Abstract
살리실옥소 α-카르복시산을 함유하는 화장품적 피부조절 조성물. 본 발명의 조성물은 피지세포로부터의 피지 분지 조절, 향상된 지성분의 조절 및 향상된 피부 촉감을 제공하고, 번들거림 및 끈적임을 예방하는 동시에, 항노화 효과도 제공하여 주름살 및 노화된 피부의 외관을 감소시키는 결과도 제공하며, 피부색을 개선시키고, 광노화된 피부를 치료하며, 피부 광채를 개선시키고, 피부 투명성 및 피부 정돈을 개선시키고, 전체적으로 건강하고 젊은 피부 외양을 제공한다.
Description
피부 외관을 향상시키는 화장품이 소비자들에게 점점더 인기가 있다. 소비자들은 자주 잔주름 및 주름살, 건조하고 처진 피부와 같은 노화 또는 광노화된 피부의 징후를 경감 또는 지연시키려고 애쓴다. 소비자들은 자주 항 노화 이외에 다른 효과도 또한 얻어려고 한다. 빈번하고 바람직하지 않은 피부 상태는 피부상에 과도한 양의 피지 때문에 생겨나는 상태인 "지성 피부"이다. 피지는 피지세포(피부에서의 피지선의 세포)에 의해 생성되어 피부 표면으로 분비되는 피부 유지이다. 지성피부는 번들거림, 바람직하지 않은 외양 및 불쾌한 촉감을 가져온다. 지성피부는 다양한 연령군에 영향을 미친다. 제조자 및 소비자 양쪽의 관점에서, 항노화 효과 및 피지 조절 효과를 모두 제공할 수 있는 화장품 활성제가 매우 요망되고 있다.
그레더 등(Grether et al.)의 미국특허 제2,116,347호에는 류마티즘의 외용 치료용으로서 살리실옥시 카르복시산 에스테르를 개시하고 있다. 이 그레더 화합물은 이 '347 화합물이 말단 카르복시기 대신에 에스테르기를 함유하고 있다는 점에서 적어도 본원에서 이용된 살리실옥시 α-카르복시산과는 차이가 있다. 따라서, 본원에 포함된 화합물은 오직 하나의 에스테르 결합을 가지는 반면, 이 그레더 화합물은 두 개의 에스테르 결합을 포함한다.
PCT 출원 WO 93/10755호는 살리실산을 주름살 방지제로서 효과가 있는 것으로 보고한다. 레베크 등(Leveque et al.)의 미국 특허 제5,262,407호는 피부 노화 에 대항하는 치료로서의 고리 아실화 살리실산의 이용을 보고한다. 고리 아실화 살리실산은 일본 특허 제4036238호(다까사고 퍼퓨머리(Takasago Perfumery) KK)에서 여드름(acne vulgaris) 치료제로서 보고된 적이 있다. 반 스콧트(Van Scott) 및 유(Yu)는 일련의 특허들에서(예를 들어 미국 특허 5,091,171호 참조) α-히드록시산의 잔주름 및 주름살의 제거를 포함하는 항노화 효과를 기술하였다. 유럽 특허 출원 제0676194호(루셀 우클라프(Roussel Uclaf)는 피부노화 치료를 위한 화장품 조성물로서의 α-히드록시산(예를 들면 젖산) 또는 이들의 에스테르 및 살리실산 또는 이들의 에스테르를 포함하는 조성물을 개시한다. EP0676194호에 개시된 살리실산 에스테르는 비-고리 에스테르화 알킬 에스테르, 즉 살리실산이 에스테르 결합을 통하여 알킬 사슬에 공유결합된 것이다.
상기한 문헌 어느 것도 본원에 포함된 살리실옥시 α-카르복시산을 개시하지 않는다. 살리실옥시 α-카르복시산은 살리실산이 산 관능기를 갖는 기에 에스테르 결합을 통하여 공유결합된 것이다. α-히드록시 카르복실산(예를 들면 젖산) 또는 살리실산이 개별적으로 사용될 때는 피지 분비를 현저하게 감소시키지 않으나, 본원발명에 사용된 살리실옥시 α-카르복시산은 피지 분비의 현저한 감소를 달성한다는 것이 본원발명의 일부로서 발견되었다. 또한, 살리실옥시 α-카르복시산의 사용은 살리실산 및 α-히드록시 카르복실산의 물리적 혼합물의 사용과 비교시 훨씬 유리한 점이 많다. 살리실옥시 α-카르복시산은 제제화하기 더 쉬운데, 이는 α-히드록시 카르복실산 및 살리실산이 개별적으로 살리실옥시 α-카르복시산 한 분자 보다 더 많은 수의 비양립성(incompatibility)을 가지기 때문이다.

물 이외에 또는 물에 더하여, 비히클들은, 액상 또는 고상 연화제, 용매, 습윤제, 농후제 및 분말 등을 포함한다. 특히 선호되는 비수성 담체는 폴리디메틸 실록산 및(또는) 폴리디메틸 페닐 실록산이다. 본원 발명의 실리콘은 25℃에서 약 10 내지 10,000,000 mm2/s(센티스토크)의 범위에 속하는 점도를 갖는 것들일 수 있다. 특히 바람직하게는, 낮은 점도 실리콘과 높은 점도 실리콘의 혼합물이다. 이들 실리콘은 제너럴 일렉트릭사로부터 비카실(Vicasil), SE 및 SF라는 상표명으로, 다우코닝사로부터 200 및 550 시리즈라는 상표명으로 구입할 수 있다. 본원 발명의 조성물에 사용될 수 있는 실리콘의 양은 조성물의 5% 내지 95 중량%의 범위이고, 바람직하게는 25% 내지 90 중량%이다.
화장품적으로 허용되는 비히클은 일반적으로 조성물의 5 내지 99.9 중량%, 바람직하게는 25 내지 80 중량%를 형성할 것이고, 다른 화장품 첨가제가 존재하지 않는 경우에는 본 조성물의 부족분을 채울것이다. 바람직하게는, 비히클 중량의 80 중량% 이상이 물이다. 바람직하게는 물의 양이 본원 조성물의 50 중량% 이상이고, 가장 바람직하게는 조성물 중량의 60 내지 80 중량%이다. 바람직한 조성물은 60% 이상, 바람직하게는 80% 이상의 물을 함유하는 수중유(oil-in water) 에멀젼이다.
임의적 피부 보호 물질 및 화장품적 첨가제
살리실옥시 α-카르복시산이 비록 선스크린 기능을 가지지만(이것이 살리실레이트 유도체이기 때문), 본원 조성물은 유해한 자외선광에의 피부 노출을 더욱 감소시키기 위하여 바람직하게는 부가적인 선스크린제를 포함한다.
선스크린제는 자외선광을 차단하기 위하여 통상적으로 사용되는 물질들 등이 있다. 예들 들면, PABA의 유도체, 신나메이트(cinnamate) 및 살리실레이트의 유도체(살리실옥시 α-카르복시산 제외)이다. 예를 들어, 옥틸 메톡시신나메이트 및 2-히드록시-4-메톡시 벤조페논(옥시벤존으로도 알려짐)이 사용될 수 있다. 옥틸 메톡시신나메이트 및 2-히드록시-4-메톡시 벤조페논은 각각 파르솔(Parsol) MCX 및 벤조페논-3라는 상표명으로 시판된다. 에멀젼 중에 사용되는 선스크린제의 정확한 양은 원하는 태양의 자외선 조사로부터의 보호 정도에 따라 달라질 수 있다.
니아신아미드, 코지산, 아르부틴, 비타민 C와 같은 피부를 밝게 하는 부가적인 활성제가 0.0001 내지 20% 양으로 또한 포함될 수 있다.
연화제와 함께 오일 또는 오일성 물질이 존재하여, 사용된 연화제의 평균 친수성-친지성 균형(HLB)에 주로 의존적인, 수중유 에멀젼 또는 유중수 에멀젼을 제공할 수도 있다. 이러한 연화제의 양은 전체 조성물의 약 0.5 내지 약 50중량%, 바람직하게는 5 내지 30 중량%이다. 연화제는 에스테르, 지방산 및 알코올, 폴리올, 및 탄화수소와 같은 일반적인 화학적 카테고리로 분류될 수 있다.

적합한 지방 알코올 및 지방산은 10 내지 20개의 탄소 원자를 갖는 화합물들이다. 특히 바람직하게는 세틸, 미리스틸, 팔미틱 및 스테아릴 알코올 및 산들과 같은 화합물들이다.
연화제로서 기능할 수 있는 폴리올 중에는 선형 및 분지쇄 알킬 폴리히드록시 화합물이 있다. 예를 들어, 프로필렌 글리콜, 소르비톨, 글리세린이 바람직하다. 또한, 폴리-프로필렌 글리콜 및 폴리에틸렌 글리콜과 같은 중합체 폴리올이 유용할 수 있다. 부틸렌 및 프로필렌 글리콜은 또한 투과 증진제로서 특히 선호된다.
연화제로서 사용될 수 있는 탄화수소 예로는, 12 내지 30개의 탄소원자를 갖는 탄화수소 사슬을 가지는 것들이다. 특정 예로는 광유, 석유젤리, 스쿠알렌 및 이소파라핀 등이 있다.
본원 발명의 화장품 조성물내의 또다른 범주의 기능적 성분으로는 농후제가 있다. 농후제는 일반적으로 조성물의 0.1 내지 20 중량%, 바람직하게는 약 0.5 내지 10 중량%의 양으로 존재할 것이다. 농후제의 예로는 B.F. 굿리치(Goodrich)사의 카르보폴(Carbopol)의 상표명으로 입수가능한 가교결합된 폴리아크릴레이트 물질이 있다. 크산탄, 카라게닌, 젤라틴, 카라야, 펙틴 및 로커스트콩검과 같은 검들이 사용될 수도 있다. 특정 상황하에서는 농후 기능이 실리콘 또는 연화제로서 기능하는 물질에 의해 수행될 수도 있다. 예를 들어, 10센티스토크 초과의 실리콘 검 및 글리세롤 스테아레이트와 같은 에스테르는 이중의 기능성을 갖는다.

제품 사용, 제품 형태 및 제품 포장
사용시, 적합한 용기 또는 도포기로부터 소량의 조성물, 예를 들어 1 내지 100 ml을 피부의 노출부분에 바르고, 필요하면, 손이나 손가락 또는 적합한 기구를 사용하여 피부에 펼치고(또는) 문지른다.
본 발명의 화장품적 피부 조절 조성물은 로션, 크림 또는 겔로서 제제화될 수 있다. 본 조성물은 그 점도에 적합하고 소비자가 의도하는 사용에 적합한 용기에 포장될 수 있다. 예를 들어, 로션 또는 크림은 병 또는 롤-볼 도포기(roll-ball applicator), 또는 고압가스-추진 에어로졸 기구(propellant-driven aerosol device) 또는 손가락 작동에 적합한 펌프를 갖춘 용기에 포장될 수 있다. 본 조성물이 크림일 때는, 간단히 튜브나 뚜껑 있는 단지와 같은 비변형 용기 또는 눌러 짜내는 용기에 저장될 수 있다. 본 발명의 조성물을 또한 인용문헌으로서 본 명세서에 포함되는 미국 특허 제5,063,507호에 기술된 바와 같은 캡슐제(실리콘-기재 무수 조성물로서 젤라틴 캡슐내)에 포함시킬 수도 있다. 따라서, 본 발명은 또한 본문에 정의된 바와 같은 화장품적으로 허용되는 조성물을 포함하는 밀폐 용기를 제공한다.

〈실시예1〉
본 실시예는 본 발명의 조성물에 포함되는 화합물인 살리실옥시 α-프로피온산의 합성을 예시한다.
재료 및 방법:
양성자 자기 공명 스펙트럼은 브루커(Bruker) AC 200 모델 분광광도계로 기록하였다. 화학적 이동(chemical shift)는 내부적 표준으로으로서의 테라메틸실란으로부터 백만분의 일(parts per million) 단위로 보고되었다. 스핀 다중도(spin multiplicity)는 s(단일), d(이중), t(삼중), q(사중), m(다중) 및 br(광역)로 나타내었다. 중수소화 NMR 용매는 지정된 위치에 99.0-99.8%의 중수소를 포함하고, 캠브리지 이소토픽 레버러토리(Cambridge Isotopic Laboratories)에서 구입하였다.
적외선 스펙트럼은 NaCl 셀을 이용하여 니콜렛 임팩트(Nicolet Impact) 모델 410 분광계상에 기록되었다. 데이터는 퀵 IR 소프트웨어(Quick IR software)를 이용하여 처리하였다. 피크 위치는 cm-1단위로 vs(매우 강함), s(강함), m(중간), w(약함) br(넓음)로 열거하였다.
기체 크로마토그래피(GC)는 휴렛-펙커드 켐스테이션(ChemStation) 소프트웨어로 제어되고 HP 7673 분사기를 가지는 휴렛-팩커드 5890 시리즈 II 기체 크로마토그래피를 이용하여 수행하였다. 사용된 휴렛-팩커드 HP-1 칼럼은 25 M ×0.22 mm이고, 가교결합된 메틸 실리콘 피복 0.33 um을 갖는다. 매개 변수는 다음과 같다. 분사 온도=250℃, 초기 오븐 온도=50℃, 초기 시간=5분, 속도=25℃/min, 최종 오븐 온도=250℃. 시료는 트리메틸 실릴 에테르/에테르로서 분석하였다. 기체 크로마토그래피/질량 분석은 피니간(Finnigan) MAT ITD 800 이온 트랩 검출기와 연결된 휴렛-팩커드 5890 시리즈 II 기체 크로마토그래피상에서 수행하였다. 25 M×0.32 mm HP-5 칼럼은 5% 가교결합된 페닐 메틸 실리콘의 0.52um 피복층을 갖는다.
시차주사열량법 실험을 듀퐁 DSC상에서 2910 셀 베이스(cell base) 및 2100 열 분석기(thermal analyst)를 가지고 수행하였다. 약 1 mg의 시료를 알루미늄 팬에 정확히 정량하여 넣고 이를 밀봉하였다. 30℃에서 평형화시킨 후, 시료를 분당 5℃의 속도로 가열하였다.
모든 용매는 시약 등급이고 구입한 상태로 사용된다. 모든 시약은 알드리크(Aldrich) 또는 시그마(Sigma) 케미칼 컴퍼니에서 구입하여 구입한 상태로 사용하였다.
단계 1: 벤질-2-브로모프로피오네이트의 합성:
2-브로모프로피온산 20.0g(0.13몰)을 5-10℃에서 무수 염화 메틸렌중에서 벤질 알코올 14.1g(0.13몰)과 교반한 후, 4-디메틸아미노피리딘(DMAP) 0.16g (1.3 e-3몰)을 첨가하였다. 온도를 약 15℃로 높인 후, 무수 염화메틸렌 중의 디시클로헥실 카보디이미드(DCC) 용액(26.7g, 0.13몰)을 적가하였다. 이 용액을 반응 온도가 35℃ 이상으로 증가되지 않도록 하는 속도로 첨가하였다. 이미드 첨가가 완료된 후, 반응을 25℃에서 수시간 동안 또는 적외선 분광법 분석시 1730 cm-1에서 카르복실산 스트레치(stretch)가 사라지는 것이 나타날 때까지 진행하였다. 완료후, 반응 혼합물을 진공하에서 여과하여 디시클로헥실 우레아(DCU) 부산물을 제거하고, 여과액을 물로 한번, 5% 초산 용액으로 한 번 추출하고(DMAP를 제거하기 위해) 다시 물로 추출하였다. 유기층을 무수 황산 마그네슘으로 건조시킨 후, 여과하고 감압하에서 농축시켰다. 벤질-2-브로모프로피오네이트와 벤질-2-클로로프로피오네이트의 혼합물의 수율이 25g(72%)이었다.

단계 2: 벤질 프로피오닐 살리실레이트의 합성
상기 혼합물 25g(0.092몰)을 2-부타논 100 ml에 용해시켰다. 침전시킨 디시클로옥엑실 우레아(부산물)을 여과하여 제거하고, 이 여과액에 200ml의 부타논을 더 첨가하였다. 이 용액을 반응 용기로 옮기고, 요오드화 칼륨 촉매 0.15g (9.2e-4몰) 및 살리실산 나트륨 14.8g (0.092몰)을 첨가하였다. 반응 용액을 환류온도로 가열하고 반응의 완료를 기체 크로마토그래피로 측정하였다. 몇시간 후 반응을 완료시키기 위하여 1/2 당량의 촉매가 추가적으로 필요하다. 완료 후, 반응 혼합물을 냉각시키고 진공하에서 여과하여 소듐 브로미드 부산물을 제거하였다. 부타온을 감압제거한 후 산물을 염화 메틸렌중에 용해시키고 물로 한 번, 포화 중탄산나트륨 용액으로 한 번 및 마지막으로 물로 추출하였다. 유기층을 단리하여 무수 황산마그네슘으로 건조시킨 후 여과하고 감압하에서 농축시켰다. 90% 벤질 프로피오닐 살리실레이트 26g(82%)을 얻었다.

콜라겐은 주된 피부 단백질이다. 콜라겐의 합성은 노화 또는 광손상으로 감소한다. 이 콜라겐의 감소 또는 파괴는 피부의 신장력을 증가시켜 주름 및 이완을 야기한다. 인간을 대상으로 하는 많은 연구들은 광손상이 심해질수록 콜라겐 유형I이 감소한다는 것을 보여주고 있다[Kligman, A., JAMA, (1969), 210, pp2377-2380; Lavker, R., J. Inv Derm., (1979), 73, 79-66; Smith J. et al., J. Inv. Derm., (1962), 39, pp 347-350; 및 Shuster, S. et al., Br. J. 슬롯 블롯을 위한 프로콜라겐 I 염색 방법.
신생 인체 피부의 섬유아세포는 클로네틱스사(Clonetics Corp., San Diego, CA)로부터 구입하였다. 세포 배양을 위한 모든 재료는 라이프 테크놀로지사(뉴욕)로부터 구입하여 패시지(passage) 5-10에서 사용하였다. DMEM(둘베코의 개질된 이글스 배지), 2 mM L-글루타민이 보강된 고-글루코스, 10% 소 태아 혈청(fetal bovine serum), 및 항생 및 항진균적 용액을 포함하는 배지에서 96웰 플레이트의 내부 48웰에 약 웰당 10,000의 밀도로 세포를 접종하였다. 그 다음 2일 동안 세포가 빽빽히 자라도록 하였다. 빽빽히 자란 후, 배지를 제거하고 세포를 혈청을 제거한 DMEM으로 세척하고, 각 웰에 혈청이 제거된 DMEM중의 시험 화합물 용액 200 μl씩을 투여하였다. 하나의 접종을 총 6개의 웰에 복제하였다. 시험 화합물은 하기 표1에 나타낸 농도로 사용하였다. 대조군은 시험 화합물을 포함하지 않았다. 24시간 후, 시험 화합물 용액 또는 대조군 용액을 제거하고, 세포를 혈청이 제거된 DMEM중의 시험 화합물 용액 100μl을 재투여하였다. 시험 화합물은 하기 표1에 나타낸 농도로 사용되었다. 24시간 후 시험 화합물 용액 또는 대조군 용액을 제거하고, 아프로티닌 대 물의 비가 1:200인 프로테아제 억제제(아프로티닌은 시그마로부터 구입)와 함께 4℃에서 주말내내 저장하였다. 이어서 시험 화합물 용액을 DMEM 으로 희석하였다(200 μl DMEM 중에 약 20 μl의 시료).
니트로셀룰로스 멤브레인 및 여과지 3장을 TRIS 완충 염수(TBS, pH 7.3)에 담구었다. 여과지를 바닥에, 멤브레인을 위에 놓고 고정하여 바이오래드(BioRad) 슬롯 블롯 장치(BioRad Labs, CA)를 설치하였다. 웰당 100 ml의 TBS를 첨가하였다. 진공을 사용하여 멤브레인을 통하여 웰을 흡입하였다. 시험 화합물 용액 또는 대조군을 와동(vortex)시키고, 웰당 100μl씩을 가하고 중력 건조시켰다. 실험중 이 시점에서 시험 용액으로부터 프로콜라겐이 멤브레인에 부착된다. 장치에서 멤브레인을 제거하고, 여분을 잘라내고, 바닥 오른쪽 귀퉁이에 방향표시를 한다. 이 멤브레인을 차단 용액(둘베코의 인산염 완충 염수중의 5% 우유 분말)중에서 교반시키면서 밤새 두었다. 이 멤브레인을 0.1% BSA를 함유하는 TBS 중의 랫 항-인간 프로콜라겐 아미노 말단 항체(Chemicon MAB1912)(항체 대 완충액/BSA의 비가 1:100) 1.5 ml과 함께 밀봉 백에서 교반하면서 실온에서 1.5 시간 동안 인큐베이션하였다. 멤브레인을 제거하고, TBS/0.1% Tween 중에서 5분 동안 3회 세척하였다. 그 다음, 이 멤브레인을 0.1% BSA를 함유하는 TBS 중의 바이오틴연결된 항-랫 퍼옥시다제-포합된 항체(Vector Labs)(항체 대 완충액/BSA의 비가 1:1000) 2 ml 중에서 밀봉 백에서 교반하면서 실온에서 1시간 동안 인큐베이션하였다.

표1의 데이타로부터, 대조군에 비해 높은 농도계 독출값에 의해 나타내지는 바와 같이, 섬유아세포 배양액에 1, 2 및 5 mM 농도의 살리실옥시 α-프로피온산의 첨가는 프로콜라겐 I 생산의 증가를 가져옴을 알 수 있다. 10 mM 농도의 살리실옥시 α-프로피온산은 섬유아세포 배양물에 손상을 주는 것으로 믿어진다.
〈실시예 3〉
본 실시예는 다양한 시험 화합물의 처리에 따른 섬유아세포에 의한 글리코스아미노글리칸의 생산을 측정한다. 글리코스아미노글리칸(GAG)는 단백질 핵에 연결되어 프로테오글리칸을 형성할 수 있는 다당류의 한 종류이다(히아루로닉 에시드(HA)는 예외). 진피의 주된 GAG는 HA 및 더마탄 설페이트(dermatan sulfate)이고, 콘드로이틴-4-설페이트 및 콘드로이틴-6-설페이트가 소량으로 존재한다. 케라티노사이트 및 피부 섬유아세포 모두에 의해 만들어지는 GAG는, 비록 피부의 건조중량의 0.2% 밖에 차지하지 않지만 세포밖 매트릭스의 핵심적 성분이다. GAG측정 방법
신생 인간 진피 섬유아세포는 클로네틱스사(Clonetics Corp., San Diego, CA)로부터 구입하였다. 세포 배양을 위한 모든 재료는 뉴욕소재 라이프 테크놀로지로부터 구입하여 패시지(passage) 5-10에서 사용하였다. 양으로 하전된 제타 프로브 멤브레인(Zeta Probe membrane)을 멸균수에 담근 후, 돗트-블롯 장치에 넣었다(둘 다 CA Hercules소재, 바이오-래드 랩제품). 각 웰에 물 100 μl를 첨가하고 진공을 이용하여 통과시켰다. 녹인 후, 시험 화합물 시료 또는 기준(미주리주 세인트 루이스 소재 시그마사로부터 구입한 소 도관으로부터의 히알루로닉 에시드 또는 콘드로이틴 설페이트) 100μl씩을 멤브레인에 가하고, 중력 여과되도록 두었다(약 1.5 내지 2시간). 이제 GAG가 멤브레인에 부착되었다. 멤브레인을 물중의 지방산이 제거된 3%(w/v) 소 혈청 알부민(시그마사)에서 1시간 동안 차단시켰다. 착색 용액으로서 pH 약 2.3의 3% 아세트산 중의 0.5% (w/v) 알시안 블루 다이(Alcian Blue dye; 오하이오주 클리브랜드 소재 ICN 바이오케미칼사 제품) 용액을 제조하였다. 멤브레인을 증류수에서 2회 세척하고, 이어서 회전 교반기상에서 15분 동안 착색 용액중에서 착색시켰다. 염료는 따라 버리고, 멤브레인은 3% 아세트산 중에서 각 15분씩 2 회 탈염색시켰다. 멤브레인을 물로 헹구고, 밤새 건조되도록 두었다. 바이오-래드 이미지 분석 농도계를 이용하여 각 스폿에서의 색상의 강도를 정량화하였다. 시험 화합물로 처리한 세포에서의 농도계 독출값의 대조군에 대한 비로서 대조군에 대한 증가 배수를 계산하였다.

〈실시예 4〉
이 실시예는 다양한 시험 화합물에 의한 피지 억제의 생체외 분석을 보고한다.
피지세포에 의한 지질생성의 생체외 측정:
인간 피지선은 남성(60세)의 코로부터 단리하여 침지 세포 배양 기술(Bajor et al, J. Invest. Dermatol. 102: 1994, P. 564)을 이용하여 배양하였다. 이들 피지세포는 성숙 인간 피지의 특징인 세포내 지질 방울을 축적한다. 젖산 및 살리실산은 시그마로부터 구입하였다.
수거하고 통과된 피지세포를 48웰 조직 배양 플레이트의 각 웰에 첨가하고, 7.5% CO2의 존재에서 37℃에서 10일 동안 배양하였다. 실험 당일, 성장 배지를 제거하고 피지세포를 인산염 완충 염수(PBS)로 세 번 세척하였다. 신선한 PBS 0.5ml양을 각 웰에 첨가하고, 시험물을 0.001% 내지 0.2% 범위의 최종 농도로 5 ml씩 첨가하였다. 각 시료에 대하여 3개씩의 웰을 이용하였다. 대조군은 PBS, 살리실옥시 α-프로피온산을 용해시키기 위해 사용된 디메틸 술폭시드(DMSO), 및 에스트로겐 유사 활성을 가지는 화합물인 페놀레드(페놀레드는 피지 생산을 감소시키고 피지세포 측정의 완전성을 확실히 하는 대조군으로서 사용되었다)로 구성된다. 모든 배양액은 37℃/7.5% CO2에서 30분 동안 배양시켰다. 방사능 표지는14C 표지된 아세트산(아머샴, 나트륨 염, 비방사능(specific activity)이 56 mCi/mmol) 100ml을 50 mM 아세트산 나트륨 완충액 10 ml에 첨가하여 제조하였다. 이어서, 50ml을 피지세포 및 시험물을 함유하는 각 웰에 첨가하였다. 배양액을 4시간 동안 배양기에 넣고 배양시켰다. 이후, 피지세포를 신선한 PBS로 3회 세척하여 결합되지 않은 활성제 및 방사능 표지를 제거하였다. 배양된 피지세포에 남아있는 방사능 표지를 벡크만 신틸레이션 계수기(scintillation counter)를 이용하여 계수하였다. 그 결과를 대조군(DMSO)과 비교하여 % 감소로서 표현하였다.

표3의 결과로부터, 0.10% 이상 농도의 살리실옥시 α-프로피온산은 일관되게 피지세포에 의한 피지 분비를 억제하는 것을 알 수 있다. 4번의 실험 중 3번에서 살리실옥시 α-프로피온산은 0.01% 미만의 농도에서도 효과가 있었다. 0.1% 에서 젖산은 같은 농도에서의 살리실옥시 α-프로피온산에서 보다 효과가 현저히 낮았다. 살리실산은 효과가 없었다.
〈실시예 5〉
본 실시예는 살리실옥시 α-프로피온산에 의한 피부색 명화(lightening) 능력의 생체외 분석을 보고한다.
B16 세포는 ATCC(메릴랜드주 락빌 소재)로부터 구입하였다. 빽빽하게 자라기 전의 B16세포를 96웰 마이크로타이터 플레이트에 웰당 5000개의 세포 농도로 접종하고, 10% 소 태아 혈청(Fetal Bovine Serum), 1% 페니실린/스트렙토마이신(페놀레드 없음)을 함유하는 DMEM(뉴욕소재 라이프 테크놀로지사)중에서 5% CO2하에서 37℃에서 밤새 배양하였다. 24시간 후, 상기 처리를 함유하는 신선한 배지로 배지를 교체하였다. 72시간 동안 세포를 배양하면, 대조군 처리에서 멜라닌이 보였다. 다음에, 각 웰로부터 멜라닌을 함유하는 배지를 깨끗한 96웰 플레이트에 옮기고 마이크로플레이트 분광광도계를 이용하여 530nm에서의 흡광도를 읽어서 정량화하였다.
멜라닌 억제가 단순히 세포 사망에 기인하는 것이 아님을 확실히 하기 위하여, 세포의 생존을 중성 적색 염료의 흡취로서 측정하였다. 배지의 제거 후, 25μg/ml의 중성 적색 염료를 함유하는 미리 따뜻하게 한 배지 200μL를 각 웰에 첨가하고 3시간 동안 배양하였다. 세포를 PBS로 2회 세척하였다. 50 H2O: 49 에탄올: 1 아세트산의 100μL를 첨가하고 실온에서 20분 동안 부드럽게 교반하여 염료를 추출하였다. 염료는 530nm에서의 흡광도를 읽음으로써 정량화하였다. 생존하는 세포만이 염료를 취할 것으로 기대되고 흡광도는 이 처리에서 살아남는 생존 세포의 수와 직접 비례한다.
각 처리에 대하여, 4개의 복제품의 독출값의 평균을 계산하고 처리하지 않은 대조군에 대한 평균의 %비율로서 나타내었다. 통계적 의미는 스튜던트(student's) t-검정을 이용하여 결정하였다(50% 초과의 생존률을 보이는 처리에 대하여서만 보고하였다).
얻어진 결과를 표4에 요약하였다. 멜라닌은 %비처리 대조군으로서 표현되므로, 이 수가 적을수록 활성은 더 좋다(중성 적색 수는 100에 가깝다.).

Claims (9)
- (a) 하기 화학식 (I)의 살리실옥시 α-카르복시산 0.0001 내지 20 중량%〈화학식 I〉(여기서, R은 수소 또는 1 내지 16개의 탄소 원자를 포함하는 알킬 라디칼이고, M은 수소, 또는 알칼리 또는 알칼리 토금속에서 선택되는 금속 양이온이다) 및(b) 화장품적으로 허용되는 비히클을 함유하는 피부 조절 조성물.
- 제1항에 있어서, 상기 살리실옥시 α-카르복시산 중의 R이 메틸기인 조성물.
- 제1항 또는 제2항에 있어서, 상기 살리실옥시 α-카르복시산 중의 M이 수소 또는 나트륨인 조성물.
- 제1항 내지 제3항 중의 어느 한 항에 따른 조성물을 피부에 바르는 것을 포함하는 지성피부를 조절하는 화장법.
- 제1항 내지 제3항 중의 어느 한 항에 따른 조성물을 피부에 바르는 것을 포함하는, 피부에서 피지세포로부터의 피지분비를 감소, 방지 또는 조절하는 화장법.
- 제1항 내지 제3항 중의 어느 한 항에 따른 조성물을 피부에 바르는 것을 포함하는, 피부에서 섬유아세포에 의한 콜라겐 및 글리코스아미노글리칸의 합성을 촉진시키는 화장법.
- 제1항 내지 제3항 중의 어느 한 항에 따른 조성물을 피부에 바르는 것을 포함하는, 노화, 광노화, 건조, 잔주름 또는 주름진 피부를 치료하는 화장법.
- 제1항 내지 제3항 중의 어느 한 항에 따른 조성물을 피부에 바르는 것을 포함하는, 피부를 유해한 UVA 및 UVB광으로부터 피부를 차단하는 화장법.
- 제1항 내지 제3항 중의 어느 한 항에 따른 조성물을 피부에 바르는 것을 포함하는, 피부를 밝게 하는 화장법.
