(材料および方法)
(マウス)
7〜10週齢の雌性C57BL/6JマウスをCLEA JAPAN INC.から購入した(日本、大阪)。MyD88 KOマウスをOriental BioService Inc.(日本、京都)から購入した。STINGのリガンド結合部位に機能欠失型変異を有する[Sauer et al., Infect. Immun.(2011)79:688−94]IL−12p40 KOおよびSTING変異マウス(Tmem173gt)を、Jackson Laboratories(Bar Harbor、ME、USA)から購入した。IRF3/7 DKOマウスを、IRF3 KO[Tang et al., PLoS One.(2013)8:1−6]およびIRF7 KOマウスから生成し、これらのうちの後者は、MEXTのNational Bio−Resource Projectを介して、RIKEN BRC(日本、茨城)によって提供された。IFNAR2 KOマウスをB&K Universalから得た。全ての動物実験を、Animal Care and Use Committe of RIMDおよび大阪大学免疫学フロンティア研究センター(IFReC)のガイドラインに従って行い、動物の使用は、大阪大学により承認された。
2’3’−cGAMP、c−di−GMPおよび3’3’−cGAMPを、Invivogen(San Diego、CA、USA)から購入した。DMXAAをSigma−Aldrich(St. Louis、MO、USA)から購入し、5%のNaHCO3に溶解した。ヤマサ(日本、千葉)は、c−di−GMPを提供してくれた。OVAを関東化学(日本、大阪)から購入し、エンドトキシンレベルを、Toxicolor(登録商標)(生化学工業、日本、東京)により1EU/mg未満として決定した。K3 CpG ODN(配列番号1)を、以前記載されたようにGene Design(Ibaraki,Osaka,Japan)によって合成された。
マウスIL−12p40、マウスIL−13、およびヒトIFNγのレベルを、BioLegend(San Diego、CA、USA)のELISAキットを用いて測定した。
Mann−WhitneyのU検定、Studentのt検定またはBonferroniの多重比較検定を含む一元分散分析を、統計分析に用いた(*p<0.05;**p<0.01;***p<0.001)。統計分析をGraphPad Prism software(La Jolla、CA、USA)を用いて行った。
麻酔後、C57BL/6Jマウスを、0および10日目に、OVA(10μg)、またはOVAおよびK3 CpG(10μg)、DMXAA(50μg)、c−di−GMP(1μg)、2’3’−cGAMPまたは3’3’−cGAMP(1μg)、あるいはK3 CpG+2’3’−cGAMP/3’3’−cGAMP/c−di−GMP/DMXAAで筋肉内に免疫化した。17日目に、OVA特異的血清IgG1およびIgG2cを、以前に記載されたELISAによって測定した[Kuroda et al., Immunity.(2011)34:514−526]。IgG2cおよびIgG1のELISAにおいて使用される二次抗体は、西洋ワサビペルオキシダーゼコンジュゲートヤギ抗マウスIgG2cおよびIgG1(Bethyl Laboratories、Montgomery、TX)であった。17日目に、脾臓細胞を回収し、単細胞懸濁液を、gentleMACS dissociator(miltenyi Biotech、Gladbach、Germany)を用いて調製した。Tris−NH4Clバッファーを用いた赤血球の溶解の後、脂肪をRPMI(1%のペニシリン/ストレプトマイシンおよび10%のウシ胎児血清(FCS)を含有する)中で培養し、全OVA(10μg/ml)あるいはMHCクラスIまたはMHCクラスIIに特異的であるOVAペプチド(それぞれOVA257、OVA323)(10μg/ml)で48時間刺激した。IFNγおよびIL−13の産生をELISAによって測定した。
アゴニストのSTINGアゴニスト、DMXAA、c−di−GMPおよび哺乳動物の2’3’−cGAMPが、2型免疫応答を誘導することが報告されていることを考慮して[Zhang et al., Mol. Cell.(2013)51:226−35;Burdette et al., Nature.(2011)478:515−8;Tang et al., PLoS One.(2013)8:1−6]、本発明者らは、次に、K3 CpGがこれらの他のSTINGアゴニストと協同する能力を調べた。マウスPBMCは、3’3’−cGAMPで刺激しただけでなく、2’3’−cGAMPおよびc−di−GMPもK3 CpGと協同して、先天性IFNγの産生を誘導した(図3A)。
全てのhPBMCの実験を、Institutional Review Board of the National Institute of Biomedical Innovationからの承認に従って行った。ヒトPBMCを、ヒトリンパ分離培地(IBL、日本)を用いて健常な血液提供者の血液から単離し、1×106個の細胞をRPMI中で培養した。PBMCをK3 CpG(10μg/ml)、cGAMP(10μM)、またはK3 CpG+cGAMPで24時間刺激し、IFNγの産生をELISAによって測定した。
K3 CpGは、1型免疫応答を誘導すると報告されているヒト化K型(B型としても知られている)CpG ODNであるが、弱くIFNを誘導するだけである[Klinman et al., Nat. Rev. Immunol.(2004)4:1−10;Verthelyi et al., J. Immunol.(2001)166:2372−2377]。他方で、cGAMPは、強い1型IFNを誘導し得、アジュバントとして機能し[Li et al., Science.(2013)341:1390−4]、他のSTINGアゴニストは、2型免疫応答を誘導することが報告された[Tang et al., PLoS One.(2013)8:1−6]。K3 CpGおよびcGAMPのこれらの公知の制限を克服するために、本発明者らは、K3 CpGおよび標準3’3’−cGAMPの組み合わせの免疫賦活性の特性をin vitroにおけるhPBMCにおいて調べた。TLR9媒介シグナル経路とSTING媒介シグナル経路との相互作用を見出すために複数のhPBMCを用いて多くのサイトカインをスクリーニングした後(データは示さず)、本発明者らは、本発明の組み合わせが、IFNγの誘導において、強力な相乗作用(K3 CpGまたはcGAMP単独での刺激よりも約10〜90倍)を示すことを見出した(図1A)。
マウスPBMCを、マウスリンパ分離培地(IBL、日本)を用いてC57BL/6Jマウスから単離し、0.5×106個の細胞をRPMI中で培養した。GM−DCの培養物を、C57BL/6Jマウスの脛骨および大腿骨由来の骨髄細胞を洗い流し、これらの細胞を、20ng/mlのGM−CSF(Pepro Tech、Rocky Hill、NJ、USA)の存在下で7日間培養するによって調製した。GM−DCを、1%のペニシリン/ストレプトマイシンおよび20%のFCSを含有するRPMI中で培養した。FL−DC培養物を、100ng/mlのヒトFlt3L(Pepro Tech)の存在下で7日間培養されたC57BL/6Jマウスの骨髄細胞から調製した。FL−DCを、1%のペニシリン/ストレプトマイシンおよび10%FCSを含有するRPMI中で培養した。
マウスにおける初期(先天性)IFNγの誘導に対する本発明のTLR9アゴニストとSTING−アゴニストとの間の相乗作用を調べるために、本発明者らは、マウスPBMC(mPBMC)をK3 CpG、cGAMP、またはこれらの組み合わせでin vitroにおいて刺激した。本発明者らがhPBMCにおいて観察したものと同様な相乗的態様において、多量のIFNγの産生を観察した。IRF3およびIRF7は、それぞれcGAMP媒介性I型IFNおよびCpG媒介性I型IFNの誘導のための必要な下流の分子であるので[Wu et al., Science.(2013)339:826−30;Kawai et al., Nat. Immunol.(2004)5:1061−8]、本発明者らは、野生型マウスまたはIRF3およびIRF7の両方を欠損したマウス(ダブルノックアウト、DKO)のどちらかに由来するmPBMCを用いて、相乗的なIFNγの産生におけるIRF3およびIRF7の役割を調べた。相乗的なIFNγの産生が、IRF3/7DKOのmPBMCにおいて抑制された(図2A)。
ヒトPBMCをK3 CpG(10μg/ml)、cGAMP(10μM)、またはK3 CpG+cGAMPで16時間刺激し、最後の4時間はブレフェルジンAの存在下で刺激した。刺激後、細胞を回収し、CD16−PerCP−Cy5.5抗体(BD Biosciences:Franklin Lake、NJ)、CD56−BV421抗体(BioLegend)、CD3−FITC抗体(BD Biosciences)およびCD8−PE抗体(Miltenyi Biotech)を用いて表面分子を染色した。固定化および透過処理された細胞を、細胞内IFNγの検出のためにIFNγ−APC(BioLegend)で染色し、BD FACSCANTO II フローサイトメータを用いて分析した。
E.G7−OVA胸腺腫細胞を、American Type Culture Collection(VA、USA)から購入し、RPMI中で培養した。1×106個の細胞を、0日目にマウスの背中の皮下に注射した。7および10日目に、マウスは、PBS(50μl)、K3 CpG(10μg)、cGAMP(10μg)、またはK3CpG+cGAMPが腫瘍内に与えられ、腫瘍の成長についてマウスを22日間モニターした。
Th1細胞およびCD8+T細胞の応答は、抗腫瘍免疫の精製に重要であるので、本発明者らは、マウス腫瘍モデルにおけるK3 CpGおよびcGAMPの組み合わせの免疫療法の潜在能力を調べた。本発明者らは、OVA発現EG−7リンパ腫細胞をマウスに皮下注射によって接種した。7日目および10日目に、マウスは、PBS、K3 CpG(10μg)、cGAMP(10μg)、またはK3 CpGおよびcGAMPの腫瘍内注射を受けた。組み合わせ処置は、PBS、K3 CpG、またはcGAMPの処置と比べて、腫瘍の成長を顕著に抑制し(図5)、これは本発明の組み合わせは、ガンに対する抗原を含まない免疫治療剤として機能し得ることを示唆する。
野生型、Tmem173gt、IRF3/7 DKO、MyD88 KOおよびIFNAR KO C57BL/6Jマウスを、OVAおよびK3 CpG、cGAMP、またはK3 CpG+cGAMPで、0および10日目に筋肉内経路を介して免疫化した。17日目に、OVA特異的血清IgG2cおよびIgG1をELISAにより測定した。また、脾臓細胞をOVAで48時間刺激し、IFNγの産生をELISAにより測定した。
本発明者らは、mPBMCにおいて先天的なIFNγの相乗的な産生が、cGAMPおよびK3 CpGのそれぞれによるI型IFNの誘導に必要とされるIRF3およびIRF7に完全に依存することを示した。cGAMPはSTINGのリガンドであり、K3 CpGは、アダプター分子MyD88を介してシグナル伝達するTLR9のリガンドであるため、本発明者らは、IRF3/7 DKOマウス、IFNα/βレセプター(IFNAR)KOマウス、MyD88 KOマウスおよびSTING変異マウスを用いて、IRF3/7、MyD88、STINGおよびI型IFNの、組み合わせによって誘導される抗原特異的IFNγの相乗的な産生における関与を評価した。血清における組み合わせによって誘導された抗原特異的IgG2cおよび脾臓によるIFNγの産生は、野生型マウスと比較して、STING変異マウス、IRF3/7 DKOマウス、MyD88 KOマウスおよびIFNAR KOマウスにおいて顕著に減少した(図4Aおよび図4B)。
本実施例は、アレルギー抑制についての実施例である。
現行のアジュバントの多くは抗体産生を誘導するTh2アジュバントであるが、これらのアジュバントはガンやアレルギーのワクチン療法には不向きであった。さらに、STINGアゴニストは副反応としてIgEを誘導するため、アレルギー性炎症を引き起こす恐れもある。しかしながら本発明では、二種類のアジュバンを組み合わせるだけでIgE誘導を抑えながら強力なTh1アジュバント効果を誘導できる(図8)。
細胞内病原体またはガンに対する効率的なワクチンは、1型免疫応答を誘導するアジュバントを必要とする。cGAMPおよびc−di−GMPなどの環状ジヌクレオチドは、膜貫通型分子STINGに直接結合し、TBK1−IRF3依存性シグナル経路を活性化してI型IFNを誘導するので、潜在的なワクチンアジュバントとして注目されてきた[Dubensky et al., Ther. Adv. Vaccines.(2013)1:131−143]。しかし、STING−アゴニストが、1型防御免疫応答よりもむしろ2型免疫応答を誘導するという証拠は[Tang et al., PLoS One.(2013)8:1−6]、それらの潜在的な治療的適用を制限することを示唆している。この研究において、本発明者らは、STING−アゴニストをTLR9リガンドのK3 CpGと組み合わせることによってこの問題を解決する。この組み合わせは、先天性IFNγおよび適応性IFNγの産生を相乗的に増強する。それは、強力な1型アジュバントとして機能し、抗体ならびにCD4+Th1細胞およびCD8+T細胞を強く誘導し、そしてマウス腫瘍モデルにおける腫瘍の成長を効率的に抑制し得る抗腫瘍剤として機能する。
本実施例では、実施例1〜9と同様の実験を行う。cGAMP、c−di−GMP、2’3’−cGAMP、DMXAAを用いて、実施例7〜9の条件で、3’3’−cGAMPに代えて、cGAMP、c−di−GMP、2’3’−cGAMP、DMXAAを用いて、同様の実験を行う。
本実施例では、実施例1〜9の実験をK3以外のCpG1826、D35 CpGのようなCpGを用いて行った。
CpG1826(配列番号2=5’-tccatgacgttcctgacgtt-3’)、およびD35 CpG(配列番号3=5’-ggtgcatcgatgcagggggg-3’)を、それぞれInvivogenから購入、およびGene Design(Ibaraki,Osaka,Japan)によって合成された。これらは、合成または購入後、凍結乾燥品を滅菌水にて溶かして使用した。
マウス(c57BL/6)の脾細胞を1×107細胞/mlに調整し、in vitroにおいて上記CpG等の刺激剤で24時間刺激した培養上清中のIFN−γをELISA法にて測定した。これらの方法は、(一般手法)に記載されるように行った。
(結果)
結果を図12に示す。示されるように、それぞれのCpGは、STINGアゴニストである3’3’−cGAMPと組み合わせた場合に脾臓細胞においてインターフェロン産生において相乗効果が生じることが実証された。したがって、本発明の作用効果は、特定のCpGに限定されず広くCpG一般において発揮されることが実証されたといえる。
