Abstract

Description
본 발명은 벤조페논을 함유하는 살균제 조성물 및 그 살균 방법에 관한 것이다.
값이 적당하고 영양분도 많으며 식료품 가게의 자판에서 쉽게 구입할 수 있기를 원하는 대중의 음식에 대한 커져가는 요구사항을 충족시키기 위해 다양한 농업기술에 의존하여 식품이 생산된다. 살균제는 전 세계적으로 이용가능한 이들 농업 기술중 하나이다. 살균제는 진균류 및 진균류의 질병으로부터 작물 및 식품을 보호하는 농약 화합물이다. 작물 및 식품은, 방제되지 않은채 남아있는다면, 작물을 황폐화시키고 수확물을 파괴시킬 수 있는 다양한 진균류에 의해 끊임없이 위협받는다.

하기 일반식의 화합물

R'이 H인 일반식 IX의 화합물은 승온(예를 들면 60-150℃)에서 탄산칼륨과 같은 염기성 화합물의 존재하에 저급 알코올에 알킬할로겐화합물(이때 알킬 부분은 치환될 수 있다)과 반응할 수 있다.
다른 위치의 히드록시 기의 경우(VIII에서 R'=OH 또는 X로 부터 얻은 반응 생성물에서와 같이)는 예를 들면 수산화 칼륨과 히드록시 화합물을 반응시킴으로써 금속과의 염이 생성어야만 한다. 그리고나서 염을 물 없이 극성 용매(예를 들면 디메틸포름아미드)내에서 임의로 치환된 알킬 할로겐화물과 반응시킨다.

디하이드록시 화합물 IX (R'=OH)을 디할로겐화합물과 반응시키는 것은 과량의 탄산칼륨과 촉매로서 소량의 산화구리가 존재하는 상태에서 약 10∼50℃ 사이, 바람직하게는 실온에서 실행된다.
아실화된 화합물을 제조하기 위해서 당해 하이드록시 화합물, 예컨대 일반식XII 화합물을

디메틸포름아미드 같은 불활성 극성용매내의 염(예컨대 칼륨)형태로 약 10˚ ∼50℃ 온도에서 임의로 치환된 산 염화물과 반응시킨다.
약 80˚ ∼120℃ 사이의 온도에서 불활성 용매가 있거나 또는 없는 상태에서 일반식 IX (R'=H)화합물을 산 무수물로 가열하면 이 화합물을 아실화시킬 수 있다.

일반식 XIV 의 5-니트로 화합물은 (R=NO2)
5-위치가 치환되지 않은 당해 화합물 (R=H)을 약 50-100℃ 에서 진한(65%) 질산으로 질화시켜 제조할 수 있다.
약 30-60℃ 에서 진한 (65%) 질산으로 진화시키면 일반식 XV 화합물을 2-위치에서 질화시킬 수 있다.
이렇게하여 만들어진 또는 달리 제조된 니트로 화합물은 높은 온도(60-100℃)에서 물/아세트산 50:1 혼합물 내에 있는 과량의 분말 철을 사용하여 예컨대 일반식 XVI의 당해 아미노 화합물로 환원될 수 있다.
환류온도에서 아미노화합물을 과량의 포름산과 반응시키면 아미노그룹이 포르밀화된다.
일반식 XIV 화합물 (R=H)을 10-30℃ 트리클로로메탄에 넣어만든 용액에 동몰량의 브롬 (예컨대 트리클로로메탄내에 들어있는)을 적가하면 이 화합물의 5-위치에 브롬화시킬 수 있다.

할로겐화 마그네슘이 알데히드와 반응할때 즉, 기타 그룹 Z1또는 Z2가 CHO 를 나타낼때 급속한 반응 생성물은 일반식 V 의 3 차 알콜이다 :
이 일반식 V 중간 생성물은 촉매가 존재하는 상태에서 Mn (IV), Mn(VII), Ce (IV) 또는 Cr (VI) 유도체, 질산 또는 산소를 적당히 이용하여 산화시키면 X 가 산소원자인 원하는 일반식 I 의 벤조페논 유도체로 쉽게 전환된다.

일반식 Ib 화합물에서, 치환체 R1, R2, R3, R6및 n은 일반식 I 및 Ia에 대해 상기한 바와 같고 m은 0 또는 1, 2 또는 3의 정수이다. 일반식 Ib의 옥심은 통상적인 알킬화 및 아실화 방법을 이용하여 O-알킬화 또는 O-아실화된 것일 수 있다.
본 발명의 방법에 따라 생상된 벤조페논의 치환체들은 수소화, 아실화, 에테르 결합의 파괴, 알킬화 또는 질화법 등과 같은 확립된 방법 또는 틀에 박힌 방법에 따라 더 유도될 수 있다.
본 발명의 일반식 Ia 화합물은 훌륭한 살균제이며 특히 농업 또는 관련분야의 식물 병변성 진균류를 제어하는데 특히 효과적인 살균제이다. 이 물은 분말 백분병 특히 에리시페 그라미니스, 포도스페라 류코트리카 또는 운실눌라 니케이터를방제하는데 유용하다. 훌륭한 작물 내성으로 인해 이 화합물은 방제가능한 진균류에 의해 오염되는 것이 바람직하지 않은 작물, 예컨대 낱알이 작은 곡물, 사과, 포도 등을 재배하는데 이용될 수 있다. 본 발명의 또 다른 일면은 목적하는 작물이 진균류 방제율에서 식물 독성이 없는 것이다.
본 발명은 또한 상술한 바와 같은 일반식 I 또는 Ia 화합물과 농업적으로 사용할 수 있는 담체로 구성된 살균 조성물을 제공한다. 이 조성물은 본 발명의 하나 이상의 화합물을 함유한다. 바람직하게, 본 발명의 조성물내에 있는 최소한 하나의 담체는 계면활성제이다. 예컨대 이 조성물은 최소한 두개의 담체를 함유하는데 이중 최소한 하나는 계먼활성제이다.
일반식 I 또는 Ia 화합물은 기술적인 물질로서 사용되지만 상기 화합물은 이것이에 제약용으로 공지된 부형약 및 보조물로 구성된 조성물로서 사용되며 예컨대 유제 농축액, 직접 또는 희석하여 분무되는 용액, 희석된 유제, 습윤가능한 분말, 가공성 분말, 더스트, 과립, 공지된 방법으로 만든 마이크로캡슐등으로 제조될 수 있다. 필요로 하는 목적 및 주어진 환경에 따라서 조성물을 스프레잉, 아토마이징, 분산, 붓기 등과 같은 적당한 형태로 사용된다.
일반식 I 또는 Ia 화합물은 단독으로 또는 하나 이상의 농약 또는 식물 생장 조절제와 합쳐져 조제되거나 또는 살포될 수 있다. 사용되는 농약에는 제초제, 살충제 또는 기타 살균제 또는 이들의 배합물이 있다. 일반식 I 또는 Ia 화합물이 다른 농약(들)과 배합되어 사용될 때 이들은 동시에 또는 후속하여 사용된다. 일반식 I 화합물과 배합되어 사용될 수 있는 사용가능한 살균제에는 4,6-디니트로-o-크레졸벤아락실, 베노밀, 캅타플, 캅탄, 카벤다짐, 크로로탈로닐, 구리, 싸이목산일, 디클로로플루아니드, 디클론, 디페노코나졸, 디메토모프, 디니콘졸, 디노켑, 디티아논, 펜피클로닐, 펜프로피오몰프, 하이막사졸, 이마잘릴, 이프로디온, 이소프로티올란, 카수가마이신, 만코제브, 메프로닐, 수은 산화물, 옥시딕실, 옥솔린산, 펜코나졸, 프로피네브, 피리페녹스, 티아벤자졸, 티람, 틀클로포스-메틸, 트리아디메폰, 트리플루미졸, 트리포린 벨리다아이신 A, 빈클로졸린, 지네브, 지람등이 있다.
본 발명의 살균 조성물은 활성 성분들을 충진제, 용매, 고체 담체 및 임의의 계면 활성 화합물(벤사이드)와 강력하게 혼합 및/또는 분쇄시키는 이미 공지된 방법에 의해 제조될 수 있다.
용매는 방향족 탄화수소, 바람직하게는 크실렌 또는 크실렌 혼합물, 치환된 나프탈렌, 디부틸 또는 디옥틸 프탈레이트같은 프탈산 에스테르같은 C8-C12방향족 탄화수소, 시클로헥산 또는 파라핀같은 지방족 탄화수소, 알콜 및 글리콜 뿐 아니라 이들의 에테르 및 에스테르, 예컨대 에탄올, 에틸렌글리콜 모노- 및 디메틸에테르, 시클로헥사논같은 케톤, N-메틸 2-피롤리돈 같은 강한 극성 용매, 디메틸설폭사이드, 알킬 포름아미드, 에폭시화된 시기물성 오일, 예컨대 에폭시화된 코코넛 또는 간장콩 오일, 물일 수 있다.
더스트 또는 분산가능한 분말로 사용될 수 있는 고체 담체는 방해석, 활석, 고령토, 몬트모릴로나이트, 아타플가이트같은 광물성 충진제일 수 있다. 크게 분산되는 실리카게 또는 크게 분산되는 중합체를 부가하면 물리적 성질을 향상시킬 수있다.
과립으로 만들기 위한 담체는 다공성 물질 예컨대 부석(pumice), 분쇄된 벽돌, 해포석, 덴토나이트일 수 있으며 비수착성 담체는 방해석 또는 모레이다. 부가로, 물로마이트 또는 분쇄된 식물 잔류물같은 미리 과립화된 여러가지 무기 또는 유기 물질을 사용할 수도 있다.
적당한 계면활성 물질은 조제된 벤조페논 화합물의 성질에 따라서 분산성, 유화성 및 습윤특성이 좋은 비이온성, 음이온성 또는 양이온성 벤사이드(tenside)일 수 있다.
적당한 벤사이드는 소위 수용성 비누 뿐만 아니라 수용성 합성 계면활성 화합물로 부르기도 한다. 비누는 일반적으로 고급 지방산(C10-20)의 알카리, 알카리 금속 또는 임의로 치환된 암모늄염, 예컨대 올레인산 또는 스테아르산의 나트륨 또는 칼륨염 또는 코코넛 오일이나 탈로우 오일같은 것으로 만든 천연 지방산 혼합물들의 나트륨 또는 칼륨염이다. 이에 더하여, 지방산의 메틸 타우린염을 사용할 수도 있다. 그러나, 소위 합성 텐사이드를 사용하는 것이 바람직한데 특히 지방설포네이트, 지방설페이트, 술폰화된 벤즈이미다졸 유도체 또는 알킬 아릴 술포네이트가 바람직하다. 지방 설페이트 또는 지방술포네이트 보통 알카리, 알카리토 또는 임의로 치환된 암모늄염으로서 사용되며 C8-22의 알킬 부분을 가지는데 여기서 알킬이란 천연 지방산에서부터 만든 지방 알콜의 혼합물의 또는 황산 도데실레이트의 또는 리그닌 슬폰산의 나트륨 또는 칼슘염 같은 아실 잔기의 알킬 부분을 의미한다. 여기에는 또한 황산 에스테르의 염, 술폰산, 지방 알콜과 산화에틸렌 부가물이 포함된다.
슬폰화된 벤즈이미다졸 유도체들은 바람직하게 2개의 술폰산 잔기와 C8-22의 지방산 잔기를 함유한다. 알킬 아릴 술포네이트는 예를 들면 도데실 벤젠 술폰산의, 디부틸 나프탈렌 술폰산의 또는 나프탈렌 술폰산과 포름알데히드의 축합물의 나트륨, 칼슘 또는 트리에틸 암모늄염이다. 이외에도 p-노닐페놀-(4-14)-산화에틸렌 부가물의 인산 에스테르 또는 포스포리피드의 염과 같은 포스페이트를 사용할 수도 있다.
비이온성 텐사이드는 바람직하게 지방족 탄화수소 잔기내에 3-10개의 글리콜 에테르 그룹과 C8-20을 가지며 알킬 페놀의 알킬 잔기내에 C6-18을 갖는 지방족 또는 지환족 알콜, 포화 또는 불포화된 지방산 및 알킬 페놀의 폴리글리콜 에테르 유도체이다. 기타 적당한 비이온성 텐사이드는 수용성의 20-250 에틸렌 글리콜 에테르 기를 함유하는 산화에틸렌과 폴리프로필렌 글리콜의 폴리부가물, 에틸렌 디아미노 폴리프로필렌 글리콜 및 알킬 부분에 C1-10을 갖는 알킬 폴리프로필렌글리콜인데 이 물질들은 보통 폴리프로필렌 글리콜 단위당 1-5개의 에틸렌 글리콜 단위를 함유한다. 비이온성 텐사이의 예로는 노닐페놀 폴리에톡시 에탄올, 캐스터 오일 폴리글리콜 에테르, 산화에틸렌과 폴리프로필렌의 폴리부가물, 트리부틸페녹시 폴리에톡시 에탄올, 폴리에틸렌 글리콜, 옥틸 페녹시 폴리에톡시에탄올이 있다. 이외에도 폴리옥시에틸렌 솔비탄 트리올레이트같은 폴리옥시 에틸렌 솔비탄의 지방산 에스테를사용하기도 한다.
양이온 텐시드는 바람직하게 4차 암모늄염이며, 상기는 8-22개의 탄소원자를 갖는 하나 이상의 알킬 잔류물을 가지며 또한 저급, 임의로 할로겐화된 알킬, 벤질 또는 히드록시알킬 잔류물을 갖는다. 염은 바람직하게 할로겐화물, 메틸 설페이트 또는 알킬 설페이트, 예를 들면 스테아릴 트리메틸암모늄 클로라이드 또는 벤질 비스(2-클로로에틸) 에틸 암모늄 브로마이드이다.
본 발명의 조성물에 일반적으로 사용되는 텐시드는 하기와 같은 문헌에 공개되어 있다; "McCutheon's Detergents and Emulsifiers Annual", MC Publishing corp., Ridgewood, NJ, USA 1981; H. Stache, "tensid-Taschenbuch", 2nd ed., C. 본 발명의 살충제 조성물은 0.1%-95%, 바람직하게 0.1%-80%의 하나 이상의 일반식 I 또는 Ia 화합물, 1%-99.9%의 고체 또는 액체 보조제 및 0%-25%, 바람직하게 0.1%-25%의 텐시드로 구성될 수 있다.

편의에 따라 본 발명의 진균제 조성물은 바람직하게 농축된 형태일 수 있으며 반면 최종-사용자는 일반적으로 휘석된 조성물을 사용한다. 상기 조성물은 활성 성분 (a.i.)0.001%의 농도로 희석될 수 있다. 사용량은 보통 0.01-10 kg a.i/ha 범위이다.
상기 조성물은 또한 다른 보조제 예를 들면, 안정화제, 소포제, 점도 조절제, 농화제, 접착제, 비료 또는 특별한 효과를 얻기 위한 다른 활성 성분으로 구성될 수 있다.

2,6-디클로로-4',5'-디메톡시-2'-메틸벤조페논 (화합물 1)

4-메틸-테라트롤 (76.1 g; 500 mmol), 2,6-디클로로벤조일 클로라이드(120.4 g; 575 mmol) 및 삼염화철 (III) (0.5 g)의 혼합물을 저어주면서 가열한다. 반응은 염화수소의 형성하에 90℃에서 시작하고, 주 반응은 95℃에서 10 분 내에 완결된다. 후속해서, 반응 혼합물을 100℃에서 30분 더 교반하고나서 65℃로 식힌다. 메탄올 (350 ml)을 첨가하면 화합물 1은 결정화되기 시작한다. 그리고나서 물/메탄올 혼합물 (1:1 부피/부피; 300 ml)을 40℃에서 서서히 첨가하고 혼합물을 30분 동은 교반하면서 실온으로 식힌다. 고체 물질을 진공 여과하여 수집하고, 메탄올/물 (3:1 부피/부피; 각각 100 ml)를 사용하여 3회 세척하고 건조시켜 융점 101.5℃인 무색 결정 148.6 g (수율 91.4%)을 얻는다.

벤조페논 유도체
A) 2,6-디클로로-4',5'-디메톡시-2'-니트로-벤조페논 (화합물 2)

실시예 1과 유사하게 제조된 2,6-디클로로-3,4'-디메톡시벤조페논 (6.22g, 20 mmol)의 일부를 15분 이내에 40℃ 가열된 질산 (65%; 40 ml) 내로 첨가한다. 맑은 용액을 40℃에서 10분 동안, 그리고나서 실온에서 1 시간 동안 교반한다. 그리고나서 서서히 응고하는 오일 형태인 반응 혼합물을 물 내로 붓는다. 이 물질을 열을 가하면서 소량의 N,N-디메틸 포름아미드 내로 용해시키고나서, 메탄올을 첨가하고 혼합물을 냉각시키고 여과하여 융점이 143℃인 노란색 결정의 화합물 2를 5.57 g (수율 78%) 얻는다.
B) 2'-아미노-2,6-디클로로-4',5'-디메톡시벤조페논 (화합물 3)

2,6-디클로로-3',4'-디메톡시-2'-니트로벤조페논 (화합물 2; 3.56 g, 10 mmol)의 일부를 70℃에서 15분 내에 물 (50 ml), 빙초산 (1 ml) 및 철 분말(3.30 g, 60 mmol)의 혼합물에 첨가한다. 반응 혼합물을 95℃에서 부가의 3 시간 동안 교반한다. 식힌 후에, 톨루엔 (50 ml)을 첨가하고 고체 물질을 진공 여과시켜 제거한다. 여과 케이크를 톨루엔을 사용하여 세척한다.

C) 2,6-디클로로-4',5'-디메톡시-2'-포르밀아미노-벤조페논 (화합물 4)

2'-아미노-2,6-디클로로-4',5'-디메톡시벤조페논 (화합물 3; 0.82 g, 2.5 mmol) 및 포름산 (30 ml)의 혼합물을 24 시간 동안 환류 온도에서 가열하고, 진공에서 증발시킨다. 잔여물을 소량의 톨루엔에 용해시키고, 화합물 4는 시클로헥산을 첨가할 때 결정화하여 융점이 152℃인 무색 결정 0.64 g(수율 72%)을 얻는다.
D) 2,6-디클로로-5'-히드록시-4'-메톡시-2'-메틸벤조페논 (화합물 5)

2,6-디클로로-4',5'-디메톡시-2'-메틸벤조페논 (화합물 1; 2.5 g, 7.7 mmol), 브롬화수소/아세트산 (33%, 10 ml) 및 빙초산 (10 ml)의 혼합물을 75℃에서 1.5 시간 동안 교반하고, 물 (100 ml) 내로 붓고 디클로로메탄 (각각 50 ml)으로 2회 추출한다. 추출물을 합하고, 건조시키고, 진공에서 농축시킨다. 결과얻은 오일을 섬광 크로마토그래피 컬럼 (실리카 겔, 30 g)의 상부 상에 바른다. 톨루엔과 톨루엔/아세톤 9:1 (각각 500 ml)을 사용하여 용출시킨다. E) 2,6-디클로로-4'-메톡시-2'-메틸-5' n-프로폭시-벤조페논 (화합물 6)

2,6-디클로로-5'-히드록시-4'-메톡시-2'-메틸벤조페논 (화합물 5; 1.0 g, 32 mmol), n-프로필 보로마이드 (0.5 g, 4 mmol), 탄산칼륨 (2.8 g, 20 mmol)) 및 에탄올 (10 ml)의 혼합물을 80℃에서 6 시간 동안 교반하고, 여과하고 여액을 진공에서 증발시킨다. 잔여물을 섬광 크로마토그래피 컬럼(실리카 겔, 30 g)상에 바른다. 톨루엔 (750 ml)을 사용하여 용출시킨 결과 서서히 결정화하는 (용융점 73 - 75℃) 갈색 오일의 화합물 6을 800 mg 얻는다.
F) 2,6-디클로로-4',5'-디메톡시-2'-메틸-벤조펜티온 (화합물 7)

2,6-디클로로-4',5'-디메톡시-2'-메틸벤조페논 (화합물 1; 3.25 g, 10.0mmol), 오황화인산 (2.22 g, 10.0 mmol) 및 톨루엔 (50 ml)의 혼합물을 110℃에서 5 시간 동안 교반하고, p-디옥산으로 처리하고, 100 ℃에서 24 시간 동안 더 교반한다. 상층액을 검정색의 타르로, 더럽혀진 반응 생성물로부터 기울여 따르고, 실리카 겔 (15 g)을 첨가하고 용매를 진공에서 증발시킨다. 섬광 크로마토그래피 컬럼을 실리카 겔 (100 g)을 사용하여 채우고 충진된 실리카 겔을 이것의 상부 상에 바른다. 컬럼을 석유 에테르/아세톤(500 ml, 98:2, 부피/부피) 및 석유 에테르/아세톤 (750 ml, 부피/부피)를 사용하여 연속해서 용출시켜서 서서히 응고하는 어두운 녹색 오일의 화합물 7을 40 mg (수율 1.2%)을 얻긴다. 오일을 시클로헥산으로 3회 가루로만들때, 이것은 융점이 142℃인 고체를 생성한다.

디클로로메탄 (50 ml) 내 염화알루미늄 (14.67 g, 0.1 몰), 2,6-디클로로 벤조일 클로라이드 (20.96 g, 0.1 몰) 및 3,4,5-트리메톡시톨루엔 (18.22 g, 0.1 몰)을 0℃에서 서서히 및 연속해서 디클로로메탄에 첨가하고, 얼음 배쓰 온도에서 1 시간 동안 및 실온에서 16 시간 동안 교반하고, 얼음 내로 붓는다.
유기 층을 분리하고, 묽은 염산 및 물로 세척하고, 건조시키고, 및 실리카 겔 (100 g)을 첨가한 후에 진공에서 농축시킨다. 섬광 크로마토그래피 컬럼을 실리카 겔 (400 g)을 사용하여 채우고 충진된 실리카 겔을 이것의 상부상에 놓는다. 석유 에테르/에틸 아세테이트 (90/10, 1 l; 80/20, 1 l; 50/50, 1 l)를 사용하여 용출시켜 융점이 161℃인 2,6-디클로로-3',4'-디메톡시-2'-히드록시-6'-메틸벤조페논 10.35g (수율 30%)을 얻는다.
b) 2,6-디클로로-3'4'-디메톡시-2'-히드록시-6'-메틸벤조페논, 칼륨 염(화합물 188)

2,6-디클로로-3',4'-디메톡시-2'-히드록시-6'-메틸-벤조페논 (10.24 g, 30mmol)의 용액을 에탄올성 수산화 칼륨 (1.98 g, 30 mmol; 에탄올 (100 ml) 내 85%) 내에 용해시키고 70℃에서 15분 동안 교반한다. 그리고나서 용매를 진공에서 증발시킨다. 잔여물을 뜨거운 에탄올 (50 ml) 내에 용해시키고, 톨루엔을 첨가하고 용매를 진공에서 증발시켜 화합물 188을 11.7 g 얻는다.

2,6-디클로로-3',4'-디메톡시-2'--히드록시-6'-메틸-벤조페논, 칼륨 염(1.13 g; 3 mmol), 1-브로모부탄 (0.69 g, 5 mmol) 및 디메틸 포름아미드 (5 ml)의 혼합물을 100℃에서 8 시간 동안 교반하고, 진공에서 농축시킨다. 잔여물을 톨루엔/물 혼합물로 흔들어서 용해시키고, 분리 후에, 유기 층을 수집하고 물로 세척하고 건조시킨다. 실리카 겔 (5 g)을 첨가한 후에, 용매를 증발시킨다. 섬광 크로마로그래피 컬럼을 실리카 겔 (25 g)로 채우고 충진된 실리카 겔을 이것의 상부 상에 놓는다. 석유에테르/에틸아세테이트 (95/5, 500 ml)를 사용하여 용출시켜 융점이 70℃인 무색 결정의 표제 생성물 0.82 g (수율 69%)을 얻는다.
