Description
[1]下記式(I)で表される構造を有する化合物を10mM〜500mMと、チオール基を有する化合物を10mM〜500mMとを含む、サンドイッチ法で用いられる検体希釈液。

≪検体希釈液≫
本発明の検体希釈液は、下記式(I)で表される構造を有する化合物を10mM〜500mMと、チオール基を有する化合物を10mM〜500mMとを含む、サンドイッチ法で用いられる検体希釈液である。
さらに、本発明の検体希釈液は、任意成分として無機還元剤を含むことも好ましい態様の一つである。本発明の検体希釈液が無機還元剤を含む場合には、無機還元剤を10mM〜800mM含むことが好ましく、100mM〜200mM含むことがより好ましい。
なお、pHの測定方法は特に限定されるものではなく、一般的なpHの測定法(例えばガラス電極法)を用いて、適切な測定条件において測定すればよい。
本発明者らは、本発明の検体希釈液を用いることにより、検出対象物質を定量する際に、定量値に対するノイズが低減でき、シグナルの感度が良好となる理由は以下の通りであると推定した。
本発明者らは測定感度が低下する原因を、チオール基を有する化合物を用いた場合には、IgM等の夾雑物のジスルフィド結合のみではなく、サンドイッチ法を行う際の固相を構成する、検出対象物質を捕捉するための捕捉物質(例えば抗PSA抗体等のIgG)中のジスルフィド結合も同時に開裂してしまうことにあると推定した。本発明ではチオール基を有する化合物に加えて、式(I)で示される構造を有する化合物を用いることにより、以下の機構により、捕捉物質におけるジスルフィド結合の開裂を抑制することができ、結果としてシグナルの感度が良好になると推定した。
式(I)で示される構造を有する化合物が、チオール基を有する化合物と相互作用することにより、複合体を形成し、複合体を形成することにより、チオールによるジスルフィド結合の開裂の反応性を低下させ、より表面にジスルフィド結合を有するIgM等の夾雑物については、開裂が起こるが、捕捉物質ではジスルフィド結合の開裂が起こらないと推定した。なお、本発明の検体希釈液は、式(I)で示される構造を有する化合物を用いない場合と比べて、チオールの臭気が劇的に低減することも、式(I)で示される構造を有する化合物が、チオール基を有する化合物と相互作用をすることにより複合体を形成していると推定する根拠の一つである。
このようなことから、サンドイッチ法で本発明の検体希釈液を用いることにより、シグナル劣化を起こさず、感度が優れた定量を行うことができると、本発明者らは考えている。
本発明の検体希釈液は、下記式(I)で表される構造を有する化合物が含まれる。本発明者らは、下記式(I)で表される構造を有する化合物が、後述するチオール基を有する化合物と相互作用することによってノイズを低減し、かつシグナル劣化を起こさないのではないかと推察している。また、検出対象物質を定量する際の各種物質(例えば、金属イオン、イオン成分、検出対象物質、捕捉物質、夾雑物)が共存する複雑な測定系において、下記式(I)で表される構造を有する化合物が、これらの物質の溶解安定性を高めることにも寄与しているため、ノイズを低減し、かつシグナル劣化を起こさないのではないかと推察している。
R2としては、例えば、−COOH、−COO-Na+、−COO-K+、−COO-NH4+であり、各R2としては同一であっても異なっていてもよい。
波線は、他の原子との結合部位であり、他の原子としては、例えば、炭素原子が挙げられる。
式(I)で表される構造を有する化合物の具体例としては、エチレンジアミン四酢酸(EDTA)、ニトリロ三酢酸(NTA)、グリコールエーテルジアミン四酢酸(GEDTA)、ジエチレントリアミン五酢酸(DTPA)、ヒドロキシエチルイミノ二酢酸(HIDA)、ジヒドロキシエチルグリシン(DHEG)、ジカルボキシメチルグルタミン酸(CMGA)、ヒドロキシエチルエチレンジアミノ三酢酸(HEDTA)、トリエチレンテトラアミン六酢酸(TTHA)、1,3−プロパンジアミン四酢酸(PDTA)、1,3−ジアミノ−2−ヒドロキシプロパン四酢酸(DPTA−OH)、これらの塩から選択される少なくとも1種が挙げられ、好ましくはエチレンジアミン四酢酸(EDTA)、ニトリロ三酢酸(NTA)、グリコールエーテルジアミン四酢酸(GEDTA)、ジエチレントリアミン五酢酸(DTPA)、ヒドロキシエチルイミノ二酢酸(HIDA)、ジヒドロキシエチルグリシン(DHEG)、ジカルボキシメチルグルタミン酸(CMGA)およびこれらの塩から選択される少なくとも1種が挙げられ、より好ましくはエチレンジアミン四酢酸(EDTA)、ニトリロ三酢酸(NTA)およびこれらの塩から選択される少なくとも1種が好ましい。前記塩としては、例えばアルカリ金属塩、およびアンモニウム塩が挙げられ、アルカリ金属塩が好ましく、溶解安定性の観点から、ナトリウム塩がより好ましい。
式(I)で表される構造を有する化合物としては、一種単独で用いても、二種以上を用いてもよい。
本発明の検体希釈液は、チオール基を有する化合物が含まれる。
上記チオール基を有する化合物としては、例えば、ジチオスレイトール(DTT)、メルカプトエタノール、チオグリコール酸、またはL−システインから選択される少なくとも1種が挙げられ、DTT、メルカプトエタノールから選択される少なくとも1種が好ましく、DTTがより好ましい。
本発明者らは、以下の機構により、チオール基を有する化合物が夾雑物、特にIgM中の−S−S−結合を還元し、開裂させると推定した。
本発明の検体希釈液は、任意で、無機還元剤を含んでいてもよい。
上記無機還元剤としては、例えば、メタ重亜硫酸ナトリウム(SM)、亜硫酸ナトリウム(SS)、ハイドロサルファイト、および二酸化チオ尿素から選択される少なくとも1種が挙げられ、メタ重亜硫酸ナトリウム(SM)、亜硫酸ナトリウム(SS)から選択される少なくとも1種が好ましい。
本発明者らは、無機還元剤が夾雑物、特に水和性の高いタンパク質の表面の−S−S−結合を還元し開裂させるため、ノイズを低減しているのではないかと推察している。
抗体を用いたタンパク質の定性・定量法の一つとして、サンドイッチ法がある。サンドイッチ法とは、ウェルプレートや磁性粒子、センサーチップ等の測定領域に、あらかじめ検出対象物質(抗原)に対する補足物質(抗体、例えば抗PSA抗体等のIgG)を固相化しておき、免疫反応によって検出対象物質を捕捉した後、続いて、検出対象物質に特異的に結合する、標識化した物質を結合させて検出する方法である。
本発明のサンドイッチ法は、例えば表面プラズモン励起増強蛍光分光法(SPFS;Surface Plasmon−field enhanced Fluorescence Spectroscopy)、または、酵素免疫測定法(ELISA;Enzyme−Linked Immuno Sorbent Assay)により行われる、サンドイッチ法である。
表面プラズモン励起増強蛍光分光法(SPFS)は、光源より照射したレーザー光などの励起光が、金属薄膜表面で減衰全反射(ATR;attenuated total reflectance)する条件において、金属薄膜表面に表面プラズモン光(粗密波)を発生させることによって、光源より照射した励起光が有するフォトン量を数十倍〜数百倍に増やして、表面プラズモン光の電場増強効果を得ている。
酵素免疫測定法(ELISA)で用いることのできる酵素としては、西洋ワサビ由来のペルオキシダーゼ(HRP)やアルカリホスファターゼ(ALP)が代表的であり、それぞれに対応した適切な基質と組み合わせて用いればよい。
本発明では、検体が生体由来物質であることが好ましく、検出対象物質および夾雑物を含む可能性のある検体を用いることが好ましい。検出対象物質および夾雑物を含む可能性のある検体は、現実にそれらを含む検体であってもよいし、現実にはそれらを含まない検体であってもよい。具体的には、例えば、検出対象物質(および夾雑物)を含む可能性の高い特定の疾患の患者に由来する検体であってもよいし、検出対象物質(および夾雑物)を含む可能性の低い健常者に由来する検体であってもよい。また、検体を採取する対象は、典型的にはヒトであるが、ヒトの疾患のモデル動物、例えばマウス、ラット、モルモット、ウサギ、ヤギ、ネコ、イヌ、ブタ、サルといったヒト以外の哺乳動物であってもよい。
本発明の実施形態の好適な一例としては、血液を検体とする。ここで血液は、全血であってもよいし、全血から調製された血清または血漿であってもよい。また、抗凝固剤、希釈液(緩衝液等)、試薬等が添加されていてもよい。
検体に含まれる検出対象物質として、糖タンパク質が好ましい。検出対象物質としての糖タンパク質としては、例えば、検体中に含まれる、病理診断に用いられるマーカー分子である。例えば、前立腺特異抗原(PSA)、α-フェトプロテイン(AFP)、癌胎児性抗原(CEA)などのがん抗原/腫瘍マーカーが好適であり、その他にもシグナル伝達物質、ホルモンなどの糖タンパク質を検出対象物質とすることも可能である。例えば、LNCaP(ヒト前立腺癌細胞培養株)から、前立腺特異抗原(PSA)を得ることができる。
本発明の試料の製造方法は、検体1μgあたり、検体希釈液を1〜100μg混合する工程を有し、前記検体希釈液が、式(I)で表される構造を有する化合物を10mM〜500mMと、チオール基を有する化合物を10mM〜500mMとを含む、サンドイッチ法で用いられる試料の製造方法である。
式(I)で表される構造を有する化合物、およびチオール基を有する化合物については、前述した通りである。
なお、試料は室温(15〜25℃)で製造することが好ましく、製造してから、1秒〜30分で測定に供することが、夾雑物由来のノイズを低減できることから好ましい。
本発明で用いる試料は、検体と、検体希釈液とから成る試料であり、検体1μgあたりの検体希釈液量が、1〜100μgである、サンドイッチ法で用いるための試料である。
本発明の試料は前述の本発明の検体希釈液を用いて調製されるため、夾雑物に由来する影響、すなわち検出対象物の定量値に対するノイズが低減でき、シグナルの感度が良好となるため好ましい。
[比較例1、実施例1]
(検体希釈液の調製)
DTT、EDTA、またはEDTAとDTTの両方を含有するリン酸緩衝生理食塩水(PBS)溶液を、表1に記載の濃度に調製して、検体希釈液を得た。なお、コントロールは、PBSのみの検体希釈液とした。
表1に、各比較例、実施例ごとの検体希釈液の種類および濃度を示す。
上記の検体希釈液100μLに、PSAフリープール血清(正常ヒトプール血清、コージンバイオ社、ELISAにてPSA濃度が1pg/mL(0.001ng/mL)以下であることを確認)20μLを加えて混合し、ノイズ成分のシグナル測定試料として用いた。
上記の検体希釈液100μLに、上記PSA含有血清20μLを加えて混合し、シグナル成分のシグナル測定試料として用いた。
SPFS用のセンサーチップに、カルボキシメチルデキストランからなる膜(CMD膜)を形成するために、25mMのMES緩衝生理食塩水および10mMのNaCl溶液(pH6.0)を0.8mL滴下し、20分間反応させた。
その後、流路にTween(登録商標)20を0.05wt%含有する、トリス緩衝生理食塩水(TBS−T)溶液を導入し、3分間ピペッティングで往復送液して測定領域を洗浄した。
蛍光標識化レクチン(Alexa Fluor 647標識WFA)を、蛍光物質ラベリングキット「Alexa Fluor(商標名)647タンパク質ラベリングキット」(インビトロジェン社製)を利用して作製した。WFA(ノダフジレクチン)「L-1350」(Vector社)100μg相当と、0.1M重炭酸ナトリウムと、Alexa Fluor 647 reactive dyeとを混合し、室温で1時間反応させた後、ゲル濾過クロマトグラフィーおよび限外濾過を行い、標識に利用されなかったAlexa Fluor 647 reactive dyeを取り除いて、蛍光標識化WFAを回収した。得られた蛍光標識化WFA溶液の吸光度を測定して濃度を定量し、PBSで希釈して1μg/mLの濃度となるよう調整した。
センサーチップ内に、上記で調製した1μg/mL蛍光標識化レクチン(Alexa Fluor 647標識WFA)溶液100μLを導入した。導入した溶液を、ピペッティングで10分間往復送液し、測定領域と反応させた。
下記式(II)を用いて、得られたシグナル値から、S/N比を算出した。

メタ重亜硫酸ナトリウム(SM)、または、亜硫酸ナトリウム(SS)を含有するPBS溶液を、表4に記載の濃度に調製して、比較例4の検体希釈液とした。
表4の結果を考察する。DTTおよびEDTAと、SMまたは、SSのどちらか1つとを組み合わせて含有する場合、ノイズ成分のシグナル値を低減する効果が非常に優れているため、感度が良好になったことが示唆された。

- 前記無機還元剤が、メタ重亜硫酸ナトリウム(SM)、亜硫酸ナトリウム(SS)、ハイドロサルファイト、および二酸化チオ尿素から選択される少なくとも1種である、請求項8に記載の検体希釈液。
- 検体が生体由来物質であり、検体に含まれる検出対象物質として糖タンパク質を含む、請求項10に記載の試料の製造方法。
- 検体と、請求項1〜9のいずれか一項に記載の検体希釈液とから成る試料であり、
検体1μgあたりの検体希釈液量が、1〜100μgである、サンドイッチ法で用いるための試料。 - 検体が生体由来物質であり、検体に含まれる検出対象物質として糖タンパク質を含む、請求項12に記載の試料。
- 請求項12または13に記載の試料を用いて、表面プラズモン励起増強蛍光分光法(SPFS;Surface Plasmon−field enhanced Fluorescence Spectroscopy)、または、酵素免疫測定法(ELISA;Enzyme−Linked Immuno Sorbent Assay)により行われる、サンドイッチ法。
