Abstract
본 발명은 신규한 피토스핑고신 유도체 및 이를 포함하는 염증성 피부질환과 자가면역질환 및 피부과다각화증의 예방 및 치료용 조성물에 관한 것으로, 좀 더 상세하게는 상기 화학식 I의 피토스핑고신 유도체 및 이를 포함하는 염증성 피부질환과 자가면역질환 및 피부과다각화증의 예방 및 치료용 조성물에 관한 것이다. 본 발명의 피토스핑고신 유도체가 염증과 자가면역질환 및 피부과다각화증에 관여하는 전사인자, 염증매개물질의 발현과 생성, 신호전달 기작, 효소의 발현과 활성 등에 관여하여 염증성 피부질환과 자가면역질환 및 피부과다각화증의 예방 및 개선, 치료 효능이 기존의 피토스핑고신 보다 뛰어나 경쟁력 있는 의약품의 제조에 효과적이다.
Description
본 발명은 신규한 피토스핑고신 유도체 및 이를 포함하는 염증성 피부질환과 자가면역질환 예방 및 치료용 조성물에 관한 것으로, 좀 더 상세하게는 화학식 I을 가지는 피토스핑고신 유도체 및 이를 포함하는 염증성 피부질환과 자가면역질환 및 피부과다각화증질환 예방 및 치료용 조성물에 관한 것이다.
피토스핑고신 (phytosphingosine)은 진균, 식물 및 인간을 포함하는 동물의 피부에서 풍부하게 존재하는 구조적으로 스핑고신 (sphingosine)의 구조골격을 가지는 지질이다. 특히 피부 표피층의 각질층을 구성하는 지질이중층 (lipid bilayer)의 구성성분인 세라마이드 (ceramide)의 전구체로 작용하여 피부의 수분손실을 방지하여 피부를 촉촉하게 하고 외부의 해로운 물질들이 피부의 진피층으로 쉽게 흡수되는 것을 막아주는 역할을 한다. 또한 체내에 흡수되어 세라마이드의 합성을 촉진하며, 여드름균 (Propionibacterium acnes)과 포도상구균 (Staphylococcus aureus)을 비롯한 여러 미생물에 대한 항균작용이 있어 여드름 등의 치료를 위한 제품에 사용된다.
피토스핑고신과 N-acetyl phytosphingosine (NAPS) 및 tetra-acetyl phytosphingosine (TAPS)와 같은 피토스핑고신 유도체는 피부각질세포의 cyclooxygenase-2 (COX-2)의 발현을 조절하고 caspase-8과 미토콘드리아 활성을 통한 세포사멸을 유도하며, 외부 자극에 대한 피부각질세포의 과증식을 억제하고 피부염증반응을 조절하여 한다. 하지만 간 (liver)과 신장 (kidney)의 손상과 어지러움과 구토를 동반하는 위장관 장애를 초래하고 세포에 대한 독성작용이 강하여 치료제로서의 장점보다는 독성으로 인한 단점이 많아 피부질환 및 기타 질환 치료제로 사용이 어려운 실정이다.
말레인산 (maleic acid)은 푸마르산 (fumaric acid)과 분자식이 같은 기하이성질체로 주스, 사이다와 과일통조림 등 신맛을 내는 산미료로 사용된다. 가열하여 생성되는 말레산무수물은 촉매를 이용하여 다양한 물질과 반응하여 불포화폴리에스테르수지, 스티렌과의 혼성중합체 및 여러 가지 합성화학약품의 제조 등에 널리 사용된다.
본 발명자들은 피토스핑고신과 무수말레인산 또는 푸마르산을 사용하여 피토스핑고신의 단점을 보완하면서 항염증 효과가 있는 피토스핑고신 유도체를 합성하고 염증성 피부질환과 자가면역질환 및 피부과다각화증질환 치료 및 예방 기능에 더 우수한 기능을 보이는 물질 및 이를 이용한 약학적 조성물을 제공하고자 이 발명을 완성하였다.
본 발명자들은 염증성 피부질환과 자가면역질환의 치료제에 대하여 연구하던 중, 피토스핑고신과 무수말레인산을 사용하여 새로운 화합물을 합성하고 신규 화합물이 염증성 피부질환과 자가면역질환 및 피부과다각화증질환 치료 및 예방 기능이 피토스핑고신보다 우수하며, 피토스핑고신의 단점을 보완한 것을 확인하여 본 발명을 완성하였다.
따라서 본 발명의 목적은 하기 화학식 I을 가지는 피토스핑고신 유도체를 제공하는 것이다.

본 발명의 다른 목적은 상기 피토스핑고신 유도체 또는 이의 약학적으로 허용 가능한 염을 유효성분으로 포함하는 염증성 피부질환과 자가면역질환 예방 및 치료용 약학적 조성물을 제공하는 것이다.
본 발명의 다른 목적은 상기 피토스핑고신 유도체 또는 이의 약학적으로 허용 가능한 염을 유효성분으로 포함하는 피부과다각화증 질환(hyperkeratotic disorder) 예방 및 치료용 약학적 조성물을 제공하는 것이다.
본 발명의 다른 목적은 (a) 피토스핑고신, 무수말레인산 또는 푸마르산을 유기용매 내에서 반응시키는 단계; 및 (b) 상기 반응물에서 제1항의 물질을 분리하는 단계를 포함하는 상기 피토스핑고신 유도체 제조방법을 제공하는 것이다.
상기의 목적을 달성하기 위해서, 본 발명은 하기 화학식 I을 가지는 피토스핑고신 유도체를 제공한다.

본 발명은 다른 목적을 달성하기 위해서, 상기의 피토스핑고신 유도체를 유효성분으로 포함하는 염증성 피부질환과 자가면역질환 예방 및 치료용 약학적 조성물을 제공한다.
본 발명은 다른 목적을 달성하기 위해서, 상기의 피토스핑고신 유도체를 유효성분으로 포함하는 피부과다각화증질환(hyperkeratotic disorder) 예방 및 치료용 약학적 조성물을 제공한다.
본 발명은 다른 목적을 달성하기 위해서, (a) 피토스핑고신, 무수말레인산 또는 푸마르산을 유기용매 내에서 반응시키는 단계: 및 (b) 상기 반응물에서 제1항의 물질을 분리하는 단계를 포함하는 상기의 피토스핑고신 유도체 제조방법을 제공한다.
이하 본 발명을 상세히 설명한다.
본 발명은 하기의 화학식 I ((E,Z)-4-oxo-4-((2S,3S,4R)-1,3,4-trihydroxyoctadecan-2-ylamino)but-2-enoic acid; YG-II-6로 명명)을 가지는 피토스핑고신 유도체를 제공한다.

하기 화학식 II로 표시되는 피토스핑고신은 피부 지질이중층을 구성하는 세라마이드의 전구체로 작용하여 피부의 수분 손실 방지와 미생물에 대한 항균작용 및 피부염증 반응을 조절하는 등 각종 방어 작용을 하여 인체에 필요한 성분이지만 강력한 독성작용에 의하여 치료제로서의 장점보다는 독성으로 인한 단점으로 질환치료제로 사용이 어렵다.

그러나 여기에 하기 화학식 III으로 표시되는 무수말레인산 또는 푸마르산을 첨가하면 피토스핑고신의 독성으로 인한 단점을 보완하면서 우수한 염증성 피부질환과 자가면역질환 예방, 치료 및 개선 기능이 우수한 본 발명의 피토스핑고신 유도체가 완성된다.

또한 본 발명의 피토스핑고신 유도체는 피토스핑고신 보다 염증성 피부질환과 자가면역질환 및 피부다각화증질환 예방 및 개선과 치료 효능이 뛰어나다.
본 발명의 일실시예에서 피토스핑고신 유도체가 TPA로 자극한 피부각질세포와 초대배양 피부각질세포에서 NF-B 활성화와 관련된 IB의 인산화와 분해를 억제하여 NF-B의 전사활성 및 인산화와 핵으로의 이동을 억제하는 것을 확인하였다. 이러한 YG-II-6의 NF-B 활성 억제작용은 합성의 전구체로 사용된 phytosphingosine보다 더욱 우수하였다 [도 1].
또한 염증매개물질에 대한 효과에서 본 발명의 피토스핑고신 유도체는 TPA 자극에 의한 산화질소 생성 및 iNOS와 COX-2의 단백질과 유전자의 발현, 그리고 대표적인 염증매개물질인 IL-1a, IL-6와 TNF-a의 발현을 효과적으로 억제하였다. 하지만 IL-1a의 발현을 제외하고는 phytosphingosine은 약하거나 억제효과를 거의 보이지 않았다 [도 2].
JAK/STAT 신호전달 경로에서 사람 유래 피부각질세포와 단핵구 세포 및 마우스 T-세포에서 TPA를 포함한 다양한 사이토카인에 의한 자극에 대하여 각각의 다른 JAK/STAT 신호전달 경로를 활성화시켰을 때 본 발명의 피토스핑고신 유도체는 이들 경로를 모두 억제하는 작용을 보였으며, 억제작용은 phytosphingosine보다 우수한 효과를 보였다[도 3].
MAP kinase에 대한 작용에서는 phytosphingosine은 아주 약하거나 억제작용을 보이지 않았지만 YG-II-6는 ERK1/2와 JNK의 인산화를 강하게 억제하였다 [도 4].
피부질환 동물모델을 이용한 염증성 피부질환에 대한 작용에서 본 발명의 피토스핑고신 유도체는 TPA 자극에 의하여 증가한 피부염증 반응인 표피의 과형성 (epidermal hyperplasia)과 각막비후증 (hyperkeratosis) 및 진피부위로 염증세포의 이동을 현저하게 억제하였으며, 세포사멸을 유도하였다 [도 5].
그리고 IL-23에 의하여 유도된 건선 유사 피부질환에서도 건선 질환의 병변인 표피의 과형성 (epidermal hyperplasia), 각막비후증 (hyperkeratosis)과 진피부위로 염증세포의 이동과 질환과 관련된 사이토카인과 키모카인의 발현을 현저하게 억제하였다 [도 6]. 본 발명의 피토스핑고신 유도체는 대조군으로 사용된 phytosphingosine보다 피부염증 및 건선을 포함하는 자가면역질환에 대하여 우수한 억제작용을 보였으며, 또한 세포에 대한 독성 작용에서는 이들과 비교하여 강한 독성작용을 보이지 않았다.
따라서 phytosphingosine과 maleic anhydride 또는 fumaric acid를 이용하여 신규 합성한 물질인 상기 화학식 I의 피토스핑고신 유도체는 전구체로 사용한 phytosphingosine보다 세포에 대한 독성작용이 약하고 약리작용은 우수하므로 염증성 피부질환과 자가면역질환의 예방, 치료 및 개선을 위한 약학적 조성물 또는 화장품 조성물의 제조에 이용할 수 있다.
본 발명은 상기의 피토스핑고신 유도체 또는 이의 약학적으로 허용가능한 염를 유효성분으로 포함하는 염증성 피부질환과 자가면역질환 및 피부과다각화증질환 (hyperkeratotic disorder) 예방 및 치료용 약학적 조성물을 제공한다.
상기 염증성 피부질환은 이에 한정하지는 않으나, 아토피 피부염, 전신 홍반성 낭창, 접촉성 피부염, 알레르기성 피부질환, 여드름, 좌창 및 두드러기 등을 포함한다.
또한 상기 자가면역질환은 이에 한정하지는 않으나, 류마티스성 관절염, 전신성 경피증, 전신 홍반성 낭창, 천식, 궤양성 대장염, 크론씨병, 베체씨병, 피부근염, 다발성 경화증, 교원병, 혈관염, 관절염, 육아종증, 장기 특이적 자가면역병변, 궤양성 대장염 및 GvHD (이식편대 숙주 반응 질환: graft-versus-host disease) 등을 포함한다.
또한 상기 과다각화증 질환은 이에 한정하지는 않으나, 그로버 질환 (일과성 극세포해리성 피부염), 티눈, 굳은 살, 사마귀, 만성 습진, 편평태선, 광선각화증, 지루성 각화증 및 어린선 등을 포함한다.
과다각화증은 피부의 바깥층이 두터워지는 현상으로, 케라틴이라는 단단한 보호 단백질을 포함하고 있다. 이 현상은 조종 마찰이나 압력, 국소 자극에 대한 정상적인 피부 보호체계의 일부이며, 손과 발에서는 굳은살이나 티눈, 구강내부에서는 백태를 형성한다. 다른 형태의 과다각화증은 만성염증, 감염, 햇빛 노출, 자극적 화학물질 등에 대한 피부 방어현상으로 발생한다. 드물게 자극받지 않는 피부에 발생하는데, 이런 형태는 출산 후에 바로 시작하기도 하고 신체의 넓은 부위에 발생하는 유전성 질환이다.
본 발명에 따른 약학적 조성물은 본 발명의 피토스핑고신 유도체 또는 이의 약학적으로 허용가능한 염을 단독으로 함유하거나 또는 하나 이상의 약학적으로 허용되는 담체, 부형제 또는 희석제를 추가로 함유할 수 있다.
상기에서 약학적으로 허용가능한 염은 생리학적으로 허용되고 인간에게 투여시 통상적인 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 것을 말하며, 상기 염으로는 약학적으로 허용 가능한 유리산(free acid)에 의하여 형성된 산 부가염이 바람직하다. 상기 유리산은 유기산과 무기산을 사용할 수 있다. 상기 유기산은 이에 제한되는 것은 아니나, 구연산, 초산, 젖산, 주석산, 말레인산, 푸마르산, 포름산, 프로피온산, 옥살산, 트리플로오로아세트산, 벤조산, 글루콘산, 메타술폰산, 글리콜산, 숙신산, 4-톨루엔술폰산, 글루탄산 및 아스파르트산을 포함한다. 또한, 상기 무기산은 이에 제한되는 것은 아니나 염산, 브롬산, 황산 및 인산을 포함한다.
약학적으로 허용되는 담체로는 예컨대, 경구 투여용 담체 또는 비경구 투여용 담체를 추가로 포함할 수 있다. 경구 투여용 담체는 락토스, 전분, 셀룰로스 유도체, 마그네슘 스테아레이트, 스테아르산 등을 포함할 수 있다. 또한, 비경구 투여용 담체는 물, 적합한 오일, 식염수, 수성 글루코스 및 글리콜 등을 포함할 수 있으며, 안정화제 및 보존제를 추가로 포함할 수 있다. 적합한 안정화제로는 아황산수소나트륨, 아황산나트륨 또는 아스코르브산과 같은 항산화제가 있다. 적합한 보존제로는 벤즈알코늄 클로라이드, 메틸- 또는 프로필-파라벤 및 클로로부탄올이 있다. 그 밖의 약학적으로 허용되는 담체로는 다음의 문헌에 기재되어 있는 것을 참고로 할 수 있다 (Remington's Pharmaceutical Sciences, 19th ed., Mack Publishing Company, Easton, PA, 1995).
본 발명의 염증성 피부질환과 자가면역질환에 대한 예방 및 치료용 약학적 조성물은 인간을 비롯한 포유동물에 어떠한 방법으로도 투여할 수 있다. 예를 들면, 경구 또는 비경구적으로 투여할 수 있다. 비경구적인 투여방법으로는 이에 한정되지는 않으나 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장내 투여일 수 있다. 바람직하게는 본 발명의 약학적 조성물은 경피투여될 수 있다. 상기에서 경피투여'란 본 발명의 약학적 조성물을 세포 또는 피부에 투여하여 본 발명 조성물에 함유된 활성성분이 피부 내로 전달되도록 하는 것을 말하다. 예컨대, 본 발명의 약학적 조성물을 주사형 제형으로 제조하여 이를 30 게이지의 가는 주사 바늘로 피부를 가볍게 단자(prick)하는 방법, 또는 피부에 직접적으로 도포하는 방법으로 투여될 수 있다.
본 발명의 약학적 조성물은 상술한 바와 같은 투여 경로에 따라 경구 투여용 또는 비경구 투여용 제제로 제형화 할 수 있다.
경구 투여용 제제의 경우에 본 발명의 조성물은 분말, 과립, 정제, 환제, 당의정제, 캡슐제, 액제, 겔제, 시럽제, 슬러리제, 현탁액 등으로 당업계에 공지된 방법을 이용하여 제형화될 수 있다. 예를 들어, 경구용 제제는 활성성분을 고체 부형제와 배합한 다음 이를 분쇄하고 적합한 보조제를 첨가한 후 과립 혼합물로 가공함으로써 정제 또는 당의 정제를 수득할 수 있다. 적합한 부형제의 예로는 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨 및 말티톨 등을 포함하는 다당류와 옥수수 전분, 밀 전분, 쌀 전분 및 감자 전분 등을 포함하는 전분류, 셀룰로즈, 메틸 셀룰로즈, 나트륨 카르복시메틸셀룰로오즈 및 하이드록시프로필메틸-셀룰로즈 등을 포함하는 셀룰로즈류, 젤라틴, 폴리비닐피롤리돈 등과 같은 충전제가 포함될 수 있다. 또한, 경우에 따라 가교결합 폴리비닐피롤리돈, 한천, 알긴산 또는 나트륨 알기네이트 등을 붕해제로 첨가할 수 있다. 나아가, 본 발명의 약학적 조성물은 항응집제, 윤활제, 습윤제, 향료, 유화제 및 방부제 등을 추가로 포함할 수 있다.
비경구 투여용 제제의 경우에는 주사제, 크림제, 로션제, 외용연고제, 오일제, 보습제, 겔제, 에어로졸 및 비강 흡입제의 형태로 당업계에 공지된 방법으로 제형화할 수 있다. 이들 제형은 모든 제약 화학에 일반적으로 공지된 처방서인 문헌 (Remington's Pharmaceutical Science, 15th Edition, 1975. Mack Publishing Company, Easton, Pennsylvania 18042, Chapter 87: Blaug, Seymour)에 기재되어 있다.
본 발명의 피토스핑고신 유도체의 총 유효량은 단일 투여량 (single dose)으로 환자에게 투여될 수 있으며, 다중 투여량 (multiple dose)으로 장기간 투여되는 분할 치료 방법 (fractionated treatment protocol)에 의해 투여될 수 있다. 본 발명의 약학적 조성물은 질환의 정도에 따라 유효성분의 함량을 달리할 수 있다. 바람직하게는 본 발명의 피토스핑고신 유도체의 바람직한 전체 용량은 1일당 환자 체중 1㎏ 당 약 0.01 ug 내지 1,000 mg, 가장 바람직하게는 0.1 ug 내지 100 mg일 수 있다. 그러나 상기 본 발명의 다당 분획물의 용량은 약학적 조성물의 투여 경로 및 치료 횟수뿐만 아니라 환자의 연령, 체중, 건강 상태, 성별, 질환의 중증도, 식이 및 배설율 등 다양한 요인들을 고려하여 환자에 대한 유효 투여량이 결정되는 것이므로, 이러한 점을 고려할 때 당 분야의 통상적인 지식을 가진 자라면 상기 본 발명의 다당 분획물을 면역기능 증진제로서의 특정한 용도에 따른 적절한 유효 투여량을 결정할 수 있을 것이다. 본 발명에 따른 약학적 조성물은 본 발명의 효과를 보이는 한 그 제형, 투여 경로 및 투여 방법에 특별히 제한되지 아니한다.
본 발명의 조성물은 면역기능 저하로 기인되는 질환의 예방 및 치료에 효과가 있으며, 상기 면역기능 저하로 기인되는 질환으로는 감기 등의 감염성 질환 및 염증성 질환, 아토피 등의 알러지 질환, 자가면역질환, 피부과다각화증, 만성피로 및 암으로 이루어진 군으로부터 선택되는 어느 하나인 것이 바람직하나 이에 한정되는 것은 아니며, 당업자에 알려진 면역기능 저하로 기인되는 질환은 모두 본 발명에 포함된다.
상기 염증성 피부질환, 자가면역질환 및 피부과다각화증은 상기에서 기재한 바와 같이 아토피 피부염, 건선, 류마티스성 관절염, 전신성 경피증, 전신 홍반성 낭창, 천식, 궤양성 대장염, 크론씨병, 다발성 경화증, 접촉성 피부염, 알레르기 피부질환, 여드름, 좌창, 두드러기, 만성 습진, 편평태선, 광선각화증, 지루성 각화증 및 어린선 등을 포함한다.
본 발명은 (a) 피토스핑고신과 무수말레인산 또는 푸마르산을 유기용매 내에서 반응시키는 단계: 및 (b) 상기 반응물에서 제1항의 물질을 분리하는 단계를 포함하는 상기의 피토스핑고신 유도체 제조방법을 제공한다.
무수말레인산은 푸마르산과 분자식이 동일한 이성질체로 유기합성 원료로서 중요하며, 가소제, 폴리에스터수지 및 섬유, 도료, 농약, 합성세제 및 피혁 등의 원료로도 쓰인다. 유기합성을 위한 유기용매에는 대표적으로 메탄올, 에탄올, 부탄올, 클로로포름, 디클로로메탄, 에틸아세트산, 헥산, 벤젠 등이 있다. 나열한 용매들과 외의 유기 용매를 한 종류만으로 쓰거나 혼합해서 쓸 수 있다.
본 발명에서는 피토스핑고신과 무수말레인산 또는 푸마르산의 유기합성을 위해서 실온에서 디클로로메탄 (dichloromethane)과 N,N-디메틸포름아마이드 (N,N-dimethylformamide)의 3:1 (v/v, 12.4 ml) 혼합액을 이용했다. 피토스핑고신과 무수말레인산 또는 푸마르산이 반응하도록 실온에서 24시간 교반 후 용매를 감압 증발로 제거하고 남은 유기물을 에테르에 녹여 결정형의 본 발명의 피토스핑고신을 추출하였다.
본 발명은 신규한 피토스핑고신 유도체 및 이를 포함하는 염증성 피부질환과 자가면역질환 및 피부과다각화증질환의 예방 및 치료용 조성물을 제공한다. 본 발명의 피토스핑고신 유도체가 염증과 자가면역질환 및 피부과다각화증질환에 관여하는 전사인자, 염증매개물질의 발현과 생성, 신호전달 기작, 효소의 발현과 활성 등에 관여하여 염증성 피부질환과 자가면역질환 및 피부과다각화증질환의 예방 및 개선, 치료 효능이 기존의 피토스핑고신 보다 뛰어나 경쟁력 있는 의약품 등의 제조에 효과적이다.

<1-1> 피토스핑고신 유도체 합성 방법
피토스핑고신 (phytosphingosine; (2S,3S,4R)-2-aminooctadecane-1,3,4-trio
l) (0.50 g, 1.58 mmol)을 실온에서 디클로로메탄 (dichloromethane)과 N,N-디메틸포름아마이드 (N,N-dimethylformamide)의 3:1 (v/v, 12.4 ml) 혼합액에 녹인 후 무수말레인산 (maleic anhydride, 162.5 mg, 1.58 mmol) 또는 푸마르산 (fumaric acid, 183.4 mg, 1.58 mmol)을 더하고 같은 온도에서 24시간 교반하였다. 반응 종료 후 용매를 감압 증발하고 남은 유기물을 에테르에 용해하여 0oC에서 24시간 동안의 재결정을 과정을 거쳐 흰색 결정형의 상기 목적 화합물 ((E,Z)-4-oxo-4-((2S,3S,4R)-1,3,4-trihydroxyoctadecan-2-ylamino)but-2-enoic acid, 177 mg, 85%, 이하 YG-II-6라 함)을 얻었다.

본 발명의 피토스핑고신 유도체과 phytosphingosine의 염증성 피부질환과 자가면역질환 치료 효능 비교 분석
<2-1> 전사인자 nuclear factor-kB의 신호전달 억제작용 확인
NF-κB는 정상적인 조건에서는 외부 자극에 대한 면역반응에 중요한 역할을 하는 전사인자로 작용하지만 과다하게 활성화되면 특정 유전자의 발현을 증가시키는데, 특히 염증성 프로스타글란딘 및 기타 에이코사노이드를 포함한 염증반응을 초래하는 다양한 매개인자들의 발현을 증가시켜 과다한 면역반응을 활성화시켜 만성적인 염증반응을 포함하는 자가면역질환과 종양의 발병 및 진행과정에 중요한 영향을 초래한다. 따라서 이들 질환과 관련되어 활성화된 NF-B의 활성을 조절하는 것은 여러 가지 생체 염증반응의 초기 과정을 조절하여 다양한 염증질환과 자가면역질환 및 암을 포함하는 질병을 완화시키거나 치료할 수 있는 중요한 표적이 된다.
본 발명에서는 먼저 NF-κB와 결합하여 복합체를 형성함으로써 NF-κB의 활성을 억제하는 단백질인 IκBα의 인산화와 분해에 대한 YG-II-6과 phytosphingosine의 영향을 확인하기 위하여 각각의 화합물을 1시간 처리한 다음 400 nM의 TPA를 15분과 30분 동안 각각 자극하였으며, NF-κB의 인산화 및 세포질과 핵에서의 발현양을 확인하기 위하여 30분과 1시간 동안 자극하였으며, 그 결과는 웨스턴 블럿으로 확인하였다. 그리고 핵으로 NF-κB의 이동 정도를 항체를 이용하여 반응시킨 다음 형광으로 표지하여 현미경으로 직접 관찰하였으며, 전사단계에서의 작용을 확인하기 위하여 NF-κB luciferase reporter가 표지된 플라스미드를 세포에 삽입하여 luciferase assay를 실시하여 luciferase 활성을 측정하였다.
그 결과, [도 1A]에서 볼 수 있듯이 본 발명에 사용된 물질인 YG-II-6와 phytosphingosine은 피부각질세포와 초대배양 피부각질세포에서 TPA 자극에 의하여 증가한 IκBα의 인산화 정도를 웨스턴 블럿으로 확인하였을 때 모두 억제하였으며, 특히 10 uM 농도에서 YG-II-6의 억제효과가 우수하였다.
TPA 자극에 의하여 인산화되고 분해된 IκBα에 의하여 자유롭게 된 NF-κB에 대하여 인산화와 핵으로의 이동 억제에 대한 정도를 웨스턴 블럿과 면역염색으로 확인한 결과인 [도 1B 및 1C]를 보면, YG-II-6와 phytosphingosine 모두 NF-κB의 인산화 정도와 핵으로의 이동을 억제하였으며, 억제 정도는 비슷하거나 10 uM 농도에서 YG-II-6의 억제효과가 우수함을 확인할 수 있다.
또한 전사단계에서 작용하는 정도를 NF-κB에 대한 luciferase 활성으로 확인하였을 때에도 [도 1D]에 나타난 것과 같이 TPA 자극에 의하여 증가된 luciferase 활성이 YG-II-6와 phytosphingosine에 의하여 모두 억제되었지만 YG-II-6의 억제효과가 더욱 우수하였다.
따라서 본 발명에 사용된 YG-II-6과 phytosphingosine은 모두 IκBα의 인산화와 분해를 억제하여 NF-κB의 활성화를 억제하였다. 하지만 신규 합성한 물질인 YG-II-6의 NF-κB 활성 억제효과가 phytosphingosine보다 더욱 우수하다는 것을 확인하였다.
<2-2> 다양한 염증매개물질의 발현과 생성 억제 효능 확인
NF-κB가 활성화되어 발현과 생성을 증가시키는 염증매개인자의 발현과 생성에 대한 YG-II-6와 phytosphingosine의 영향을 확인하기 위하여 물질을 24시간 처리하여 iNOS에 의하여 생성되는 산화질소 (nitric oxide, NO)의 양은 Griess 시약으로, 그리고 염증유발인자로 작용하는 효소인 COX-2와 iNOS의 발현을 웨스턴 블럿으로 확인하였으며, 그리고 화합물을 6시간 처리하여 사이토카인을 포함하는 다양한 염증매개인자들의 발현에 대한 영향을 실시간 중합효소반응으로 측정하였다.
측정 결과 YG-II-6와 phytosphingosine은 모두 산화질소의 생성을 억제하였으며, 특히 10 uM 농도에서는 YG-II-6의 효과가 더욱 우수하였다. 이는 [도 2A]에서 확인할 수 있다. 또한 결과 [도 2B]를 보면, 염증반응과 관련된 효소인 iNOS와 COX-2의 단백질 발현에 대한 작용에서도 YG-II-6는 10 uM에서 이들 효소의 발현을 강하게 억제하였지만 phytosphingosine은 10 uM에서 아주 약한 억제작용을 보였다.
NF-κB 활성화에 의해 발현과 생성이 증가되는 대표적인 염증매개 사이토카인인 IL-1a, IL-1b, IL-6와 TNF-a 및 효소인 COX-2의 RNA 발현에 대한 작용 결과는 [도 2C]에 모두 나와 있다. YG-II-6는 모두 우수한 억제작용을 보였지만 phytosphingosine은 IL-1a를 제외하고는 YG-II-6와 비교하여 상대적으로 약한 억제작용을 보였다. 따라서 YG-II-6는 NF-κB가 활성화되는 것을 억제하여 염증매개인자들의 발현과 생성을 억제함으로써 염증반응을 억제할 수 있음을 확인할 수 있었으며, 대조물질인 phytosphingosine보다 우수한 억제작용을 보였다.

신호전달 억제작용 확인
JAK/STAT 신호전달 경로는 다양한 사이토카인과 같은 다양한 종류의 자극에 의하여 활성화되어 세포내 신호를 전달하여 세포분화, 성장 및 생존 등 다양한 역할을 하게 된다. 하지만 비정상적인 상태에서 생성되는 염증매개인자와 같은 다양한 인자들에 의하여 활성화되면 류마티스 관절염을 포함하는 만성염증질환과 더불어 암 질환 등 다양한 질환을 초래하게 된다. 따라서 질환과 관련된 비정상적으로 활성화된 NF-κB와 더불어 비정상적인 상태에서 활성화되는 JAK/STAT 신호전달도 억제하거나 조절하는 것 또한 염증과 자가면역질환의 예방과 치료에 중요한 역할을 한다.
화합물 YG-II-6와 phytosphingosine의 JAK/STAT 신호전달에 대한 영향을 확인하기 위하여 다양한 세포를 이용하였으며, 이들 세포에 각각의 농도에 해당하는 화합물을 1시간 처리한 다음 피부각질세포와 초대배양 피부각질세포를 400 nM의 TPA로 3시간 [도 3A], 피부각질세포를 100 U/ml의 IFN-g로 15분간 [도 3B], 마우스 CD4+ T-세포를 100 U/ml의 IFN-g로 15분간 [도 3C], 50 ng/ml의 IL-6로 10분간 [도 3D, 위], 20 ng/ml의 IL-4로 10분간 [도 3E],100 ng/ml의 IL-2로 1분간 [도 3F, 위], 사람 말초혈액 단핵구 세포를 50 ng/ml의 IL-6로 10분간 [도 3D, 아래], 또는 100 ng/ml의 IL-2로 1분간 [도 3F, 아래] 각각 자극하였다. JAK과 STAT의 활성화 정도를 확인하기 위하여 세포의 단백질을 분리하고 전기영동하여 웨스턴 블럿으로 인산화 정도를 확인하였다.
사람유래 피부각질세포와 사람피부에서 분리한 초대배양 피부각질세포를 400 nM의 TPA로 자극하였을 때 [도 3A]에 나와 있듯이 STAT3은 인산화가 증가하였지만 YG-II-6은 STAT3의 인산화를 효과적으로 억제하였다. 하지만 phytosphingosine은 억제작용을 거의 보이지 않았다.
사람유래 피부각질세포를 IFN-g로 자극하였을 때에도 [도 4B]를 보면, YG-II-6는 TPA 자극에서와 비슷하게 STAT1과 STAT3의 인산화를 억제하였다. 또한 이들 STAT의 상위신호전달 단계에서 작용하는 JAK1과 JAK2의 인산화도 억제하였다. 하지만 phytosphingosine은 이들에 대하여 별다른 억제작용을 보이지 않았다. 그리고 마우스 CD4+ T-세포에서도 IFN-g 자극에 의한 STAT1과 STAT3의 인산화에 대한 작용을 YG-II-6는 효과적으로 억제하였으며, 이들에 대한 억제작용은 phytosphingosine의 작용보다 우수하였다 [도 3C].
대표적인 염증신호전달 사이토카인인 IL-6 자극에 의한 JAK1, JAK2와 TYK2 경로를 통하여 활성화되는 STAT1과 STAT3도 마우스 CD4+ T-세포에서 YG-II-6는 효과적으로 억제하였지만 phytosphingosine은 STAT1에 대해서는 YG-II-6와 비슷하지만 STAT3에 대해서는 별다른 억제작용을 보이지 않음을 [도 3D, 위]에서 확인할 수 있다. 그리고 사람혈액에서 분리한 말초혈액 단핵구 세포에서도 YG-II-6는 STAT3의 인산화를 phytosphingosine보다 우수하게 억제하였다 [도 3D, 아래].
JAK의 다른 종류의 하나인 JAK3 경로를 통한 STAT의 억제작용을 확인하기 위하여 마우스 CD4+ T-세포를 IL-4로 자극하여 JAK1과 JAK3에 의하여 활성화되는 STAT6의 인산화에 대한 영향을 확인하였을 때 YG-II-6는 phytosphingosine보다 우수한 억제작용을 보였으며, phytosphingosine은 별다른 억제작용을 보이지 않았다 [도 3E].
또한 마우스 CD4+ T-세포와 사람 말초혈액 단핵구 세포를 IL-2로 자극하여 활성화된 JAK1과 JAK3 경로를 통하여 하위 신호전달 경로인 STAT5의 인산화 정도를 확인하였을 때에도 YG-II-6는 STAT5의 인산화를 phytosphingosine보다 비슷하거나 우수한 억제작용을 보였다 [도 3F].
JAK/STAT 신호전달에 대한 상기의 결과들을 종합해 볼 때 YG-II-6는 phytosphingosine보다는 상대적으로 우수한 억제작용을 하는 것을 알 수 있다.

MAP
kinase
에 대한 억제 효과 확인
MAP kinase 경로는 세포의 분화, 성장 및 생존 등을 조절하는 중요한 역할을 하지만 염증질환을 포함하는 다양한 질환에서 활성화되는 것이 확인된다.
이들 MAP kinase에 대한 화합물의 영향을 확인하기 위하여 각각의 화합물을 사람유래 피부각질세포에 1시간 처리한 다음 400 nM의 TPA를 15분간 자극하여 웨스턴 블럿으로 이들의 인산화 정도를 확인하였다.
확인 결과 [도 4]에서 볼 수 있듯이 YG-II-6는 ERK1/2와 JNK의 인산화를 효과적으로 억제하였지만 p38의 인산화에는 별다른 영향을 주지 못하였다. 하지만 phytosphingosine은 p38의 인산화에 대하여 YG-II-6보다 우수한 억제작용을 보였지만 ERK1/2와 JNK에 대하여는 아주 약하거나 별다른 억제작용은 보이지 않았다.

TPA
자극에 의한
피부염증
질환과 세포사멸에 대한 효과 확인
피부자극으로 초래되는 염증반응 동물모델을 위하여 TPA를 마우스 피부에 자극하였다. 먼저 C57BL/6 마우스의 등에 있는 털을 제거하고 다음날 각각의 화합물 시료 100 ul를 털을 제거한 등 부위에 도포하였다. 그리고 1시간 후에 아세톤에 녹인 8.1 mM 농도의 TPA 100 ul로 시료를 도포한 부위에 자극하였다. 그리고 24시간 후에 다시 동일한 방법으로 한 번 더 시료와 TPA를 도포하고 1시간 후에 마우스의 피부를 채취하였다. 채취한 피부는 피부의 두께와 부종의 정도를 측정하고 hematoxylin and eosin (H&E) 염색을 통하여 조직병리학적인 변화를 관찰하였다. 그리고 피부에서 세포의 사멸 정도를 fluorescein isothiocyanate (FITC)로 염색하여 확인하였다.
또한 세포의 사멸에 대한 화합물의 효과를 확인하기 위하여 피부각질세포와 초대배양 피부각질세포를 시료를 처리하고 TPA로 24시간 자극하여 FITC와 TUNEL assay를 진행하여 평가하였다. 세포사멸과 관련된 단백질의 발현에 대한 화합물의 영향을 확인하기 위하여 농도의 시료를 각각 처리하고 TPA로 24시간 자극하여 단백질 발현 정도를 웨스턴 블럿으로 확인하였다.
TPA 자극에 의하여 마우스 피부는 전형적인 염증반응을 초래하였다. 즉 피부각질세포의 증식으로 피부가 두꺼워지는 표피의 과형성 (epidermal hyperplasia = 극세포증, acanthosis)과 각막비후증 (hyperkeratosis) 및 진피부위로 염증세포의 이동이 현저하게 증가되었다.
하지만 [도 5A와 5B]를 보면, 화합물 시료의 처리에 의하여 염증반응과 관련된 이러한 조직병리학적 변화는 효과적으로 억제되었는데, 특히 YG-II-6의 억제작용이 phytosphingosine과 비교하여 현저하게 우수하였다.
세포사멸에 대한 시료의 효과를 확인하기 위하여 채취한 피부조직에 대한 FITC 염색과 시료를 처리한 세포에서는 FITC와 TUNEL assay를 진행한 결과인 [도 5C 및 5D]를 확인한 바, YG-II-6와 phytosphingosine은 모두 TPA를 자극한 마우스의 피부조직에서 세포사멸을 유도하였으며, 이들의 효과는 YG-II-6가 더 우수함을 알 수 있다.
세포사멸과 관련된 단백질 발현에서는 YG-II-6와 phytosphingosine은 caspase-3와 caspase-9의 단편화 증가 및 cellular inhibitor of apoptotic protein (cIAP)-1과 cIAP-2, Bax, Bcl-2, 그리고 Cyclin A의 발현을 감소시켰다. 이는 [도 5E]에서 확인할 수 있다.
상기의 결과들을 종합해본 바, YG-II-6는 TPA 자극에 의한 피부 염증반응의 억제와 세포사멸 작용에 대하여 phytosphingosine보다 우수함을 알 수 있다.

IL-23을 마우스 귀에 주입하면 자가면역질환의 하나인 건선과 유사한 질환이 유도된다. 따라서 건선질환에 대한 화합물의 영향을 확인하기 위하여 IL-23을 이용하여 건선유사 질환을 유도하였다. 즉 500 ug의 IL-23를 10 ul 용량으로 C57BL/6 마우스의 귀에 2일에 1회 주사기로 피내에 주사하였으며, 12일간 모두 7회를 주입하였다. 그리고 마지막으로 자극 후 24시간이 경과한 다음날에 귀의 두께를 측정하였으며 마우스의 귀를 채취하여 조직촬영과 RNA를 분리하여 건선질환과 관련된 유전자의 발현을 실시간 중합효소 반응으로 확인하였다.
[도 6A와 6B]를 보면, 마우스 귀의 두께가 PBS를 투여한 정상군과 비교하여 IL-23 자극에 의하여 현저하게 두꺼워졌음을 알 수 있다.
H&E 염색에 의한 조직검사 결과 건선 질환의 병변으로 예상되는 표피의 과형성 (epidermal hyperplasia = 극세포증, acanthosis), 각막비후증 (hyperkeratosis)과 진피부위로 염증세포의 이동이 현저하게 증가되었지만 YG-II-6는 이러한 조직병리학적인 건선질환 병변의 변화를 효과적으로 억제하였다. 이는 [도 6B]에서 확인할 수 있다.
그리고 IL-23 자극에 의하여 건선 질환과 관련된 다양한 사이토카인 (cytokine)과 키모카인 (chemokine) 등 다양한 질환매개 염증인자들의 발현이 현저하게 증가하였지만 YG-II-6는 이러한 유전자의 발현을 현저하게 억제하였다. 다양한 염증인자들의 발현 억제 결과를 모아놓은 [도 6C]를 보면, 이러한 효과를 확인할 수 있다.
이상 살펴본 바와 같이, 본 발명은 신규한 피토스핑고신 유도체 및 이를 포함하는 염증성 피부질환과 자가면역질환 및 피부과다각화증에 대한 예방 및 치료용 조성물을 제공한다. 본 발명의 피토스핑고신 유도체가 염증과 자가면역질환 및 피부과다각화증에 관련된 전사인자, 염증매개물질, 신호전달 기작, 효소 등의 발현과 생성 및 활성화를 억제하는 반응에 관여하여 산업상 이용가능성이 우수하다.
Claims (9)
- 제1항의 피토스핑고신 유도체 또는 이의 약학적으로 허용 가능한 염을 유효성분으로 포함하는 염증성 피부질환 예방 및 치료용 약학적 조성물.
- 제2항에 있어서, 상기 염증성 피부질환은 아토피 피부염, 전신 홍반성 낭창, 접촉성 피부염, 알레르기 피부질환, 여드름, 좌창 및 두드러기로 이루어진 군에서 선택되는 것을 특징으로 하는 조성물.
- 제1항의 피토스핑고신 유도체 또는 이의 약학적으로 허용 가능한 염을 유효성분으로 포함하는 자가면역질환 예방 및 치료용 약학적 조성물.
- 제4항에 있어서, 상기 자가면역질환은 류마티스성 관절염, 전신성 경피증, 전신 홍반성 낭창, 건선, 천식, 궤양성 대장염, 베체씨병, 크론씨병, 다발성 경화증, 피부근염, 교원병, 혈관염, 관절염, 육아종증, 장기 특이적 자가면역병변, 궤양성 대장염 및 GvHD (이식편대 숙주 반응 질환: graft-versus-host disease)으로 이루어진 군에서 선택되는 것을 특징으로 하는 조성물.
- 제1항의 피토스핑고신 유도체 또는 이의 약학적으로 허용 가능한 염을 유효성분으로 포함하는 과다각화증 질환 (hyperkeratotic disorders) 예방 및 치료용 약학적 조성물.
- 제6항에 있어서, 상기 과다각화증 질환은 그로버 질환 (일과성 극세포해리성 피부염), 티눈, 굳은 살, 사마귀, 만성 습진, 편평태선, 광선각화증, 지루성 각화증 및 어린선으로 이루어진 군에서 선택되는 것을 특징으로 하는 약학 조성물.
- (a) 피토스핑고신과 무수말레인산 또는 푸마르산을 유기용매 내에서 실온에서 24시간 교반하는 단계; 및
(b) 상기 (a)단계에서 교반된 물질을 감압 증발하여 용매를 제거하는 단계;
를 포함하는 제1항의 피토스핑고신 유도체 제조방법. - 제8항에 있어서, (a)단계의 유기용매가 디클로로메탄 (dichloromethane)과 N,N-디메틸포름아마이드 (N,N-dimethylformamide)의 3:1 (v/v) 혼합액인 것을 특징으로 하는 제조방법.
