<抗体とストレプトアビジン変異体との融合タンパク質>
本発明の融合タンパク質は、N末端側からC末端側に、配列番号2又は配列番号7に記載のアミノ酸配列と、リンカー配列と、配列番号1に記載のアミノ酸配列(但し、C末端の6個のヒスチジンからなる配列アミノ酸配列は一部又は全部が欠失していてもよい)とをこの順に有する融合タンパク質である。本発明の融合タンパク質は、N末端側からC末端側に、抗デスモグレイン抗体のアミノ酸配列と、リンカー配列と、配列番号1に記載のアミノ酸配列(但し、C末端の6個のヒスチジンからなる配列アミノ酸配列は一部又は全部が欠失していてもよい)とをこの順に有する融合タンパク質でもよい。
配列番号2に記載のアミノ酸配列は、scFv型の抗CEACAM抗体のアミノ酸配列である。配列番号7に記載のアミノ酸配列は、scFv型の抗Her2抗体のアミノ酸配列である。
配列番号1に記載のアミノ酸配列は、ステレプトアビジン変異体のアミノ酸配列であり、具体的には、国際公開WO2015/125820の実施例3(国際公開WO2015/125820の配列番号4)(本願明細書の配列番号1)に記載されているストレプトアビジン変異体LISA314-V2122である。
上記の通り、本発明の融合タンパク質は、上記の通り、がん細胞を認識する抗体とストレプトアビジン変異体との融合タンパク質である。
リンカー配列の具体例としては、4〜14個のグリシン残基及び1個のシステイン残基からなる配列を挙げることができる。リンカー配列としては、例えば、(Gly)m−Cys−(Gly)n(m及びnはそれぞれ独立に1〜13の整数を示す)で示されるアミノ酸配列を使用することができる。リンカー配列の具体例としては、Gly- Gly-Gly- Gly-Ser- Gly- Gly- Gly- GlyまたはGly- Gly- Gly-Gly- Gly-Ser- Gly- Gly- Gly- Gly- Glyを挙げることができるが特に限定されない。
本発明の融合タンパク質は、そのN末端に、分泌シグナル配列を有していてもよい。分泌シグナル配列を有する融合タンパク質としては、配列番号4又は配列番号9に記載のアミノ酸配列(但し、C末端の6個のヒスチジンからなる配列アミノ酸配列は一部又は全部が欠失していてもよい)を有する融合タンパク質を挙げることができる。
本発明の融合タンパク質は、がん治療剤またはがん診断剤として有用である。
本発明によれば、(1)本発明の融合タンパク質、及び(2)後記する式(1)で示される化合物又はその塩と、診断用物質又は治療用物質とのコンジュゲートを含む、がんの治療または診断キットが提供される。
ビオチン改変二量体は、下記式(1)で示される化合物又はその塩であり、好ましくは下記式(2)で示される化合物又はその塩である。ビオチン改変二量体は、国際公開WO2015/125820号に記載されている化合物を使用することができる。

ビオチン改変二量体に、診断用物質又は治療用物質を結合させることにより、ビオチン改変二量体と診断用物質又は治療用物質とのコンジュゲートを調製することができる。診断用物質又は治療用物質としては、蛍光色素、化学発光剤、放射性同位元素、金属化合物等からなる増感剤、金属化合物等からなる中性子捕捉剤、フタロシアニン染料、低分子化合物、マイクロあるいはナノバブル、タンパク質などを挙げることができる。好ましくは、フタロシアニン染料を使用することができる。
フタロシアニン染料は、好ましくはシリコンフタロシアニン染料である。IRDye(登録商標)700DXのようなフタロシアニン染料の具体例は、例えば、米国特許第7,005,518号に記載されている。
X及びYの具体例としては、
(i)X及びYがともに親水性基である場合、
(ii)X及びYの一方が親水性基であり、X及びYの他方が−OH、又は水素原子である場合、
(iii)X及びYが、−OH、又は水素原子である場合、
を挙げることができる。
光免疫療法とは、体内で特定の細胞を破壊するために光増感剤及び照射光を使用する治療法である。光増感剤は、特異的な波長の光に晒されるとき、付近の細胞のアポトーシス、ネクローシス、及び/又は自食作用を誘発できる細胞傷害性の活性酸素種を生じる。例えば、特許6127045号公報には、細胞を死滅させる方法であって、 細胞表面タンパク質を含む細胞を、治療有効量の1または複数の抗体−IR700分子と接触させるステップであって、該抗体が該細胞表面タンパク質に特異的に結合するステップと;該細胞に、660〜740nmの波長で、かつ少なくとも1Jcm−2の線量で照射するステップと;該細胞に照射した約0〜8時間後に、該細胞を1または複数の治療剤と接触させ、これにより、該細胞を死滅させるステップとを含む方法が記載されている。特表2017−524659号公報には、疾患又は病態を患っている対象において細胞毒性を誘発する方法であって、(a)対象の細胞に特異的に結合するプローブにコンジュゲートしたIRDye(登録商標)700DXなどのフタロシアニン染料を包含する治療的に有効な薬剤を該対象に投与し;そして、(b)細胞死を誘発するのに有効な量で適切な励起光を前記細胞に照射することを含む、方法が記載されている。
治療有効量は、上記のコンジュゲートと融合タンパク質のそれぞれについて、60キログラム当たり少なくとも0.5ミリグラム(mg/60kg)、少なくとも5mg/60kg、少なくとも10mg/60kg、少なくとも20mg/60kg、少なくとも30mg/60kg、少なくとも50mg/60kgである。例えば、静脈内投与される場合、1mg/60kg、2mg/60kg、5mg/60kg、20mg/60kg、または50mg/60kgの用量など、例えば、0.5〜50mg/60kgである。別の例では、治療有効量は、少なくとも100μg/kg、少なくとも500μg/kgまたは少なくとも500μg/kgなど、少なくとも10μg/kg、例えば、腫瘍内投与または腹腔内投与される場合、100μg/kg、250μg/kg、約500μg/kg、750μg/kg、または1000μg/kgの用量など、例えば、10μg/kg〜1000μg/kgである。一例では、治療有効量は、局所用溶液で投与される10μg/ml、20μg/ml、30μg/ml、40μg/ml、50μg/ml、60μg/ml、70μg/ml、80μg/ml、90μg/ml、または100μg/mlなど、20μg/ml〜100μg/mlの間など、少なくとも500μg/mlなど、少なくとも1μg/mlである。

V2122は、国際公開WO2015/125820の実施例3(国際公開WO2015/125820の配列番号4)に記載されているストレプトアビジン変異体である。V2122のアミノ酸配列(C末端に6×Hisタグを有する配列)を配列表の配列番号1に記載する。
図2より精製されたCEA-V2122は約150kDaの4量体を主たる成分であることが確認された。
CEA-V2122と抗原CEACAM5との親和性を表面プラズモン共鳴(SPR)測定装置:Biacore T200 (GE Healthcare Life Sciences社) を用いて実施した。具体的にはRecombinant Human CEACAM-5/CD66e Protein, CF (R&D SYSTEMS社) を Sensor Chip CM5 (GE Healthcare Life Sciences社) にアミンカップリングキット (GE Healthcare Life Sciences社) を用いて固定化操作を行い、リガンドの最終固定化量は279RUとなった。また、精製されたCEA-V2122は 1E-08 M から 6.25E-10 M までの2倍希釈系列をアナライトとして調整した。相互作用解析は Single-Cycle Kinetics 解析にてデータの取得を行った。得られたデータを Biacore T200 Evaluation Software, version 2.0 にて Bivalent Analyze モードによるカーブフィッティングを実施しka1=3.208E+5, kd1=3.461E-7の値を得た。また、Bivalent AnalyzeではKD=kd1/ka1で評価できることからKD=kd1/ka1= 3.461E-7/3.208E+5=1.078E-12の評価値を得た。それらの結果を図3に示す。
図3に示されるセンサーグラム、KD値よりCEA-V2122はCEACAM5に強く結合することが確認された。
図4に示されるセンサーグラム、KD値よりCEA-V2122はビオチン改変体に強く結合することが確認された。
CEACAM5発現陽性のがん細胞株の染色のために、精製されたCEA-V2122 タンパク質を100 μg 使用しFITC標識を行なった。具体的には、Fluorescein Labeling Kit - NH2 (同仁化学研究所) を用いて操作手順書の用法用量に従い標識を実施し、得られた産物をCEA-V2122-FITC とした。具体的なCEACAM5発現陽性のがん細胞株の染色は次のとおりである。 CELLSTAR, μClear, 96ウエルプレート (Greiner社) に2.0x104cell/well となるようにCEACAM5陽性のヒト胃がん由来MKN-45細胞とCEACAM5陰性のヒト結腸がん由来DLD1細胞を播種し一晩培養した。次に 20 nM CEA-V2122-FITCと1μM Hoechist を含む培養液を100 μL/well となるように添加し4℃にて30分間反応ののち、In Cell Analyzer 6000 (GE Healthcare Life Sciences社) にて画像の取得を行なった。その結果を図5および図6に示す。
光活性化化合物標識ビオチン改変体、コンパウンド1、コンパウンド2およびコンパウンド3を用いて細胞傷害性試験を実施した。これらのコンパウンドは出願番号:特願2018-149295 に記載されている化合物である。コンパウンド1、コンパウンド2およびコンパウンド3を図7に示す。具体的にはMKN45細胞を細胞培養用96ウェルプレートに細胞数が 5x103 cells/well、培養液が 50 μL/well となるように播種し一晩培養した。CEA-V2122と光活性化化合物標識ビオチン改変体の複合体溶液は、CEA-V2122と各コンパウンドとのモル比が1:2となるように混合し室温で10分間インキュベーションをおこない、CEA-V2122の最終濃度10μg/mLとなるように培養液で濃度調製を行なった。希釈系列は20μg/mLを開始濃度とし、4倍希釈系列を4系列(5.0 μg/mL, 1.25 μg/mL, 0.312 μg/mL, 0.078μg/mL)を5系列の複合体希釈系列液とした。また、複合体を含まない培地のみをゼロコントロールとした。
図8に示されるように、CEA-V2122とコンパウンド1、コンパウンド2、コンパウンド3との複合体の濃度依存的に細胞傷害性を示すことが確認された。
ヌードマウスにMKN45細胞を皮下に移植したゼノグラフトモデルマウスの作製を行なった。4週例のヌードマウス雌を購入し、1週間馴化後、1匹あたり細胞数 2x10^5 cells にて皮下に細胞を移植した。移植後、約10日に実験例4に記載されているCEA-V2122と光活性化化合物標識ビオチン改変体コンパウンド1の複合体を100 μg を尾静脈から投与した。波長690 nm の光を発生するLED光源(USHIO OPTO SMICONDUCTORS, INC., LD690D-66-60-550.), T-Cube LED Driver(THORLABS, NC0713145)およびT-CUBE 15V POWER SUPPLY(THORLABS, TPS001) を使用し、投与6時間後に690 nm の光を230 J/cm^2 となるように照射した。再度、複合体投与24時間後に690 nm の光を230 J/cm^2 となるように照射した。光源として腫瘍の体積の変化のグラフを図9に示す。複合体投与5日後のマウス個体の写真を図10に示す。また、複合体投与7日目にマウス個体を安楽死させ、解剖を行い腫瘍部分を病理解析した。具体的な手法は次のとおりである。マウス皮下腫瘍とその周囲組織を一塊として摘出し、4%パラホルムアルデヒド溶液(和光純薬, 163-20145)に室温で一晩浸漬して組織を固定した。固定後の皮下腫瘍組織に対して約3~5mm厚の割を入れてパラフィン包埋ブロックを作成し、ミクロトームによって4μm厚の薄切病理標本を作成した。
図11及び図12よりCEA-V2122とコンパウンド1との複合体を投与し、690 nm の光を照射することでゼノグラフトモデルマウスの腫瘍の壊死を誘導できることが確認された。
V2122は、国際公開WO2015/125820の実施例3(国際公開WO2015/125820の配列番号4)に記載されているストレプトアビジン変異体である。V2122のアミノ酸配列を配列表の配列番号1に記載する。
scFv-V2122は、上記のV2122に、HER2 (ERBB2) に対する一本鎖抗体(scFv)を結合したものである。このscFv型の抗HER2抗体は、Zhang H, et al.,Therapeutic potential of an anti-HER2 single chain antibody-DM1 conjugates for the treatment of HER2-positive cancer. Signal Transduct Target Ther. 2017 May 19;2:17015. doi: 10.1038/sigtrans.2017.15に記載されているscFv配列である。scFv型の抗HER2抗体のアミノ酸配列を配列表の配列番号7に記載する。また、scFv型の抗HER2抗体とV2122とをアミノ酸リンカー(GGGGGSGGGGG)(配列番号18)で結合したHER2-V2122の構造図を図13、アミノ酸配列を配列表の配列番号8に示す。
図14より精製されたHER2-V2122は約150kDaの4量体を主たる成分であることが確認された。
HER2-V2122と抗原CEACAM5との親和性を表面プラズモン共鳴(SPR)測定装置:Biacore T200 (GE Healthcare Life Sciences社) を用いて実施した。具体的にはErbB2/Fcキメラ、ヒト、組換え体、キャリアフリー (R&D SYSTEMS、1129-ER-050) を Sensor Chip CM5 (GE Healthcare Life Sciences社) にアミンカップリングキット (GE Healthcare Life Sciences社) を用いて固定化操作を行い、リガンドの最終固定化量は279RUとなった。また、精製された HER2-V2122 は 1E-08 M から 6.25E-10 M までの2倍希釈系列をアナライトとして調整した。相互作用解析は Single-Cycle Kinetics 解析にてデータの取得を行った。得られたデータを Biacore T200 Evaluation Software, version 2.0 にて Bivalent Analyze モードによるカーブフィッティングを実施しka1=3.857E+5, kd1=1.710E-6の値を得た。また、Bivalent AnalyzeではKD=kd1/ka1で評価できることからKD=kd1/ka1=1.710E-6/3.857E+5=4.433E-12の評価値を得た。それらの結果を図15に示す。
図15に示されるセンサーグラムおよび計算されたKD値よりHER-V2122はErbB2を認識し強く結合することが確認された。
光活性化化合物標識ビオチン改変体、コンパウンド1、コンパウンド2およびコンパウンド3を用いて細胞傷害性試験を実施した。これらのコンパウンドは出願番号:特願2018-149295 に記載されている化合物である。コンパウンド1、コンパウンド2およびコンパウンド3を図7に示す。具体的には 10% FBSを含むMcCoy’s 培地にて培養したSK-BR-3細胞を細胞培養用96ウェルプレートに細胞数が 5.0x103 cells/well、培養液が 50 μL/well となるように播種し一晩培養した。CEA-V2122と光活性化化合物標識ビオチン改変体の複合体溶液は、CEA-V2122と各コンパウンドとのモル比が1:2となるように混合し室温で10分間インキュベーションをおこない、HER2-V2122の最終濃度10μg/mLとなるように培養液で濃度調製を行なった。希釈系列は20 μg/mLを開始濃度とし、4倍希釈系列を4系列(5.0 μg/mL, 1.25 μg/mL, 0.312 μg/mL, 0.078μg/mL)を5系列の複合体希釈系列液とした。また、複合体を含まない培地のみをゼロコントロールとした。

Claims (12)
- N末端側からC末端側に、配列番号2又は配列番号7に記載のアミノ酸配列と、リンカー配列と、配列番号1に記載のアミノ酸配列(但し、C末端の6個のヒスチジンからなる配列アミノ酸配列は一部又は全部が欠失していてもよい)とをこの順に有する、融合タンパク質。
- リンカー配列のアミノ酸数が5から15アミノ酸である、請求項1に記載の融合タンパク質。
- リンカー配列が、4〜14個のグリシン残基及び1個のシステイン残基からなる、請求項1又は2に記載の融合タンパク質。
- リンカー配列が、Gly- Gly-Gly- Gly-Ser- Gly- Gly- Gly- GlyまたはGly- Gly- Gly-Gly- Gly-Ser- Gly- Gly- Gly- Gly- Gly
である、請求項1から3の何れか一項に記載の融合タンパク質。 - 配列番号3又は配列番号8に記載のアミノ酸配列(但し、C末端の6個のヒスチジンからなる配列アミノ酸配列は一部又は全部が欠失していてもよい)を有する、融合タンパク質。
- さらに分泌シグナル配列を有する、請求項1から5の何れか一項に記載の融合タンパク質。
- 配列番号4又は配列番号9に記載のアミノ酸配列(但し、C末端の6個のヒスチジンからなる配列アミノ酸配列は一部又は全部が欠失していてもよい)を有する、融合タンパク質。
- 配列番号3又は配列番号8に記載のアミノ酸配列(但し、C末端の6個のヒスチジンからなる配列アミノ酸配列は一部又は全部が欠失していてもよい)を有する融合タンパク質をコードする核酸。
- 配列番号4又は配列番号9に記載のアミノ酸配列(但し、C末端の6個のヒスチジンからなる配列アミノ酸配列は一部又は全部が欠失していてもよい)を有する、融合タンパク質をコードする核酸。
- 請求項1から7の何れか1項に記載の融合タンパク質を含む、がん治療剤またはがん診断剤。
- (1)請求項1から7の何れか1項に記載の融合タンパク質、及び(2)下記式(1)で示される化合物又はその塩と、診断用物質又は治療用物質とのコンジュゲートを含む、がんの治療または診断キット。
X1a、X1b、X2a及びX2bはそれぞれ独立にO又はNHを示し、
Y1及びY2はそれぞれ独立にC又はSを示し、
Z1及びZ2はそれぞれ独立にO、S又はNHを示し、
V1及びV2はそれぞれ独立にS又はS+−O−を示し、n1及びn2はそれぞれ独立に0又は1の整数を示し、
L1及びL2はそれぞれ独立に、2価の連結基を示し、
L3は、診断用物質又は治療用物質と結合できる官能基を末端に含む基であり、
L4は、3価の連結基を示す。) - 診断用物質又は治療用物質が、フタロシアニン染料である、請求項11に記載のキット。
