Abstract
염기성 조건에 의해 활성이 유도되는 프로모터의 제어하에서 이종 단백질의 형질발현을 수행할 수 있는 약독해진 박테리아는 백신으로서 사용 가능하다. 이때, 적합한 프로모터는 nirB 프로모터이다.
Description
본 발명은 이종 단백질을 형질발현할 수 있는 약독해진 박테리아를 함유하는 백신에 관한 것이다.
유독한 살모넬라 균주는 생채내에서 생존 및 성장하는데 필요한 유전자내로 특정 돌연변이를 도입시킴으로써 약독해질 수 있다. 자가 제한적이고, 임상적으로 중요치 않은 감염을 일으키는 약독해진 변이주는 살모넬라 감염에 대한 생균 경구 백신으로 사용가능하다. Ty21a는 galE내 돌연변이 및 기타 알려지지 않은 약독화 손상을 보유한 살모넬라 티피(Salmonella typhi)의 약독해진 변이주로서, 많은 국가에서 생균 경구 장티푸스 백신으로 인정받아 사용되고 있다.
더욱 최근에는, 상이한 유전자에서 개별적인 특징적 돌연변이를 보유한 살모넬라속으로 유전적으로 규정된 균주는 몇몇 표적이 되는 종에서 실험용 경구 백신으로 시험된 바 있다. 예를 들면, 수종의 방향족 화합물을 필요로 하는 영양요구균인 살모넬라 아로(Salmonella aro) 돌연변이주는 마우스, 양, 소, 치킨에게 있어 효과적인 경구 백신인 것으로 밝혀진 바 있으며, 더욱 최근에 이들은 지원자내에서 약독해져 면역원이 되는 것으로도 또한 밝혀졌다. 살모넬라 이중 아로 돌연변이주는 EP-A-0322237에 개시되어 있다. 살모넬라 cya crp 이중 돌연변이주도 또한 효과적인 경구 백신이다.
약독해진 살모넬라균은 살모넬라증에 대한 백신으로서 뿐 아니라, 경구 면역계에 대한 이종 항원의 캐리어로서도 고려할 수 있다. 그 이유는 살로넬라균이 경구 경로를 통해 전이될 수 있고 또한 전신 및 국소적 세포성 및 항체 응답반응을 자극할 수 있는 강력한 면역원이기 때문이다. 박테리아, 바이러스 및 기생충으로부터의 이종 항원이 살모넬라 백신에 의해 숙주로 전이될 수 있다.
항원 전이를 위해 상기 생균 백신을 이용하는 경우 가능한 심각한 단점중 하나는 생체내에서의 외래 항원 형질발현의 안정성과 연관된 문제이다. 박테리아내에서 다수의 카피 플라스미드로부터 외래 단백질이 비조절적으로 높은 수준으로 발현될 경유, 대개 세포로부터 그 플라스미드 또는 형질발현된 유전자의 급속한 소실이 초래된다. 이러한 문제점은 적절한 생물체량이 얻어졌을 때 trp 또는 lac과 같은 유도성 프로모터 시스템에 의해 유전자 형질발현의 조절된 유도가 이루어지도록 함으로써 발효기내에서 통제가능하다. 이들 프로모터는 박테리아가 예방접종후 자가-제한 성장시 숙주 조직내에서 성장할 때 외부로부터 적용된 PP나 IPTG 같은 유도인자에 의해서도 유도될 수 없음이 명백하다.
생균 박테리아 벡터로 예방접종시 생체내 플라스미드 불안정성은 실제로 많은 연구인에 의해 보고된 바 있다(Maskell et al., Microb. Path 2, 295-305, 1987; Nakayama et al., Bio/technology 6, 693-697, 1988; Tite et al., Immunology 70, 540-546, 1990). 이러한 문제점을 극복하기 위하여 박테리아의 염색체로부터 이종 항원을 형질발현하는데 통합 시스템을 사용하는 것을 포함하는 수많은 연구가 이루어졌다(Hone et al., Microbiol. Path. 5, 407-418, 1988; Strugnell et al., Gene 88, 57-63, 1990). 그러나, 이러한 방안은 때로는 형질발현 수준이 너무나 낮기 때문에 일부 항원을 사용하는 경우에만 적합하다(Maskell et al., 1987). 나카야마 등은 생체내 형질발현 안정화를 위해 형질 발현 플라스미드에 필수 유전자를 연결시키는 방안에 대해 기재하고 있다. 이는 매우 효과적인 방안이지만, 플라스미드 부재 변이주의 산출을 방지하는 것이 아니라 단순히 이들이 생존하지 않음을 확인해 줄 뿐이다. 또한, 살모넬라 백신 균주내 외래 항원의 안정하지만 구성적인 높은 수준의 형질발현은 성장 속도를 감속시켜서 생균 백신의 면역원성에 잠재적으로 영향을 미친다.
본 발명에 따르면, 약학적으로 허용되는 담체나 희석제 및 활성 성분으로서, 병원성 유기체의 항원 결정부를 포함하는 이종 단백질을 암호하는 DNA 서열에 작동가능하게 연결되어 있고 그 활성이 혐기성 조건에 의해 유도되는 프로모터를 함유하는 약독해진 박테리아를 포함하는, 병원성 유기체에 의한 감염에 대해 숙주를 예방 치료하는데 사용하기 위한 백신이 제공된다.
이종 단백질의 안정한 형질발현은 생체내에서 수득될 수 있다. 적합한 박테리아, 예를 들면 그램-네가티브 박테리아가 사용될 수도 있다. 살모넬라와 같은 일부 그램-네가티브 박테리아는 진핵 세포를 침입하여 그 안에서 성장하며 점막 표면에 서식한다.
즉, 약독해진 박테리아는 살모넬라속, 보르데텔라속, 비브리오속, 헤모필루스속, 니세리아속 및 예르시니아속중에서 선택될 수도 있다. 대용으로, 약독해진 박테리아로 장독소원인 에세리시아 콜리의 약독해진 균주도 가능하다. 특히 하기의 종들이 언급될 수 있다 : 에스. 티피-인체 장티푸스의 원인균; 에스. 티피뮤리움-수종의 동물종내 살모넬라증의 원인균; 에스. 엔테리티디스-인체내 식중독의 원인균; 에스.콜레라수이스-돼지내 살모넬라증의 원인균; 보르데텔라 페르투시스-백일해의 원인균; 헤모필루스 인플루엔자-수막염의 원인균; 니세리아 고노로에아-임질의 원인균; 및 예시니아-식중독의 원인균.
박테리아의 약독화는 박테리아의 방향족 아미노산 생합성 경로에 관여하는 유전자의 비-복귀성(non-reverting) 돌연변이에 의한 것일 수 있다. 방향족 아미노산 생합성 경로에서 분지점 화합물인 코리스메이트의 합성에는 10개 이상의 유전자가 관여하고 있다. 이들중 몇가지 예를 들면 aroA (5-에놀피루빌쉬키메이트-3-포스페이스 합성효소), aroC (코리스메이트 합성효소), aroD (3-디히드로퀴네이트 디히드라타제) 및 areE (쉬키메이트 디히드로게나제)는 박테리아 게놈상에서 광범위하게 상이한 위치에 자리하고 있다. 따라서 돌연변이는 aroA, aroC, aroD 또는 aroE 유전자에서 일어날 수 있다.
그러나, 약독해진 박테리아는 방향족 아미노산 생합성 경로에 관여하는 별개의 두 유전자 각각에서 비-복귀성 돌연변이를 보유하는 것이 바람직하다. 이러한 박테리아는 EP-A-0322237에 기재되어 있다. 적합한 이중의 aro 돌연변이주는 aroA aroC, aroA aroD 및 aroA aroE 돌연변이주 박테리아이다. 그러나 다른 조합의 aroA, aroC, aroD 및 aroE 유전자 돌연변이를 가진 다른 박테리아도 사용가능하다. 특히 바람직한 것은 살모넬라 이중 aro 돌연변이주, 예를 들면 에스. 티피 또는 에스. 티피뮤리움의 이중 aro 돌연변이주, 특히 aroA aroC, aroA aroD 및 aroA aroE 돌연변이주이다.
대안으로, 약독해진 박테리아는 1종 이상의 다른 유전자의 조절과 관련된 유전자의 비-복귀성 돌연변이를 보유할 수도 있다(EP-A-0400958). 돌연변이는 ompR 유전자 또는 조절에 관여하는 다른 유전자에서 일어나는 것이 바람직하다. 조절에 관여하며, 주위환경의 자극에 대해 응답반응하는 것으로 공지된 다른 유전자가 다수 있다(Ronson et al., Cell 49, 579-581).
이러한 유형의 약독해진 박테리아는 제2유전자내에 제2의 돌연변이를 보유할 수 있다. 바람직한 제2유전자는 필수적인 생합성 경로에 관여하는 효소를 암호하는 유전자, 특히 방향족 화합물의 생합성에 관여하는 프리-코리스메이트 경로에 관여하는 유전자이다. 따라서 aroA, aroC 또는 aroD 유전자에서 제2의 돌연변이가 일어나는 것이 바람직하다.
다른 유형의 약독해진 박테리아는 주위환경의 스트레스에 대한 응답반응으로 생성되는 단백질을 암호하는 박테리아 DNA 또는 주위환경의 스트레스에 대한 응답반응으로 생성되는 단백질을 암호하는 DNA의 형질발현을 조절하는 단백질을 암호하는 박테리아 DNA에서 비-복귀성 돌연변이의 존재에 의해 약독화가 일어나는 것이다. 이러한 박테리아는 WO 91/15572에 기재되어 있다. 비-복귀성 돌연변이로는 결실, 삽입, 역위 또는 치환이 가능하다. 결실 돌연변이는 트랜스포존(transposon)을 사용하므로써 산출될 수 있다.
주위환경의 스트레스에 대한 응답반응으로 생성되는 단백질의 일례로는 열충격 단백질(42℃ 이상의 온도 상승에 대한 응답반응으로 생성됨); 영양소 결핍 단백질 (생존을 위해 미생물이 요구하는 정도 이하 수준의 포스페이트 또는 질소 같은 필수 영양소에 대한 응답반응으로 생성됨); 독성 스트레스 단백질(염료, 산 또는 가능하다면 식물 삼출물 같은 독성 화합물에 대한 응답반응으로 생성됨); 또는 물질대사파괴 단백질(물질대사를 파괴하는 것과 같이, 삼투조절에 대한 미생물 능력에 영향을 미치는 예컨대 이온 레벨 또는 비타민 또는 보조인자 레벨의 변동에 대한 응답 반응으로 생성됨)이 포함된다.

생균 백신의 형태로 사용하기 위해서는, 약독해진 박테리아가 유독 상태로 복귀되지 않아야 한다. 단일 DNA 서열에서 돌연변이시 이러한 일이 일어날 가능성은 적은 것으로 간주된다. 그러나, 별개의 두 DNA 서열 각각에서의 돌연변이의 존재로 인해 약독해진 박테리아에서 복귀가 일어날 위험성은 미미한 것으로 간주된다. 따라서, 바람직한 약독해진 박테리아는 (1) 주위환경이 스트레스에 대한 응답반응으로 생성되는 단백질을 암호하는 DNA 서열 또는 주위환경의 스트레스에 대한 응답반응으로 생성되는 단백질을 암호하는 DNA의 형질발현을 조절하는 단백질을 암호하는 DNA 서열내 돌연변이 존재에 의해 및 (2) 제2 DNA 서열내 돌연변이 존재에 의해 약독화가 일어난 것이다.
제2 DNA 서열은 필수 영양요구성 경로에 관여하는 효소를 암호하거나 또는 그 생성물이 삼투적으로 응답반응하는 유전자 즉, ompR의 조절을 제어하는 서열이 바람직하다(Infect and Immun 1989 2136-2140). 가장 바람직한 것은 방향족 아미노산 생합성 경로에 관여하는 DNA 서열에서, 더욱 구체적으로는 aroA, aroC 또는 aroD를 암호하는 DNA 서열에서 돌연변이가 일어나는 경우이다.
약독해진 박테리아는 당업자에게 공지된 방법에 의해 DNA 서열내로 돌연변이를 도입함으로써 작제될 수 있다(Maniatis, Molecular Cloning and Laboratory Manual, 1982). 비-복귀성 돌연변이는 하이브리드 트랜스포존 TnphoA를, 예를 들면 에스. 티피뮤리움 균주내로 도입시켜 산출할 수 있다. TnphoA는 주변세포질 단백질 또는 막 단백질 알칼린 포스파타제를 효소적 활성 단백질 융합시켜 산출할 수 있다. TnphoA 트랜스포존은 카나마이신 내성을 암호하는 유전자를 보유한다. 카나마이산 내성의 형질도입주는 적절한 선별용 배지상에서 콜로니를 성장시켜 선별한다.
대안 방법은 DNA 서열을 벡터 예컨대, 플라스미드 또는 코스미드내로 클로닝키시고, 클로닝된 DNA 서열내로 선별용 마커(marker) 유전자를 삽입하여 불활성화를 유발시키는 것을 포함한다. 불활성화된 DNA 서열과 상이한 선별용 마커를 보유한 플라스미드는 공지 기술에 의해 유기체내로 도입할 수 있다(Maniatis, Molecular Cloning and Laboratory Manual, 1982). 그후, 적당한 선별 과정에 의해, 비활성화된 DNA 서열이 미생물의 염색체내로 재조합되어 야생형 DNA 서열이 대립형질 교체로 공지된 과정으로 비-작용성이 된 돌연변이주를 규명하는 것이 가능하다. 특히, 이때 사용되는 벡터는 미생물내에서 불안정하여 자발적으로 소실되는 것이 바람직하다. 플라스미드상의 돌연변이된 DNA 서열과 야생형 DNA 서열은 유전적 교차에 의해 교체될 수 있다. 또 다른 방법은 돌연변이 부위에서 외래 DNA를 백신 균주내로 도입시키는 과정과 항생제 내성 마커를 균주내로 도입시키는 과정을 제외시키는 것이다.
약독해진 박테리아가 형질발현할 수 있는 이종 항원은 병원성 유기체의 항원 결정부를 포함할 수 있다. 항원은 바이러스, 박테리아, 진균, 효모 또는 기생충으로부터 유래될 수 있다. 따라서, 이종 단백질은 통상적으로 바이러스, 박테리아, 진균, 효모 또는 기생충으로부터 유래된 항원성 서열을 포함한다. 더욱 특히, 항원성 서열은 HIV-1 또는 HIV-2와 같은 인체 면역 결핍성 바이러스(HIV), A형 또는 B형 간염 바이러스, 타입 2 또는 타입 14와 같은 인체 리노바이러스, 단순 포진 바이러스, 폴리오바이러스 타입 2 또는 3, 구제역(foot-and-mouth disease) 바이러스, 인플루엔자 바이러스, 코스삭키 바이러스, 세포 표면 항원 CD4 및 클라미디아 트라코마티스 유형으로부터 유래될 수 있다. 항원은 HIV, 예를 들면 HIV-1 또는 HIV-2로부터의 CD4 수용체 결합 부위를 포함할 수 있다. 기타 사용가능한 항원에는 이. 콜리 열 불안정성 독소 B 서브유니트(LT-B), 이. 콜리 K88 항원, 비. 페르투시스로부터의 p.69 단백질, 테타누스 독소 단편 C 및 흡충, 미코플라스마, 회충, 촌충, 광견병 바이러스 및 로타바이러스의 항원이 포함된다.
이종 단백질의 형질발현을 조절하는데 사용하기 바람직한 프로모터는 nirB 프로모터이다. nirB 프로모터는 이. 콜리로부터 분리되는데, 이때 이것은 니트라이트 환원효소 유전자 nirB(Jayaraman et al., J. Mol. Biol. 196, 781-788, 1987) 및 nirD, nirC 및 cysG(Peakman et al., Eur. J. Biochem. 191, 315-323, 1990)를 포함하는 오페론의 형질발현을 조종한다. 이것은 니트라이트에 의해 및 산소 결핍시 활성적이 되는 주위 환경의 산소압 변화에 의해 조절된다. (Cole, Biochim. Biophys. 결실 및 돌연변이 분석에 의해, 혐기성 조건에 대해 단독으로 응답반응하는 프로모터 부분을 분리하고, 다른 혐기적으로 조절되는 프로모터와 비교하여, 공통적인 FNR-결합 부위를 규명하였다(Bell et al., Nucl. Acids. Res. 17, 3865-3874, 1989; Jayaraman et al., Nucl. Acids Res. 17, 135-145, 1989). 또한 추정적인 FNR-결합 부위와 -10 상동성 영역간에는 거리가 중요한 것으로 밝혀졌다(Bell et al., Molec. Microbiol. 4, 1753-1763, 1990). 따라서, 혐기성 조건에 단독으로 응답반응하는 nirB 프로모터 부분만을 사용하는 것이 바람직하다. 본 명세서에서 사용된 nirB 프로모터는 혐기성 조건하에서 암호 서열의 형질발현을 촉진시킬 수 있는 프로모터 자체 또는 그것의 일부 또는 유도체를 지칭한다. 실제 사용되는 것으로 nirB 프로모터를 포함하는 서열은

본 발명에 따라 사용되는 약독해진 박테리아는 nirB 프로모터와 같이, 이종 단백질을 암호하는 DNA 서열에 작동가능하게 연결되어 있고, 활성이 혐기성 조건에 의해 유도되는 프로모터를 포함한 DNA 구조체로 약독해진 박테리아를 형질전환시켜 작제할 수 있다. 전기 침투(electroporation)와 같은 적절한 형질전환 기술을 사용할 수도 있다. 이렇게 하여, 박테리아에 대해 이종인 단백질을 형질발현 할 수 있는 약독해진 박테리아가 수득될 수 있다. 약독해진 박테리아의 배양물은 호기성 조건하에서 성장시킬 수 있다. 따라서, 이종 단백질의 형질발현을 최소로 하면서 백신 제제를 위한 충분량의 박테리아가 작제된다.
DNA 구조체는 통상적으로 이종 단백질을 암호하는 DNA 서열에 작동가능하게 연결된 nirB 프로모터를 포함한 복제성 형질발현 벡터이다. 단백질의 형질발현을 조절하는 기존의 프로모터 대신에 이종 단백질을 암호하는 유전자를 사전에 혼입한 형질발현 벡터내에 nirB 프로모터를 삽입할 수 있다. 형질발현 벡터는 물론 벡터가 삽입되는 약독해진 박테리아에 상용가능한 것이어야 한다.
형질발현 벡터에는 nirB 프로모터 이외에, 전사 종결 부위 및 해독 출발 및 중지 코돈을 비롯하여 적절한 전사 및 해독 제어 인자가 제공되어 있다. 적절한 리보좀 결합 부위가 제공되어 있다. 벡터는 통상, 복제 기점 및 필요에 따라서는 항생제 내성 유전자와 같은 선별용 마커 유전자를 포함한다. 벡터는 플라스미드일 수 있다.
백신은 환자에게 경구 투여하기 위해 동결건조 형태, 예를 들면 캅셀 형태로 제시되는 것이 유리하다. 이러합 캅셀에는 예를 들면 유드라게이트(Eudragate) "S", 유드라게이트 "L", 셀룰로오스 아세테이트, 셀룰로오스 프탈레이트 또는 히드록시프로필메틸 셀룰로오스를 함유한 장용 피복제가 제공될 수 있다. 이러한 캅셀은 그 자체로서 또는 다른 방도로 사용될 수 있는데, 동결건조된 물질은 투여 이전에, 예를 들면 현탁액으로서 재구성될 수 있다. 재구성은 유기체가 생존하기 적합한 pH하에 완충액중에서 실행하는 것이 유리하다. 약독해진 박테리아 및 백신을 위의 산도로부터 보호하기 위해서는, 백신 투여전에 매번 중탄산 나트륨 제제를 투여하는 것이 유리하다. 대안으로, 백신은 비경구 투여, 비강내 투여 또는 유선내 투여용으로 제조될 수도 있다.
본 발명의 약독해진 박테리아는 숙주, 특히 인체 숙주 그러나 또한 가능하다면 동물 숙주의 예방적 치료에 사용될 수 있다. 따라서 본 발명에 따른 유효량의 약독해진 박테리아를 투여하므로써 미생물, 특히 병원체에 의해 유발되는 감염을 예방할 수 있다. 그 후, 박테리아는 미생물에 대한 항체를 생성시킬 수 있는 이종 단백질을 형질발현한다. 사용된 투여량은 숙주의 크기 및 체중, 백신 제제의 유형 및 이종 단백질의 특성에 따라 좌우된다. 그러나, 약독해진 에스. 티피의 경우, 70kg의 성인 숙주에 대해 1회분당 109내지 1011개의 에스. 티피 유기체의 경구 투여량을 함유하는 투여용량이 일반적으로 편리하다.
하기 실시예에서는 본 발명을 설명한다. 첨부한 도면은 다음과 같다; 제1도 내지 제4도는 BALB/c 마우스의 간, 비장 파이어판 및 장간막 림프절 각각에서 생체내 성장할 수 있는 에스. 티피뮤리움 분리물의 능력을 나타낸 것이다. x-축은 감염후 경과 시일을 나타내며 y-축은 기관당 log10생존 유기체수를 나타내고, △는 분리물 BRD509를, □는 분미룰 BRD847을, ○는 분리물 BRD743을 의미하며, ―는 암피실린 부재를 및 ----는 암피실린 첨가를 나타낸다.
제5도는 마우스 혈청의 항-테타누스 독소 단편 C 역가를 나타낸 것이다. x-축은 마우스 혈청에 사용된 박테리아의 유형을 나타낸다. x-축은 마우스를 공격하는데 사용된 박테리아의 유형을 나타낸다. 투여 횟수는 괄호내에 제시되어 있다. y-축은 492nm에서의 흡광 판독치를 의미한다.
pTETnir15의 작제
pTETtac115에서 lacI 유전자 및 tac 프로모터를 포함하는 EcoRI-ApaI 영역(1354bp)를 하기 쌍의 올리고 1 및 2로 대체하여 형질발현 플라스미드 pTETnir15를 작제하였다(Makoff et al., Nucl. Acids Res. 17 10191-10202, 1989);
파마시아(pharmacia) 유전자 조립기로 올리고뉴클레오티드를 합성하고 생성된 플라스미드를 서열분석에 의해 확인하였다(Makoff et al., Bio/Technology 7, 1043-1046, 1989).
pTETnir15를 보유한 SL1334 aroA aroD의 작제
nirB 프로모터의 제어하에서 테타누스 독소 단편 C를 형질발현하는 살모넬라 백신 균주를 작제하기 위해, 중간체 균주 에스. 티피뮤리움 LB5010(r-m+)(Bullas 및 Ryo. J. Bact. 156. 471-474, 1983)을 pTETnir15로 형질전환시켰다. 항생제 선별후, 콜로니를 항-테타누스 독소 단편 C 혈청으로 면역블로팅하여 단편 C를 형질발현하는 콜로니를 검색하였다. 니트로 셀룰로오스 필터상에서 밤새 콜로니를 호기 배양한 후, 면역 블로팅에 앞서 4시간 동안 혐기성 조건하에서 배양하여 콜로니를 유도하였다. 단편 C를 안정하게 형질발현하는 균주 하나를 사용하여 플라스미드 DNA를 작제하였다. 이를 사용하여 전기침투에 의해 BRD509로 명명된 에스. 티피뮤리움 SL1344 aroA aroD의 분리체를 형질전환시켰다. 단편 C를 안정하게 형질발현하는 균주(전술한 바와 같이 면역블로팅에 의해 검색)를 생체내 연구용으로 선택하여 명명하였다.
BALB/c 마우스에 있어 BRD743과 BRD 847의 생체내 역학 비교
BALB/c 마우스에 경구 투여한 후, BRD743(pTET85를 보유한 BRD509) 및 BRD847의 생체내에서 성장할 수 있는 능력을 비교하였다. pTETtac115에서 lacI 유전자를 가진 1.2kb EcoRI 단편을 결실시켜 pTET85를 작제하였다(Makoff et al., Nucl. Acids Res. 17, 10191-10202, 1989). 그 결과 살모넬라 균주에서 단편 C의 구성적 형질발현이 이루어졌다. 간, 비장, 파이어판 및 장간막 림프절에서 박테리아의 수를 계수하였다. 마우스로부터 분리한 박테리아를, 단편 C를 형질발현하는 플라스미드를 보유한 유기체 백분율의 자표로서 암피실린을 함유하는 평판상에서 성장할 수 있는 능력에 대해 또한 평가하였다. 그 결과는 제1도 내지 제4도에 제시되어 있다.
유사한 초기 유기체 수(5×109)를 사용하여 마우스를 감염시켰을 때, BRD743 및 BRD847은 둘다 검사한 쥐 조직을 침입하여 그안에서 존속할 수 있지만 BRD509 보다는 낮은 수준임이 밝혀졌다. 그러나 흥미로운 특징은 BRD743으로 감염된 마우스로부터 수득한 암피실린 내성의 유기체 수는 급속히 감소했으며, 원상복귀된 모든 유기체는 14일째까지 암피실린 감수성이라는 점이다. 이를 통해, 생체내에서 선별 결과 살모넬라 백신 균주로부터 pTET85가 급속히 소실됨을 알 수 있다. 대조적으로, 암피실린 존재 및 부재하에서의 BRD847에 대한 계수는 감염 모니터 시간동안 거의 동일하였다. 이는 에스. 티피뮤리움 백신 균주내의 pTETnir15가 장기간동안 생체내에서 단편 C를 형질발현하는 가능성이 있는 유기체를 생성하므로 면역원성면에서 확실한 추가의 잇점을 가지고 있음을 입증한다.
pTET85(BRD743) 또는 pTETnir15(BRD847)를 보유한 살모넬라 균주를 사용하는 BALB/c 마우스의 면역화
20 마리의 마우스 그룹을 마우스 1 마리당 5×109세포의 BRD743, BRD847 또는 BRD509에 의해 경구적으로 항온처리하였다. 25일째에 모든 마우스로부터 혈청을 채취하여 ELISA로 항-테타누스 항체에 대해 분석하였다. BRD847로 예방접종시킨 모든 마우스에서는 25일째에 항-단편 C항체가 검출되었으나 반면, BRD743 또는 BRD509로 예방접종시킨 마우스에서는 항-단편 C항체가 검출되지 않았다(제5도). 25일째에 각 그룹에서 10 마리의 마우스를 유사한 량의 동종 유기체로 경구 접종시켜 추가면역시켰다. 46일째에 이들 마우스로부터 채취한 혈청의 ELISA 분석 결과, 항-단편 C 응답반응은 BRD743 및 BRD847로 접종시킨 그룹에서 증대됨이 밝혀졌다. BRD847로 추가면역시킨 마우스에 대한 역가는 BRD743으로 추가면역시킨 마우스의 경우보다 상당히 높았다. BRD509로 경구적으로 추가면역시킨 마우스는 단편 C에 대해 검출가능한 항체 응답반응을 산출하는데 실패했다.
BRD847 및 BRD743으로 경구 면역화시킨 마우스의 테타누스 독소 공격
BRD743, BRD847 및 BRD509로 경구적으로 예방접종시킨 마우스에 면역화용 균주를 1 또는 2회 투여한 후, 테타누스 독소 공격에 대한 면역성에 관하여 시험하였다. 20마리의 마우스 그룹에 5×109유기체를 단 1회 경구 투여하고 25일째에 10 마리의 마우스 그룹을 50% 치사하는 500 투여량의 테타누스 독소로 공격하였다(표 1 참조). BRD847로 예방접종시킨 마우스는 1회 경우 투여후 공격에 대해 완전 보호되었으나 반면, BRD743으로 예방접종시킨 마우스는 일부만이 보호되었다(2/10 생존). 25일째에 10마리의 나머지 마우스 그룹에 유기체(5×109)를 2차 투여하고 46일째(1차 투여후)에 공격하였다. BRD847로 면역시킨 마우스는 테타누스 독소 공격후 완전 보호되었으나, 반면 BRD743으로 면역시킨 마우스는 일부만이 보호되었다(5/10). BRD509로 1 또는 2회 투여하며 면역시킨 테타누스 독소로 공격한 마우스는 모두 죽었다. BRD847은 마우스에 있어 테타누스 독소 공격에 대한 효과적인 단일 투여용 경구 백신이다. 마우스 그룹에 105개의 BRD847 및 BRD743 유기체를 1 및 2회 정맥내 투여한 후, 또한 테타누스 독소로 공격하였다. 1 또는 2회 백신 균주 투여후, 모든 마우스는 테타누스 독소의 공격에 대해 완전 보호되었다.

- 병원성 유기체의 항원 결정부를 포함하는 이종 단백질을 암호하는 DNA 서열에 작동가능하게 연결되어 있고 그 활성이 혐기성 조건에 의해 유도되는 mirB 프로모터를 함유하는 약독해진 살모넬라 균주인 약독해진 박테리아 활성 성분으로 포함하며 약학적으로 허용되는 담체나 희석제를 포함하는, 병원성 유기체에 의한 감염에 대해 숙주를 예방 치료하는데 사용하기 위한 백신.
- 제1항에 있어서, 상기 약독해진 박테리아가 살모넬라 티피 또는 살모넬라 티피뮤리움의 약독해진 균주인 백신.
- 제1항에 있어서, 상기 박테리아의 약독화가 방향족 아미노산 생합성 경로에 관여하는 유전자의 비-복귀성 돌연변이에 의한 것인 백신.
- 제1항에 있어서, 상기 박테리아의 약독화가 환경적인 스트레스에 대한 응답반응으로 생성되는 단백질을 암호하는 박테리아 DNA 또는 환경적인 스트레스에 대한 응답반응으로 생성되는 단백질을 암호하는 DNA의 형질발현을 조절하는 단백질을 암호하는 박테리아 DNA의 비-복귀성 돌연변이에 의한 것인 백신.
- 제3항에 있어서, 상기 약독해진 박테리아가 방향족 아미노산 생합성 경로에 관여하는 별개의 두 유전자 각각의 비-복귀성 돌연변이를 보유한 백신.
- 제5항에 있어서, 박테리아가 aroA aroC, aroA aroD 또는 aroA aroE 돌연변이주인 백신.
- 제1항에 있어서, 상기 항원 결정부가 바이러스, 박테리아, 진균, 효모 또는 기생충으로부터 유래된 것인 백신.
- 제7항에 있어서, 상기 이종 단백질이 보르데텔라 페르투시스로부터의 P69 단백질 또는 테타누스 독소 단편 C인 백신.
- 미생물에 의한 감염에 대해 인체 또는 동물 숙주를 예방 치료하는 방법에 사용하기 위한, 제1항에 정의된 약독해진 박테리아.
- 제1항에 정의된 약독해진 살모넬라 박테리아.
