Abstract
상기와 같은 본 발명은 앰버 서프레서 tRNA의 안티코돈을 더 잘 인식하도록 유전적으로 변이된 효모균의 티로실-티알엔에이 합성효소(tyrosyl-tRNA synthetase, TyrRS)로서,
위 돌연변이 TyrRS는 320번, 321번 아미노산 잔기가 모두 알라닌(alanine)으로 치환되었거나; 또는 320번 잔기가 알라닌(alanine), 321번 잔기가 히스티딘(Histidine)으로 각각 치환된 것임을 특징으로 한다.
Description
도 2는 fMam tRNA의 안티코돈을 인식하는 부위에 위치한 효모 TyrRS의 320 및 321 아미노산 잔기를 PCR 방법으로 무작위로 변화시킨 돌연변이 라이브러리 제작을 위한 도면이다.
도 3은 1차 PCR 결과를 젤에서 전기영동한 결과(A)와 2개의 1차 PCR 산물을 합친 후 최종 2차 PCR 결과를 젤에서 전기영동한 결과(B)이다.
도 4는 라이브러리 제작을 위한 최종 PCR 산물의 염기서열 분석결과이다.
도 5는 라이브러리 탐색을 통해 높은 앰버 서프레션 활성을 나타내는 클론을 선별한 후, 선별된 클론이 앰버 서프레션에 의해 나타나는 클로람페니콜(chloramphenicol) 저항성을 조사한 결과이다.
도 6은 선별된 2개의 돌연변이 TyrRS가 대장균에서 wild type TyrRS에 비해 앰버 서프레션 활성이 얼마나 높아졌나를 β-galactosidase 활성분석을 통해 조사한 결과이다.
Claims (2)
- 야생 효모균(Saccharomyces cerevisiae)의 유전적으로 변이된 티로실-티알엔에이 합성효소(tyrosyl-tRNA synthetase, TyrRS)로서, 위 변이된 티로실-티알엔에이 합성효소(tyrosyl-tRNA synthetase, TyrRS)는 앰버 서프레서 tRNA의 안티코돈을 더 잘 인식하도록 (ⅰ) 320번과 321번 아미노산 잔기인 프롤린과 아스파르트산이 모두 알라닌(alanine)으로 치환되었거나; 또는 (ⅱ) 320번 잔기인 프롤린은 알라닌(alanine)으로 치환되고 321번 잔기인 아스파르트산은 히스티딘(Histidine)으로 치환된 것;임을 특징으로 하는,
효모균(Saccharomyces cerevisiae)의 티로실-티알엔에이 합성효소. - 제1항에 있어서,
효모균(Saccharomyces cerevisiae)의 티로실-티알엔에이 합성효소(TyrRS)에 의해 천연 또는 비천연 아미노산이 결합하는 앰버 서프레서 tRNA는 효모균(Saccharomyces cerevisiae)의 tRNATyr CUA, 또는 효모균의 TyrRS가 인식할 수 있는 대장균 (E.coli)의 돌연변이된 initiator tRNA (fMamtRNA)인 것을 특징으로 하는,
효모균(Saccharomyces cerevisiae)의 티로실-티알엔에이 합성효소.
