Abstract
본 발명은 소포카르핀, 탄제리틴, 우스닌산, 이들의 약학적으로 허용되는 염 또는 이들의 조합을 유효성분으로 포함하는 파골세포 분화 억제용 조성물, 골 관련 질환 예방 또는 치료용 조성물 및 상기 조성물을 이용하여 파골세포로의 분화를 억제하는 방법에 관한 것이다. 본 발명의 약학 조성물은 파골세포의 분화를 억제할 수 있는 화합물을 유효성분으로 포함하므로, 파골세포의 과도한 분화로 인하여 유발되는 각종 질환의 예방 또는 치료에 널리 활용될 수 있을 것이다.
Description
본 발명은 골 관련 질환 예방 또는 치료용 약학조성물에 관한 것으로, 보다 구체적으로 본 발명은 소포카르핀, 탄제리틴, 우스닌산, 이들의 약학적으로 허용되는 염 또는 이들의 조합을 유효성분으로 포함하는 파골세포 분화 억제용 조성물, 골 관련 질환 예방 또는 치료용 조성물 및 상기 조성물을 이용하여 파골세포로의 분화를 억제하는 방법에 관한 것이다.
골의 재형성은 기계적 스트레스와 호르몬 조절에 반응하여 골을 유지하도록 살아가면서 지속적으로 발생한다. 통상적인 골량을 유지하기 위하여, 밀접하게 결합된 과정을 통해 골은 파골세포에 의하여 재흡수되고 새로운 골기질이 조골세포에 의해 축적된다. 많은 일반적인 골 질환에서 보듯이, 이러한 과정에서의 불균형은 파골세포에 의한 과다한 골 재흡수 및 골의 연약성의 증가를 유도할 수 있다.
파골세포는 그의 주위에 존재하는 T-cell, fibroblast, osteoblast 등에서 분비 되는 호르몬에 의해 조혈모전구체 또는 골수성전구체로부터 분화되는 것으로 알려져 있다. 파골세포의 분화과정에 있어서, RANKL에 의한 신호전달과정이 중요한 역할을 수행하는 것으로 알려져 있다. RANKL은 조골세포, 활성화된 T세포 및 기질세포에서 생성되고, M-CSF(macrophage colony-stimulating factor)와 함께 파골세포의 형태변화, 생존 및 골 흡수를 활성화 시킨다. RANKL이 수용체인 RANK에 결합되면, RANK의 세포질 도메인으로 TRAF6(TNF receptor-associated factor 6)가 모여들고, 이에 따라 NF-κB와 JNK(c-Jun N-terminal kinase), p38(p38 MAP kinase), 및 ERK(extracellular signal-regulated kinase)를 포함하는 MAPK(mitogen-activated protein kinase) 신호전달 경로가 활성화된다. 활성화된 NF-κB는 하류에 있는 c-Fos와 NFATc1의 발현을 유도하는데, 이들은 RANKL에 의한 파골세포 분화에서 핵심적인 역할을 수행하는 것으로 알려져 있다. 아울러, 이러한 파골세포의 분화과정은 IL-1, Il-3, PGE 등의 염증성 사이토카인에 의해 촉진되는 것으로 알려져 있다.
최종적으로 분화된 파골세포는 다핵화 세포(multi-nulceated)의 형태를 띄고, 뼈를 흡수할 부위에 결합하여 액틴(actin), 빈컬틴(vinculin) 등을 이용하여 실링존(sealing zone)을 형성하여 주위와 밀폐하게 된다. 이후 이곳에 수소 이온을 펌핑하여 pH를 낮추어 하이드록시아파타이드 성분을 녹이게 되고 뼈의 10% 정도를 차지하는 콜라겐 성분을 분해하기 위해서 MMP-9 혹은 카뎁신 K와 같은 단백질분해효소를 분비하게 된다. 이같이 최종 분화된 파골세포는 타르타르산이 있는 조건에서 활성을 보이는 phosphatase(tartrate-resistant acid phosphatase: TRACP)를 특이적으로 발현하는 것으로 알려져 있으므로, TRACP의 활성을 측정함으로써 파골 세포분화를 확인할 수 있다.
상술한 바와 같은, 파골세포는 생체내의 골대사에 있어서 중요한 역할을 수행하지만, 다양한 원인에 의하여 과다하게 분화될 경우, 골조직의 장애를 초래하거나 또는 골조직을 파괴하는 질환이 유발될 수 있는데, 이러한 질환으로는 성장기 발육부진, 골절, 과도한 파골세포의 골 흡수에 의한 골다공증(osteoprosis), 류마티스성 관절염(rheumatoid arthritis), 치주질환(periodontal disease), 파제트병(Paget disease), 전이성 골암(metastatic bone cancers) 등을 들 수 있다.
이러한 배경하에서, 본 발명자들은 파골세포의 과도한 생성으로 인하여 유발되는 골다공증 등의 질환을 보다 효과적으로 치료할 수 있는 방법을 개발하기 위하여 예의 연구 노력한 결과, 소포카르핀, 탄제리틴, 우스닌산, 이들의 약학적으로 허용되는 염 또는 이들의 조합이 파골세포의 분화를 억제할 수 있어, 상기 화합물을 사용하면 파골세포의 과도한 분화에 의하여 유발되는 질환을 예방 또는 치료할 수 있음을 확인하고, 본 발명을 완성하였다.
본 발명의 하나의 목적은 소포카르핀, 탄제리틴, 우스닌산, 이들의 약학적으로 허용되는 염 또는 이들의 조합을 유효성분으로 포함하는 골 관련 질환 예방 또는 치료용 조성물을 제공하는 것이다.
본 발명의 다른 목적은 소포카르핀, 탄제리틴, 우스닌산, 이들의 약학적으로 허용되는 염 또는 이들의 조합을 유효성분으로 포함하는 파골세포 분화 억제용 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 조성물을 이용하여 파골세포의 분화를 억제하는 방법을 제공하는 것이다.
본 발명자들은 골관련 질환중에서 파골세포의 과도한 생성으로 인하여 유발되는 골다공증 등의 질환을 보다 효과적으로 치료할 수 있는 방법을 개발하기 위하여 다양한 연구를 수행하던 중, 파골세포의 분화를 억제할 수 있는 화합물을 개발하고자 다양한 화합물을 대상으로 하여 파골세포의 분화를 억제할 수 있는 화합물을 스크리닝하였다. 그 결과, 소포카르핀(sophocarpine), 탄제리틴(tangeritin) 및 우스닌산(usnic acid)이 파골세포 전구체 세포가 파골세포로 분화되는 것을 억제할 수 있음을 확인하였다. 아울러, 상기 화합물의 파골세포 분화억제 효과를 파골세포에서 특이적으로 발현되는 탈인산화효소인 TRACP(tartrate-resistant acid phosphatase)의 활성을 대상으로 확인한 결과, 상기 3종의 화합물 각각을 파골세포 전구체 세포에 처리하고, 파골세포로 분화시킬 경우, 상기 화합물의 처리농도가 증가할 수록 TRACP의 활성이 저하됨을 확인하였다.
아울러, 상기 화합물의 파골세포 분화억제 활성이 어떠한 세포내 작용기작에 관련된 것인지를 확인한 결과, 소포카르핀, 탄제리틴 및 우스닌산의 파골세포 분화억제 효과는 RANKL-RANK 복합체의 신호전달과정을 억제함에 의하여 유발됨을 알 수 있었다.
상술한 바와 같이, 상기 3종의 화합물은 파골세포의 분화를 억제할 수 있으므로, 파골세포의 과도한 분화에 의하여 유발되는 질환의 예방 또는 치료를 위하여 사용될 수 있다.
상기 목적을 달성하기 위한 일 실시양태로서, 본 발명은 소포카르핀, 탄제리틴, 우스닌산, 이들의 약학적으로 허용되는 염 또는 이들의 조합을 유효성분으로 포함하는 파골세포 분화 억제용 조성물 또는 골 관련 질환 예방 또는 치료용 약학 조성물을 제공한다.
본 발명의 용어 "소포카르핀(sophocarpine)" 이란, 고삼(Sophora flavescens)으로부터 분리된 하기 화학식 1의 구조와 약 246의 분자량을 갖는 알칼로이드 화합물을 의미하는데, DMSO, 물, 에탄올 등의 용매에 대하여 동등한 수준의 용해도(≥49 mg/mL)를 나타낸다. 생물학적으로는 HERG(human ether-a-go-go-related gene)에 대한 농도의존적인 억제효과를 나타내는 것으로 알려져 있다.
본 발명의 용어 "탄제리틴(tangeritin)" 이란, 다양한 감귤류 식물로부터 분리된 하기 화학식 2의 구조와 약 372의 분자량을 갖는 화합물을 의미한다. 생물학적으로는, 식물의 세포벽을 강화시키고, 병원균에 대한 식물의 방어기전에 관여하며, 파킨슨병에 대한 보호효과를 나타내는 것으로 알려져 있다.
본 발명의 용어 "우스닌산(usnic acid)" 이란, Lecanorales 과에 속하는 옆고리송라(Usnea diffracta) 등의 지의류로부터 분리된 하기 화학식 3의 구조와 약 344의 분자량을 갖는 다이벤조퓨란 유도체 화합물을 의미한다. 생물학적으로는, 항생제로서의 활성을 나타내고, 빛으로부터 지의류를 보호하는 활성을 나타내며, 쓴맛을 나타내어 섭식동물로부터 지의류를 보호하는 활성을 나타내는 것으로 알려져 있다.
본 발명의 용어 "약학적으로 허용되는 염"이란, 양이온과 음이온이 정전기적 인력에 의해 결합하고 있는 물질인 염 중에서도 약제학적으로 사용될 수 있는 형태의 염을 의미하는데, 통상적으로 금속염, 유기 염기와의 염, 무기산과의 염, 유기산과의 염, 염기성 또는 산성 아미노산과의 염 등이 될 수 있다. 예를 들어, 금속염으로는 알칼리 금속염(나트륨염, 칼륨염 등), 알칼리 토금속염(칼슘염, 마그네슘염, 바륨염 등), 알루미늄염 등이 될 수 있고; 유기 염기와의 염으로는 트리에틸아민, 피리딘, 피콜린, 2,6-루티딘, 에탄올아민, 디에탄올아민, 트리에탄올아민, 시클로헥실아민, 디시클로헥실아민, N,N-디벤질에틸렌디아민 등과의 염이 될 수 있으며; 무기산과의 염으로는 염산, 브롬화수소산, 질산, 황산, 인산 등과의 염이 될 수 있고; 유기산과의 염으로는 포름산, 아세트산, 트리플루오로아세트산, 프탈산, 푸마르산, 옥살산, 타르타르산, 말레인산, 시트르산, 숙신산, 메탄술폰산, 벤젠술폰산, p-톨루엔술폰산 등과의 염이 될 수 있으며; 염기성 아미노산과의 염으로는 아르기닌, 라이신, 오르니틴 등과의 염이 될 수 있고; 산성 아미노산과의 염으로는 아스파르트산, 글루탐산 등과의 염이 될 수 있다.
본 발명의 용어 "파골세포(osteoclast)"란, 파골세포 전구체(osteoclast precursor)로부터 분화되는 세포로서 파골세포 전구세포들은 M-CSF 및 RANKL 존재 하에서 파골세포로 분화되며, 융합을 통해 다핵 파골세포(multinucleated osteoclast)를 형성한다. 파골세포는 αvβ3 인테그린(integrin) 등을 통해 골(bone)에 결합하며 산성 환경을 조성하는 한편 각종 콜라게네이즈(collagenase) 및 프로테아제(protease)를 분비하여 골 흡수(bone resorption)를 일으킨다. 파골세포는 완전히 분화된 세포로 증식하지 않으며 약 2주간의 수명이 다하면 세포 사멸(apotosis)를 일으킨다.
본 발명의 용어 "골 관련 질환"이란, 조골세포 및 파골세포의 불균형으로 인해 유발되어 골조직의 장애를 초래하거나 또는 골조직을 파괴하는 질환을 의미하는데, 상기 골 관련 질환은 특별히 이에 제한되지 않으나, 성장기 발육부진, 골절, 과도한 파골세포의 골 흡수에 의한 골다공증(osteoprosis), 류마티스성 관절염(rheumatoid arthritis), 치주질환(periodontal disease), 파제트병(Paget disease), 전이성 골암(metastatic bone cancers) 등이 될 수 있다.
본 발명의 용어 "골다공증"이란, 남아 있는 뼈에는 구조상으로 아무런 이상이 없으면서 뼈를 형성하는 무기질과 기질의 양이 과도하게 감소되어 뼈에 스펀지 처럼 작은 구멍이 많이 나서 무르고 쉽게 부러지는 증상을 나타내는 질환을 의미하는데, 골조소증 또는 골소공증이라고도 한다.
본 발명의 용어 "골 관련 질환의 치료"란 상기 골 관련 질환의 발병에 의한 증세를 호전하거나 이롭게 변경하는 모든 행위를 의미하는데, 골 질환 증상의 감소, 개선, 그 증상의 고통 경감, 골 질환 발생율 감소 또는 치료결과를 증가시키는 환자의 어떠한 다른 변화를 모두 포함한다.
본 발명의 일 실시예에 따르면, 파골세포 전구체 세포를 파골세포로 분화시키기 전에 소포카르핀(sophocarpine), 탄제리틴(tangeritin) 및 우스닌산(usnic acid)을 처리하면, 파골세포의 분화가 억제됨을 세포형태(도 1) 및 세포 염색(도 2)을 통해 확인하였고, 상기 각 화합물을 서로 다른 농도로 처리한 결과 상기 화합물의 처리농도가 증가할 수록 파골세포의 분화억제 수준이 증가함을 확인하였다(도 3).
따라서, 소포카르핀, 탄제리틴 또는 우스닌산은 단독으로 또는 조합되어 파골세포의 과도한 분화에 의하여 유발되는 질환의 예방 또는 치료를 위하여 사용될 수 있다.
한편, 본 발명의 골 관련 질환 예방 또는 치료용 약학 조성물은 약학적 조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형제 또는 희석제를 추가로 포함할 수 있다. 구체적으로, 상기 약학 조성물은, 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 본 발명에서, 상기 약학 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 상기 적소두 추출물과 이의 분획물들에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트(calcium carbonate), 수크로스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 마그네슘 스티레이트, 탈크 같은 윤활제들도 사용된다. 경구를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는 데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 일 실시예에 따른 약학 조성물에 포함된 상기 소포카르핀, 탄제리틴, 우스닌산, 이들의 약학적으로 허용되는 염 또는 이들의 조합의 함량은 특별히 이에 제한되지 않으나, 최종 조성물 총중량을 기준으로 0.0001 내지 50 중량%, 보다 바람직하게는 0.01 내지 5 중량%의 함량으로 포함할 수 있다.
또한, 상기 약학 조성물에 소포카르핀, 탄제리틴 및 우스닌산 중 둘 이상의 조합을 유효성분으로 포함할 경우, 상기 조합에 포함된 소포카르핀 및 탄제리틴의 함량비는 바람직하게는 1 내지 100 : 1 내지 100(w/w)이 될 수 있고, 소포카르핀 및 우스닌산의 함량비는 바람직하게는 1 내지 100 : 1 내지 100(w/w)이 될 수 있으며, 탄제리틴 및 우스닌산의 함량비는 바람직하게는 1 내지 100 : 1 내지 100(w/w)이 될 수 있고, 소포카르핀, 탄제리틴 및 우스닌산의 함량비는 바람직하게는 1 내지 100 : 1 내지 100 : 1 내지 100(w/w/w)이 될 수 있다.
상기 본 발명의 약학 조성물은 약학적으로 유효한 양으로 투여될 수 있는데, 본 발명의 용어 "약제학적으로 유효한 양"이란 의학적 치료 또는 예방에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료 또는 예방하기에 충분한 양을 의미하며, 유효 용량 수준은 질환의 중증도, 약물의 활성, 환자의 연령, 체중, 건강, 성별, 환자의 약물에 대한 민감도, 사용된 본 발명 조성물의 투여 시간, 투여 경로 및 배출 비율 치료기간, 사용된 본 발명의 조성물과 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 약학 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다. 그리고 단일 또는 다중 투여될 수 있다. 상기 요소를 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하다.
본 발명의 약학조성물의 투여량은 예를 들어, 상기 소포카르핀, 탄제리틴, 우스닌산, 이들의 약학적으로 허용되는 염 또는 이들의 조합을 포함하는 본 발명의 약학 조성물을 사람을 포함하는 포유동물에 하루 동안 1 내지 20 ㎎/㎏, 보다 바람직하게는 1 내지 10 ㎎/㎏으로 투여할 수 있고, 본 발명의 조성물의 투여빈도는 특별히 이에 제한되지 않으나, 1일 1회 투여하거나 또는 용량을 분할하여 수회 투여할 수 있다.
상기 목적을 달성하기 위한 또 다른 실시양태로서, 본 발명은 상기 소포카르핀, 탄제리틴, 우스닌산, 이들의 약학적으로 허용되는 염 또는 이들의 조합을 유효성분으로 포함하는 약학 조성물을 약제학적으로 유효한 양으로 골 관련 질환이 발병될 가능성이 있거나 또는 발병된 개체에 투여하는 단계를 포함하는 파골세포 분화를 억제하는 방법 또는 골 관련 질환을 예방 또는 치료하는 방법을 제공한다.
상술한 바와 같이, 본 발명에서 제공하는 소포카르핀, 탄제리틴, 우스닌산, 이들의 약학적으로 허용되는 염 또는 이들의 조합은 파골세포 분화 억제용 조성물 또는 골 관련 질환 예방 또는 치료용 약학 조성물의 유효성분으로서 사용할 수 있으므로, 상기 조성물은 파골세포 분화를 억제하거나 또는 골 관련 질환을 예방 또는 치료하는데 사용될 수 있다.
본 발명에서 용어 "개체"란 골 관련 질환이 발병될 가능성이 있거나 또는 발병된 쥐, 가축, 인간 등을 포함하는 포유동물을 제한 없이 포함한다.
본 발명의 골 관련 질환을 치료하는 방법에 있어서, 상기 약학 조성물의 투여 경로는 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여도 투여될 수 있다. 본 발명의 약학 조성물은 특별히 이에 제한되지 않으나, 목적하는 바에 따라 복강내 투여, 정맥내 투여, 근육내 투여, 피하 투여, 피내 투여, 경구 투여, 비내 투여, 폐내 투여, 직장내 투여될 수 있다. 다만, 경구 투여시에는 위산에 의하여 상기 화합물이 변성될 수 있기 때문에 경구용 조성물은 활성 약제를 코팅하거나 위에서의 분해로부터 보호되도록 제형화 되어야 한다. 또한, 상기 조성물은 활성 물질이 표적 세포로 이동할 수 있는 임의의 장치에 의해 투여될 수 있다.
본 발명의 약학 조성물은 파골세포의 분화를 억제할 수 있는 화합물을 유효성분으로 포함하므로, 파골세포의 과도한 분화로 인하여 유발되는 각종 질환의 예방 또는 치료에 널리 활용될 수 있을 것이다.

도 3은 파골세포 분화 억제 화합물의 처리농도에 따른 파골세포 분화억제 효과를 나타내는 그래프이다.

이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예
1: 파골세포 분화 억제 화합물의 효과
파골세포 전구체 세포를 파골세포로 분화시키기 위해서 B-Bridge International 의 Primaty precursor osteoclast culture kit을 이용하였다. 구체적으로, 2 X 106개의 파골세포 전구체 세포를 10ml의 Culture Medium(M-CSF 10 ng/ml, RANKL 10 ng/ml)에 희석하고 96웰 플레이트에서 배양하였다. 상기 배양된 전구체 세포에 소포카르핀(sophocarpine), 탄제리틴(tangeritin) 및 우스닌산(usnic acid)을 20ppm의 농도로 각각 처리하고, 37℃ CO2 배양기에서 4일 동안 배양하였으며, 이후 배양액을 교체하고 다시 3일 동안 추가로 배양하여 파골세포를 분화시켰다. 이때, 대조군으로는 상기 화합물을 처리하지 않고 분화시킨 파골세포를 사용하였다.
배양이 종료된 후, TRACP 염색 kit(TAKARA)를 사용하여 파골세포를 염색하였다. 구체적으로, 배양된 파골세포에 고정용액(Citrate buffer (pH 5.4) , 60% acetone , 10% methanol)을 처리하여 고정시키고, 5분동안 실온에서 방치한 다음, 상기 세포를 증류수로 세척하였다. 이어, 상기 세포에 0.5 M sodium tartrate buffer (pH 5.2)이 10% 첨가된 NABP/FRVLB(Naphthol-AS-BI-phosphate/Fast Red Violet LB (diazonium salt) premix 용액을 처리하고, 37℃에서 45분동안 반응시켜서 상기 세포를 염색하고, 염색이 종료된 후 증류수로 세척하였으며, 이를 현미경으로 관찰하고, 염색된 세포의 수를 계수하였다(도 1 및 2).
도 1은 각 화합물(소포카르핀, 탄제리틴 및 우스닌산)이 처리된 파골세포를 나타내는 현미경사진이고, 도 2는 소포카르핀, 탄제리틴 및 우스닌산의 파골세포의 분화억제 효과를 나타내는 그래프이다. 도 1 및 2에서 보듯이, 대조군에 비하여 소포카르핀, 탄제리틴 및 우스닌산을 처리한 실험군에서는 분화된 파골세포가 존재하지 않음을 확인하였다.
따라서, 상기 소포카르핀, 탄제리틴 및 우스닌산은 파골세포의 분화를 억제할 수 있음을 알 수 있었다.
실시예
2: 파골세포 분화 억제 화합물의 처리농도에 따른 효과
상기 실시예 1에서 사용한 각 화합물의 처리농도에 따른 파골세포 분화억제 효과를 파골세포에서 특징적으로 나타나는 TRACP 활성을 이용하여 확인하였다.
구체적으로, 상기 실시예 1에서 사용한 각 화합물을 5 및 20ppm의 농도로 각각 처리하는 것을 제외하고는, 상기 실시예 1의 방법에 따라 배양 및 염색과정을 수행하였다. 이어, 염색된 세포를 분광광도계에 적용하여, 상기 세포에서 적색으로 염색된 정도를 측정하고, 이를 이용하여 TRACP 활성을 산출하였다(도 3).
도 3은 파골세포 분화 억제 화합물의 처리농도에 따른 파골세포 분화억제 효과를 나타내는 그래프이다. 도 3에서 보듯이, 3가지 화합물(소포카르핀, 탄제리틴 및 우스닌산)을 처리한 실험군에서는 모두 처리농도에 비례하여 파골세포의 분화억제 효과가 향상됨을 알 수 있었다.
실시예
3: 파골세포 분화 억제 화합물의 처리농도에 따른
RANK
수용체 억제효과
파골세포의 분화에 핵심적인 역할을 수행하는 RANKL-RANK 복합체의 신호전달과정은 NF-κB를 매개로 하여 수행되므로, 상기 파골세포 분화 억제 화합물이 RANKL-RANK 복합체의 신호전달과정에 어떠한 영향을 미치는지 확인하고자 하였다.
먼저, CHO 세포를 24웰 플레이트에 접종하고, 70~80% 정도의 포화도(confluent)가 될 때 까지 배양한 다음, RANK 수용체를 발현시키는 벡터(gencopoeia) 및 NF-κB-luciferase를 발현시키는 벡터(promega)를 도입하여 형질전환체를 제작하였다. 이때, 배지로는 10% FBS 및 1% 항생제를 포함하는 RPMI1640 배지를 사용하고, 배양조건은 37℃ 및 5% CO2로 하였으며, 도입된 각 벡터의 양은 각 웰당 250ng이었고, 도입수단으로는 lipofectamine/plus reagent (invitrogen)를 사용하였다.
상기 제작한 형질전환체에 10ng/㎖ RANKL과, 5 또는 20ppm의 화합물(소포카르핀, 탄제리틴 및 우스닌산)을 처리하고, 16시간동안 추가로 배양하였다. 그런 다음, 각 세포를 PBS로 세척하고, Reporter lysis buffer(Promega)를 가하여 각 세포를 용해시켰으며, 상기 세포용해물에 luciferase substrate(Promega)를 가하고 반응시킨 다음, 분광광도계에 적용하여 흡광도를 측정하고, 이를 이용하여 RANKL-RANK 복합체의 신호전달과정을 통한 NF-κB 발현억제활성을 산출하였다(도 4).
도 4는 파골세포 분화 억제 화합물의 처리농도에 따른 RANKL-RANK 복합체의 신호전달과정 억제효과를 나타내는 그래프이다. 도 4에서 보듯이, 3가지 화합물(소포카르핀, 탄제리틴 및 우스닌산)을 처리한 실험군에서는 모두 처리농도에 비례하여 RANKL-RANK 복합체의 신호전달과정을 통한 NF-κB 발현이 억제됨을 확인하였다.
따라서, 소포카르핀, 탄제리틴 및 우스닌산의 파골세포 분화억제 효과는 RANKL-RANK 복합체의 신호전달과정을 억제함에 의하여 유발됨을 알 수 있었다.
Claims (9)
- 소포카르핀(sophocarpine), 우스닌산(usnic acid), 이들의 약학적으로 허용되는 염 및 이들의 조합으로 구성된 군으로부터 선택되는 화합물을 유효성분으로 포함하는 골 관련 질환 예방 또는 치료용 약학조성물.
- 제1항에 있어서,
상기 골 관련 질환은 성장기 발육부진, 골절, 과도한 파골세포의 골 흡수에 의한 골다공증(osteoprosis), 류마티스성 관절염(rheumatoid arthritis), 치주질환(periodontal disease), 파제트병(Paget disease) 및 전이성 골암(metastatic bone cancers)으로 구성된 군으로부터 선택되는 하나 이상인 것인 조성물.
- 제1항에 있어서,
상기 화합물의 함량은 조성물 총중량을 기준으로 0.0001 내지 50 중량%인 것인 조성물.
- 제1항에 있어서,
상기 조성물은 탄제리틴을 추가적으로 포함하고, 소포카르핀 및 탄제리틴을 1 내지 100 : 1 내지 100(w/w)으로 포함하는 것인 조성물.
- 제1항에 있어서,
소포카르핀 및 우스닌산을 1 내지 100 : 1 내지 100(w/w)으로 포함하는 것인 조성물.
- 제1항에 있어서,
상기 조성물은 탄제리틴을 추가적으로 포함하고, 탄제리틴 및 우스닌산을 1 내지 100 : 1 내지 100(w/w)으로 포함하는 것인 조성물.
- 제1항에 있어서,
상기 조성물은 탄제리틴을 추가적으로 포함하고, 소포카르핀, 탄제리틴 및 우스닌산을 1 내지 100 : 1 내지 100 : 1 내지 100(w/w/w)으로 포함하는 것인 조성물.
- 소포카르핀(sophocarpine), 우스닌산(usnic acid), 이들의 약학적으로 허용되는 염 및 이들의 조합으로 구성된 군으로부터 선택되는 화합물을 유효성분으로 포함하는 파골세포 분화 억제용 조성물.
- 제8항의 조성물을 파골세포 전구체(osteoclast precursor)에 처리하는 단계를 포함하는, 시험관 내(in vitro) 파골세포의 분화를 억제하는 방법.
