제조예 1: 산양삼 추출조성물 제조 Preparation Example 1: Preparation of wild ginseng extract composition

산양삼은 주식회사 자연애 바이오랩 농업회사법인으로부터 구입하여 사용하였다. Wild ginseng was purchased and used from Jayeonae Biolab Agricultural Corporation.

산양삼 분말 5 g에 물을 100ml 첨가한 후 추출온도를 80℃로 설정하여 각각 2시간 및 4시간 환류냉각장치에서 추출하여 추출조성물을 얻었다 (실시예 1 및 2). 진세노사이드 분석은 고압액체크로마토그래피(HPLC, high press liquid chromatograph)로 분석하였다. 분석 컬럼은 C18 TSKgel ODS-100Z을 사용하여 시료주입량 20 ㎕, 온도는 30℃, 측정파장은 203 nm, 유속은 1.0 ㎖/min으로 하였고 이동상으로는 A용액은 HPLC water, B용액은 아세토나이트릴을 사용하였다. HPLC 분석 조건은 이동상 용액은 0분때 A용액 100% : B용액 0%로 흘려주고 5분때에는 A용액 80% : B용액 20%로 흘려주고 12분때 A용액 70% : B용액 30%, 45분때 A용액 65% : B용액 35%, 60분때 A용액 10% : B용액 90%, 65분때에 A용액 8% : B용액 92%로 이동상을 흘려주었다. 실시예 1 및 실시예 2의 진세노사이드 HPLC 크로마토그램을 각각 도 1 및 도 2에 나타냈고, 분석된 진세노사이드 Rg1 및 Rb1 함량을 표 1에 나타내었다.Ginsenoside analysis was performed using high press liquid chromatography (HPLC). 도 1 및 도 2에 나타낸 바와 같이, 본 발명에 따른 실시예 1 및 2의 산양삼 추출조성물에서는 진세노사이드 Rg1 및 Rb1 피크가 현저히 높아진 것을 확인할 수 있다. 또한, 표 1에 나타낸 바와 같이, 실시예 1 및 2의 산양삼 추출조성물은 진세노사이드 Rg1이 각각 17.85 mg/g 및 18.31 mg/g, 진세노사이드 Rb1이 각각 12.22 mg/g 및 13.64 mg/g으로 나타나, 다른 추출조건으로 추출된 비교예 1 내지 비교예 10의 산양삼 추출조성물에 비하여 현저히 증진되었음을 확인할 수 있다. 반면에 열수의 온도가 실시예들에 비하여 5℃ 낮거나 (비교예 3 및 4) 5℃ 높더라도 (비교예 5 및 6) 추출조성물에서 진세노사이드 Rg1 및 Rb1의 함량이 현저히 떨어졌다 (표 1). 항산화 활성은 DPPH 라디칼 소거활성 및 ABTS 라디칼 소거활성을 측정하여 분석하였다.Antioxidant activity was analyzed by measuring DPPH radical scavenging activity and ABTS radical scavenging activity.

<DPPH 라디칼 소거활성><DPPH radical scavenging activity>

실시예 1 및 실시예 2의 분석시료 각각을 100 ㎕를 96 웰-플레이트에 분주하고, 0.5 mM DPPH 에탄올 용액 (1,1-diphenyl-2-picrylhydrazyl) 50 ㎕를 첨가하여 혼합한 후 25℃에서 30분 동안 반응시킨 후 517 nm에서 흡광도를 측정하였다. DPPH 라디칼 소거활성의 음성 대조구는 시료 대신 PBS를 사용하여 동일한 방법으로 진행하여 흡광도의 차이를 다음과 같은 식에 의해 백분율(%)로 산출하였으며, 그 결과를 도 3에 도시하였다. 7 mM ABTS 5ml와 140 mM K2S2O8 (potassium persulfate) 88 ㎕를 혼합한 후 상온에서 암실에서 16시간 반응시켜 ABTS 양이온 용액을 제조하였다. 이 용액을 414 nm에서 흡광도 값이 1.5가 되도록 PBS로 희석하고 조제된 희석용액 190 ㎕와 각각 실시예 1 및 실시예 2의 분석시료 10 ㎕를 혼합한 후 상온에서 6분간 반응시킨 후 734 nm에서 흡광도를 측정하였다. ABTS 라디칼 소거활성의 음성 대조구는 시료 대신 PBS를 사용하여 동일한 방법으로 진행하여 흡광도의 차이를 상기 식에 의해 백분율(%)로 산출하였으며, 그 결과를 도 4에 도시하였다. 7mM ABTS 5ml with 140mM K 2 S 2 O 8 (potassium persulfate) After mixing 88 ㎕, the ABTS cation solution was prepared by reacting for 16 hours in the dark at room temperature. 시험예 3. 이화학적 특성 분석Test Example 3. Physicochemical property analysis

제조예 1에서 제조된 각각의 추출조성물의 pH, 산도 및 가용성 고형물 함량을 분석하였다. pH는 각각의 추출조성물 1 mL에 증류수 49 mL를 비이커에 옮겨 담아 pH meter (MP 200, Jenway, Lodon, UK)를 사용하여 측정하였다. 산도 측정은 pH를 측정한 시료에 0.1 N NaOH 용액으로 pH 값이 8.3±0.2까지 중화시킨 후 젖산 양으로 환산하여 %로 나타내었다. 가용성 고형물은 각 추출조성물 1 mL를 굴절당도계 (Sccharometer, W.S.R.O-90, Atago, Co., Tokyo, Japan)로 당도를 측정하였다. 그 결과를 표 2에 나타냈다. 본 발명에 따라 제조된 산양삼 추출조성물은 pH는 약간 증가하였고, 산도 및 가용성 고형물은 크게 변화가 없었다. pH가 높을 경우에는 식품위해미생물 등의 증식이 가능하고, 너무 낮을 경우에는 과다 산이 생성되면 신맛이 강화하여 기호성에 문제가 발생할 수 있는데, 본 발명의 산양삼 추출조성물은 적당한 pH와 산도로 기호성이 좋다. Claims (3)

삭제delete 삭제delete 진세노사이드 Rg1 및 Rb1이 강화된 음료의 제조방법으로, 상기 방법은
i) 산양삼은 흐르는 물에 세척한 후, 물기를 제거하고, 50 ~ 55℃의 드라이오븐에서 이틀간 열풍건조하고 이를 분쇄기로 분쇄하여 80 ~ 100 메쉬(mesh)의 체에 걸러 분말화하는 단계;
ii) 산양삼 분말에 20배(v/w)의 80℃ 열수를 혼합하여 환류냉각 추출을 2 시간 수행하여 산양삼 추출조성물을 얻는 단계; 및
iii) 산양삼 추출조성물 96.04 중량%, 산양삼배양근 3.84 중량%, 비타민 C 0.06 중량% 및 자몽종자추출물 0.06 중량%을 혼합하는 단계;를 포함하고,
상기 산양삼 추출조성물은 진세노사이드 Rg1을 17.8 mg/g 이상 및 진세노사이드 Rb1을 12 mg/g 이상 함유하는 것인 제조방법.
