Description
本発明は高血圧の経口治療に有効なメチルドー
パ含有液体組成物に関する。 メチルドーパ（Methyldopa）〔Ｌ−３−（３，
４−ジヒドロキシフエニル）−２−メチルアラニ
ン）〕は公知の抗高血圧剤である。この剤は高血
圧症患者の治療のために通常は経口投与される。
一般に使用されている投薬製剤形状は錠剤であ
る。たとえば子供や老人などの場合では、少なく
とも錠剤形製剤と等価な生体内有効性を有するも
のであればメチルドーパを甘味液体製剤形で投与
した方が有利な場合がある。 今回、本発明によつてメチルドーパを含有する
液状薬剤組成物が開発された。すなわち、本発明
はメチルドーパとスクロースとを含有する高血圧
治療のための液体薬剤組成物を提供することであ
る。 本発明による人の高血圧症治療のための薬剤組
成物はメチルドーパとスクロースとを含有する水
性懸濁物よりなる。 その水性懸濁物は水性懸濁物１mlにつき約15乃
至100mgのメチルドーパを含有しうる。好ましく
は水性懸濁物１mlにつき25乃至100mg、最も好ま
しくは約50mgのメチルドーパを含有する。 その水性懸濁物はさらにメチルドーパの１mgに
対し約１乃至24mgのスクロースを含有する。好ま
しくは水性懸濁物はメチルドーパの１mgに対し約
１乃至12mgのスクロースを含有する。より好まし
いメチルドーパ：スクロースの重量比は約１：２
乃至１：10であり、最も好ましいのは約１：10で
ある。 上記必須成分すなわちメチルドーパとスクロー
スとのほかに、本発明の組成物はさらに薬物学的
に有効な懸濁物を調製するために従来慣用されて
いる他の成分を含有することができる。このよう
な成分を例示すれば酸化防止剤たとえば重亜硫酸
ナトリウム、ジナトリウムエデテート
（disodium edetate）およびクエン酸、粘稠／沈
殿防止剤たとえばキサンテンゴムおよびビーゴム
Ｋ（VeegumK）、芳香剤、防腐剤、接着剤などで
ある。本発明の懸濁物を調製するためには従来慣
用の方法および装置が使用される。 本発明の懸濁物のすぐれた特徴はそのメチルド
ーパの生体内有効性すなわち効能が従来の錠剤形
状で投与されるメチルドーパと等価あるいはそれ
以上であることである。薬物学的に有効な懸濁物
を調製するために従来非常によく使用されている
ソルビトールを本発明の懸濁物中のスクロースの
代りに使用すると、そのメチルドーパの生体内有
効性は錠剤として投与された場合のメチルドーパ
のそれより実質的に低くなつてしまう。これは誠
に驚くべきことである。なぜならばソルビトール
はたとえばビタミン類など他の薬剤活性化合物の
生体内有効性を高めると報告されているからであ
る（Ｃ＆Ｅ、News1958年２月24日号、第59〜60
頁参照）。 メチルドーパの生体内有効性（bioavail−
ability）は生体内で次のようにして判定した。
すなわち、メチルドーパ500mgを標準錠剤形態な
らびに懸濁液形態で投与した被験者の血漿および
尿中のメチルドーパおよび複合メチルドーパの濃
度を測定した。これらの測定の結果は各種投与製
剤形態でのメチルドーパの生体内有効性が以下の
ごとくであることを示した。 スクロース含有懸濁物 ＝または＞錠剤＞ソルビトール含有懸濁物 以下に、これら懸濁物及び錠剤の配合処方例に
おける、具体的薬効数値データを示す。

- 【特許請求の範囲】 １ メチルドーパとスクロースとを含有する水性
懸濁物からなる高血圧の経口治療用薬剤組成物。 ２ メチルドーパ１mgにつきスクロース約１乃至
12mgを含有する特許請求の範囲第１項による薬剤
組成物。 ３ メチルドーパ１mgにつきスクロース約２乃至
10mgを含有する特許請求の範囲第１項による薬剤
組成物。 ４ 水性懸濁物１mlにつきメチルドーパ約50mgを
含有する特許請求の範囲第３項による薬剤組成
物。 ５ 水性懸濁物１mlにつきスクロース約500mgを
含有する特許請求の範囲第３項による薬剤組成
物。 ６ 水性懸濁物１mlにつきメチルドーパ約50mgそ
してスクロース約500mgを含有する特許請求の範
囲第３項による薬剤組成物。 ７ 錠剤形状のメチルドーパ製剤と効能において
等価であることを特徴とする特許請求の範囲第１
項の組成物。
