진세노사이드 분석은 고성능액체크로마토그래피(UPLC, Ultra performance liquid chromatograph)로 분석하였다. 분석 컬럼은 WATERS Acquity BEH C18(2.1 mm × 50 mm, 1.7 ㎛)을 사용하였고 이동상 용매는 0.001% 증류수 중의 인산(phosphoric acid in distilled water; solution A)와 0.001% 아세토니트릴 중의 인산 (phosphoric acid in acetonitrile; solution B)로 분석하였다. 이동상 조건은 A 용매 기준으로 0-5분-80%, 15분-70%, 26분-50%, 35분-80%를 유지하였다. 각각의 분석시료는 2 ㎕를 주입하였고 이동상 속도는 40℃에서 0.6 ㎖/min으로 설정하였다. 검출기는 PDA detector를 사용하여 203 nm에서 검출하였다. 각 실험은 3회 반복하여 평균값을 산출하여 그 결과를 표 2에 나타냈고, 진세노사이드 HPLC 크로마토그램을 도 3에 나타냈다(실시예 1 - 검은색(sample 2), 비교예 1 - 붉은색(sample 1)). 총 폴리페놀 함량은 액상차 조성물 시료를 96-well 플레이트에 분주한 후 1 N Folin-Ciocalteu (Sigma, USA) 시약을 넣고 실온에서 3분간 반응시킨 다음 10% Na2CO3 용액을 80 ㎕ 첨가하여 1시간 반응시키고 나서 Multimicroplate reader-SpectraMax M5 (Molecular Devices, USA)로 765 nm에서 흡광도를 측정하였다. 총 폴리페놀의 함량은 0∼200 ㎍/㎖의 갈산를 이용하여 표준 곡선을 작성하고 시료 1 g당 갈산의 함량(GA)으로 산출하여 표 3에 나타내었다. 상기 숙성 산양산삼 열수추출액은 산양산삼 전초를 100℃에서 1시간 증숙하고 68℃에서 3일간 중온 숙성 후 55℃ 2일간 건조하여 숙성 산양산삼을 제조한 후, 숙성 산양산삼에 190~200배(w/w)의 물을 첨가하여 95℃에서 8시간 동안 열수 추출한 후 여과하여 얻는 것이고,
상기 산양산삼 액상차 조성물은 진세노사이드 Rb1, Rg1 및 Rg3을 각각 56 ㎍/㎖ 이상, 40 ㎍/㎖ 이상, 및 34 ㎍/㎖ 이상으로 함유하는 것을 특징으로 하는 조성물.
