본 발명에 따른 조성물의 살균 활성을 확인하기 위해 먼저 시험 균주인 대장균(E. coli KCTC 2571)과 포도상구균(Staphylococcus aureus KCTC 2513)을 32℃에서 48hr 동안 페트리디쉬에서 배양하였다. 배양된 균들의 초기 균수는 대장균 2.57×104 CFU/mL, 포도상구균 3.67×105 CFU/mL로 나타났다. 시험 균액은 멸균된 생리식염수 9 mL를 이용하여 앞서 배양시킨 균을 순차적으로 희석시켜 최종 균수가 103 CFU/mL이 되도록 제조하였다. 그 후 2개의 유리병에 실시예에 따른 항균 조성물과 멸균된 생리식염수 100 g, 시험 균액 1 mL씩 취하여 혼합한 시료를 준비하였다. 준비된 시료 1 mL를 9 mL 레틴 배지(letheen broth)에 취하고 균수가 102 CFU/mL가 되도록 희석시킨 후, 희석시킨 균을 다시 1 mL씩 새로 준비한 페트리디쉬에 분주한 후 시험배지 20 mL를 붓고 혼합하여 겔화시켰다. 1분 후 시료 1 mL를 9 mL 레틴 배지(letheen broth)에 취한 후, 이를 다시 1 mL씩 페트리디쉬에 분주하고 시험배지 20 mL를 붓고 혼합하여 겔화시키고 3분, 5분 및 10분을 주기로 반복하여 균의 살균 상태를 관찰하였다.
