Abstract
본 발명은 5-아미노레불린산 수화염화물(5-aminolevulinic acid hydrochloride, ALA)과 그 유도체들을 유효성분으로 포함하는 여드름성 피부염 개선 또는 예방용 조성물에 관한 것으로 여드름 원인균에 대한 탁월한 항균효과를 나타낸다.
Description
본 발명은 5-아미노레불린산 수화염화물(5-aminolevulinic acid hydrochloride, ALA)과 그 유도체를 유효성분으로 포함하는 여드름성 피부염 개선 또는 예방용 조성물에 관한 것이다.
일반적으로 염증반응은 생체의 세포나 조직에 어떠한 기질적 변화를 가져오는 자극이 가해질 때 생체를 보호하기 위한 방어 시스템이다. 정상적인 염증반응은 생체 방어기작으로 작용한다. 그러나 염증반응이 종결되지 않고 지속으로 진행되면 질병을 일으키게 된다.
염증성 질환의 대표적인 경우로 여드름을 들 수 있는데, 이러한 여드름은 피지의 과다 분비로 인한 것으로 알려져 있다. 피지는 피지선에서 분비되는 물질로서 정상적인 양만큼 분비되면 피부 외부의 유해 물질이 각질 세포 사이를 침투하는 것을 막아주는 기능을 하지만 사춘기, 월경 전후, 임신 초기나 피임약 복용, 스트레스 등의 환경하에서는 과다 분비되어 여드름의 원인균이 증식하기 용이한 상태가 되고 증식한 여드름균은 약해진 피부를 통해 화농성 염증을 유발하게 된다.
상기한 여드름은 증상에 따라 4단계로 나눌 수 있다. 1단계는 과다 분비된 피지가 피지선 주변의 모공에 쌓이면서, 면포를 형성하게 되는 면포성 여드름인데, 이 단계에서는 잘 관리해주면 염증으로의 발전을 억제할 수도 있다. 2단계인 구진성 여드름 단계에서는 여드름균이 번식되어 붉은 여드름이 나타나는데, 이때 적절한 관리가 되지 않을 경우 피부에 흉터가 생길 수 있다. 3단계인 화농성 여드름 단계에서는 염증 반응이 더 심해지면서 내부에 고름 주머니(농포)가 생기는데 치료 후에도 흉터가 남게 될 수 있으므로 전문적인 치료가 필요하다. 마지막 4단계 결절성 여드름은 고름 주머니가 피부 안에서 터져 여드름 환부 주변에 더 많은 염증을 유발시키기 때문에 보다 전문적인 치료를 요하며 치료 시기도 장기화 될 수 있다.
일반적인 여드름 치료는 국소 제제 도포 요법 및 경구 약물 복용 요법과 최근 시행되고 있는 레이저 시술, 박피술 등 수술 요법 등이 있다. 국소 제제 도포 요법은 여드름의 원인균으로 알려진 프로피오니박테리움 아크네스(propionibacterium acnes)에 대한 항균 작용 목적으로 아젤라익 애씨드(azelaic acid), 살리실산 등의 항균제를 사용하거나 면포 용해 작용을 위해 벤조일퍼록사이드(benzoyl peroxide) 등을 외용 연고 타입으로 사용한다.
이러한 연고 타입으로로 가장 많이 상용화되고 있는 것은 스테로이드 제제이다. 그러나 스테로이드 제제는 장기적 사용 시 부신억제, 체액의 저류와 같은 부작용을 수반하게 되는 경우가 많다. 따라서 안전하게 섭취할 수 있으며, 염증을 억제할 수 있는 물질 및 천연물에 대한 연구가 필요한 실정이다.
이에 본 발명자는 부작용이 적으며 염증 억제 효능이 우수한 물질 및 천연물을 탐색하던 중, 5-아미노레불린산 염화수화물(5-aminolevulinic acid hydrochloride)과 고본 및 고삼을 혼합한 조성물이 여드름 원인균인 P. acnes(Propionibacterium acnes)에 대해 탁월한 항균 활성을 보였으며, tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), interleukin-1β (IL-1β)와 같은 pro-inflammatory cytokine의 발현을 억제할 뿐만 아니라, inducible nitric oxide synthase (iNOS)와 cyclooxygenase-2 (COX-2)의 발현을 억제함으로써 염증 인자인 nitric oxide (NO)및 prostaglandin E2 (PGE2)의 생성을 억제하는 효능을 확인하여 본 발명을 완성하였다.
아미노레불린산을 포함한 약학 조성물에 관한 선행문헌을 살펴보면, 공개특허공보 10-2014-0056370의 아미노레불린산 염산염용 경피 치료 시스템을 참고할 수 있다. 상기 선행문헌은 활성 성분에 불침투성인 지지층, 활성 성분-함류 중합체 매트릭스 및 제거가능한 보호층을 포함하되, 상기 활성성분은 5-아미노레불린산 염산염이고, 상기 중합체 매트릭스의 기초 중합체는 접착성 폴리아크릴레이트인, 경피 치료 시스템에 관한 것이다. 그러므로 본원발명과는 용도 및 효과가 상이하다.
천연물을 포함한 여드름 개선용 조성물에 관한 선행문헌을 살펴보면, 등록특허공보 10-0993676의 여드름 예방 또는 치료용 조성물을 참고할 수 있다. 상기 선행문헌은 석류풀, 개면마, 상산 및 이의 혼합물로 이루어진 군으로부터 선택된 추출물을 함유하는 여드름 예방 또는 치료용 조성물에 관한 것이다. 그러므로 본원발명과는 구성 및 효과가 상이하다.
본 발명은, 5-아미노레불린산 염화수화물(5-aminolevulinic acid hydrochloride)과 그 유도체를 유효성분으로 포함하는 여드름 치료용 조성물 및 염증 개선 또는 예방용 조성물을 제공하는 것을 해결하고자 하는 과제로 삼는다.
상기 과제를 해결하기 위하여, 본 발명은 5-아미노레불린산 수화염화물 (5-aminolevulinic acid hydrochloride)과 그 유도체들을 유효성분으로 함유하는 여드름성 피부염 개선 또는 예방용 조성물을 과제 해결을 위한 수단으로 제공한다.
또한, 본 발명의 예방용 조성물은, 5-아미노레불린산 수화염화물 (5-aminolevulinic acid hydrochloride)과 그 유도체들은 NO(Nitrite oxide)와 PGE2 (prostaglandin E2)생성을 억제하고, TNF-α, IL-1β 그리고 IL-6의 발현을 억제하며, iNOS 및 COX-2 단백질 발현이 억제함으로써 여드름성 피부염증 발생을 억제되는 특징이 있다.
또한, 본 발명의 예방용 조성물은 5-아미노레불린산 수화염화물 (5-aminolevulinic acid hydrochloride)의 유도체, 고본 및 고삼을 1 : 0.5 : 0.5의 중량비로 포함할 수도 있다.
본 발명에서는 5-아미노레불린산 염화수화물(5-aminolevulinic acid hydrochloride)의 약자를 ALA로, 5-Aminolevulinic acid hexyl ester hydrochloride의 약자를 HAL로, Methyl-aminolevulinic acid의 약자를 MAL로 표기하였다.
하기 화학식 1로 표시되는 5-아미노레불린산 염화수화물은 C-5의 지방족 화합물로서 각종 유기체의 tetrapyrrole 생합성(prophyrin, chlorophyll, heme, vitamin B12 등) 과정에서 발견되는 중간물질의 하나이다.
본 발명의 여드름 치료용 조성물 및 염증 개선 또는 예방용 조성물은 여드름균인 P. acnes(Propionibacterium acnes) 에 대한 항균 활성이 탁월하다.
뿐만 아니라 LPS와 P. acnes lysates에 의해 염증 유발된 대식세포에서 NO와 PGE2 생성을 억제하고, 사이토카인의 생성 및 iNOS 및 COX-2 단백질 발현이 억제된다.
또한, 본 발명은 RAW 264.7 대식세포 2.0 X 105 cells/㎖ 기준으로 25~100 μg/ml 함유하여 LPS에 의해 염증 유발된 대식세포에서 NO와 PGE2 생성을 강하게 억제할 수 있고, 염증관련 사이토카인 TNF-α, IL-1β, 그리고 IL-6의 생성을 억제하며, 관련 단백질인 iNOS 및 COX-2 단백질 발현이 억제되는 효과를 나타낸다.

본 명세서 및 청구 범위에 사용된 용어나 단어는 통상적이거나 사전적인 의미로 한정해서 해석되어서는 아니되며, 발명자는 그 자신의 발명을 가장 최선의 방법으로 설명하기 위해 용어의 개념을 적절하게 정의할 수 있다는 원칙에 입각하여 본 발명의 기술적 사상에 부합하는 의미와 개념으로 해석되어야만 한다.
따라서 본 명세서에 기재된 실시예, 참조예 및 도면에 기술된 사항은 본 발명의 가장 바람직한 일 예에 불과할 뿐이고 본 발명의 기술적 사상을 모두 대변하는 것은 아니므로, 본 출원 시점에 있어서 이들을 대체할 수 있는 다양한 균등물과 변형예들이 있을 수 있음을 이해하여야 한다.
5-ALA는 포피린(porphyrin) 중간물질 및 비타민의 제조에서 전구물질(intermediates)로 사용되는 환경 친화적이면서 부작용이 적은 생체물질이다. 특히, 5-ALA는 자외선 각화증, 건선, 사마귀 및 피부암과 같은 피부 질환에 사용된다. 이중, 특히 5-ALA는 세포 내에서 포피린의 생합성 과정을 통해 프로토포피린 PpIX(Protoporphyrin IX)라는 광민감제로 전환된다. 이때 외부로부터 5-ALA가 많이 투여되면 인체 내 과량의 heme이 생성되어 PpIX을 heme으로 전환시키는 효소를 저해하여 세포에 PpIX가 많이 축척된다. 이렇게 생산된 PpIX는 특정 파장의 빛과 에너지에 의해 주위의 산소와 반응하여 산소를 생산하여 상해된 세포의 사멸을 유발하여 질병을 치료할 수 있다.
그러나 5-ALA는 친수성이 강해 세포 내로 효과적으로 침투하기 곤란한다. 따라서, 이를 해결하는 방법으로 5-ALA를 메틸 에스터(methyl ester) 및 헥실 에스터(hexyl ester) 유도체를 제조하여 세포 침투성을 증가시켜 사용하였다.
한편, 고본(Ligusticum tenuissimum)은 쌍떡잎식물의 산형화목의 미나리과의 여러해살이풀로 주로 한국의 경남, 경북 지역에 분포되어 있다. 크기는 30~80 cm이며 8~9월에 흰색 꽃을 피운다. 한방에서는 가을에 뿌리를 캐서 말린 것을 두통, 관절통, 치통, 복통 또는 습진 등에 처방한다.
고삼(Sophora flavescens)은 쌍떡잎식물의 장미목 콩과의 여러해살이풀로 주로 한국, 일본, 중국과 같은 동북아시아 지역에 분포되어 있다. 크기는 80~110 cm이며 6~7월에 연한 노란색 꽃을 피운다. 한방에서는 뿌리를 말린 것을 소화불량, 신경통, 간염 또는 치질 등에 처방한다.
따라서, 본 발명은 여드름성 피부염 개선을 위한 염증 억제 효능을 향상시키기 위해 ALA, HAL 및 MAL에서 선택되는 화합물에 고본 및 고삼을 첨가하여 사용하였다.
실시예 1. 고본 분말 제조
경상북도 영양군에서 구입한 고본 100g을 생약분쇄기로 분말화한 다음, 70% 에탄올 1L를 첨가하여 48시간 침지시켜 고본 추출물을 추출하였다. 그런 다음, 거즈를 이용하여 찌거기를 걸러준 뒤 원심분리기를 이용하여 4℃에서 2500rpm으로 20분간 2번 원심분리하여 맑은 상층액을 분리하였다. 분리된 상층액의 순도를 높이기 위해 20~25um pore size를 가지는 정량여과지 (Whatman no.41)로 여과하였다. 여과액을 감압농축기를 이용하여 농축시킨 후, 동결건조기로 -70℃에서 동결건조하여 고본 분말을 획득하였다.
실시예 2. 고삼 분말 제조
경상북도 산천군에서 구입한 고삼 100g을 상기 실시예1과 동일한 방법으로 실시하여 고삼 분말을 획득하였다.
실시예 3. ALA, 고본 및 고삼 혼합조성물의 제조
ALA 5g, 고본 분말 2.5g 및 고삼 분말 2.5g을 배합하여 ALA, 고본 및 고삼 혼합조성물을 제조하였다.
실시예 4. HAL, 고본 및 고삼 혼합조성물의 제조

실시예 5. MAL, 고본 및 고삼 혼합조성물의 제조
MAL 5g, 고본 분말 2.5g 및 고삼 분말 2.5g을 배합하여 MAL, 고본 및 고삼 혼합조성물을 제조하였다.
실험예 1. 여드름 균에 대한 항균활성
실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 여드름 균에 대한 항균효능을 실험하기 위하여 P. acnes에 대해 액체배지희석법을 사용하여 최소억제농도 (Minimum Inhibitory Concentrations, MICs) 및 최소살균농도 (Minimum Bactericidal Concentrations, MBCs)를 확인하는 실험을 실행하였다.
액체배지희석법은 24 well plat에 각 조성물을 농도별로 처리한 후 배양된 균을 106~107 CFU/mL 농도로 처리한 후 혐기성 배양기에서 24~48시간 내에 육안과 흡광도를 통해 균의 억제정도를 확인하여 최소억제농도를 입증하며, 이후의 최소사멸농도를 확인하기 위해 최소억제농도에서부터 그 위 농도를 한천배지에 접종하여 혐기성 배양기에서 1-2일간 배양 후 집락을 계수하였다.
그 결과, 실시예 3의 ALA, 고본 및 고삼 혼합조성물의 0.1% 농도에서 P. acnes가 20% 이상 사멸하였고, 0.2% 농도에서는 P. acnes가 70% 이상이 사멸하였으며, HAL, 고본 및 고삼 혼합조성물의 0.05% 농도에서 P. acnes가 65% 이상 사멸하였고, MAL, 고본 및 고삼 혼합조성물의 0.2% 농도에서 P. acnes가 30% 이상 사멸하였다.

실험예 2. 세포생존율
실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물이 세포의 생존률에 미치는 영향을 측정하기 위하여 MTT 분석을 수행 하였다.
본 발명에서는 96 well 기준으로 RAW 264.7 대식세포 2.0 *?*5 cells/㎖에 분주배양 하였다. 분주 배양하고, 24시간 뒤에 LPS (0.5 μg/mL)와 P. acnes lysates (1 μg/mL)와 각 조성물을 농도별로 처리한 후 24시간 배양하였다. 배양된 세포를 MTT용액을 첨가하고 다시 3-4시간 배양한 후 formazan 생성물은 DMSO로 용해했다. Formazan 용해액을 96-well plate에 loading한 후, spectrophotometer를 이용하여 540 nm에 흡수되는 양을 측정하였다. 세포의 생존율은 어떠한 처리도 가하지 않은 naive cell (나이브 만능성 줄기세포)과의 비율로 나타내었다.
도 1은 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 세포독성에 대한 영향을 실험한 결과를 나타낸 도면이다.
그 결과, 실시예 3의 ALA, 고본 및 고삼 혼합조성물은 400 μg/mL농도까지도 독성을 나타내지 않았으며, 실시예 4의 HAL. 고본 및 고삼 혼합조성물은 50 μg/mL농도까지 독성을 나타내지 않았고, 실시예 5의 MAL, 고본 및 고삼 혼합조성물은 400 μg/mL농도까지 독성을 나타내지 않는 것을 확인하였다.
따라서, 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 0~100 μg/mL농도에서는 대식세포 생존률에 영향이 미치지 않음으로 독성이 없음을 알 수 있다.
실험예 3. NO와 PGE
2
생성억제
실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물이 염증성 프로세서인 iNOS에 의해 생합성되는 NO를 측정하기 위하여 RAW 264.7 macrophage cell에 LPS (0.5 μg/mL)와 P. acnes lysates (1 μg/mL)로 자극한 뒤 조성물을 농도에 따라서 NO의 생성을 조사하였다.
LPS와 P. acnes lysates에 의해 유도된 NO 생성 억제에 대한 각 조성물의 영향을 측정하기 위하여 ELISA 분석을 수행하였다. RAW 264.7 세포를 10% FBS가 첨가된 DMEM 배지를 사용하여 2.0 *?*5 cells/ml로 조절한 후 24 well plate에 분주 배양하고, LPS 또는 P. acnes lysates를 처리한 후 각 조성물을 농도별로 처리하여 24시간 배양하였다. 배양액에서 NO를 측정하기 위해 Griess 시약[1% (w/v) sulfanilamide, 0.1% (w/v) naphylethyleneduamine in 2.5% (v/v) phosphoric acid]를 사용하여 배양액에 녹아있는 NO2 -의 형태로 측정하였다. 세포배양액 100 uL와 Griess시약 100 uL를 혼합하여 96 well plate에서 20분간 반응시킨 후 540nm에서 흡광도를 측정하였다.
실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물이 염증성 프로세서인 COX-2에 의해 생합성되는 PGE2를 측정하기 위하여 RAW 264.7 macrophage cell에 LPS 또는 P. acnes lysates로 자극한 뒤 조성물을 농도에 따라서 PGE2의 생성을 조사하였다.
RAW 264.7 세포를 DMEM 배지를 이용하여 1.5*?*5 cells/mL로 조절한 후 24 well plate에 접종하고, 5% CO2 항온기에서 18시간 전 배양하였다. 이후 배지를 제거하고 각 조성물을 농도별로 1시간 동안 처리한 후, 그람-음성 박테리아 내독소인 LPS 또는 P. acnes lysates로 RAW 264.7 대식세포를 자극한 후 세포부유액을 원심분리하여 세포들을 침전시켜 상층액을 수집하고, 상층액내 PGE2 생성량을 EIA kit (R&D Systems Inc., Minneapolis,MN, USA)를 이용하여 사용자 매뉴얼에 기재된 방법대로 정량해 분석하였다.
도 2는 LPS에 의해 유발된 NO와 PGE2 생성에 대한 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 억제 효능을 나타낸 도면이다.
그 결과, 대식세포에 LPS를 처리하여 염증을 유발시킨 후 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물을 농도별로 처리한 결과, NO와 PGE2의 생성이 조성물의 처리 농도에 의존적으로 감소하는 것을 확인하였다.
도 6은 P. acnes lysates 처리된 대식세포에서 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 NO 및 PGE2 억제 효능을 나타낸 도면이다.
그 결과, 대식세포에 P. acnes lysates를 처리하여 염증을 유발 시킨 후 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물을 농도별로 처리한 결과, NO와 PGE2의 생성이 농도의존적으로 감소하는 것을 확인하였다.
실험예 4. TNF-α, IL-1β 및 IL-6 발현억제
LPS 또는 P. acnes lysates로 유도한 대식세포에서 염증 발생 시, 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물이 전염증성 사이토카인의 생성에 미치는 영향을 조사하기 위하여 ELISA 방법과 Mouse ELISA kit (BD pharmingen)를 이용해 전염증성 사이토카인의 단백질 수준을 측정하였다.
Cell culture시 나온 상등액을 사용하며 TNF-α, IL-1β 및 IL-6 검출시약을 이용하여 측정하였다. 각 검출시약의 capture antibody를 96 well 플레이트에 50 μl씩 넣은 후 다음날까지 4℃에 보관하였다. 각 well을 wash buffer (1x PBS + 0.05% tween-20)을 이용하여 세척 후 assay buffer (100μL) 처리하고 인큐베이터에 1시간 보관하였다. 이후에 다시 세척세포의 상등액과 standard solution (50μL/well)를 플레이트에 넣은 뒤 인큐베이터에 2시간 동안 보관 후 세척하였다. 그 후 antibody 와 enzyme ragent를 섞은 후 50 μL/well 인큐베이터에 1시간 보관하고 마지막 세척을 하였다. 마지막으로 TMB substrate reagen tset 50 μl/well 넣고, 어두운 곳에 30분 동안 차광한 뒤 stop buffer (0.5M H2SO4)을 50 μl/well 주입하고, 플레이트를 450 nm파장에서 측정하였다.
도 3은 LPS에 의해 유발된 TNF-α 생성에 대한 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 억제 효능을 나타낸 도면이다.
도 4는 LPS에 의해 유발된 IL-1β 생성에 대한 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 억제 효능을 나타낸 도면이다.
도 5는 LPS에 의해 유발된 IL-6 생성에 대한 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 억제 효능을 나타낸 도면이다.
대식세포에 LPS를 처리하여 염증을 유발시킨 후 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물을 농도별로 처리한 결과, 사이토카인인 TNF-α, IL-1β 및 IL-6의 생성이 농도의존적으로 감소하는 것을 확인하였으며, 25~200 μ g/mL 의 농도에서 감소하는 것을 확인하였다.
도 7은 P. acnes lysates 처리된 대식세포에서 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 TNF-α, IL-1β 및 IL-6 억제 효능을 나타낸 도면이다.
대식세포에 P. acnes lysates를 처리하여 염증을 유발시킨 후 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물을 농도별로 처리한 결과, 사이토카인인 TNF-α, IL-1β 및 IL-6의 생성이 농도의존적으로 감소하는 것을 확인하였으며, 25~200 μg/mL 의 농도에서 감소하는 것을 확인하였다.
실험예 5. iNOS 및 COX-2 발현억제
실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 염증 예방을 확인하기 위하여 P. acnes lysates 자극에 의해 유도되는 iNOS와 COX-2 효소 단백질의 발현에 미치는 영향을 측정하였다.
100 mm dish에 2 X 106 cells/well 배양하고 시료를 농도별로 처리하였다. 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물을 농도별로 처리하여 1시간 전처리한 후 P. acnes lysates (1 μg/mL)를 사용하여 염증을 유발시킨다. 각 처리군 별로 cell을 모은 뒤 원심분리기를 이용하여 세포만 모으고, lysis buffer (50 mM Tris-HCl(PH 7.4),150 mM NaCl,5 mM EDTA, 0.1% TritonX-100, protease 및 phosphatase inhibitor cocktail)를 이용하여 단백질만 취하였다. 단백질은 BCA protein assay를 이용하여 측정하였다.
각 샘플의 단백질양을 20 μg로 정하였고 그 뒤, 10% sodium dodecylsulphate (SDS) polyacrylamide gels을 이용하여 전기연동기기를 통하여 내린 뒤 poly vibylidene fluoride membrane (PVDF; Biorad)에 electrophoretically transfer 기기를 사용하여 단백질을 이전시켰다. 그런 다음, membrane에 5% skim milk 용액을 사용하여 블로킹시키고 antibodies against COX-2와 iNOS (Cell Signaling Technology, USA)의 각각 보고자 하는 항체를 사용하였다.
1차 항체를 처리하여 4℃에서 overnight 한 후 TBST (TBS에 0.05 % tween-20가 들어있는 용액)를 사용하여 세척하였다. Horseradish peroxidase-conjugated의 2차 항체에 1시간 붙인 뒤, 다시 TBST를 이용하여 세척하였다. 마지막으로 chemiluminescence방식을 이용하여 단백질 밴드를 확인하였다.
염증 관련 인자인 PGE2와 NO 생성에 관여하는 COX-2와 iNOS의 효소를 확인하기 위하여 이들 효소의 세포 내 발현을 측정하였다. RAW 264.7 macrophage cell에 조성물을 농도별로 1시간 전처리한 후 P. acnes lysates를 처리한 후, 세포 내 단백질을 분리를 하여 웨스턴 블롯 분석 방법을 통하여 이들 단백질의 발현을 확인하였다. 이들 효소 단백질 발현은 densitometry assay를 통하여 그래프로 정량화하였다.
도 8은 P. acnes lysates 처리된 대신세포에서 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 iNOS 단백질 발현 억제 효능을 나타낸 도면이다.
도 9는 P. acnes lysates 처리된 대식세포에서 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 COX-2 단백질 발현 억제 효능을 나타낸 도면이다.
Western blot을 통한 단백질 발현 변화를 확인한 결과, P. acnes lysates를 처리하였을 때 iNOS와 COX-2 단백질 발현이 증가하는 것을 확인할 수 있었으며, 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물을 농도별로 처리시 농도의존적으로 단백질 발현이 현저히 감소하였으며, 25~200 μg/ml의 농도에서 감소한 것을 확인할 수 있다.
대식세포는 외부 자극에 의한 염증 반응에서 TNF-α, IL-6 및 IL-1β와 같은 전염증성 사이토카인의 발현이 유도되고, iNOS와 COX-2에 영향을 받는 유전자의 발현을 자극하여 NO 및 PGE2의 염증 인자를 생성한다.
일반적으로 iNOS에 의해 생성된 NO는 박테리아를 죽이거나 종양을 제거하고, 혈압을 조절하거나 신경 전달을 매개하는 등 다양한 역할을 한다. 그러나 과도하게 생성된 NO는 염증을 유발시켜 조직의 손상, 유전자 변이, 신경 손상 등을 유발하고, 혈관 투과성을 증가시켜 부종 등의 염증반응을 촉진시킬 뿐만 아니라 염증 매개체의 생성을 촉진하여 염증을 악화한다. 또 다른 염증인자인 PGE2는 통증과 발열에 주로 관여하는 염증 매개체로서 염증반응과 면역반응에 관여하며, 혈관신생 (angiogene sis)을 촉진하는 등 암 발생에도 관여하고 있다. 이러한 역할을 가진 PGE2는 염증반응이 시작되면서 생성되는 COX-2에 의해 발현된다.
따라서, 본 발명에서 상기 실험예 1 내지 4를 통하여 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물의 염증 억제 효능을 확인할 수 있다.
실험예 3와 5에서 확인한 바와 같이 inducible nitric oxide synthase (iNOS)와 cyclooxygenase-2 (COX-2)의 발현을 억제함으로써 염증 인자인 nitric oxide (NO)및 prostaglandin E2 (PGE2) 의 생성을 억제한다.
또한, 대식세포는 외부 자극에 의한 염증 반응에서 발현된 tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), interleukin-1β (IL-1β)와 같은 pro-inflammatory cytokine의 발현을 억제하는 효능을 보유함으로써, 염증을 예방 또는 개선하는 조성물이다.
특히, 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물는 25~100 μg/mL 에서 독성이 없고, Tumor necrosis factor-α(TNF-α), interleukin-6(IL-6), interleukin-1β(IL-1β)와 같은 pro-inflammatory cytokine의 발현을 억제 효능이 실시예 4의 HAL, 고본 및 고삼 혼합조성물 및 실시예 5의 MAL, 고본 및 고삼 혼합조성물에 비하여 낮은 농도에서 억제율이 높은 효능을 보유하고 있다.
또한, inducible nitric oxide synthase (iNOS)와 cyclooxygenase-2 (COX-2)의 발현을 억제하는 효능도 실시예 4의 HAL, 고본 및 고삼 혼합조성물 및 실시예 5의 MAL, 고본 및 고삼 혼합조성물에 비하여 낮은 농도에서 억제율이 높은 효능을 보유하고 있으므로, 실시예 3의 ALA, 고본 및 고삼 혼합조성물는 실시예 4의 HAL, 고본 및 고삼혼합조성물 및 실시예 5의 MAL, 고본 및 고삼 혼합조성물에 대비하여 염증 예방 또는 개선 효능이 더 뛰어난 것으로 판단된다.
한편, 본 발명은 화장품 조성물로 이용되기 위하여, 용도에 맞게 본 발명에 따른 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물을 적용하여 제공할 수 있다. 그 자체 또는 화장품으로 제조되는 크림, 에센스, 로션, 스킨 제형으로 제조될 수 있으며, 화장품에서 사용되는 유화제, 담체, 부형제, 희석제와 혼합하여 제조되어 사용될 수 있다. 이 경우에 각 제형을 제조하기 위하여 첨가제가 추가될 수 있음은 물론이다.
상기 실시예와 실험예의 결과에 의하여 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물은 RAW 264.7 대식세포 2.0 X 105 cells/㎖ 기준으로 25~100 μg/ml 함유하는 것을 특징으로 하는 화장품 조성물을 제조할 수 있다.
상기와 같은 본 발명에 따른 실시예 3 내지 5에 따른 ALA, 고본 및 고삼 혼합조성물, HAL, 고본 및 고삼 혼합조성물 및 MAL, 고본 및 고삼 혼합조성물을 이용한 화장료 조성물은 염증 예방 또는 개선용 화장료 조성물로서, 다양하게 이용될 수 있다.
Claims (3)
- 5-아미노레불린산 헥실 에스터 하이드로클로라이드(5-Aminolevulinic acid hexyl ester hydrochloride), 고본 및 고삼을 유효성분으로 함유하는 여드름성 피부염 개선 또는 예방용 화장료 조성물에 있어서,
5-아미노레불린산 헥실 에스터 하이드로클로라이드, 고본 및 고삼은 1 : 0.5 : 0.5의 중량비로 혼합되며,
5-아미노레불린산 헥실 에스터 하이드로클로라이드, 고본 및 고삼 혼합물은 RAW 264.7 대식세포 2.0 X 105 cells/㎖ 기준으로 25~100 μg/ml 함유하고,
상기 여드름성 피부염은 프로피오니박테리움 아크네스(propionibacterium acnes)에 의해 유발되는 것을 특징으로 하는 여드름성 피부염 개선 또는 예방용 화장료 조성물.
