발명에서 생체촉매반응 또는 생체변환반응(biotransformation)에 의해서 제조된 선형 다당류라 함은, 적당한 조건 하에서 이른바 생체촉매, 즉 효소를 이용하여 올리고머 당(oligomeric saccharides)(예:단당류 및/또는 이당류)와 같은 단위체 염기단위를 촉매반응 시켜서 선형 다당류를 제조하는 것을 말한다. 특히 다당류 합성효소(polysaccharide synthase) 및/또는 전분 합성효소(starch synthase) 및/또는 글리코실 전이효소(glycosyl transferase) 및/또는 알파-1,4-글루칸 전이효소 및/또는 글리코겐 합성효소(glycogen synthase) 및/또는 아밀로수크라제(amylosucrase) 및/또는 포스포릴라아제(phosphorylase)에 의하여, 폴리(1,4-알파-D-글루칸)을 제조하는 것이 바람직하다.
마찬가지로 발효(fermentation)로부터 선형 다당류를 제조하는 방법을 생각해 볼 수 있다. 이러한 발명에서의 선형 다당류는 곰팡이, 조류(algae) 또는 미생물과 같은 천연유기체를 사용하는 효소공정에 의하거나, 일반적인 유전공학적 방법을 이용하여 곰팡이, 조류 또는 미생물과 같은 천연유기체의 변형물을 이용하는 효소공정에 의하여 얻어지는 것들이다.
게다가 가지(branching)를 포함하는 비선형(nonlinear) 다당류로부터, 본 발명에서 기술된 천천히 방출되는 정제 조제용 선형 다당류를 제조할 수 있다. 즉 비선형 다당류를 효소로 처리한 후, 이들 선형 중합체를 예를 들면, 효소[예:아밀라제, 이소아밀라제(isoamylase), 글루코노히드로라제(gluconohydrolase), 풀루라나제(pullulanase)]등으로 분해시킨 후 가지를 제거하는 것이다.
본 발명에서 사용된 선형 다당류의 분자량 MW는 103g/mol 내지 107g/mol의 넓은 범위에 존재하는 데, 분자량 MW는 2 ×103g/mol 내지 5 ×104g/mol, 특히 3×103g/mol 내지 2 ×104g/mol인 것이 바람직하다. 사용된 선형 다당류인 폴리(1,4-알파-D-글루칸)의 경우, 이와 동일한 범위가 사용되었다. 분자량 분포 또는 다분산도(polydisperity) MW/Mn는 다당류 제조방법에 따라 넓은 범위 내에서 변화할 수 있다. 다분산도가 1.01 내지 50 정도, 특히 1.5 내지 15정도가 바람직하다. 이러한 경우 다분산도는 분자량이 바이모덜(bimodal)하게 분포됨에 따라 증가하는데, 이는 정제 조제의 특성에 악영향을 미치지 않는다.
본 발명에 의한 선형 다당류 및 미세입자(microparticles)의 형태를 띤 선형 다당류와 비선형 다당류와의 혼합물은 배제되지 아니하였다. "활성화합물의 방출을 조절함(controlled release of active compound)"이라는 것은, 환경에 대응되는 통계학적 편차 범위내에서 생물학적 유기체에 유용한 약물을 복용하는 특정시기 및/또는 기간 후에, 활성화합물이 방출되는 것을 말한다.
이러한 정의는 극단적인 경우도 포함하고 있다. 즉 조제물(formulation)에 존재하는 모든 활성화합물이 0에 가까운 시간 내에 즉시 방출되는 경우와, 조제물에 존재하는 모든 활성화합물을 방출하는데 필요한 오랜 시간, 적어도 특정 기간 동안에 치료효과를 얻기 위하여 필요한 최소한의 량/복용양도 포함한다.
그러므로 여기서의 천천히 방출되는 조제물의 경우, 방출을 늦추는 조제물 또는 저장부 조제물(depot formulation)에 대해서 유사하게 참조할 필요가 있다. "활성화합물(active compound)"이란, 넓은 의미에서의 (특히 인간과 가축분야) 생물학적 활성물질 및 물질의 화합물로서, 특히 약효를 나타내기 위한 것이라고 간주된다. 구체적으로 말하면 다음과 같다. 즉 진통제, 협심증 치료제(anginal preparation), 항알레르기제, 항히스타민제, 항염증제, 기관지확장제, 기관지경련치료제(bronchospasmolytics), 이뇨제, 항콜린제(anticholinergics), 항부착분자(antiadhesion molecules), 세포 조절제(cytokine modulator), 생물학적 활성 엔도누클리아제(endonuclease), 재조합 인간 디옥시리보뉴크레아제(recombinant human DNase), 신경전달물질, 류코트린(leukotriene) 억제재, 혈관활성 장 펩타이드(vasoactive intestinal peptide), 엔도테린(endothelin) 길항제, 중추신경자극제(analeptics), 국소마취제, 마취제, 간질방지제(antiepileptics), 항경련제(anticonvulsant), 항파키슨병제(antiparkinson agents), 항구토제(antiemetics), 호르몬계를 조절 또는 자극하는 화합물, 심혈관계(cardiovascular system)를 조절 또는 자극하는 화합물, 기도계(respiratory tract system)를 조절 또는 자극하는 화합물, 비타민, 미량원소, 항산화제, 세포성장억지제(cytostatics), 항대사물질(antimetabolites), 항감염제(antiinfectives), 면역조절물질, 면역억제재, 항생제, 단백질, 펩타이드, 호르몬, 생장호르몬, 생장요소(growth factors), 크산틴, 백신, 스테로이드 및 베타2모방제 등이다.
본 발명에서 말하는 "치료효과"라 함은, 치료에 효과적인 일정양의 활성화합물이 원하는 목표지점에 도달해서 활성을 나타내고 생리학적 반응을 일으키는 것을 말한다. 완화 및/또는 치료효과도 여기에 포함된다.
본 발명에서 말하는 "생체적합(biocompatible)" 이라 함은, 사용된 다당류가 완전히 생분해(biodegradation)되어서 체내에 농축물(concentration)이 생기지 않는 것을 의미한다. 생분해라 함은, 중합체를 분해 또는 파괴하는, 생체에서 일어나는 어떤 처리공정을 말한다. 특히 가수(hydrolytic) 또는 효소처리공정이 이러한 분야에 포함된다. 다당류와 이의 분해산물(즉, 대사산물)의 생체적합성의 경우, 사용된 다당류의 천연적으로 동일한 성질은 별로 중요하지 않다. 따라서 본 발명에 의하여 사용된 다당류는 치료, 진단 또는 예방용으로 적당하다. 본 발명에서 의미하는 "약제적으로 용인된(pharmaceutically acceptable)"이란 용어는, 활성화합물용 매개체(vehicle), 즉 보조제 또는 이른바 부형제(excipient)가 유기체에 심각한 부작용을 일으키지 않고 생물체에 흡수될 수 있는 것을 말한다.
활성화합물과 혼합 또는 균질화된 선형 다당류를, 공지된 방법(예:ball mill)에 의하여 개시물질과 혼합함으로써 정제를 조제할 수 있다. 활성화합물은 약 50%의 농도를 가질 수 있는데, 1% 내지 20%, 더 나아가 5% 내지 15% 사이의 농도가 바람직하다. 추가적으로 보조제(auxiliary) 및 첨가제(additive)를 사용할 수 있다. 본 발명에서 전체 조성물(보조제 및 첨가제가 있으면 이들을 포함해서)에 대한 활성화합물과 다당류의 합은 최소한 50% 는 되어야 하나, 70% 내지 100%가 바람직하며, 특히 85% 내지 98%가 가장 바람직하다. 보조제의 조성은 넓은 범위 내에서 변화할 수 있는데, 조성물의 비는 활성화합물과 선형 불수용성 다당류간의 상호작용에 따라 변화한다.
정제 생산과 선삽입(preinserted) 혼합공정에서 사용된 보조제는 용매, 특히 쉽게 증발되는 용매가 바람직하다.
정제생산을 위한 본 발명에서의 다당류의 모 구조(parent structure)는 비정형(amorphous) 또는 결정구조 또는 세립체(grain)인데, 이들은 합성 또는 미세분자에서 직접 얻을 수 있다. 이에 대해서는 특허출원(German Patent Office, ref.:197 37 481.6)에 기술되어 있다. 정제의 원료 또는 원혼합물을 제조하기 위해서는 간단한 혼합공정을 사용하는 것이 바람직하다. 이러한 정제의 제조공정은 정제의 성질에 영향을 끼친다. 예를 들면, 스프레이 기술(spraying technique), 즉 유동성 베드공정(fluidized bed process) 또는 본 발명에서 사용되는 다당류 현탁액의 코팅등을 이용하여, 활성화합물을 다당류의 모 구조에 결합시키는 것이 가능하다. 여기서는 흡수 공정(absorption process) 또는 스프레이-건조 기술(spray-drying technique)을 이용할 수 있는데, 흡수공정에서는 용액내의 활성화합물을 흡수하기 위하여(즉 스폰지 특성) 미세분자의 다공질(porous) 구조를 이용한다. 여기서 선형 다당류와 활성화합물의 용액, 현탁액 또는 에멀션(emulsion)은 공지된 스프레이 기술을 이용하여 건조된다. 용액의 경우, 이에 상응하는 유기용매를 이용한다, 고온 또는 고압 및 과임계공정(supercritical process)을 사용하면 짧은 시간동안 필요한 용해도를 얻을 수 있다.
정제 제조에 사용되는 압력은 넓은 범위 내에서 변화할 수 있다. 천천히 방출되는 효과(slow-release effect)를 추가적으로 얻기 위하여, 본 발명에서 기술된 다당류에 대해서 압력을 변동(variations)시킨다. 압력은 1MPa 내지 103MPa 범위 내에서 변동될 수 있다 (105Pa = 1bar). 10MPa 내지 300MPa를 사용하는 것이 바람직하며, 특히 100MPa 내지 250MPa 범위의 압력을 사용하는 것이 가장 바람직하다.
다음의 실시예들 및 도는 본 발명을 더욱 자세하게 설명하기 위한 것이며, 본 발명은 아래 실시예들에서 기술된 생산물과 구체적인 예에 한정된 것은 아니다.
다음 예들은 미세입자의 제조에 특히 관련된 것인데, 이는 명확하게 참조할 수 있는 특허출원(German Patent Office, ref.: 197 37 481.6)에 기술되어 있다. 게다가 특히 유용한 폴리(1,4- 알파-D-글루칸)제조방법은 WO 95/31553에 기술되어있다.
실시예 1
효소인 아밀로수크라제(amylosucrase)를 이용한 생체촉매공정에서 폴리(1,4-알파-D-글루칸)의 생체외(in vitro)생산.
20% 강화 자당(strength sucrose)용액 10ℓ를 살균(스팀 살균)된 15ℓ용기에 가한다. 발효에 의하여 얻어진 아밀로수크라제를 포함한 효소 추출물을 일정량 자당 용액에 가한다. 효소 활성도는 16단위(unit)이다. 여기서 1 단위라 함은, 1분간 효소 1㎎에 대한 자당 1μ㏖의 반응을 말한다. 실험기기는 살균화된 KGP 교반기(stirrer)를 구비하고 있다. 용기는 밀봉되어 40℃에 보관된 후 교반된다. 일정시간 경과 후 하얀 침전물이 형성된다. 180시간이 지나면 반응은 종료된다. 침전물은 여과된 후 저분자량 당(sugar)을 제거하기 위하여 여러 번 세척한다. 필터에 남아있는 잔류물을 진공상태의 건조 오븐(drying oven)내에서 30℃ 내지 40℃ 사이에서 건조시켰는 데, 여기서 막 펌프(membrane pump)(Vacuubrand GmbH & Co, CVC 2)를 이용하여 진공상태를 만들었다. 질량은 685g이다(수율 69%).
실시예 2
겔 투과 크로마토그래피(gel permeation chromatography)에 의하여 실시예1의 아밀로수크라제를 이용하여 합성된 폴리(1,4-알파-D-글루칸)의 특성.
실시예 1에서 구해진 폴리(1,4-알파-D-글루칸) 2㎎을 상온에서 디메틸 술폭사이드(DMSO, p.a. from Riedel-de-Haen)로 용해시킨 후 여과시켰다(2mm 필터). 용액의 일부분을 겔 투과 크로마토그래피 칼럼에 주입한다. DMSO를 용리제(eluent)로 사용한다. 신호의 강도는 RI 탐지기(RI detector)에 의하여 측정하며, 풀루란(pullulan) 표준(중합체 표준 시스템)으로 측정하였다. 유속은 분당 1.0㎖이다. 측정결과 분자량의 수평균(numerical mean, Mn)은 2700g/mol이고, 분자량의 중량평균(weight mean, MW)은 11,700g/mol이었다. 즉 분산도(disperity)는 4.3이었다.
실시예 3
폴리(1,4-알파-D-글루칸) 미세입자의 조제.
폴리(1,4-알파-D-글루칸) 400g을 60℃의 디메틸 술폭사이드(DMSO, p.a. from Riedel-de-Haen) 2ℓ에서 1.5 시간동안 용해시켰다. 그 후 상온에서 1시간동안 용액을 휘저었다. 적하 깔대기(dropping funnel)를 통하여 2시간동안 상기 용액을 재증류수(double-distilled water)에 가해주면서 동시에 휘저었다. 혼합물은 6 ℃에서 40시간동안 저장되었다. 미세한 입자를 가진 현탁액(fine suspension)이 형성되었다. 우선, 상층액(supernatant)을 천천히 기울여 따라서 (decantation) 입자들을 분리하였다. 침강물(sediment)을 슬러리(slurry)한 후 소량을 원심분리 시킨다(초 원심분리기 RC5C: 5000r.p.m의 속도로 각 5분간). 고체 잔류물을 재증류수로 슬러리한 후 총 3번 다시 원심분리 시킨다. 고체는 따로 모으고, 현탁액 약 1000㎖ 정도를 냉동-건조시킨다 (Christ Delta 1-24 KD). 흰색의 고체 283g이 분리된다(실시예 3a: 수율 71%). 모아진 상층액은 18℃에서 밤새도록 저장한다. 전술한 바와 같은 작업(working up)을 실행한다. 추가적으로 흰색의 고체 55g이 분리된다(실시예 3b: 수율 14%). 총 수율은 85%이다.
실시예 4
실시예 3에서 만들어진 미세입자의 탈황반응(desulfurization).
입자에 남아있는 디메틸 술폭사이드(DMSO)를 제거하기 위하여, 다음과 같이 처리한다. 즉 실시예 9에서 구해진 아밀로오스 입자 100g을 탈이온수 1000㎖에 가한다. 이 혼합물을 천천히 저으면서 24시간 동안 그대로 방치한다. 입자들은 실시예 9에서 기술한 방법에 의하여 제거된다[초원심분리기 RC5C: 5000r.p.m의 속도로 각 5분간. 냉동건조 후, 최종 중량 98.3g 구함(수율 98%)]. 원소분석에 의하여 황의 양을 측정하면 다음과 같다(시험 법 연소와 IR탐지):

실시예 3에서 구해진 입자의 황 함량 : < 0.01%
실시예 5
실시예 3에서 구해진 미세입자를 전자현미경으로 조사함.
입자의 특성을 알아보기 위해서, 전자 현미경 사진 스캐닝(scanning electron micrographs, SEMs)을 시행하였다. 도 1 및 도 2는 구형이고, 모양과 크기 및 표면 거침도(surface roughness)가 매우 균일한 입자의 사진을 보여주고 있다.
실시예 6
실시예 3에서 구해진 입자의 크기 분포(size-distributions)를 조사함.
실시예 1 및 실시예 9에서 구해진 입자의 크기 분포의 특성을 알아보기 위하여, Mastersizer를 이용한 조사가 행해졌다(Malven Instruments). 용적농도(volume concentration)가 0.012% 내지 0.014%이고, 밀도가 1.080g/㎤인 Fraunhofer 모드 (평가: multimodal, 숫자)에서 조사가 행해졌다.

실시예 7
폴리(1,4-알파-D-글루칸)을 포함한 미세입자로부터 정제를 제조하는 일반적인 공정.
정제 보조제[tablet auxiliary, 폴리(1,4-알파-D-글루칸)] 270mg과 활성화합물 30mg을, 세포파멸기[ball mill(Retsch MM2000)]에서 10분 동안 진폭 100으로 빻는다 (생산자의 정보). 균질화된 물질 250mg을 얻은 후 압축기(pressing tool)(Perkin Elmer, 피스톤 지름:13mm)로 옮긴다. 압축기를 압착기(press)[ Perkin Elmer, 수압기] 밑에 위치시킨다. 그 후 2톤의 압력으로 10분간 상기 물질을 압착한다. 기구의 압력을 완화시킨 후 완성된 정제를 조심스럽게 떼어낸 후 특성(예: 안정도 측정 또는 방출실험)을 더 알아보기 위하여 저장한다.
다음에서는 비교를 위해서, 미세결정질 셀룰로스[Avicel(등록상표)], 감자 전분[Toffena(등록상표)-Sudstarke)] 및 폴리아크릴산[Eudragits(등록상표)- Rohm]과 같은 공지된 정제 조제물질(즉 비교예)로부터, 정제를 제조한 것이다.
실시예 8
시간변화에 따른 활성화합물 방출의 측정
실시예 7에 따라 조제된 정제의 방출정도를 다음과 같이 측정하였다. 즉 50㎖ 짜리 Erlenmeyer 프라스크 내에 있는 물(탈이온수) 25㎖에 정제를 가한다. 플라스크의 입구는 파라필름(Parafilm)으로 밀봉한다. 플라스크를 교반기(shaker)(IKA Labortechnik; KS 125 basic)에 고정시킨다. 교반기는 분당 약 150 정도로 동작한다.
일정 시간 경과 후 샘플-약 1.5㎖-를 상기 용액의 상층액으로부터 제거한다. 상기 용량의 충분한 양을 1회용 큐벳(cuvette)[Sarstedt No. 67,741]으로 옮긴 후 분광계(spectrometer)[Kontron Instruments, Uvikon 860]로 측정한다. 각 활성화합물 또는 모델 물질에서 발생하는 존재하는 최대 흡수량(absorption maxima)이 사용(apply)된다.
실시예 9
추가적인 활성화합물의 최대 흡수량.
추가적인 활성화합물의 최대 흡수량은 실시예 10과 같은 방법으로 측정되었다. 언급된 모든 활성화합물은 천천히 방출되는 효과를 관측할 수 있는 비교 결과를 보여줬는데, 그 결과 다양한 응용이 가능하다는 결론을 내릴 수 있다.

비타민 B12100㎎을 10㎖ 짜리 눈금 플라스크에 가하고, 보정표지(calibration mark)까지 탈이온수를 채워서, 1% 농도의 원액(stock solution)을 제조한다. 연속적인 희석화 과정을 통하여, 상기 원액으로부터 농도가 0.005%, 0.01% 및 0.02%인 용액을 제조하고, 분광계(Kontron Instruments, Uvikon 860)로 측정한다. 최대 흡수점이 λ= 549 ㎚에서 흡광(extinctions)된다. 직선으로부터의 편차가 뚜렷한 경우에 한하여 측정점(measurement points)이 추가로 필요하다. 이러한 보정직선은, 활성화합물 방출실험에서의 상층액의 농도를 측정하는데 있어서 개시점 (starting point)으로 작용한다. 다른 활성화합물과 모델 물질의 보정직선을 작성할 때에도, 이와 유사한 방법으로 데이터를 수집한다.

실시예 11
폴리(1,4-알파-D-글루칸)과 이의 미세입자를 사용하여 제조된 정제로부터의 비타민 B12의 방출실험.
실시예 10에서 기술된 바와 같이, 특정시간 경과 후에 생체외(in-vitro) 방출실험에서의 상층액에 대한 흡광을 측정한다. 측정된 흡광에 의하여 보정곡선으로부터 이에 상응한 농도값을 얻기 위해서는, 상층액을 1:10의 비율로 희석시키는 것이 필요할 수도 있다. 그러므로 상기 요소를 고려해야한다. 표 3에서는, 값들이 요약된 형태로 표시되어 있다. 이에 상응하는 도 4에서는, 상기 값들이 그래프 내에서 비교된다. 실시예 8에 의하면 최대 가능농도는 0.1%로서, 명확한 설명을 위해서 100% 값을 가지며, 따라서 그 결과 어느 시간 경과 후에 어느 정도의 반응이 완결되었는지를 바로 측정할 수 있다.

실시예 12
비교예인 미세결정질 셀룰로스[Avicel(등록상표)] 및 감자 전분[Toffena(등록상표)]를 이용하여 생산된 정제로부터의, 비타민 B12방출에 관한 실험
표 4에 기재된 결과는 실시예 11에서 기술된 방법에 의하여 측정되고 계산되었다. 도 8은, 비교물질인 미세결정질 셀룰로스[Avicel(등록상표)] 및 감자 전분[Toffena(등록상표)]에 대하여, 본 발명에 따라 기술된 정제 보조제인 폴리(1,4-알파-D-글루칸) 및 폴리(1,4-알파-D-글루칸)의 미세입자의 결과를 비교형태로 보여주고 있다. 여기서 천천히 방출되는 효과를 뚜렷하게 식별할 수 있다.

실시예 13
폴리(1,4-알파-D-글루칸) 및 폴리(1,4-알파-D-글루칸)의 미세입자로 구성된 테오필린(theophylline)정제로부터의 방출에 관한 실험.
여러 가지 정제 보조제로 구성된 정제로부터의 테오필린 방출에 관한 실험이, 실시예 12와 유사한 방법으로 수행되었다. 사용된 정제 보조제는 폴리(1,4-알파-D-글루칸) 및 폴리(1,4-알파-D-글루칸)의 미세입자이다. 여기서 폴리(1,4-알파-D-글루칸)은 적절한 작업(working-up) 공정(실시예 1 참조) 후에 생체촉매반응에 의하여 직접 얻을 수 있다. 그 결과는 도 6에 나타나 있다. 각각에 대하여 중복 측정을 하였는데, 이는 실험결과의 높은 재현성(reproducibility)을 입증하고 있다.
실시예 14
비교예인 c)미세결정질 셀룰로스[Avicel(등록상표)], d)Eudragit RS(등록상표) 및 e)Eudragit RL(등록상표)로 구성된 테오필린 정제로부터의 방출에 관한 실험.
비교 물질인 정제 보조제에 대한, 정제로부터의 테오필린 방출에 관한 실험이 실시예 13과 유사한 방법으로 수행되었다. 사용된 정제 보조제는 다음과 같다: 미세결정질 셀룰로스[Avicel(등록상표)], Eudragit RS(등록상표) 및 Eudragit RL(등록상표)
그 결과는 도 8에 나타나 있다. 각각에 대하여 중복 측정이 행해졌다. 도 6에서는, 실시예 13으로부터의 방출범위(release profile)가 비교할 수 있게 병렬로 놓여져 있다.

실시예 15
인공 위액(artificial gastric juice)내에서, 비교예인 폴리(1,4-알파-D-글루칸), 폴리(1,4-알파-D-글루칸)의 미세입자 및 미세결정질 셀룰로스[Avicel(등록상표)]로 구성된 테오필린 정제로부터의 방출에 관한 실험.
인공 위액 내에서, 정제 보조제인 폴리(1,4-알파-D-글루칸)(미세입자 포함) 및 미세결정질 셀룰로스[Avicel(등록상표)]를 함유한 정제로부터의 테오필린 방출실험이, 실시예 8과 유사한 방법으로 행해졌다. [여기서 인공 위액은 NaCl 2g, 펩신 3.2g, 농축(concentrated) 염산(HClaq) 7㎖에 탈이온수를 가하여 전체 부피가 1ℓ가 되게한 것이다]. 자연적인 환경상태를 반영하는 매질(medium)을 사용한 경우라도, 방출물질이 천천히 방출되는 효과를 다시 관측할 수 있다.
도 7은 여러 가지 정제 보조제{a)폴리(1,4-알파-D-글루칸), b)폴리(1,4-알파-D-글루칸)의 미세입자 및 c)미세결정질 셀룰로스[Avicel(등록상표)](비교예)}로부터 테오필린이 방출되는 것을 보여주고 있다
실시예 16
폴리(1,4-알파-D-글루칸) 및 이것의 미세입자로 구성된 Ramorelix(등록상표) 정제로부터의 방출에 관한 실험.
Ramorelix(등록상표) 정제로부터의 방출에 관한 실험은 전술한 실시예들에 따라 행하여졌다. Ramorelix(등록상표)는 다음과 같은 아미노산 배열(구조)를 갖는 LHRH 길항제(antagonist)이다: 1-[N-아세틸-3-(2-나프틸)-D-알라닐-p-클로로-D-페닐알라닐-D-트립토필-L-세릴-L-티로실-O-(6-디옥시-알파-L-마노피라노실)-D-세릴-L-루실-L-아르기닐-L-프로릴] 세미카아바지드 아세트산.
그러나 방출 매질로서 탈이온수 대신에 인공 위액을 사용하였다. 인공 위액의 처방은 다음과 같다: 염하나트륨(NaCl) 2g, 펩신 3.2g, 농축(concentrated) 염산(HClaq) 7㎖에 탈이온수를 가하여 전체 부피가 1ℓ가 되게 하였다. 용액의 pH는 1.2이다.
실시예 17
비교예로서 미세결정질 셀룰로스[Avicel(등록상표)]로 구성된 Ramorelix(등록상표) 정제의 방출에 대한 실험.
실시예 8과 같은 방법으로 방출실험을 행하였다. 사용된 방출위액은 인공 위액이다. 도 8은 여러 가지 정제 보조제{a)폴리(1,4-알파-D-글루칸), b)폴리(1,4-알파-D-글루칸)의 미세입자 및 c)미세결정질 셀룰로스 [Avicel(등록상표)](비교예)}로부터 Ramorelix(등록상표)가 방출되는 것을 보여주고 있다
실시예 18
다당류의 용해도 측정.
폴리(1,4-알파-D-글루칸) 100㎎을 재증류수 5㎖에 가하였다. 반응용기를 자석 젓개(magnetic stirrer)로 휘저으면서 천천히 가열하였다. 20℃의 간격을 두면서 단계별로 가열한 후 눈으로 관찰하였다. 40℃, 60℃, 80℃ 및 100℃에서 어떤 변화도 관측되지 않았다. 이러한 관측결과에 따라, 화합물은 "불수용성"의 특성을 갖는다고 말할 수 있다.
실시예 19
다당류의 용해도 측정과 독일 약전(German Pharmacopeia, GP)에 따른 분류.
폴리(1,4-알파-D-글루칸) 564㎎을, 1.3bar이고 130℃인 오토클레이브(autoclave)내의 재증류수 약 0.5ℓ에서 1.5시간 동안 가열한다(Certoclave apparatus). 반응용기의 무게는 미리 측정하였다. 오토클레이브의 압력을 감소시킨 후 상온까지 냉각시킨다. 내용물의 무게를 재보니 501.74g이다. 24시간이 더 지난 후 혼합물을 원심분리 시키고, 상층액을 기울여 따른다. 고체 잔류물을 건조시킨 후 무게를 재어보니 468㎎이다. 이로부터 용해된 양이 96㎎인 것으로 계산되었다. 사용된 용매를 기준으로 할 때, 폴리(1,4-알파-D-글루칸) 1㎎ 당 물 5226㎎이 필요한 것으로 계산된다. 독일 약전의 분류에 따르면, 상기 1 물질부(1 part of the substance)를 용액에 용해시키려면 1000 내지 10000 용매부가 필요하므로, 상기 물질은 "매우 용해되지 않은(very poorly soluble)" 것으로 분류된다. 용해도 분류의 7단계["매우 잘 녹는"(단계1)에서 "사실상 녹지 않는"(단계7)까지] 중에서, 상기 물질은 단계 6에 속한다.
실시예 20
다당류의 용해도 측정과 독일 약전(GP)에 따른 분류.
본 실험은 실시예 19와 같은 방법으로 수행된다. 유일한 차이점은 오토클레이브 처리 후 삽입되여, 상온까지 냉각시키는 냉각 처리공정(cooling process)이다. 혼합물은 5℃에서 3시간동안 저장된다.
폴리(1,4-알파-D-글루칸) 526㎎을 측정하여 재증류수 약 480 ㎖에 첨가하였다. 열처리한 결과 최종적으로 468.09g이 얻어진다. 건조된 침강물의 무게는 488㎎ 이다. 따라서 폴리(1,4-알파-D-글루칸) 38㎎이 용해되었다. 이는 12,318 용매부에 대한 물질 1mg의 비에 해당된다. 따라서 상기 물질은 이러한 처리법에 의하면 독일 약전상의 단계 7에 해당되어, 사실상 녹지 않는 것이라고 분류할 수 있다. 왜냐하면, 1 물질부에 대해 10,000 용매부 이상이 필요하기 때문이다.
본 발명은 바람직한 실시예와 함께 기술되고 있다. 다수의 대안적인 변경, 변형 및 사용은 앞서 진술한 설명의 관점에서 당업자들에게 자명할 것이다.
Claims (12)
- 전체적으로 또는 부분적으로 천천히 방출되는 물질로서 적어도 하나의 불수용성 선형 다당류를 포함하고 있는, 천천히 방출되는 정제(slow-release tablet).
- 제 1항에 있어서, 전체적으로 또는 부분적으로 적어도 하나의 불수용성 선형 다당류를 포함하고 있고, 생명공학적 공정(biotechnological process)으로 조제되는, 천천히 방출되는 정제.
- 제 1항에 있어서, 전체적으로 또는 부분적으로 적어도 하나의 불수용성 선형 다당류를 포함하고 있고, 생체촉매 공정(biocatalytic process)으로 조제되는, 천천히 방출되는 정제.
- 제 1항에 있어서, 전체적으로 또는 부분적으로 적어도 하나의 불수용성 선형 다당류를 포함하고 있고, 발효공정(fermentation process)으로 조제되는, 천천히 방출되는 정제.
- 선형 불수용성 다당류로부터 얻을 수 있는 천천히 방출되는 정제
- 제 1항 내지 제5항 중 한 항에 있어서, 전체적 또는 부분적으로 적어도 하나의 불수용성 선형 폴리(1,4-알파-D-글루칸)을 포함하고 있는, 천천히 방출되는 정제.
- 제 1항 내지 제6항 중 하나 이상의 항에 있어서, 전체적 또는 부분적으로 미세입자(microparticles)로서 존재하는 불수용성 선형 다당류를 포함하고 있는, 천천히 방출되는 정제.
- 제 1항 내지 제 7항 중 하나 이상의 항에 있어서, 생물기능에 적합하고 및/또는 약제용으로 적합한, 천천히 방출되는 정제.
- 활성 화합물의 방출을 조절하기 위한 제 1항 내지 제 8항 중 하나 이상의 항에 있어서의 천천히 방출되는 정제의 용도.
- 제 1항 내지 제 9항 중 하나 이상의 항에 있어서, 하나 이상의 활성 화합물과 적당한 첨가제 및 보조제를 추가적으로 포함한, 천천히 방출되는 정제.
- 제 1항 내지 제 10항 중 하나 이상의 항에 있어서, 약제적 조성물로서 치료효과를 얻는 천천히 방출되는 정제.
- 조성물로서, 바람직하게는 경구투여 형태로서 제 1항 내지 제 11항 중 한 항에 있어서의 천천히 방출되는 정제의 용도.
