Description
Claims (14)
- 핵산을 포함하는 시료로부터 핵산을 분리하는 방법으로서,실란화된 고상 물질에 시료를 접촉시켜서 핵산을 캡쳐하는 단계; 및핵산이 캡쳐된 상기 고상 물질을 pH 9 내지 pH 14의 염기성 용액으로 처리하는 단계를 포함하는 것을 특징으로 하는 핵산의 분리 방법.
- 제 1 항에 있어서, 염기성 용액으로 처리한 후, 40 ℃ 내지 100 ℃의 온도로 가열하는 단계를 더 포함하는 것을 특징으로 하는 핵산의 분리 방법.
- 제 1 항 또는 제 2 항에 있어서, 상기 핵산을 포함하는 시료가, 이중가닥 DNA, 단일가닥 DNA, 플라스미드 DNA, 및 RNA로 구성된 군으로부터 선택된 한 가지 이상을 포함하는 것을 특징으로 하는 핵산의 분리 방법.
- 제 1 항 또는 제 2 항에 있어서, 상기 고상 물질이, 실리카, 글래스, 플라스틱, 실리콘, 산화 철, 산화 알루미늄, 산화 티탄, 또는 산화 지르코늄인 것을 특징으로 하는 핵산의 분리 방법.
- 제 1 항 또는 제 2 항에 있어서, 상기 고상 물질이, 비드, 필라, 또는 평판 형태인 것을 특징으로 하는 핵산의 분리 방법.
- 제 1 항 또는 제 2 항에 있어서, 상기 실란화된 고상 물질이, 그 표면에 음이온 교환체를 포함하는 것을 특징으로 하는 핵산의 분리 방법.
- 제 6 항에 있어서, 상기 음이온 교환체가, 모노알킬아미노, 디알킬아미노, 4차 알킬아미노기를 갖는 것을 특징으로 하는 핵산의 분리 방법.
- 제 6 항에 있어서, 상기 음이온 교환체가, 상기 실란화된 고상 물질의 표면에 아미드 결합으로 연결된 베타인인 것을 특징으로 하는 핵산의 분리 방법.
- 핵산을 포함하는 시료로부터 핵산을 증폭하는 방법으로서,실란화된 고상 물질에 시료를 접촉시켜서 핵산을 캡쳐하는 단계;핵산이 캡쳐된 상기 고상 물질을 pH 9 내지 pH 14의 염기성 용액으로 처리하는 단계; 및,상기 처리 결과물에 핵산증폭용액을 가하여 핵산 증폭반응을 수행하는 단계를 포함하는 것을 특징으로 하는 핵산의 증폭 방법.
- 제 9 항에 있어서, 염기성 용액으로 처리한 후, 40 ℃ 내지 100 ℃의 온도로 가열하는 단계를 더 포함하는 것을 특징으로 하는 핵산의 증폭 방법.
- 제 9 항 또는 제 10 항에 있어서, 상기 염기성 용액 처리 후의 용액부피에 대해 실란은 0.0001%(v/v) 이상 내지 1%(v/v) 미만으로 함유되어 있는 것을 특징으로 하는 핵산의 증폭 방법.
- 제 9 항 또는 제 10 항에 있어서, 상기 실란화된 고상 물질이, 그 표면에 음이온 교환체를 포함하는 것을 특징으로 하는 핵산의 증폭 방법.
- 제 12 항에 있어서, 상기 음이온 교환체가, 모노알킬아미노, 디알킬아미노, 4차 알킬아미노기를 갖는 것을 특징으로 하는 핵산의 증폭 방법.
- 제 12 항에 있어서, 상기 음이온 교환체가, 상기 실란화된 고상 물질의 표면에 아미드 결합으로 연결된 베타인인 것을 특징으로 하는 핵산의 증폭 방법.
