Abstract
본 발명은 자가포식소체를 검출하는 프로브에 관한 것으로, 보다 구체적으로, 본 발명은 LC3에 결합할 수 있는 단백질인 TP53INP2의 LC3-interacting region (LIR) 부위를 이용하여 자가포식소체를 특이적으로 검출하는 프로브 및 이의 용도에 관한 것이다.
Description
본 발명은 in vivo에서 자가포식체 결합 단백질을 검출하는 방법에 관한 것으로, 보다 구체적으로, 핵위치신호(NLS, nuclear localization signal)를 포함하는 프로브를 이용하여, 자가포식체 결합 단백질인 LC3/GABARAP family와 LIR(LC3-interacting region)모티프의 결합 여부를 확인하는 방법에 대한 것이다.
자가포식(autophagy) 현상은 기아상태 생존, 감염성 세균으로부터의 보호 및 신경붕괴조절과 같은 세포 기능을 하는데 중요한 역할을 하는데, 이는 진화적으로 보존된 과정으로, 이스트에서 포유류에 이르기까지 모든 진핵세포에서 나타난다.
지금까지 밝혀진 바에 의하면, 자가포식은 유도단계(induction/nucleation), 세포막 변형단계(membrane elongation/closure) 및 포식단계(vesicle trafficking/maturation/fusion/degradation)의 세 가지 단계로 크게 구분된다고 알려져 있다. 초기에는, 세포질 및 세포 내 기관들은 오토파고좀(autophagosome)이라는 이중막 결합 구조로 퇴화하고, 상기 퇴화된 오토파고좀은 리소좀(lysosome)과 융합하여 오토리소좀(autolysosome)을 형성하며, 상기 오토리소좀에 의해 퇴화된 물질이 가수분해되어 재사용된다. 그러나, 이러한 과정에 내재한 분자적 작용기작은 아직 명확하게 밝혀지지 않았기 때문에 이를 규명하기 위한 연구가 활발히 진행중이다.
효모, 초파리 등을 대상으로 수행된 다양한 유전학/생화학적 연구를 통하여 세포 내에서 자가포식(autophagy) 현상을 일으키는데 필요한 단백질들이 발굴되었고, 이들의 상관관계를 분석하기 위한 연구가 수행되었다. 이러한 자가 포식에 대한 연구가 근래 다양한 동물모델 및 질병의 유전자 분석 연구를 통하여 자가포식과정이 당뇨, 고혈압등의 대사성 질환, 퇴행성 뇌질환, 감염성 질환, 그리고 암등의 매우 다양한 질환에서 중요한 역할을 하고 있음이 보고됨으로써 그 중요성이 부각되고 있다. 따라서 자가포식(autophagy) 작용에 대한 정보를 수득하는 방법은 새로운 작용점을 타겟으로 하는 질병치료제 개발에 매우 중요한 역할을 할 수 있을 것이다.
자가포식체에서 기능을 하는 핵심 단백질로서, LC3/GABARAP 단백질들이 알려져 있다. 효모에서는 ATG8 한 종류가 알려져 있고, 포유류에서는 LC3A, LC3B, LC3C, GABARAP, GABARAP-L1, GABATRAP-L2 6종류가 알려져 있다. 이 단백질들은 평상시에는 세포질에 존재하는데, 자가포식체가 생성이 될 때 지질인 PE가 결합해서 자가포식체 막에 결합한다. 이 후 많은 단백질들이 LC3나 GABARAP 단백질들에 결합해서 자가포식체로 위치해서 기능을 수행하게 된다. LC3나 GABARAP 단백질에 결합하는 단백질들은 LIR(LC3-interacting region) 부위를 갖고 있는데 공통적으로 W/F/Y-X-X-I/L/V 서열을 갖고 있다. 상기 LIR 부위가 LC3나 GABARAP 단백질에 실제 결합성이 잇는지 알아보기 위해 현재는 GST pulldown (in vitro)과 co-immunoprecipitation (in vivo)을 이용하여 확인이 가능하며, 이는 실험이 복잡하고, 검출 효율이 좋지 못한 단점을 가진다. 이와 같은 문제를 개선하기 위하여 본 발명자들은 in-vivo에서 대량으로 자가포식체 결합을 확인할 수 있는 방법에 대한 본 발명을 완성하게 되었다.

본 발명의 목적은 in vivo에서 직접 자가포식체와 LIR 모티프의 결합성을 확인하기 위하여, 자가포식체 결합 단백질인 LC3/GABARAP 패밀리 단백질에 결합하는 LIR(LC3-interacting region) 모티프 서열; 핵위치신호 서열; 및 리포터 형광 단백질의 유전자를 포함하는 자가포식체 검출용 프로브를 제공하는 것이다.
본 발명의 다른 목적은 자가포식체 결합 단백질인 LC3/GABARAP 단백질과 결합성을 가지는 새로운 LIR 모티프를 검출하는 방법으로서, 상기 프로브를 이용하는 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 프로브에 결합된 리포터 형광 유전자의 핵 내 발현량 및 세포질에 존재하는 LC3 단백질과 결합된 리포터 형광 유전자의 발현량의 비율을 통하여 자가 포식작용에 대한 정보를 제공하는 방법을 제공하는 것이다.
상기 본 발명의 과제를 해결하기 위하여, 본 발명은 본 발명의 목적은 in vivo에서 직접 자가포식체와 LIR 모티프의 결합성을 확인하기 위하여, 자가포식체 결합 단백질인 LC3/GABARAP 패밀리 단백질에 결합하는 LIR(LC3-interacting region) 모티프 서열; 핵위치신호 서열; 및 리포터 형광 단백질의 유전자를 포함하는 자가포식체 검출용 프로브를 제공한다.
상기 LC3/GABARAP 패밀리 단백질은, 이에 제한되는 것은 아니나 LC3A, LC3B, LC3C, GABARAP, GABARAP-L1 및 GABARAP-L2로 이루어진 군으로부터 선택되는 1 이상인 것이다.
또한, 상기 LC3/GABARAP 패밀리 단백질에 결합하는 LIR(LC3-interacting region) 모티프는 ATG13, FYCO1, NBR1, Nix/Bnip3L, p62, TP53INP1, TP53INP2/DOR, ULK1, ULK2, Optineurin, FUNDC1, TBC1D5 LIR1, TBC1D5 LIR2, c-Cbl, β-catenin, Dvl2, MAP15K, Bnip3, ATG4B, Tax1bp1/T6BP, Stbd1, NDP52, Calreticulin, Clathrin HC, FIP200 및 TBC1D25로 이루어진 군으로부터 선택되는 LIR 단백질에 포함된 것일 수 있으며, 상기 모티프 내의 특정 서열이 LC3/GABARAP 패밀리 단백질과 결합성을 가지도록 유도한다. 이에 제한되는 것은 아니나, 본 발명의 실시예에서 제작된 프로브는 TP53INP2 또는 Fyco1 단백질의 LIR 모티프 서열을 포함하도록 설계하였다.
본 발명의 상기 프로브는, 핵위치신호 서열을 포함한다. “핵 위치 신호”(nuclear localizationsignal, NLS)는 단백질이나 핵산과 같은 특정 물질을 세포 핵 내로 운반하는 역할을 하는 아미노산 서열을 의미하며, 본 발명의 프로브가 핵으로 이동하여 위치할 수 있게 하는 역할을 수행한다. 이에 제한되는 것은 아니나, 본 발명의 일 실시예에서 핵위치 신호는 서열번호 3의 아미노산 서열을 포함하는 것으로서(3xNLS), 도 8의 벡터로부터 유래된 것이다.
본 발명의 일실시예에 있어서, 상기 리포터 단백질의 예로는 녹색 형광 단백질(green fluorescent protein; GFP), 변형된 녹색 형광 단백질(modified green fluorescent protein; mGFP), 증강된 녹색 형광 단백질(enhanced green fluorescent protein; EGFP), 적색 형광 단백질(red fluorescent protein; RFP), 변형된 적색 형광 단백질(modified red fluorescent protein; mRFP) 등이 있으며, 바람직하게는 증강된 녹색 형광 단백질(EGFP) 또는 변형된 적색 형광 단백질(mRFP)이 사용될 수 있다. 본 발명의 프로브에서 상기 리포터 단백질을 포함함으로 인하여 세포를 유지한 상태에서 직접 LC3/GABARAP 패밀리 단백질과 LIR 모티프의 결합 여부를 확인할 수 있다.
특히, LIR과 핵위치신호가 결합된 프로브와 세포질에 존재하는 LC3/GABARAP 패밀리 단백질에 각각 결합하는 리포터 단백질은 서로 다른 색을 띄도록 제작할 수 있으며, 이를 통하여 핵 내 위치하는 LC3/GABARAP 패밀리 단백질 및 세포질에 위치하는 LC3/GABARAP 패밀리 단백질의 C/N ratio를 통해 LC3/GABARAP 패밀리 단백질과 결합하는 LIR에 대한 정보를 제공할 수 있다.
본 발명의 일실시예에 있어서, 상기 자가포식체 검출용 프로브는, 서열번호 1의 아미노산 서열로 이루어진 FYCO1 유전자의 LIR (LC3-interacting region) 부위(“LIR(FYCO1)”) 또는 서열번호 2의 아미노산 서열로 이루어진 TP53INP2 유전자의 LIR 부위(“LIR(TP53INP2)”); 리포터 단백질로서 mRFP; 및 서열번호-의 3xNLS 가 결합된 구조체일 수 있으며, 본 명세서에서 이에 제한되지는 않으나, 상기 프로브는 LIR(Fy or TP)-mRFP-3xNLS 로 표시될 수 있다.
본 발명의 일실시예에 있어서, 상기 프로브는 3xNLS의 영향으로 인하여 핵 내에 위치하며, 세포질 내 에 존재하는 EGFP-LC3/GABARAP과 LIR의 결합력으로 인하여 핵 내로 유입될 수 있고, 그 결과, 세포질과 핵 내에서 관찰되는 리포터 단백질의 비율(C/N ratio)을 계산함으로써, 자가소포체 검출에 사용될 수 있다.
또한, 이에 제한되는 것은 아니나, 상기 검출은 GST pulldown-assay와 같이 in-vitro에서 수행될 수 있고, in-vivo에서도 수행될 수 있다. 기존의 검출 방법은 GST pulldown-assay를 통한 분석이었으나, 이는 in-vivo에서는 수행되기 어려운 단점이 있었다. 그러나, 본 발명의 프로브 및 검출 방법을 이용하면, 세포를 유지한 상태에서도 검출이 가능하므로, in-vivo에서도 자가소포체의 검출이 가능한 장점을 가진다.
또한, 본 발명은 상기 프로브 또는 조성물을 이용하되, 상기 리포터 형광 단백질의 세포질/핵의 비율(c/n ratio)을 통해 세포 내 자가포식체의 발현여부를 확인하는 단계를 포함하는 것을 특징으로 하는, 자식작용에 대한 정보 제공 방법을 제공한다.
본 발명은 종래 자가포식체 검출 방법의 문제점을 해결하여 보다 정확하고 간편한 방법으로 자가포식체를 검출할 수 있게 함으로써 자식작용과 관련된 생체 내의 다양한 정보를 수득하여 활용할 수 있게 한다.
도 1은 LIR (Fyco1 and TP53INP2)-mRFP-3xNLS와 EGFP-mATG8, 및 이의 돌연변이의 결합을 나타낸 모식도이다. 여기서, ATG8은 포유동물 유래 Atg8 단백질이며, LIR은 LC3-결합부위, NLS는 핵위치 신호(서열)을 의미한다.

본 명세서에서 사용되는 용어에 대한 정의는 다음과 같다.
"자가포식(Autophagy)은 라이소좀-의존 방식으로 세포내 구성물질을 변동, 조정하는 과정으로, 결함이 있는 거대 단백질 복합체 및 세포내 소기관을 제거하여, 세포 성장, 생존 및 항상성에 중요한 역할을 한다. 자가포식은 손상된 단백질 및 세포 소기관의 세포내 변동을 통한 세포 항상성의 유지를 위한 중요한데, 그 결과, 자가포식현상이 저하될 경우, 변성단백질(misfolded protein)의 축적이 초래되어 이로 인해 신경변성질환이 발생한다(Komatsu et al., (2006), Nature, 441, 880-884).
"자가포식 관련 질환"이란, 자식작용이 관여하여 유발되거나 또는 악화되는 질환을 의미한다. 특별히 이에 제한되지 않으나, 바람직하게는 악성종양, 신경퇴행성 질환, 허혈성 질환, 당뇨병, 폐섬유화증 등이 될 수 있고, 보다 바람직하게는, 유방암, 폐암, 섬유육종, 대장암 등의 악성종양; 알츠하이머병, 파킨슨병, 헌팅턴병 등의 신경퇴행성 질환; 협심증, 심근경색 (myocardiac infarction), 심부전(cardiac failure), 허혈성 대장염, 허혈성 급성 신부전증, 급성 허혈성 뇌졸중 등의 허혈성 질환; 제1형 당뇨병, 제2형 당뇨병 등의 당뇨병, 폐섬유화증 등이 될 수 있다.
"진단"은 병리 상태의 존재 또는 특징을 확인하는 것을 의미한다. 그 중에서도 본 발명은 자가포식 관련 질환에 대한 정보를 제공하는 것에 유용하다. 본 발명에서는 자가포식체 수의 증감 특성은 유용한 지표(진단 마커)로 사용될 수 있다.
"진단용 마커 또는 프로브(탐침)(diagnosis marker or probe)"란 자가포식체를 다른 세포와 구분하여 진단할 수 있는 물질로, 자가포식 관련 질환이 있는 경우 자가포식체가 증가 또는 감소 양상을 보이는 폴리펩타이드 또는 핵산(예: mRNA 등), 지질 , 당지질, 당단백질, 당(단당류, 이당류, 다당류 등) 등과 같은 유기 생체 분자 등을 포함한다.
"프로브"는 mRNA와 특이적 결합을 이룰 수 있는 짧게는 수개 염기 내지 길게는 수백 개 염기에 해당하는 RNA 또는 DNA 등의 핵산 단편을 의미한다. 프로브는 올리고 뉴클레오티드 프로브, 단일가닥 DNA 프로브, 이중가닥 DNA 프로브, RNA 프로브 등의 형태로 제작될 수 있다. 적당한 프로브의 선택 및 혼성화 조건은 당해분야에 공지된 것을 기초로 변형할 수 있다.
"대상" 또는 "환자"는 인간, 소, 개, 기니아 피그, 토끼, 닭, 곤충 등을 포함하여 치료가 요구되는 임의의 단일 개체를 의미한다. 또한, 임의의 질병 임상 소견을 보이지 않는 임상 연구 시험에 참여한 임의의 대상 또는 역학 연구에 참여한 대상 또는 대조군으로 사용된 대상이 대상에 포함된다.
"조직 또는 세포 샘플"은 대상 또는 환자의 조직으로부터 얻은 유사한 세포의 집합체를 의미한다. 조직 또는 세포 샘플의 공급원은 신선한, 동결된 및/또는 보존된 장기 또는 조직 샘플 또는 생검 또는 흡인물로부터의 고형 조직; 혈액 또는 임의의 혈액 구성분; 대상의 임신 또는 발생의 임의의 시점의 세포일 수 있다. 조직 샘플은 또한 1차 또는 배양 세포 또는 세포주일 수 있다.
"핵산"은 임의의 DNA 또는 RNA, 예를 들어, 조직 샘플에 존재하는 염색체, 미토콘드리아, 바이러스 및/또는 세균 핵산을 포함하는 의미이다. 이중가닥 핵산 분자의 하나 또는 두개 모두의 가닥을 포함하고, 무손상 핵산 분자의 임의의 단편 또는 일부를 포함한다.
"유전자"는 단백질 코딩 또는 전사시에 또는 다른 유전자 발현의 조절시에 기능적 역할을 갖는 임의의 핵산 서열 또는 그의 일부를 의미한다. 유전자는 기능적 단백질을 코딩하는 모든 핵산 또는 단백질을 코딩 또는 발현하는 핵산의 일부만으로 이루어질 수 있다. 핵산 서열은 엑손, 인트론, 개시 또는 종료 영역, 프로모터 서열, 다른 조절 서열 또는 유전자에 인접한 특유한 서열 내에 유전자 이상을 포함할 수 있다.
"단백질"은 또한 기준 단백질과 본질적으로 동일한 생물 활성 또는 기능을 보유하는, 단백질의 단편, 유사체 및 유도체를 포함하는 것이다.
"형질전환, 형질감염 또는 트랜스펙션"은 세포외부 DNA가, 수반물질이 있고 없는 상태로 숙주 세포로 들어가는 과정을 말한다. "트랜스펙션된 세포"란 세포 외부 DNA가 세포 내로 도입되어 세포 외부 DNA를 가지고 있는 세포를 가리킨다. DNA는 세포로 도입되어 핵산이 염색체에 삽입되거나 혹은 염색체 외 물질로 복제될 수 있다. 그리고 외부 DNA 등이 도입된 세포 등을 '형질전환체'라고 한다.
" 벡터" 라는 용어는 숙주 세포에 삽입되어 숙주 세포 게놈과 재조합되고 이에 삽입되거나, 또는 에피좀으로서 자발적으로 복제하는 컴피턴트 뉴클레오티드 서열을 포함하는 임의의 핵산을 의미한다. 이러한 벡터로는 선형 핵산, 플라스미드, 파지미드, 코스미드, RNA 벡터, 바이러스 벡터 등이 있다.
"표지" 또는 "라벨"는 직접 또는 간접적으로 시약, 예를 들어 핵산 프로브 또는 항체에 컨쥬게이팅 되거나 융합되고 컨쥬게이팅 되거나 융합된 시약의 검출을 용이하게 하는 화합물 또는 조성물을 의미한다. 표지는 그 자체가 검출될 수 있거나 (예를 들어, 방사성 동위원소 표지 또는 형광 표지), 효소 표지의 경우에, 검출가능한 기질 화합물 또는 조성물의 화학적 변형을 촉매 할 수 있다.
"약"이라는 것은 참조 양, 수준, 값, 수, 빈도, 퍼센트, 치수, 크기, 양, 중량 또는 길이에 대해 30, 25, 20, 25, 10, 9, 8, 7, 6, 5, 4, 3, 2 또는 1% 정도로 변하는 양, 수준, 값, 수, 빈도, 퍼센트, 치수, 크기, 양, 중량 또는 길이를 의미한다.
본 명세서를 통해, 문맥에서 달리 필요하지 않으면, "포함하다" 및 "포함하는"이란 말은 제시된 단계 또는 원소, 또는 단계 또는 원소들의 군을 포함하나, 임의의 다른 단계 또는 원소, 또는 단계 또는 원소들의 군이 배제되지는 않음을 내포하는 것으로 이해하여야 한다.
이하, 본 발명에 대하여 구체적으로 설명한다.
본 발명은 자가포식체(autophagosome)를 검출하는 새로운 프로브(probe)에 관한 것으로, 더욱 구체적으로는 LC3에 결합할 수 있는 단백질인 LIR 부위 및 핵위치 신호 서열(NLS)을 이용하여 자가포식체를 특이적으로 검출하는 프로브 및 이의 용도에 관한 것이다.
자가포식현상은 리소좀(lysosome)을 통해 세포구성물이 제거되는 과정으로 생리적으로 자가포식은 세포소기관과 단백질을 생성/분해하는데 균형을 잡는 역할을 한다. 자가포식은 질병이 발생하는 과정을 매개하고, 세포내 신호전달과 단백질 손상 과정에서 일어나는 활성산소족과 활성질소족(ROS/RNS)의 정보교환(crosstalk)에 관여하는 것으로 밝혀지고 있다.
자가포식은 기질이 어떻게 리소좀에 이르는지에 따라 세 가지로 분류된다: (i) 대자가포식(macroautophagy) - 일반적으로 일컫는 자가포식을 대자가포식이라 한다. 자가포식소체(autophagosome)를 형성하는 방법이다. (ii) 소자가포식 - 리소좀이 직접 세포 구성물을 둘러싸고 소화한다. (iii) 샤페론 매개 자가포식 - 샤페론이 결합한 단백질 복합체는 LAMP-2A수용체에 결합해 표적 단백질을 리소좀에 데려가 분해한다
포유류 세포에서, 영양분이 감소하면 TOR(target of rapamy-cin)가 억제되어 atg13이 탈인산화(dephosphorylated)된다. 탈인산화된 atg13은 atg1 상동단백질(homologue)과 결합하여 활성화한다. 활성화된 atg1은 FIP200과 유도결합(recruit)한다. 이 단백질 복합체는 분리막을 연장하여 자가포식체( autophagosome) 구조가 완성되도록 유도한다.
자가포식체 구조를 이루기 위해서는 beclin-1-class III PI3K 복합체가 필요하고, LC3(light chain 3)-II가 자가포식체의 막에 삽입되어야 한다. LC3는 유비퀴틴(ubiquitin)과 유사한 단백질이며 LC3는 해독후변형(post translational modifications)되어 LC3-II가 된다. LC3는 Atg4에 잘려나가 LC3-I이 된 후 C말단에 글라이신(Glycine) 잔기를 노출시킨다. 이후 LC3-I과 PE(phosphatydilethanolamine) 가 결합하여 LC3-II가 된다. 이 과정은 자가포식체가 확장하는 것에 필수적이다
자가포식의 전형적인 특징은 자가포식을 유도하는 신호, 표적, 그리고 세포의 유형에 따라 다양한 크기(0.5 - 1.5 um)를 가지는 자가포식체의 형성이다. 효모를 이용한 유전적 스크린을 통하여 자가포식체 형성의 여러 가지 과정을 조절하는 35개 이상의 자가포식에 관련된(autophagy-related, Atg) 유전자들이 발견되었다.
그러므로 형성된 자가포식체를 검출함으로써 자가포식 관련 질환에 대한 다양한 정보를 수득할 수 있고, 본 발명은 이러한 자가포식체 검출용 프로브에 관한 것이다.
자가포식작용이 실제로 일어나고 있는지 확인하기 위해서 성숙하고 있는 각단계의 자가포식체를 전자현미경으로 관찰하는 것이 가장 전통적인 방법이다. 또 다른 방법으로 수명이 긴 단백질의 분해 속도를 파동추적(PULSE-CHASE) 방법으로 측정하는 것이 일반적이면서도 보조적이다. LC3에 기반한 방법도 자가포식을 측정하는 데에 널리 이용되고 있어왔다. LC3는 PE와 결합하여 LC3-II로 변환된다. LC3-I과 II는 전기영동에서 이동성의 차이로 구분할 수 있다. 자가포식체가 형성될 때 LC3-II가 자가포식체막에 삽입되는 것이 일관되게 일어나는 중요한 단계이며 LC3의 양은 세포 내 자가포식체의 상대적인 양을 반영한다. LC3-II 단백질은 자가포식체와 결합하면 세포면역염색법으로 관찰할 때 소낭과 같은 작은 점으로 보이게 된다.
기존에는 자가포식체 형성에 직접적으로 관여하는 LC3에 GFP 또는 RFP를 붙인 RFP/GFP-LC3가 일반적으로 자가포식체를 표지하기 위해 사용되었다. 이는 RFP가 산성조건에 저항성이 있는 반면 GFP는 산성에서 약해지는 원리를 이용한다. GFP와 RFP가 연속으로 결합된 LC3은 리소좀과 결합하지 않은 자가포식체에서 GFP와 RFP 빛을 동시에 내지만, 자가포식체(autolysosome)가 성숙하면 RFP만 발산하게 된다.
그러나, LC3 자체가 자가포식체 형성에 직접관여하기 때문에, LC3 과발현에 의한 다양한 부작용은 피할 수가 없었다.
또한 위와 같은 방법은 in-vitro에서만 수행될 수 있는 방법이어서, 실제 자가포식체를 이용한 질병의 진단이나 검출 등에 활용되기에 번거로운 문제점도 있었다.
본 발명은 이러한 문제를 해결할 수 있는, 특정 단백질의 LIR(LC3-interacting region) 부위와 핵 위치 신호(NLS) 서열을 포함하는, 새로운 자가포식소체 검출용 프로브 및 이의 이용에 관한 것이다.
상기 핵 위치화 신호(NLS) 펩타이드는 NLS 기능과 관련된 서열을 최소 서열로 포함하며 다른 서열을 추가적으로 포함할 수 있다. 본 명세서에서 용어 “핵 위치화 신호(nuclear localizationsignal, NLS)"는 단백질이나 핵산과 같은 특정 물질을 세포 핵 내로 운반하는 역할을 하는 아미노산 서열을 의미한다. 본 발명의 펩타이드는 본 발명이 속하는 기술분야에서 통상적으로 사용되는 방법에 의해 제조될 수 있다.
본 발명의 일 실시예에서 상기 핵 위치화 신호(NLS) 펩타이드는 인간 세포로부터 유래된 것일 수 있으며, 보다 구체적으로는 서열번호 3의 펩타이드 서열을 포함할 수 있다.
일 양태로서 본 발명은 자가포식소체 검출용 프로브에 관한 것이다. 상기 프로브 구조체는 세포 내 자가포식소체 수준을 확인하여 자식작용에 대한 다양한 정보 제공을 위해 사용될 수 있다.
본 발명의 프로브는 일 구체예로서, TP53INP2 (Tumor Protein P53 Inducible Nuclear Protein 2) 또는 FYCO1(FYVE And Coiled-Coil Domain Containing 1) 유전자의 LIR(LC3-interacting region) 부위; 3xNLS 펩타이드; 및 리포터 형광 단백질을 함유하는 것을 특징으로 한다.
상기 TP53INP2 및 FYCO1 펩타이드 서열은 동물 유래, 바람직하게는 인간의 공지된 유전자 서열정보로부터 발현된 아미노산 서열로부터 얻을 수 있으며, 상기 NLS 펩타이드는 인간세포로부터 유래된 것일 수 있다. 상기 유전자 또는 단백질들은 야생형(wild type) 뿐만 아니라 이의 결실, 삽입, 비보전적 또는 보전적 치환 또는 이들의 조합에 의한 변이체를 포함한다.
상기 단백질 등을 코딩하는 유전자는 당업계에 공지된 염기서열을 참조하여 이의 말단 부분에 상보적인 올리고뉴클레오타이를 삭제한 후 이를 프라이머로 하고 특정한 세포의 지노믹 DNA나 cDNA를 주형으로 하여 PCR을 수행함으로써 수득할 수 있다.
본 발명은 상기 단백질과 기능적 동등물인 경우를 모두 포함한다. '기능적 동등물'이란 아미노산의 부가, 치환 또는 결실의 결과, 원래 아미노산 서열과 적어도 70% 이상, 바람직하게는 80% 이상, 더욱 바람직하게는 90%이상, 더 더욱 바람직하게는 95% 이상의 서열 상동성을 갖는 것으로, 실질적으로 동질의 생리활성을 나타내는 단백질을 말한다. '실질적으로 동질의 생리활성'이란 자식작용 메커니즘에 관여하는 각 단백질들의 활성을 의미한다.
기능적인 LIR들은 다양한 단백질들에서 발견된다. 그 중 몇몇은 자가포식 수용체인 반면 다른 단백질들은 자가포식의 기질이기도 하다. 중요한 것은 Rab guanosine triphosphatase (GTPase)-활성화 단백질(GTPase-activating proteins, GAPs), Atg3, Atg4B, 그리고 Atg1/ULK1 복합체의 요소들과 같이 점점 더 많은 자가포식 조절 단백질들이 Atg8/LC3/GABARAP와 상호작용하는 것으로 밝혀지고 있다.
본 발명의 일 구체예에서는 다양한 LIR 모티프 중에서, 특히, TP53INP2 유전자의 LIR(LC3-interacting region) 부위를, 핵 위치 신호 서열인 3xNLS와 결합함으로써 더욱 간편하게 LC3-양성의 자가포식소체(autophagosomes)활성을 확인할 수 있다.
상기 LIR(LC3-interacting region) 부위는 예를 들어, TP53INP2 유전자 또는 FYCO1 유전자로부터 유래된 염기서열에 의해 발현되는 것으로서, 서열번호 1 또는 서열번호 2로 이루어진 아미노산 서열을 갖는 것일 수 있다. 특히, 상기 TP53INP2 의 LIR(LC3-interacting region) 부위는 트립토판-류신-이소류신-이소류신(Trp-Leu-Ile-Ile; WLII)을 포함할 수 있으며, 상기 FYCO1의 LIR 부위는 페닐알라닌-아스파틱산-이소류신-이소류신(Phe-Asp-Ile-Ile; FDII)을 포함할 수 있다. 상기 아미노산 서열은 야생형(wild type)을 포함하여, 결실, 삽입, 비보전적 또는 보전적 치환 또는 이들의 조합에 의한 변이체를 포함한다.
또한, 본 발명은 자가포식소체의 세포 내 수준을 검출하기 위해서 리포터 유전자로서 형광 단백질을 융합시켜 발현시킬 수 있다. 상기 리포터 형광 단백질 유전자는 그 업스트림(upstream)의 프로모터 활성에 따라 유전자가 발현되어 세포 내에서 그 발현 여부를 확인할 수 있다. 리포터 형광 단백질의 예로는 녹색 형광 단백질(GFP), 변형된 녹색 형광 단백질(modified green fluorescent protein), 증강된 녹색 형광 단백질(enhanced green fluorescent protein; EGFP), 적색 형광 단백질(RFP), 변형된 적색 형광 단백질(mRFP), 청색 형광 단백질(BFP), 증강된 청색 형광 단백질(EBFP), 황색 형광 단백질(YFP), 증강된 황색 형광 단백질(EYFP), 남색 형광 단백질(CFP), 및 증강된 남색 형광 단백질(ECFP) 및 레닐라 루시퍼라제(Renila luciferase), 적색 형광 단백질(DsRed) 등을 포함하나 상기 예에 제한되는 것은 아니다. 바람직하게는 녹색 형광 단백질(GFP), 변형된 녹색 형광 단백질(modified green fluorescent protein), 증강된 녹색 형광 단백질(enhanced green fluorescent protein; EGFP), 적색 형광 단백질(RFP), 변형된 적색 형광 단백질(mRFP)을, 가장 바람직하게는 증강된 녹색 형광 단백질(EGFP) 및 변형된 적색 형광 단백질(mRFP)을 리포터 단백질로서 사용한다.
다른 관점에서, 본 발명은 상기 자가포식소체 검출용 프로브를 함유하는, 자식작용에 대한 정보 제공을 위한 자가포식소체 검출용 재조합 벡터에 관한 것이다.
상기 벡터는 예를 들어, 상기 프로브에 함유되어 있는 유전자 각각을 포함할 수도 있고 이들이 융합된 재조합 단백질을 코딩하는 서열을 포함할 수도 있다.
상기 "벡터"란 적당한 숙주세포에서 목적 단백질을 발현할 수 있는 발현 벡터로서, 유전자 삽입물이 발현되도록 작동가능하게 연결된 필수적인 조절 요소를 포함하는 유전자 작제물을 말한다. 벡터는 플라스미드, 파지 입자 또는 간단하게 잠재적 게놈 삽입물일 수 있다. 적당한 숙주로 형질전환되면, 벡터는 숙주 게놈과 무관하게 복제하고 기능할 수 있거나, 또는 일부 경우에 게놈 그 자체에 통합될 수 있다. 플라스미드가 현재 벡터의 가장 통상적으로 사용되는 형태이므로, 본 발명의 명세서에서 "플라스미드(plasmid)" 및 "벡터(vector)"는 때로 상호 교환적으로 사용된다. 플라스미드 벡터는 (a) 숙주세포당 수백 개의 플라스미드 벡터를 포함하도록 복제가 효율적으로 이루어지도록 하는 복제 개시점, (b) 플라스미드 벡터로 형질전환된 숙주세포가 선발될 수 있도록 하는 항생제 내성 유전자 및 (c)외래 DNA 절편이 삽입될 수 있는 제한효소 절단부위를 포함하는 구조를 지니고 있다. 적절한 제한효소 절단부위가 존재하지 않을지라도, 통상의 방법에 따른 합성 올리고뉴클레오타이드 어댑터 (oligonucleotide adaptor) 또는 링커 (linker)를 사용하면 벡터와 외래 DNA를 용이하게 라이게이션 (ligation)할 수 있다.
상기 "작동 가능하게 연결된"이란, 본 발명의 프로모터 서열의 다운스트림에 유전자가 연결될 때, 해당 유전자의 발현이 가능한 형태로 연결되는 것을 의미하며, 상기 목적을 달성하기 위하여 임의의 서열이 추가로 포함될 수 있다. 상기 임의의 서열을 예로 들면, 오퍼레이터(operator) 서열, 인프레임(in frame)을 위한 서열 등이 있으며, 또한 본 발명의 유전자 이외에 인핸서 (enhancer)와 같은 시스 요소 (cis element), 스플라이싱 시그널 (splicing signal), 폴리 A 추가 시그널 (poly A addition signal), 선택 마커 (selection marker), 라이보좀 결합 서열 (ribosome binding sequence, SD sequence) 등을 추가로 포함할 수 있다. 선택 마커의 예로, 클로람페니콜 저항 유전자, 암피실린 저항 유전자, 디하이드로폴레이트 환원효소(dihydrofolate reductase), 네오마이신 저항 유전자 등이 사용될 수 있으나, 상기 예들에 의해 작동 가능하도록 연결되기 위한 추가적 구성요소가 제한되는 것은 아니다.
다른 관점에서, 본 발명은 상기 프로브 또는 상기 벡터를 포함하는, 자가포식소체 검출 및 자식작용에 대한 정보 제공을 위한 자가포식소체 검출용 조성물에 관한 것이다.
본 발명의 일시시예에 있어서, 상기 검출은 리포터 형광 단백질의 발현 수준을 측정하는 방법으로 수행될 수 있다. 상기 '발현 수준 측정'이란, mRNA 또는 이의 단백질 발현 수준을 측정하는 것을 모두 포함한다. mRNA 발현수준 측정을 위한 분석 방법으로는 역전사중합효소반응(RT-PCR), 경쟁적 역전사 중합효소반응(Competitive RT-PCR), 실시간 역전사 중합효소반응(Realtime RT-PCR), RNase 보호 분석법(RPA; RNase protection assay), 노던 블랏팅(Northern blotting), DNA 칩 등이 있으나, 이에 제한되는 것은 아니다.
이 때, 사용되는 프라이머는 적절한 완충용액 및 온도에서 중합반응(즉, DNA 중합효소 또는 역전사효소)을 위한 시약 및 상이한 4가지 뉴클레오사이드 트리포스페이트의 존재하에서 DNA 합성이 개시할 수 있다. 본 발명의 프라이머는, 각 마커 유전자 특이적인 프라이머로 7개 내지 50개의 뉴클레오타이드 서열을 가진 센스 및 안티센스 핵산이다. 프라이머는 DNA 합성의 개시점으로 작용하는 프라이머의 기본 성질을 변화시키지 않는 추가의 특징을 혼입할 수 있다. 상기 프라이머는 기타 널리 공지된 방법을 사용하여 화학적으로 합성할 수 있고, 당해 분야에 공지된 많은 수단을 이용하여 변형시킬 수 있다.
상기 단백질 발현수준 측정은 상기 유전자의 단백질에 대하여 특이적으로 결합하는 항체를 이용하여 단백질의 양을 확인할 수 있다. "항체"란 항원성 부위에 대해서 지시되는 특이적인 단백질 분자를 의미한다. 다클론 항체, 단클론 항체 및 재조합 항체를 모두 포함한다. 이를 위한 분석 방법으로는 웨스턴 블랏, 엘라이자(enzyme linked immunosorbent assay,ELISA), 방사선면역분석(RIA: Radioimmunoassay), 방사 면역 확산법(radioimmunodiffusion), 오우크테로니(Ouchterlony) 면역확산법, 로케트(rocket) 면역전기영동, 조직면역 염색, 면역침전 분석법(Immunoprecipitation Assay), 보체 고정 분석법(Complement Fixation Assay), 유세포분석(Fluorescence Activated Cell Sorter, FACS), 단백질 칩(protein chip) 등이 있으나 이에 제한되는 것은 아니다.
또한, 본 발명은 다른 측면에서 상기 재조합 벡터로 형질 전환된 세포를 제공한다. 상기 형질 전환 방법은 당업계에 공지된 방법, 예를 들어 이에 한정되지는 않으나, 열 충격 방법(heat shock method), 칼슘 포스페이트 침전법, 전기 천공법(electroporation), 유전자 총(gene gun), 실리콘 탄화물 위스터(silicon carbide whiskers), 초음파 처리(sonication), 및 PEG(polyethylenglycol)를 이용한 침전법, 자연 도입 방법 등이 있으나, 상기 예에 의해 본 발명의 벡터를 도입하는 방법이 제한되는 것은 아니다.
또한, 본 발명은 상기 재조합 벡터로 대상 세포를 형질 전환하는 단계; 및 상기 형질 전환체의 리포터 형광 단백질의 C/N ratio를 정량하는 단계를 포함하는 자가포식소체 수준을 확인함으로써 자식작용에 대한 정보를 제공하는 방법을 제공한다.
상기 C/N ratio는, 다음과 같은 의미를 가진다. 자가포식소체의 수준에 따라 발현되는 리포터 형광 단백질의 양이 결정되며, 세포 내 자가포식소체의 수준이 높을수록 발현되는 리포터 형광 단백질의 양이 많아진다. 특히 본 발명에서는 상기 자가포식소체 관련 단백질인 LC3/GABARAP 에 돌연변이를 일으켜 자가포식작용에 관여하지 않고, 세포질 내에 존재하도록 하였으며, 상기 LC3/GABARAP과 핵 내에 위치하고 있는 프로브의 LIR 부위가 서로 결합하여, LC3/GABARAP를 핵 안으로의 유입되도록 하였다. 세포질과 핵에서 확인되는 LC3/GABARAP의 형광표지자의 비율을 다양한 방식으로 측정하여 자식작용과 관련된 다양한 정보를 수득할 수 있는 것이다.
본 발명의 일 구체예에 있어서 형질감염(형질전환)에 이용되는 세포주는 바람직하게는 동물 세포, 보다 바람직하게는 포유동물 세포주인 것이 바람직하다. 본 발명에서 이용될 수 있는 배지는 동물세포의 배양에 통상적으로 이용되는 어떠한 배지도 가능하며, 예를 들어, Eagles's MEM (Eagle's minimum essential medium, Eagle, H. Science 130:432(1959)), α-MEM (Stanner, C.P. et al., Nat. New Biol. 230:52(1971)), Iscove's MEM (Iscove, N.et al., J. Exp. Med. 147:923(1978)), 199 배지 (Morgan et al., Proc. Soc. Exp. Bio. 본 발명에서는, 상기 형질감염된 세포주에서 형광 단백질의 세포질 및 핵내 발현 비율(C/N ratio)을 비교하여 자식작용 관련 자가포식소체 수준을 확인할 수 있고 이를 비교 분석하여 비율이 낮은 것으로 측정되면 자식작용이 많은 것으로 판별한다.
상기와 같이 본 발명의 자가포식소체 프로브를 이용할 경우 자가포식소체 검출을 효과적으로 할 수 있으므로, 본 발명은 자가포식 작용 관련 질환에 대한 정보를 제공하는데 이용될 수 있다.
최근에는 신경변성질환인 알츠하이머병과 파킨슨병, 헌팅턴병 그리고 암, 당뇨병, 염증반응 등과 같은 다양한 질병과 병리학적 현상에서 자가포식의 중요한 역할이 밝혀지고 있다. 자식작용은 특정 신경변성질환에서 세포보호능을 통해 신경세포의 세포사멸을 억제하는 것으로 알려져 있다. 단백질 응집체의 축적은 정상적인 신경세포의 기능을 방해하며 세포독성을 나타낸다. 자식작용은 신경변성질환에서 나타날 수 있는 다음과 같은 비정상적 단백질의 분해와 연관성을 나타낸다. 헌팅턴병을 일으키는 헌팅턴 단백질(huntington, Htt) 내의 글루타민 다량체의 확장, 그리고 파킨슨병을 유도하는 α-synuclein, 전측두엽치매와 연관하는 tau 단백질, 루게릭병에서의 Cu-ZnSOD의 돌연변이가 그 예이다. Atg7이 결핍된 마우스에서는 신경세포의 기능이상과 세포사멸이 일어난다. 자식작용은 이러한 단백질 응집의 해소뿐만 아니라 신경세포의 항상성을 유지시켜줌으로써 세포를 보호한다. 또한 신경변성질환 환자의 뇌조직에서도 자기소화액포(autophagic vacuole)가 축적되어 있음이 보고되어 있다. 알츠하이머병에서 presenilin-1 단백질의 돌연변이는 자가포식소포체의 성숙과 리소좀을 통한 분해능을 억제하며, presenilin-1 단백질의 손실은 자가포식소포체의 산성화를 방해하여 단백질 분해능을 방해한다
또한, 상기 자식작용은 암 질환에도 관여한다. 자식작용은 종양형성에서 두 가지 역할을 담당한다. 첫째는 종양억제자로서의 세포사멸유도이고, 둘째는 종양형성기전에서의 세포사멸억제이다. 종양형성의 주요원인은 방사능과 산화적 스트레스, 노화 등의 요소로부터 기인하는 DNA 손상과 돌연변이이다. 미토콘드리아 DNA는 복구기전과 히스톤 단백질에 의한 보호의 부족으로 손상에 보다 취약하다. 미토콘드리아는 전자전달계 활동의 부산물로 다량의 활성산소를 발생시킬 수 있으며 이러한 활성산소의 조절 부재는 미토콘드리아 DNA를 손상시키고 내막과 외막의 산화를 초래할 수 있다. 또한 미토콘드리아의 기능장애는 보다 많은 활성산소를 축적시켜 그 결과 세포의 산화적 스트레스와 다른 미토콘드리아의 손상을 일으킨다. 자식작용은 손상된미토콘드리아를 제거하여 산화적 스트레스를 완화시킴으로써 발암현상을 억제한다. 한편, 종양형성에서 자식작용은 제한적 영양상태와 저산소 상태에서 그리고 방사능 노출 등에서 손상단백질과 세포소기관을 제거함으로써 암세포를 생존시킬 수 있다.
또한, 자식작용은 당뇨병 질환에도 관여한다. 당뇨병은 췌장 β세포의 기능장애와 인슐린저항성에 의해 발생한다. 산화적 스트레스가 유발한 미토콘드리아의 기능장애는 당뇨병의 발전에 중요한 역할을 담당하며 손상된 미토콘드리아는 자식 작용에 의해 제거된다. Atg7-/-마우스는 췌장 β세포의 급격한 감소를 보이며 이에 따라 인슐린분비량의 감소를 나타낸다. 또한, Atg7돌연변이 마우스는 저인슐린혈증과 과혈당증를 나타낸다. 췌장 β세포의 기능장애에 이어 자식작용의 기능장애는 인슐린저항성과 연관되어 있다. 소포체스트레스는 인슐린수용체의 세포막발현을 약화시켜 인슐린저항성을 일으킨다. 인슐린저항성은 순환적으로 자식작용을 억제하여 결국 기능장애 상태의 단백질과 미토콘드리아 등의 세포소기관의 분해를 방해하여 췌장 β세포의 사멸을 유도하게 된다. 따라서 자식작용은 췌장 β세포의 기능과 세포항상성에 중요한 기전이며 인슐린감수성에 영향을 미칠 수 있다. 노화의 경우에도 당내성과 연관이 있기 때문에 자식작용의 활성은 노화와도 밀접한 연관이 있을 것으로 사료된다.
또한, 자식작용은 심혈관계 질환에도 관여한다. 정상적인 심혈관계 기능을 위해 자식작용은 반드시 필요한 세포 기전이나 비정상적인 자식작용은고혈압과 심장기능부전을 일으킬 수 있다. 심근질환의 하나인 다논병(danon disease)의 경우, 자가포식소포체와 리소좀의 결합에 관여하는 LAMP-2 단백질의 결핍에 의해 일어난다. 심혈관계질환 관련 질병모델에서 자식작용은 병리학적 조건에 따라 세포보호 또는 세포사멸의 효과를 나타낸다. 관상동맥혈전증에서 심장근육세포는 자식작용을 일으킨다. 교차대동맥협착(transverse aortic constriction, TAC) 모델에서 Atg5 단백질의 결핍은 세포사멸을 증가시키나 Beclin1 단백질의 결핍은 심장기능의 악화로부터 세포를 보호한다. 즉, Beclin1은 Bcell lymphoma 2 (Bcl-2) 단백질과 상호작용하여 항 아폽토시스효 과를 억제하여 세포사멸을 유도한다. 파킨슨병 연관유전자로 알려진 Parkin은 p62와의 상호작용을 통해 심장에서 미토콘드리아 특이적 자식작용을 일으킨다.
또한, 자식작용은 염증반응에도 관여한다. 자식작용은 선천성 면역반응의 중요한 요소이며 적응성 면역반응에서도 다양한 기능을 담당하고 있다. 자식작용은 자가포식용해소체 경로를 통해 침입한 병원체의 성장을 제한한다. 세균 그리고 바이러스, 기생충 등은 자식작용의 표적이다. 자식작용은 특정 단일가닥 RNA 바이러스의 탐색을 가능하게 한다. 이 과정에서 엔도좀내로 이동된 바이러스 복제 중개체는 병원체 형태 인식수용체(pathogen pattern-recognition receptor, PRR)와 TLR7에 의해 인지된다. 또한 자식작용은 전 염증시토카인의 생성과 관련이 있다.형질세포양수지상세포(plasmacytoid dendritic cell, pDC)에서 자식작용은 제1형 인터페론(infection-initiated type I interferon, IFN)의 생성에 필요한 기전이다. 그러나 ATG5 결핍 마우스의 배아 섬유아세포는 단일가닥 RNA 바이러스의 감염에 대해 제1형 인터페론의 생성을 촉진하였으며, 이 경우에는 자식작용이 시토카인의 생성을 억제한다는 것을 보여준다. 따라서 자식작용은 선천성면역반응에서 전 염증반응과 항 염증반응에서 두 가지 상반되는 역할을 담당한다.
이처럼, 자식작용은 신경변성질환 그리고 암, 당뇨병, 염증반응 등과 같은 다양한 질병으로 진행하는 기작에 관여하므로, 본 발명의 자가포식소체 검출 프로브를 이용하여 관련 정보를 수득 및 제공할 수 있을 것이다.
이처럼, 본 발명은 LIR(Fy or TP)-mRFP-3xNLS를 구조체를 이용하여 자가포식체가 세포질과 핵에 존재하는 비율인 C/N ratio를 확인함으로써, 자식작용에 대한 다양한 정보를 제공하는 모든 용도를 포함한다.
즉, 자가포식체의 수준을 특정 형광 리포터 검출을 통해 쉽게 확인할 수 있는 특징이 있어, 자가포식체 검출과 관련된 용도로 효과적으로 사용할 수 있고, 나아가 자식작용 관련 질환의 새로운 치료제를 발굴하는 데 유용할 것이다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
달리 지시되지 않는 한, 핵산은 좌측에서 우측으로 5'→3' 배향으로 기록된다. 명세서 내에서 열거된 수치 범위는 범위를 정의하는 숫자를 포함하고, 정의된 범위 내의 각각의 정수 또는 임의의 비-정수 분획을 포함한다.
달리 정의되지 않는 한, 본원에서 사용된 모든 기술적 및 과학적 용어는 본 발명이 속하는 분야의 당업자가 통상적으로 이해하는 것과 동일한 의미를 갖는다. 본원에 기술된 것들과 유사하거나 등가인 임의의 방법 및 재료가 본 발명을 테스트하기 위한 실행에서 사용될 수 있지만, 바람직한 재료 및 방법이 본원에서 기술된다.
실험예
1. 세포 배양, 형질감염, 약물 처리 및 세포
이미징
HEK293T 세포 또는 MEFs를 세포배양 배지(Dulbecco's modified Eagle's medium [DMEM]: 10% [v/v] FBS 및 페니실린/스트렙토마이신 보충)에서 5% (v/v) CO2 를 함유하는 습한 대기 및 37℃에서 성장시켰다. 일차 신경세포 배양을 수행하였다.
혈청 기아(serum starvation)를 위해, HeLa 세포 또는 MEFs를 FBS 없이 세포 배양 배지에서 24 h 동안 배양시켰다. 상기 세포들을, 칼슘 포스페이트 방법 또는 리포펙타민 2000(Life Technologies, Carlsbad, CA, USA) 중 어느 하나를 이용하여 DNA 구조물로 형질 감염시키고 24시간 동안 배양하였다.
형광 이미지들을 공초점 레이저-스캐닝 현미경(Radiance 2000, Zeiss)으로 수득하고 NIH Image J software (National Institutes of Health, Bethesda, MD, USA)로 분석하였다.
자식작용(autophagy)을 유도하기 위해, HeLa 세포, WT MEFs, atg5-/- MEFs, 또는 배양된 뉴런들을, 10 nM 라파마이신으로 세포 유형 및 실험에 따라 10 mM NH4Cl 존재 또는 부존재 하에서 4시간 동안 배양하였다.

특히, 상기 34개의 LIR 모티프 중 FYCO1 유래 또는 TP53INP2 유래 LIR 모티프를 실험에 사용하여 LIR 모티프가 자식작용 유도 동안 LC3-양성 자가포식체를 검출할 수 있는지를 결정하였다. 즉, 상기 제조한, FYCO1, TP53INP2의 LIR 부위를 이용한 LIR(Fyco1)-mRFP-3xNLS 및 LIR(TP53INP2)-mRFP-3xNLS를 센서 후보들로 검출 효율을 비교하였다.
본 발명의 LIR 서열과 LC3 및 GABARAP의 상호 작용에 대하여 in vitro에서 확인하였다. GST pull-down assay를 통하여 GFP-표지된 LIR에 대한 LC3 및 GABARAP의 결합 여부를 확인하기 위하여, 겔에 고정된 GST와 융합된 LC3 및 GABARAP과 GFP 표지된 LIR의 결합 여부를 웨스턴 블럿으로 확인하여다. 그 결과, 도2의 A 및 C에서 보듯이, LIR 서열은, LC3 도메인 또는 GABARAP 도메인과 결합을 이루어 밴드를 나타내는 것을 확인하였다. B,D는 상기 A,C의 결과를 수치화한 것이다.
상기 B 및 D의 결과에서, LC3 family 중에서도 각 LIR 서열에 더 특이성을 가지는 종류를 확인할 수 있으며, Fyco1 유래의 LIR서열에 대해서는 LC3A 및 LC3B 가 높은 결합력을 가지고, TP53INP2 유래의 LIR 에 대해서는 GATE16이 비교적 높은 결합력을 가짐을 확인하였다.

2-2. 핵위치신호 서열과 LIR 모티프의 융합
본 발명의 자가포식체 관련 단백질인 LC3 family를 핵위치신호 서열과 결합된 LIR 모티프가 실제로 핵 내 유입을 유도할 수 있는지 여부를 확인하였다.
이를 위하여, FYCO1 유전자로부터 LIR 서열(서열번호 1)을 코딩하는 염기서열을 분리하였으며, 여기에 mRFP로 표지하고, 핵위치신호 서열(서열번호 3)를 융합하여 최종적으로 LIR(Fyco1)-mRFP-3xNLS를 형성하였다.
또한, 상기 LIR과 LC3 family의 결합 여부를 확인하기 위하여, LIR 서열 중, 보존된 부위인 "Phe-Asp-Ile-Ile;FDII" 에서 마지막 I(Ile)를 A(Alanine)으로 치환한 돌연변이를 유발하였다.
같은 방법으로, TP53INP2 유전자로부터 LIR 서열(서열번호 2)을 코딩하는 염기서열을 분리하여, 형광단백질 및 핵위치신호 서열을 융합하여 LIR(TP53INP2)-mRFP-3xNLS를 형성하고, TP53INP2 유래의 LIR 중 보존된 부위인 "Trp-Leu-Ile-Ile;WLII"의 W(Trp) 및 네 번째 I(Ile)를 각각 A(Alanine)으로 치환한 돌연변이를 유발하였다.
위와 같이 제작된 프로브는 도 3 및 도 6에 모식화하였다.
3. LIR-mRFP-3xNLS의 핵 내 위치화
상기 실시예 1에서 제작한 프로브를 이용하여, 상기 LIR 서열과 LC3 family 단백질 사이의 상호작용이, LC3 family 단백질의 핵 내 유입을 유도하는지 여부를 확인하기 위하여, 실험을 수행하였다.
여기서 LC3 family 단백질은 자가포식체 내부로 유입되지 않고, 세포질에 머물러있도록 하기 위하여, 돌연변이를 유발하였다. EGFP로 표지된 LC3 family(LC3/GABARAP)의 C-말단의 글리신(Gly) 잔기를 알라닌(Ala) 잔기로 치환하였다.
도 4,5,7 및 8의 결과는, 각 LC3 family가 가지는 LIR 에 대한 친화도에 따라, 세포질에서 핵 내부로 유입되는 LC3 family를 영상으로 확인한 결과(도 4 및 도 7) 및 이의 양을 수치화하여 그래프로 나타낸 것(도 5및 도8)이다. 도 4 및 도 7에서, EGFP의 경우, 세포질 전체에 넓게 분포하고 있다가, NLS 결합된 LIR 프로브와 함께 발현 시, 핵 내부로 유입되어 있는 것을 직접 확인할 수 있었다. 결합에 대한 친화도는 앞서 검토한 바와 같이, 각 LC3 family에 따라 조금씩 상이하나, 결합을 이룬 경우에는 대부분 핵 내로 유입되는 것을 확인하였다.
이러한 결과들을 통해 본 발명의 프로브를 이용한 자가포식소체 검출법은 세포 내에서 직접 자가포식소체의 LC3/GABARAP과 LIR 모티프의 상호작용을 이용한 방법으로서, 특히 in-vivo에서도 LC3/GABARAP과 LIR 모티프의 결합성을 확인할 수 있는 방법으로 유용하게 활용될 수 있다.
