Description
본 발명은 아미노 또는 카복시 말단에 결합된 약 2 내지 약 36개 아미노산 잔기 또는 이들의 유사체의 길이를 갖는 아미노산 잔기의 제1 세트, 및 3 내지 8개의 소수성 아미노산 잔기 또는 이들의 유사체를 포함하는 제2 세트를 포함하고, 항미생물 활성을 갖는, 항미생물성 펩타이드에 관한 것이다.
다수의 감염은 사람과 같은 포유동물의 면역계에 의해 성공적으로 치료된다. 그러나, 일부 경우, 국소적 또는 전신적 급성 감염을 유발시킬 수 있는 세균, 진균 또는 바이러스가 항상 제거되지는 않는다. 이는 주산기, 화상 또는 중환자실에서 그리고 면역손상된 사람에 있어서 심각한 관심사이다. 국소적 급성 감염은 집중적 이병률을 야기시킨다. 예를 들어, 슈도모나스 아에루기노사(Pseudomonas aeruginosa)는 심한 세균성 각막염의 주요 원인이며, 감염은 현재의 항미생물제로는 성공적으로 치료하기가 어렵다. 다른 경우에, 상피 표면에서의 지속적인 세균 잔류는 만성 질환을 유발시키거나 악화시킬 수 있다. 사람에 있어서, 이는 만성 피부 궤양, 아토피성 피부염 및 다른 종류의 습진, 여드름 또는 비뇨생식기 감염에 의해 예시된다. 예를 들어, 그람양성균인 스타필로코커스 아우레우스(Staphylococcus aureus)에 의한 전이증식 또는 감염이 아토피성 피부염의 유발 또는 악화 요인임이 현재 상당히 입증되고 있다. 전체 아토피성 피부염 환자의 약 90%는 에스.아우레우스에 의해 전이증식되거나 감염되며, 반면에 건강한 사람의 5%만이 이러한 세균이 번식한다. 만성 궤양은 피.아에루기노사 및 에스.아우레우스와 같은 다양한 세균의 의해 전이증식되거나 감염되며, 이는 이들 궤양의 치료 지연을 유도한다.
증후성 감염은 다양한 약제에 의해 치료될 수 있다. 일부 질환은 또한 예를 들어 백신에 의해 치료될 수 있다. 그러나, 백신은 항상 최고의 치료 선택은 아니며, 특정 미생물에 대해서는 백신이 사용될 수 없다. 방어가 이용될 수 없는 경우에는, 질환의 치료가 수행된다. 종종 치료는 항생제의 사용에 의해 수행된다. 그러나, 지난 수년 동안, 다수의 미생물은 항생제에 대한 내성을 갖게 되었다. 아마도, 가까운 장래에 내성 문제가 증가할 것이다. 또한, 몇몇 사람들은 특정 항생제에 대한 알레르기가 발생했으며, 이에 의해 특정 항생제를 효과적으로 사용할 가능성이 감소하였다.
다양한 유기체의 상피 표면은 지속적으로 세균에 노출된다. 최근 수년 동안, 항미생물성 펩타이드에 근거한 선천성 면역계는 감염되기 쉬운 생물학적 경계에서 세균의 초기 제거에 중요한 역할을 하고 있다[참조: Lehrer, R. L, and Ganz, T. (1999) Curr Opin Immunol 11: 23-27, Boman, H. G.(2000) Immunol . Rev. 173, 5-16]. 항미생물성 펩타이드는 일반적으로 세균의 막을 투과함으써 세균을 사멸시 키는 것으로 여겨지며, 그에 따라 특정 분자 미생물 목표물의 결여는 내성 발생을 최소화시킨다.

미국 특허 제6,503,881호에는 양이온성 펩타이드가 항미생물성 펩타이드로서 사용하려는 인돌리시딘 유사체임이 기술되어 있다. 양이온성 펩타이드는 동물 및 식물을 포함하는 상이한 종으로부터 유래된다.
미국 특허 제5,912,230호에는 항진균성 및 항세균성 히스타틴계 펩타이드가 기술되어 있다. 당해 펩타이드는 천연 사람 히스타틴의 아미노산 서열의 특정 부분을 기재로 한다. 또한, 진균 및 세균 감염의 치료 방법이 기술되어 있다.
미국 특허 제5,717,064호에는 메틸화된 리신-풍부 용해성 펩타이드가 기재되어 있다. 용해성 펩타이드는 트립신 분해 저항성이고 비천연적이다. 용해성 펩타이드는 생체내 투여에 적합하다.
미국 특허 제5,646,014호에는 항미생물성 펩타이드가 기술되어 있다. 펩타이드는 누에 혈림프로부터의 항미생물성 분획으로부터 분리되었다. 펩타이드는 에스케리키아 콜라이(Escherichia coli), 스타필로코커스 아우레우스(Staphylococcus aureus) 및 바실러스 세레우스(Bacillus cereus)와 같은 다수의 세균 균주에 대한 탁월한 항미생물 활성을 나타낸다.
WO2004016653호에는 아주로시딘의 20 내지 44개 서열에 근거한 펩타이드가 기술되어 있다. 상기 펩타이드는 이황화 브릿지에 의해 결합된 루프 구조를 함유 한다.
미국 특허 제6,495,516호 및 관련 특허에는 살균성 55 kDa 단백질인 살균성/투과성 증가 단백질(BPI)에 근거한 펩타이드가 기술되어 있다. 펩타이드는 항미생물 효과를 나타낼 뿐만 아니라, LPS 중화 능력을 갖는다.
WO 01/81578호에는 많은 질환에 사용될 수 있는 G-커플링된 단백질 수용체 관련 단백질을 암호화하는 많은 서열이 기술되어 있다.
현재, 세크로핀, 디펜신, 마가이닌 및 카텔리시딘을 포함하여 700 가지가 넘는 상이한 항미생물성 펩타이드 서열이 공지되어 있다 (www.bbcm.univ.trieste.it/~tossi/search.htm).
오늘날 이용할 수 있는 비교적 많은 항미생물성 펩타이드가 있다 하더라도, 항생제 및/또는 다른 항미생물제에 대해 내성 또는 저항성이 있는 미생물들을 제거하기 위해 사용될 수 있는 새로운 항미생물성 펩타이드의 요구가 여전히 증가하고 있다. 더욱 중요하게는, 사람과 같은 포유동물에 투여될 때에 알레르기를 일으키지 않는 새로운 항미생물성 펩타이드가 요구된다.
세균 및 포유동물 막에 대한 AMP의 잠재적 용해성 및 다른 성질로 인해, 새로운 펩타이드를 설계하는 데에 있어서의 과제 중 하나는 세균 또는 진균 세포와 같은 미생물에 대한 높은 특이성, 즉 높은 치료 지수(최소 용혈 농도/최소 항미생물 활성; MHC/MEC)를 갖는 AMP의 개발에 의존한다.
피.아에루기노사, 이.파에칼리스(E. faecalis), 프로테우스 미라빌리스(Proteus mirabilis), 스트렙토코커스 피오게네스(Streptococcus pyogenes) 및 에스.아우레우스와 같은 다양한 세균은 모두 카텔리시딘 LL-37과 같은 다수의 항미생물성 펩타이드를 분해하는 프로테아제를 분비한다. 이와 같이, 프로테아제 내성 항미생물성 펩타이드가 치료적 관점에서 중요하다. 또한, 많은 항미생물성 펩타이드는 문제의 발병에 자주 중요한 역할을 하는 세균, 예를 들어 에스.아우레우스 및 피.아에루기노사와 같은 미생물을 공격하는 데에 매우 효율적이지 않으며, 증가된 효과를 나타내도록 최적화시킬 필요가 있다.
발명의 개요
본 발명은 상응하는 펩타이드에 비해 항미생물 활성이 증가된 신규의 개선된 항미생물성 펩타이드에 관한 것이다. 놀랍게도, 항미생물 활성을 증가시키기 위해 특정 수의 아미노산이 필요하며, 즉, 아미노산 잔기가 3개 미만이거나 8개를 초과하는 경우에, 항미생물 활성이 감소함이 밝혀졌다. 이러한 접근법은 친수성의 고도로 양으로 하전된 펩타이드에 특히 적합한데, 그 이유는 이들의 막 파괴성이 높기 때문이다. 펩타이드를 하나 또는 다수의 소수성 아미노산으로 변형시킴으로써, 세균의 지질막에 대한 이들의 결합 능력이 향상되며, 더 높은 펩타이드 결합은 미생물의 막의 결함 형성을 향상시키고, 미생물의 치사율을 더 높일 수 있다. 그러나, 이것은 단지 이론일 뿐이며, 작용 방식은 상이할 수 있거나 상이한 작용 방식의 조합일 수 있다.

또다른 측면에서, 본 발명은 치료 또는 진단에서, 항미생물성 펩타이드 또는 항미생물성/약제학적 조성물 또는 제품의 용도에 관한 것이다.
최종 측면에서, 본 발명은 세균, 바이러스, 기생충, 진균 및 효모로 이루어진 그룹으로부터 선택된 미생물에 의해 유발되는 미생물 질환 또는 감염의 치료용 약제의 제조를 위한, 항미생물성 펩타이드, 항미생물성/약제학적 조성물 또는 상기 항미생물성 펩타이드를 포함하는 제품의 용도에 관한 것이다.

약제학적 조성물은 또한 렌즈 소독제 및 저장 용액과 같은 세정 용액 중에 사용되거나, 요로 카테터 사용 또는 중심정맥 카테터의 사용과 관련하여 세균 감염을 예방하기 위해 사용될 수 있다.
또한, 조성물은 수술후 감염의 예방을 위해 깁스, 접착제 또는 봉합재에 사용되거나 상처 드레싱에 혼입될 수 있다.
항미생물성 펩타이드는 또한 항세균성 표면 또는 화장품을 생성시키 위해 중합체, 직물 등에 사용될 수 있고, 퍼스널 케어 제품(비누, 샴푸, 치약, 항여드름제, 선크림, 탐폰, 기저귀 등)이 약제학적 조성물로 보충될 수 있다.
최종적으로, 본 발명은 치료학적 유효량의 상기 정의된 약제학적 조성물을 환자에게 투여함을 포함하여, 미생물 감염되거나 알레르기로 고통받는 포유동물을 치료하는 방법에 관한 것이다.

항미생물성
펩타이드
펩타이드. 펩타이드는 펩타이드 합성 플랫폼(PEPscreen®, Custom Peptide Libraries, SigmaGenosys)에 의해 생성되는 시그마-게노시스(Sigma-Genosys)사 제품이다. 수율은 약 1 내지 6mg이었고, 펩타이드 길이는 20개 아미노산이었다. 이들 펩타이드를 대상으로 MALDI-ToF 질량 분석을 수행하였다. 20량체의 평균 조 순도는 약 60%이었다. 펩타이드를 96-웰 시험관대 내에 동결건조 상태로 공급하였다. 생물학적 시험 이전에, PEP스크린 펩타이드를 dH20(5mM 원료)로 희석시키고 -20℃에서 저장하였다. 이 원액을 후속 실험을 위해 사용하였다.

방사상 확산 검정
방사상 확산 검정(RDA)을 본질적으로 문헌[참조: Lehrer, R. I., Rosenman, M., Harwig, S. S., Jackson, R. & Eisenhauer, P. 용혈 검정

Claims (23)
- (a) 서열번호 10 및 12로부터 선택되는 제1 세트의 아미노산 서열, 및(b) V, L, I, F, Y 및 W로 이루어진 그룹으로부터 선택된 3개 내지 8개의 소수성 아미노산 잔기들로 이루어진 제2 세트의 아미노산 잔기들을 포함하며, 상기 제2 세트의 아미노산 잔기들이 상기 제1 세트의 아미노산 서열의 아미노 또는 카르복시 말단에 결합되어, 단독의 제1 세트의 아미노산 서열과 비교하여 항미생물 활성이 증가된 항미생물성 펩타이드.
- 제1항에 있어서, 상기 제2 세트가 동일한 소수성 아미노산 잔기들 또는 상이한 소수성 아미노산 잔기들의 혼합물을 포함하는 것인, 항미생물성 펩타이드.
- 제1항에 있어서, 상기 제2 세트가 F(3-8), W(3-8), I(3-8), Y(3-8) 및 V(3-8)로 이루어진 그룹으로부터 선택되는 것인, 항미생물성 펩타이드.

- 제1항에 있어서, 아미노산중 어느 하나가 상응하는 D-아미노산으로 대체될 수 있는 것인, 항미생물성 펩타이드.
- 제1항에 따른 항미생물성 펩타이드, 및 허용가능한 완충제, 희석제, 담체, 보강제 또는 부형제를 포함하는, 세균, 바이러스, 기생충, 진균 및 효모로 이루어진 그룹으로부터 선택되는 미생물에 의해 유발되는 감염의 치료 또는 예방을 위한 항미생물성 조성물.
- 제7항에 있어서, 상기 항미생물성 조성물이 과립제, 산제, 정제, 피복정제, 카테터 및 바늘의 피복물, 캡슐제, 좌제, 시럽제, 에멀젼제, 겔제, 연고제, 분산액제, 현탁액제, 크림제, 에어로졸제, 점적제의 형태 또는 주사가능한 형태인, 항미생물성 조성물.
- 제1항에 따른 항미생물성 펩타이드, 또는 제7항에 따른 항미생물성 조성물을 포함하는 제품으로서, 상기 제품이 붕대, 깁스, 봉합재, 비누, 탐폰, 기저귀, 샴푸, 치약, 항여드름 화합물, 선크림, 직물, 접착제, 상처 드레싱, 세정 용액, 콘택트렌즈 또는 이식물로 이루어진 그룹으로부터 선택되는 제품.
- 제1항 내지 제6항 중의 어느 한 항에 따른 항미생물성 펩타이드를 포함하는, 아토피성 피부염, 농가진, 만성 피부 궤양, 감염성인 급성 및 만성 상처 및 화상 상처, 여드름, 외이염, 진균 감염, 폐렴, 지루성 피부염, 칸디다 간찰진, 칸디다 질염, 구강인두 칸디다증, 안구 감염 및 비강 감염으로 이루어진 그룹으로부터 선택되는 질환 치료용 약제학적 조성물.
