Abstract
Description
[1]配列番号:1または18に記載のアミノ酸配列からなる配列を有するペプチドに結合し、かつ
(1)B細胞表面上のIgE抗体に結合する、および/または
(2)56℃で加熱処理したIgE抗体に結合する、
単離されたモノクローナル抗体またはその抗原結合フラグメント。

可変領域のうち、直接抗原と接触する領域は特に変化が大きく、相補性決定領域(complementarity−determining region: CDR)と呼ばれる。CDR以外の比較的変異の少ない部分をフレームワーク(framework region: FR)と呼ぶ。軽鎖と重鎖の可変領域には、それぞれ3つのCDRが存在し、それぞれN末端側から順に、重鎖CDR1〜3及び軽鎖CDR1〜3と呼ばれる。
「ヒトキメラ抗体」は、非ヒト由来の抗体において、非ヒト由来の抗体の定常領域がヒトの抗体の定常領域に置換されている抗体である。ヒトキメラ抗体では、ADCC活性を高める観点では、例えば、定常領域に用いるヒトの抗体のサブタイプはIgG1とすることができる。
「イヌキメラ抗体」は、非イヌ由来の抗体において、非イヌ由来の抗体の定常領域がイヌの抗体の定常領域に置換されている抗体である。イヌキメラ抗体では、ADCC活性を高める観点では、例えば、定常領域に用いるイヌの抗体のサブタイプは、特に限定されないが例えばIgGbとすることができる。
「ネコキメラ抗体」は、非ネコ由来の抗体において、非ネコ由来の抗体の定常領域がネコの抗体の定常領域に置換されている抗体である。
「哺乳動物キメラ抗体」は、哺乳動物のある種について、当該種以外の抗体において、当該種以外の抗体の定常領域が当該種の抗体の定常領域に置換されている抗体である。哺乳動物キメラ抗体では、ADCC活性を高める観点では、例えば、定常領域に用いる哺乳動物の抗体のサブタイプはIgG1あるいはイヌにおいてはIgGbとすることができる。
霊長類化抗体、霊長類キメラ抗体も同様の意味で用いられる。
本明細書では、「B細胞膜表面上のIgE抗体」とは、B細胞に結合したIgE抗体を意味する。IgE抗体は、B細胞表面にFcε受容体を介して結合し得る。IgE抗体はまた、IgE抗体を産生するB細胞の膜表面に膜結合タンパク(たとえば、マウスではM1タンパク、ヒトではM1’タンパク)によって発現しうる。B細胞膜表面上のIgE抗体は、B細胞受容体であり得る。B細胞は、刺激(例えば、CD40への刺激とIL−4またはIL−13による刺激)により活性化していることができる。CD40は、抗CD40抗体などの公知の様々な方法によって刺激することができる。また、抗CD40抗体の代わりにC4b−binding protein(C4BP)を使うこともでB細胞上のCD40を刺激することもできる。さらに、CD40によるB細胞刺激はB細胞内のTRAF(TNF−receptor associated factor)分子を活性化させるが、その活性化刺激はLMP1、CD40以外のTNF受容体(CD120a、CD120bなど)、CD27、CD30、CD267、CD269、B cell activation factor (BAFF/Blys/CD257)−receptor(CD268)、Toll様受容体(TLR)によってもCD40に対する刺激と同等の効果を得ることができる。あるいは、TNFファミリーのサイトカイン(TNFやAPRIL/CD256など)をB細胞に接触させても誘導できる。さらにヒトB細胞においては、B細胞の腫瘍化増殖を誘導するエプスタイン・バール・ウイルス(EBV、Epstein−Barr Virus)をB細胞に感染させてもよい。また、CD40刺激を使用せずにB細胞のIgE産生を誘導する方法として、IL−4、BAFF、および抗IgM抗体を同時にB細胞に接触さえる方法を用いることもできる。IL−4の代わりとして、その構造が類似したサイトカインのIL−13を用いることもできる。このように、様々な公知の方法を適宜用いることによって、当業者は、刺激されたB細胞を得ることができる。本発明の抗体は、このようにして刺激したB細胞表面上のIgEに結合し得る。
本明細書では、「肥満細胞表面上のIgE抗体」とは、肥満細胞に結合したIgE抗体を意味する。IgE抗体は、肥満細胞にFcε受容体を介して結合し得る。
本発明によれば、6C12は、加熱変性したIgE抗体とB細胞膜表面上のIgE抗体に結合性を示したが、加熱変性前のIgE抗体と肥満細胞表面上のIgE抗体に対しては反応性を示さなかった。このことから、B細胞膜表面上のIgE抗体は、少なくとも6C12のエピトープ部分において加熱変性したIgE抗体と同様の構造を取るが、加熱変性前のIgE抗体や肥満細胞表面上のIgE抗体とは異なる構造を取ると考えられる。
本明細書では、「肥満細胞表面上のIgE抗体」とは、肥満細胞に結合したIgE抗体を意味する。IgE抗体は、肥満細胞にFcε受容体を介して結合し得る。
本発明によれば、配列番号:18に記載のアミノ酸配列(DWIEGETYYC)を有するペプチドに結合する抗体が提供される。本発明によれば、配列番号:18に記載のアミノ酸配列(DWIEGETYYC)を有するペプチドに結合する抗体であって、B細胞膜表面上のIgE抗体に結合する抗体が提供される。いくつかの態様では、配列番号:18に記載のアミノ酸配列(DWIEGETYYC)を有するペプチドに結合する抗体は、配列番号:12〜17のいずれか1以上のペプチドには、配列番号:18に記載のアミノ酸配列(DWIEGETYYC)を有するペプチドよりも弱い親和性で結合する。
本発明によれば、加熱した病原性IgE若しくはB細胞表面上のIgEまたは配列番号:11に記載のアミノ酸配列(HHHHHHGGSGGSDWIEGETYYC)を有するペプチドとの結合に関して、配列番号:8に記載のアミノ酸配列を有する重鎖可変領域と、配列番号:9に記載のアミノ酸配列を有する軽鎖可変領域とを有する抗体と競合する、単離されたモノクローナル抗体が提供される。本発明によれば、加熱した病原性IgE若しくはB細胞表面上のIgEまたは配列番号:18に記載のアミノ酸配列(DWIEGETYYC)を有するペプチドとの結合に関して、配列番号:8に記載のアミノ酸配列を有する重鎖可変領域と、配列番号:9に記載のアミノ酸配列を有する軽鎖可変領域とを有する抗体と競合する、単離されたモノクローナル抗体が提供される。
配列番号:2に記載のアミノ酸配列を有する重鎖CDR1、配列番号:3に記載のアミノ酸配列を有する重鎖CDR2、及び配列番号:4に記載のアミノ酸配列を有する重鎖CDR3を含む重鎖可変領域と、
配列番号:5に記載のアミノ酸配列を有する軽鎖CDR1、配列番号:6に記載のアミノ酸配列を有する軽鎖CDR2、及び配列番号:7に記載のアミノ酸配列を有する軽鎖CDR3を含む軽鎖可変領域と
を有する。
(i)配列番号:8に記載の重鎖可変領域のアミノ酸配列を有する重鎖と、配列番号:9に記載の軽鎖可変領域のアミノ酸配列を有する軽鎖とを有し得る。本発明によれば、また、(ii)IgE抗体に結合する抗体は、配列番号8に記載の重鎖のアミノ酸配列を有する重鎖と、配列番号9に記載の軽鎖のアミノ酸配列を有する軽鎖とを有し得る。
本発明によればまた、IgE抗体に結合する抗体は、上記(i)または(ii)に記載されたIgE抗体に結合する抗体と、IgE抗体との結合に関して競合する抗体であり得る。本発明によればまた、IgE抗体に結合する抗体は、上記(i)または(ii)に記載されたIgE抗体に結合する抗体と、配列番号1、11、または18のアミノ酸配列を有するペプチドとの結合に関して競合する抗体であり得る。
IgE抗体に結合する抗体を製造する方法であって、
配列番号:1に記載のアミノ酸配列(NTNDWIEGETYYC)を有するイヌのペプチドを動物(例えば、非ヒト動物、非ヒト哺乳動物、鳥類)に免疫することを含む、方法が提供される。
本発明のある態様では、本発明の医薬組成物は、オマリズマブと併用されうる。
哺乳動物から得られた生体試料を加熱して、本発明のIgE抗体に結合する抗体と反応するIgE抗体を得ることと(または、本発明のIgE抗体に結合する抗体と反応する構造変化を生じる条件下で哺乳動物から得られた生体試料を加熱することと)、
加熱した生体試料と本発明のIgE抗体に結合する抗体またはその抗原結合フラグメントとを接触させることと、
を含む、方法が提供される。本発明の方法では、本発明のIgE抗体に結合する抗体は、加熱したIgE抗体に結合するが、臨床上のアレルギー症状を呈する検体の非加熱IgE抗体に対しての親和性が低く、これによって、血中に存在し得る非病原性IgE抗体による診断のノイズを抑制することができる。
本実施例では、抗IgE抗体の作製を試みた。以下、断りの無い限り、ヒトIgE抗体のアミノ酸配列は、GenBank登録番号:AH005278において登録されたアミノ酸配列であり;イヌIgE抗体のアミノ酸配列は、GenBank登録番号:L36872において登録されたアミノ酸配列であり;ネコIgE抗体のアミノ酸配列は、GenBank登録番号:AF162134において登録されたアミノ酸配列であり;ラットIgE抗体のアミノ酸配列は、GenBank登録番号:K02901において登録されたアミノ酸配列であり;マウスIgE抗体のアミノ酸配列は、GenBank登録番号:LC387253において登録されたアミノ酸配列である。
精製した6C12をビオチン標識し、ELISAシステムを確立して各動物種のIgE抗体に対する反応性を検討した。ELISA法は、96穴ELISA用ブラックマイクロプレート(グライナー社)に任意の濃度でIgE抗体およびIgG抗体をリン酸緩衝液に希釈して固相化した。固相化に使用したIgE抗体およびIgG抗体は次のとおりである。イヌIgE(Bethyl Laboratories社)、ヒトIgE(ミリポア社)、ラットIgE(インビトロジェン社)、マウスIgE(BD Phamingen社)、また別途作製された、カイコ体液より精製した組換型ネコIgEタンパク(Griot−Wenket et al., 2000, Vet. Immunol. Immunopathol., 75: 59−69参照)、イヌIgG(Cappel社)、ヒトIgG(ミリポア社)、及びネコIgG(Bethyl Laboratories社)を用いた。固相化は4℃で一晩行った。
本実施例では、得られた抗IgEモノクローナル抗体とIgE抗体の構造変化との関係を調べた。
IgEは、血中の遊離形態に加えて、B細胞表面および肥満細胞表面に発現している。本実施例では、得られた抗IgE抗体の細胞特異性を評価した。
本実施例では、組換えマウス抗体(マウスIgG1κ)および組換えイヌキメラ抗体を作製した。
本発明の抗IgE抗体6C12は、臨床的なアレルギー症状を呈する検体のIgE抗体に対しては、非加熱では反応せず、加熱すると反応を示した。これに対して、実験的にアレルゲンを皮下注射して感作した検体のIgE抗体に対しては加熱前後で結合性を示した。これにより、上記実施例では、6C12は、肥満細胞に結合可能なIgE抗体(病原性IgE抗体)には加熱前には結合できず、肥満細胞に結合不能なIgE抗体(加熱したIgE抗体および実験的に感作した検体のIgE抗体)には結合できると考えられた。

No.4:非病原性IgE抗体を含む検体から得られたIgE抗体 50 ng/mL
No.5:非病原性IgE抗体を含む検体から得られたIgE抗体 100 ng/mL
No.6:非病原性IgE抗体を含む検体から得られたIgE抗体 300 ng/mL
をそれぞれ図12の上段に示される箇所にそれぞれ投与した(1日目)。
リンパ腫を疑うイヌ(13歳、ヨークシャテリア種、去勢オス)の血液から末梢血単核球をフィコール比重遠心法で定法にしたがって分離した後、ウシ胎児血清を10%含有するRPMI−1640を用いて6か月間以上継代培養を行ったところ、継続的に増殖する腫瘍細胞を得た。この腫瘍細胞の細胞表面抗体の種類についてイヌ用の各種抗体を用いてフローサイトメトリー法で検討した。検討に使用した抗体は、抗イヌIgMヤギポリクローナル抗体(Novus Biologicals社)、抗イヌIgGヒツジポリクローナル抗体(AbD Serotec社)、抗イヌIgAヤギポリクローナル抗体(AbD Serotec社)、抗イヌIgEヤギポリクローナル抗体(Bethyl Laboratories社)および6C12で、それらいずれもビオチン標識されたものを用いた。これら抗体を1×106個/mlの細胞濃度に調整した腫瘍細胞浮遊液50μlに500μg/mlの濃度で添加して4℃で30分間反応させた後、洗浄して次にFITC標識ストレプトアビジン(BioLend社)を500μg/mlの濃度で添加し、さらに4℃で30分間反応させた。染色した細胞の検出はFACS Canto II(Becton, Dikinson and Company社)を用いて行い、解析直前に各細胞浮遊液に0.2μLで加えたPropidium Iodide(Propidimu Iodide Staining Solution、Becton, Dikinson and Company社)に染色された細胞を死細胞として除去した後、使用した抗体に染色された細胞集団を検出した。細胞集団の解析はFACS Diva software(Becton, Dikinson and Company社)を用いた。その結果、この腫瘍細胞はその細胞表面にIgEのみを発現し、その他の抗体であるIgM、IgG、IgAは発現していない細胞であることがわかった。よって、本腫瘍細胞をイヌIgE産生B細胞腫瘍細胞株とした。図13に示されるように、IgE抗体は、IgE産生B細胞の細胞表面に有効に結合した。
実験的放出(ER)は、各濃度の抗体溶液、エフェクター細胞、および標的細胞を混合した後に放出されるLDHの量を表し、
エフェクターによる自発的放出(ESR)は、エフェクター細胞が自発的に放出するLDH量を表し、
標的自発的放出(TSR)は、標的細胞が自発的に放出するLDH量を表し、
標的最大放出(TMR)は、溶解緩衝液で細胞を完全に破壊して得られる、標的細胞に含まれる全LDH量を表し、
培地バックグラウンド(CMB)は、培地中にバックグラウンドとして含まれるLDH量を表し、
体積補正用コントロール(VCC)は、培地中に溶解緩衝液を添加したときのLDH量を表す。
抗ヒトIgEモノクローナル抗体を固相化したPVDFメンブランプレートのウェルに培養後の細胞を8.4×104細胞/wellで添加し、24時間反応させた後、洗浄し、その後ビオチン標識した抗ヒトIgEモノクローナル抗体、ストレプトアビジン結合アルカリフォスファターゼを加えた後、基質を反応させてIgE産生B細胞のスポットを発色させた。肉眼的に認識可能な大きさの発色スポットを数えた。
1条件3ウェルアッセイとして平均値を出して評価した。結果は表5に示される通りであった。
本実施例では、ダニアレルゲンであるDer f 2に対するIgE抗体(Der f 2−IgE)をPNGaseF、EndoH、またはノイラミニダーゼで処理して、その糖鎖を分解し、分解された糖鎖を有するDer f 2−IgEに対する6C12抗体の親和性を確認した。
図15、17、および19に示されるように、6C12抗体は、病原性Der f 2−IgEに対して加熱処理依存的に結合した。病原性Der f 2−IgEを酵素処理すると、図15、17、および19に示されるように、レクチン(糖鎖結合タンパク質)との結合性が喪失したことから、糖鎖が分解された。
ここで、6C12抗体が、酵素処理した非病原性Der f 2−IgEおよび病原性Der f 2−IgEに結合するかを確認したところ、図16および18に示されるように、PNGaseFまたはEndoHで処理した病原性Der f 2−IgEに対しては、6C12抗体が親和性を示した。これに対して、図20に示されるように、ノイラミニダーゼで処理した病原性Der f 2−IgEに対しては、6C12は親和性を示さなかった。
6C12抗体は、表1に示されるように、免疫原として用いたペプチド配列のうちヒトと他の動物とで保存された領域に結合することが考えられる。

Claims (15)
- 配列番号:1または18に記載のアミノ酸配列からなる配列に対応するIgE抗体の重鎖のアミノ酸配列を有するペプチドに結合し、かつ
(1)B細胞表面上のIgE抗体に結合する、および/または
(2)56℃で加熱処理した遊離形態のIgE抗体に結合する、
単離されたモノクローナル抗体またはその抗原結合フラグメント。 - 配列番号:1または18に記載のアミノ酸配列からなる配列に対応するIgE抗体の重鎖のアミノ酸配列を有するペプチドに結合し、かつ
(1)B細胞表面上のIgE抗体に結合する、
請求項1に記載の抗体またはその抗原結合フラグメント。 - 配列番号:1または18に記載のアミノ酸配列からなる配列に対応するIgE抗体の重鎖のアミノ酸配列を有するペプチドに結合し、かつ
(2)56℃で加熱処理した遊離形態のIgE抗体に結合する、
請求項1に記載の抗体またはその抗原結合フラグメント。 - 請求項1に記載の抗体であって、
(1)B細胞表面上のIgE抗体に結合し、かつ
(2)56℃で加熱処理した遊離形態のIgE抗体に結合する、
抗体またはその抗原結合フラグメント。 - 肥満細胞表面上のIgE抗体に対するよりも、B細胞表面上のIgE抗体に対して強い親和性を有する、請求項2または4に記載の抗体またはその抗原結合フラグメント。
- 加熱処理前の遊離形態のIgE抗体に対するよりも、56℃で加熱処理した遊離形態のIgE抗体に対して強い親和性を有する、請求項3または4に記載の抗体またはその抗原結合フラグメント。
- 配列番号:2に記載のアミノ酸配列を有する重鎖CDR1、配列番号:3に記載のアミノ酸配列を有する重鎖CDR2、及び配列番号:4に記載のアミノ酸配列を有する重鎖CDR3を含む重鎖可変領域と、
配列番号:5に記載のアミノ酸配列を有する軽鎖CDR1、配列番号:6に記載のアミノ酸配列を有する軽鎖CDR2、及び配列番号:7に記載のアミノ酸配列を有する軽鎖CDR3を含む軽鎖可変領域と
を有する、請求項1〜6のいずれか一項に記載の抗体またはその抗原結合フラグメント。 - 配列番号:8に記載のアミノ酸配列を有する重鎖可変領域と、配列番号:9に記載のアミノ酸配列を有する軽鎖可変領域とを有する、請求項7に記載の抗体またはその抗原結合フラグメント。
- 請求項1〜8のいずれか一項に記載の抗体のイヌ化抗体またはその抗原結合フラグメント。
- 請求項1〜8のいずれか一項に記載の抗体のネコ化抗体またはその抗原結合フラグメント。
- 請求項2、4、5、7及び8のいずれか一項に記載の抗体またはその抗原結合フラグメントを含む、医薬組成物。
- 請求項3、4、6、7及び8のいずれか一項に記載の抗体またはその抗原結合フラグメントを含む、加熱した遊離形態のIgE抗体の検出に用いるための組成物。
- 哺乳動物においてアレルギー症状またはその発症リスクを検査する方法であって、
哺乳動物から得られた生体試料を加熱して、請求項3、4、6、7及び8のいずれか一項に記載の抗体と反応する遊離形態のIgE抗体を含む生体試料を得ることと、
加熱した生体試料と請求項3、4、6、7及び8のいずれか一項に記載の抗体またはその抗原結合フラグメントとを接触させることと、
を含む、方法。 - 請求項13に記載の方法であって、
加熱前の哺乳動物の生体試料と請求項3、4、5、7及び8のいずれか一項に記載の抗体またはその抗原結合フラグメントとを接触させることをさらに含む、方法。 - 請求項14に記載の方法であって、
哺乳動物から得られた生体試料に対する請求項3、4、5、7及び8のいずれか一項に記載の抗体またはその抗原結合フラグメントの反応性を生体試料の加熱前後で比較することをさらに含む、方法。
