Description
Claims (6)
- 다음의 단계를 포함하는 표적 유전자 변형된 원핵세포 생산방법:(a) 코리네박테리아 복제원점(replication origin), 선별마커 및 표적 유전자에 상동인 서열을 포함하는 표적 유전자 변형 벡터를 준비하는 단계;(b) 상기 벡터에서 복제원점을 제거하고 원형으로 연결하는 단계;(c) 상기 복제원점이 제거된 벡터를 원핵세포에 삽입하여 형질전환 미생물을 준비하는 단계;(d) 상기 형질전환 미생물을 선별마커가 함유된 배지에서 배양하는 단계; 및(e) 상동 재조합에 의하여 표적 유전자가 변형된 세포를 선택하는 단계.
- 제1항에 있어서, 단계 (a)의 벡터가 수탁번호 KCCM 10618로부터 유래되는 것인 방법.
- 제1항에 있어서, 표적 유전자가 트랜스케토라제 (tkt: transketolase), 포스포프럭토키나제 (pfk: phosphofructokinase) 또는 말레이트 디하이드로게나제 (mdh: malate dehydrogenase)인 방법.
- 제1항에 있어서, 원핵세포가 에스케리키아(Escherichia), 브레비박테리움 (Brevibacterium) 또는 코리네박테리움 (Corynebacterium)인 방법.
- 제1항의 방법으로 생산된 표적 유전자 변형된 원핵세포.
- 수탁번호 KCCM 10618의 벡터.
