Abstract
본 발명은 항염증 활성을 나타내는 신규 화합물에 관한 것으로, 본 발명의 화합물은 염증전구물질 (pro-inflammatory cytokines)의 생성에 결정적인 역할을 하여 염증성 질환을 유발하는 것으로 알려진 p38 MAPK에 대하여 우수한 억제효능을 가지므로, 염증성 질환의 치료제로 유용하게 사용될 수 있다.
Description
본 발명은 항염증 활성을 나타내는 신규 화합물에 관한 것으로, 본 발명의 화합물은 특히 p38 MAP 키나아제 억제 효능이 우수하여 염증성 질환의 치료제로 유용하게 사용될 수 있다.
p38은 MAP 키나아제 (mitogen-activated protein kinases, MAPK) 패밀리에 속하는 세린/트레오닌 인산화 효소의 일종이다. MAPK 경로는 여러 가지 세포 외부의 자극에 의해 활성화되어 세포의 특정 기질의 인산화를 통해 세포 내 여러 가지 반응이 일어나도록 전환하는데, MAPK의 하위 종류로는 p38 외에도 JNK, ERK등이 있다. MAPK는 인슐린 (insulin), 유사분열물질 (mitogen), 성장인자 (growth factor), 면역세포의 활성화 자극 등에 의해 활성화되는 단백질 인산화 효소이며 세포 표면에 있는 수용체의 신호를 세포 안으로 전달하는 신호전달경로에서 중요한 위치를 차지한다.
p38 신호전달 체계는 신진 대사 및 세포 사멸과 같은 과정을 조절하며 상위 단계의 인산화 효소가 하위 단계의 인산화 효소에게 인산기를 전달하는 일련의 연쇄적인 인산화 과정을 거친다. p38 MAPK로는 p38-α, p38-β, p38-γ. p38-δ 등이 밝혀졌으며 삼투압 (osmotic shock), 염증 사이토카인 (inflammatory cytokines), LPS (lipopolysaccharides), 자외선 (UV), 성장인자 (GF) 등에 의해 활성화된다.
염증은 여러 가지 원인에 의해서 발생하는 것으로 알려져 있으나, 여러 연구에 의하면 다양한 스트레스 자극에 의해 MAPK가 활성화될 수 있고, 특히 p38 MAPK는 TNF-α, IL-1β, IL-6 등과 같은 염증전구물질 (pro-inflammatory cytokines)의 생성에 결정적인 역할을 하여, 염증성 질환을 유발하는 것으로 알려져 있다 (NATURE REVIEWS, Drug discovery (2003), Vol.2, 717-726). 이에 p38 MAPK의 억제제로서 몇몇 저분자 화합물 (small molecule)이 개발되어왔고 (한국등록특허 제10-1879481호 등), p38의 여러 가지 저분자 억제제는 낮은 농도에서 사람 단핵세포에서 IL-1 및 TNF 합성을 억제함으로써 염증 조절자 역할을 하며, 이러한 사이토카인의 억제는 염증 반응을 조절, 완화 또는 경감시킬 수 있는 것으로 밝혀졌다.
이와 관련하여, 본 발명의 발명자들은 p38 MAPK 억제를 통해 항염증 활성을 갖는 신규한 화합물을 연구한 결과, 본 발명을 완성하였다.

본 발명은 염증성 질환의 효과적인 치료를 위해서, p38 MAP 키나아제를 효과적으로 억제하여 우수한 항염증 효능을 갖는 신규 화합물을 제공하는 것을 목적으로 한다.
상기 목적을 달성하기 위하여, 본 발명은 하기 화학식 1의 화합물 또는 이의 약학적으로 허용가능한 염을 제공한다.

정의
본 명세서에서 "약학적으로 허용 가능한 염"은 약학적으로 허용 가능하고 모 화합물 (parent compound)의 바람직한 약리 활성을 유지하는 염을 의미한다. 상기 염으로는 약학적으로 허용되는 것이면 특별히 한정되지 않으며, 예를 들어 염산, 황산, 질산, 인산, 불화수소산, 브롬화수소산, 포름산 아세트산, 타르타르산, 젖산, 시트르산, 푸마르산, 말레산, 숙신산, 메탄술폰산, 벤젠술폰산, 톨루엔술폰산, 나프탈렌술폰산 등을 사용할 수 있다.

본원에서 사용되는 용어 "할로" 및 "할로겐"은 할로겐을 의미하고, 클로로, 플루오로, 브로모, 및 요오도를 포함한다.

본 발명에 따른 화합물은 용이하게 입수가능한 출발 물질로부터 당업자에게 익히 공지된 아래 기재된 특정 합성 프로토콜에 대한 변형을 이용해서 제조될 수 있다.
[제조예]
본 발명에 따른 화합물의 합성 방법에 사용된 기기 및 시약
본 발명의 화합물 합성에 사용된 모든 시작물질과 시약들은 Aldrich, Alfa Aesar, TCI 등으로부터 구입하였으며 별도의 정제과정 없이 사용하였다. 반응에 사용된 각종 초자들은 80℃ 오븐에서 건조시킨 상태로 사용하였으며 공기 및 수분을 차단하기 위해 아르곤 가스나 질소 가스를 주입하여 반응을 진행하였다.
본 발명의 칼럼 크로마토그래피 (Flash column chromatography)는 실리카겔 60 (230-400 mesh)를 이용하여 헥세인 (Hexane), 에틸 아세테이트 (Ethyl acetate), 다이클로로메테인 (Dichloromethane), 메탄올 (Methanol) 등의 용매들을 적절히 조절하여 사용하였다. 분석용 얇은막 크로마토그래피 (Thin-layer chromatography)는 0.25 mm 실리카겔 플레이트를 사용하였다.
본 발명의 1H-NMR 스펙트럼과 13C-NMR 스펙트럼은 Bruker Avance 400 (400 MHz for 1H; 100 MHz for 13C) spectrometer 또는 VARIAN VNMRS 500 (500MHz for 13C) spectrometer를 이용해 분석하였다. 1H과 13C NMR 화학 시프트 값은 TMS (tetramethylsilane)를 내부 기준으로 사용하여 ppm단위로 표현되었다. NMR 신호들은 m (multiplet), s (singlet), d (doublet), t (triplet), q (quartet), quin (quintet), bs (broad singlet), bd (broad doublet), dd (doublet of doublets), dt (doublet of triplets), dq (doublet of quartets) 등으로 표기되었고 결합 상수는 헤르츠 (Hz)단위로 표현되었다.
본 발명의 저해상도 질량 스펙트럼 (Low resolution mass spectra)과 고해상도 질량 스펙트럼 (High resolution mass spectra)은 VG Trio-2 GC-MS 또는 JEOL JMS-700 기기를 이용해 측정하였다.
일 실시태양에서, 본 발명의 중간체 화합물 16 내지 17은 하기 반응식 1에 따라 제조될 수 있다.

상업적으로 구입 가능한 4-브로모벤조일 클로라이드를 이용하여 상기 반응식 1에 따라 합성을 진행하였다. 반응식 1-a에 따라, 웨인렙 아마이드 (weinreb amide)를 형성시켜 화합물16을 수득하고, 적절한 알킬 그리냐르 시약을 이용하여 화합물 17a 내지 17f를 얻었다.
일 실시태양에서, 본 발명의 중간체 화합물 18은 하기 반응식 2에 따라 제조될 수 있다.

상업적으로 구입 가능한 1-(tert-부톡시카보닐)피롤리딘-3-카복실산 혹은 시클로펜텐 카복실산을 출발물질로 하여 EDC 커플링을 통해 웨인렙 아마이드를 형성시킨 후 n-부틸리튬 조건에서 디브로모벤젠을 치환반응하여 화합물 19 및 20을 수득하였다. 또한 화합물 20을 이용한 히드로 붕소화 반응을 통해 히드록시시클로펜틸 유도체 화합물 21 혹은 알콕시 수은화 반응을 통해 히드록시에톡시시클로펜틸 유도체 화합물 22를 합성하여 각각 부분입체 이성질체 혼합물 상태로 수득하였다. 또한 화합물 22를 THF에 녹인 후, t-BuOK를 적가하고 MeI를 첨가해 화합물 23을 수득하였다.

N-시클로프로필-4'-(2-히드록시-2-메틸프로파노일)-6-메틸-[1,1'-바이페닐]-3-카르복사마이드 (N-cyclopropyl-4'-(2-hydroxy-2-methylpropanoyl)-6-methyl-[1,1'-biphenyl]-3-carboxamide, 화합물 1)의 합성

N-시클로프로필-4'-(3-(2-메톡시에톡시)시클로펜테인-1-카보닐)-6-메틸-[1,1'-바이페닐]-3-카르복사마이드 (N-cyclopropyl-4'-(3-(2-methoxyethoxy)cyclopentane-1-carbonyl)-6-methyl-[1,1'-biphenyl]-3-carboxamide, 화합물 12)의 합성
(4-브로모페닐)(3-(2-메톡시에톡시)시클로펜틸)메탄온 (96.7 mg, 0.296 mmol)을 출발물질로 하여 N-시클로프로필-4-메틸-3-(4,4,5,5-테트라메틸-1,3,2-디옥사보롤란-2-yl)벤즈아미드 (134 mg, 0.444 mmol), 테트라키스(트리페닐포스핀)팔라듐 (35 mg, 0.030 mmol) 및 탄산칼륨(123 mg, 0.888 mmol)을 아이소프로필알콜 (3.5 mL)에 녹인 후 90 oC로 가열하였다. 얇은막 크로마토그래피로 출발물질이 없어진 것이 확인되면 온도를 상온으로 낮추고 2N-염산을 이용하여 반응을 중지시킨 뒤 셀라이트로 여과하여 여액을 아세트산에틸과 물로 추출을 진행하였다. 얻어진 유기층에 황산마그네슘을 넣어 탈수한 후 감압 여과한 여액의 용매를 제거하여 농축시켰다. 이후 컬럼 크로마토그래피를 통해 화합물 12를 최종적으로 수득하였다 (12 mg, 10%); 1H-NMR (400 MHz, CDCl3) δ 8.03 (d, J = 8.2 Hz, 2H), 7.68-7.65 (m, 1H), 7.58 (d, J = 1.68 Hz, 1H), 7.42-7.40 (m, 2H), 7.33 (d, J = 7.9 Hz, 1H), 6.25 (s, 1H), 4.11-4.10 (m, 1H), 3.99-3.98 (m, 1H), 3.61-3.55 (m, 4H), 3.41 (s, 3H), 2.91-2.90 (m, 1H), 2.29-2.27 (s, 3H), 2.19-2.04 (m, 4H), 1.91-1.85 (m, 2H), 0.89-0.83 (m, 2H), 0.62-0.61 (m, 2H).

N-시클로프로필-4'-(2-히드록시프로파노일)-6-메틸-[1,1'-바이페닐]-3-카르복사마이드 (N-cyclopropyl-4'-(2-hydroxypropanoyl)-6-methyl-[1,1'-biphenyl]-3-carboxamide, 화합물 13)의 합성
중간체 화합물 N-시클로프로필-6-메틸-4'-프로피오닐-[1,1'-바이페닐]-3-카르복사마이드 (26.0 mg, 0.085 mmol), 브롬화제이구리(2 mg, 0.1eq)를 DMSO 1ml에 녹인 후 아르곤 기체 치환 뒤 90℃에서 반응하였다. 얇은막 크로마토그래피로 출발물질이 사라진 것을 확인하면 다이클로로메테인과 물로 묽혀 반응을 종결하였다. 다이클로로메테인과 물로 추출을 진행한 뒤 유기층을 황산마그네슘으로 탈수한 후 감압여과 한 여액의 용매를 농축하여 제거하였다. 이후 컬럼 크로마토그래피를 통해 화합물 13을 최종적으로 수득하였다 (7.7 mg, 28%); 1H-NMR (400 MHz, CDCl3) δ 7.99 (d, J = 8 Hz, 2H), 7.66 (dd, J = 7.8 Hz, 1H), 7.61 (m, 1H), 7.45 (d, J = 8.4 Hz, 2H), 7.34 (d, J = 8 Hz, 1H), 6.25 (ds, 1H), 3.82-3.78 (m, 1H), 2.93-2.86 (m, 1H), 2.30 (s, 3H), 1.50 (d, J = 7.2 Hz, 3H), 0.90-0.84 (m, 2H), 0.63-0.59 (m, 2H).

N-시클로프로필-6-메틸-4'-(피롤리딘-3-카보닐)-[1,1'-바이페닐]-3-카르복사마이드 (N-cyclopropyl-6-methyl-4'-(pyrrolidine-3-carbonyl)-[1,1'-biphenyl]-3-carboxamide, 화합물 14)의 합성
중간체 화합물 tert-부틸 3-(5'-(시클로프로필카바모일)-2'-메틸-[1,1'-바이페닐]-4-카보닐)피롤리딘-1-카복실레이트 (103 mg, 0.230 mmol)을 다이클로로메테인 (5 mL)에 녹인 후 트리플루오로아세트산 (1 mL)를 적가하여 상온에서 반응하였다. 얇은막 크로마토그래피로 출발물질이 없어진 것이 확인되면 온도를 올려 용매를 완전히 제거한 후 염산 (2.0 M in ether)를 부어 침전을 형성하였다. 에터를 이용해 세척한 후 침전을 탄산수소나트륨 포화 수용액에 녹여 염기성화 한 후 아세트산에틸로 추출을 진행하였다. 얻어진 유기층에 황산마그네슘을 넣어 탈수한 후 감압 여과한 여액의 용매를 제거하여 농축시켰다. 이후 컬럼 크로마토그래피를 통해 화합물 14를 최종적으로 수득하였다 (33mg, 41%); 1H-NMR (400 MHz, CDCl3 + MeOD) δ 8.11 (d, J = 8.3 Hz, 2H), 7.75-7.72 (m, 1H), 7.70 (d, J = 1.6 Hz, 1H), 7.52 (d, J = 8.3 Hz, 2H), 7.37 (d, J = 8.0 Hz, 1H), 4.34-4.29 (m, 1H), 3.65-3.61 (m, 1H), 3.47-3.42 (m, 1H), 3.35-3.33 (m, 1H), 3.32-3.25 (m, 2H), 2.87-2.84 (m, 1H), 2.50-2.43 (m, 1H), 2.32 (s, 3H), 2.22-2.16 (m, 1H), 0.84-0.79 (m, 2H), 0.68-0.64 (m, 2H).
[실시예 15]
N-시클로프로필-4'-(1-(2-메톡시에틸)피롤리딘-3-카보닐)-6-메틸-[1,1'-바이페닐]-3-카르복사마이드 (N-cyclopropyl-4'-(1-(2-methoxyethyl)pyrrolidine-3-carbonyl)-6-methyl-[1,1'-biphenyl]-3-carboxamide, 화합물 15)의 합성
중간체 화합물 N-시클로프로필-6-메틸-4'-(피롤리딘-3-카보닐)-[1,1'-바이페닐]-3-카르복사마이드 (화합물 14) (19 mg, 0.055 mmol)와 탄산칼륨 (15 mg, 0.110 mmol)를 디메틸포름아마이드 (2 mL)에 녹인 후 2-브로모에틸 메틸 에터 (0.008 mL)를 적가하였다. 반응 온도는 50 oC로 가열하였다. 얇은막 크로마토그래피로 출발물질이 없어진 것이 확인되면 온도를 상온으로 낮춘 후 아세트산에틸을 이용하여 희석시켰다. 아세트산에틸과 물로 추출을 진행하여 얻어진 유기층에 황산마그네슘을 넣어 탈수한 후 감압 여과한 여액의 용매를 제거하여 농축시켰다. 이후 컬럼 크로마토그래피를 통해 화합물 15를 최종적으로 수득하였다 (4 mg, 19%); 1H-NMR (400 MHz, CDCl3) δ 8.01 (d, J = 8.2 Hz, 2H), 7.68-7.65 (m, 1H), 7.59 (d, J = 1.8 Hz, 1H), 7.42 (d, J = 8.2 Hz, 2H), 7.34 (d, J = 7.9 Hz, 1H), 6.22 (s, 1H), 4.11-4.04 (m, 1H), 3.57 (t, J = 5.4 Hz, 2H), 3.38 (s, 3H), 3.28 (bs, 1H), 3.07 (bs, 1H), 2.93-2.87 (m, 1H), 2.80 (bs, 2H), 2.57-2.51 (m, 1H), 2.29 (s, 3H), 2.25-2.20 (m, 1H), 2.04-2.00 (m, 2H), 0.89-0.83 (m, 2H), 0.63-0.59 (m, 2H).
[실험예 1]
Calculated LogP (CLogP)를 통한 지질친화도 평가
Chemdaw 소프트웨어를 이용하여 화합물 1부터 15까지 구조식을 기입한 후 CLogP 값을 확인하여 그 결과를 하기 표 1에 나타내었다.

기존의 약물을 조사하여 작성된 Lipinski's rule of five에 따르면 CLogP가 5 미만일 경우 경구투여되기 쉬운 약물의 특징을 가진다. 따라서 화합물 1 내지 15 화합물이 경구투여 약물로 개발가능한 것을 알 수 있다.
[실험예 2]
p38α MAPK(mitogen-activated protein kinase) 저해능 평가
p38 MAPK를 저해하는 능력을 확인하기 위해 화합물 1부터 15를 희석하여 농도에 따른 저해효과 확인하였다. 간략하게, 1000, 333, 111, 37.0, 12.3, 4.12, 1.37, 0.457, 0.152, 0.0495 nM농도로 화합물을 희석한후 각각 peptide/kinase 혼합물 (최종 7.07-28.3 ng p38 alpha, 2 μM Serine/Threonine 15, 50 mM HEPES pH 7.0, 0.01% NaN3)과 ATP 용액을 넣었다. 1시간 상온에서 반응시킨 후 development 시약을 넣고 1시간 상온에서 다시 반응시켰다. 445 nm와 520 nm 파장에서 Kinase 저해능을 확인하여, 그 결과를 하기 표 2에 나타내었다.

본 발명의 화합물의 대부분이 100 nM이하의 농도에서 p38 alpha를 50% 억제하는 강력한 저해제임을 확인하였다.
[실험예 3]
세포에서의 p38 MAPK 단백질 억제능 평가
p-p38 MAPK의 억제를 확인하기 위해 p-p38 MAPK 활성에 따라 염증을 유발하는 macrophage를 암세포화시킨 Raw264.7 세포에 각 화합물을 처리한 후 p38 MAPK의 억제를 확인하였다. 간략하게, Raw264.7 세포를 10% 소태아혈청 및 penicillin/streptomycin (100 U/ml)이 보충된 Dulbecco's modified Eagle Medium에서 배양하였다. 배양접시에서 Raw264.7 세포에 본 발명에 따른 화합물 1 내지 15를 10 μM씩 처리하여 CO2 배양기에서 24 시간 배양하였다. 단백질 면역블랏을 통해 각 화합물이 세포의 p-p38 MAPK의 억제에 대한 영향을 확인하였다.
실험 조건에 따라 세포의 배양이 끝난 후, 차가운 phosphate buffered saline (PBS)로 세척하였다. 이후 세포를 수거하여 1200 rpm에서 3분동안 원심분리하였다. 침전물을 2X sample buffer (1M Tris-HCl pH 6.8, 50 % Glycerol, 10 % SDS, 2-mercaptoethanol, 1 % bromophenol blue)로 세포를 용해하고 100 ℃에서 10 분 동안 끓였다. 제조된 단백질 샘플을 Sodium Dodecyl sulfate poly acrylamide gel electrophoresis 하고 PVDF membrane (Millipore)으로 옮겼다. PVDF membrane의 단백질과 항체 사이의 특이적 결합 반응을 위해 5 % bovine serum albumin (BSA, RMbio) 용액에서 blocking을 수행하였다. 이어서 BSA 용액을 폐기하고 membrane을 PBS-T (0.5 % tweene 20 in PBS)로 세척하였다. Membrane의 단백질과 antibody의 결합 반응을 4 ℃에서 밤새 수행하고 PBS-T로 다시 세척하였다. 마지막으로, Horse radish peroxidase (HRP)이 접합된 2차 항체 (Cell signaling Technology)를 PBS-T에 희석하고 실온에서 2 시간 동안 반응시켰다. Luminata Forte HRP substrate (Millipore)을 사용하여 membrane의 단백질양을 확인하였다.
그 결과를 하기 표 3에 나타내었다.

면역 블랏 결과 합성한 본 발명의 화합물은 10 μM 농도에서 세포 내의 p38 인산화를 효과적으로 억제하는 것을 확인하였다.
[실험예 4]
세포 염증반응에 따른 Nitric Oxide억제능 평가
p-p38 MAPK의 억제에 따른 염증억제 효과를 확인하기 위해 염증반응에 관여하는 macrophage를 암세포화시킨 Raw264.7 세포에 각 화합물을 처리하여 p38 MAPK의 억제에 따른 염증억제를 확인하였다. 간략하게, Raw264.7 세포를 10% 소태아혈청 및 penicillin/streptomycin (100 U/ml)이 보충된 Dulbecco’s modified Eagle Medium에서 배양하였다. 배양접시에서 Raw264.7 세포에 본 발명에 따른 화합물 1 내지 15를 10 μM씩 처리하여 CO2 배양기에서 24 시간 배양하였다.
실험 조건에 따라 세포의 배양이 끝난 후, 상층액을 취득하여 1200 rpm에서 3분동안 원심분리하였다. 원심 분리한 상층액을 Griess 시약 (1% sulfanilamide, 0.1% N-(1-Naphthyl)ethylenediamine dihydrochloride, 5% phosphoric acid) 와 1:1로 섞은 후 차광하여 10분 반응시킨다. 550 nm 파장으로 Nitric Oxide의 농도를 확인하였다.
그 결과를 하기 표 4에 나타내었다.

본 발명의 화합물은 염증반응의 최종 산물인 Nitric Oxide의 발생을 억제하여, 본 발명의 화합물은 p38 억제를 통해 염증반응을 억제하는 것을 확인하였다.
