ゲリペプチドー毒素混成タンパク質
本発明は線状二二一マペプチドを井有ジスルフィド含有結合によって毒素涌片に
カップルさせて成る選択された1乳動物の細胞を治療処置するのに有用な1群の
混成タンパク質(hybrid protein ) に関し、より詳細には、
下垂体1瘍におけるような下垂体細胞を治療処置するのに有用、かつ高血圧症及
び対lI痺患者の筋肉けいれんを臨床処置するのに有用なホルモン−毒素混成タ
ンパク質に関する。より詳細には、本発明はネーチャー(二二一パイオマジー)
、233巻、8−11(1971)にウチダ等が記載するような突然変異体C,
ジフテリア鳳菌株Cv (β tox −45) の早期終了tox遺伝子生成
物(自生毒素の分子量62,000に対して分子量45,000’)である交さ
反応物質45 (CRM 45 )として知られる毒素にカップルさせた甲状腺
刺激ホルモン放出ホルモン(TRH)から成るキメラに関する。
生体内の特異の選択Ka胞(標的細胞)を他のa胞に影響を与えることなくそこ
なったり或は破壊することのできる治療剤を提供することが長い間の目標であっ
た。選択された群の細胞に特異的に結合することのできるリガンドにカップルさ
せた毒素から成る混成タンパク質は従来提案されてきたが、これらの混成タンパ
ク質は種々の理由から治療剤としての役に立たなかった。ジフテリア毒素A鎖を
ヒト胎盤性催乳ホルモンにジスルフィド結合を通して橋かけすることによってカ
ップルさせて成る混成タンパク質を合成することが提案された。しかし、J。
Btol 、 Ch@w、、252巻、1515−1522(1977)チャン
等に報告されているように、キメラは填乳ルセプターを含有する細胞に結合した
が、それらの4鹿におけるタンパク質の合成を抑制しなかった0リチンA毒素を
ジスルフィド結合を通して同様に僑かけすることによってヒト絨毛膜ゴナトド冒
ビンホルモン蹟にカップルさせて成るキメラが報告された;特異性を有すると言
われているが、その結合能力は報告されなかったし、かつラットライジツヒ踵瘍
S胞におけるタンノぐり質合成を顕著に抑制するのに極めて高い濃度が必要とさ
れ、エンドシト−シスによる非特異的入り込みと標的細胞の一胞質漢を横切るキ
メラの毒性部分の移送による特異的入り込みとを区別するのを極めて木蝋にさせ

1032(1979)瀾じ欠点がシスタミンを橋かけ剤として用いてインシュリ
ンにカップルさせたジフテリア人から成る混成物に見出された。ミスキミンズ等

橋かけ剤によって表皮成長因子(gGF )にカップルさせたりチン人から成る
混成物が作られたが、EGFによって付与される待合特性は選択された特異細胞
に制約されずに広範囲の細胞刑を包含する。フーリー等、セル、22巻、563
−570(1’980)。
線状二二一マペプチドを共有ジスルフィド含有結合によってCRM 45にカッ
プルさせて成る混成タンパク質又はキメラが、尊素活注(′4胞タンパク質合成
を抑制する)及び01M45の高い治窄比をも保搏しながら線状ニューペプチド
の@合特異憔を高文に保持することを今見出した。その上、遊1のCRMは′S
素膜レセプターに結合する能力が欠けており全ての@乳動物細胞に対し毒性がほ
とんど頌い1従って、カップ1Jングした後のCRM 45は選択された線状ニ
ューロペプチドが待合するそれらの細胞に対してのみ毒性がある。CRM 45
分子の自生構造体は疎水性アミノ巻シーケンスを含有し、該アミノ酸シーケンス
はそれが結合する細胞の脂質膜を混成分子が通り抜けるのを容易にさせるものと
考えられる。本発明の混成タンパク質の一部を形成することができる線状ニュー
ロペプチドの中には、甲状W#激ホルモン放出ホルモン(TRH) 、ソマトス
タチン、黄体形成ホルモン放出ホルモン、5ell−エンケファリン、meet
〜エンケファリン、二二一マテンシン、P物質、オキシトシン、ガストリン、コ
ルチコトロピン放出因子、コレシストキニン、モチリン、ボンベシンがある。こ
れらの中でTRHが好ましい、というのはTRHレセプターを運ぶセル型の数は
少いから、混成TRH−CRM 45についての標的細胞の範囲は極めて限られ
、下垂体のそれら(甲状礫刺激ホルモン及びプ豐ラクチンを作るそれら)の内の
ある傭、かつ一層低い程度まで視床下部の神経単位のある種及び畢丸のある橿の
細胞にほとんど独占的に限られるからである。混成TRH+ CRM 45はそ
の他の近い関係の細厄、例えば黄体形成ホルモンを作る下垂体1@泡に結合しな
い。従って、この混成物は下垂庫i!l唱、特にプロラクチノマス、チマトロピ
ノマス、ソマトトマピノマスの治療に有用であり癖かつ混成づが3′Ji勘性機
能及び自律神経系活性であって、共にTRHレセプターを運ぶものを制御するそ
れらの二ニーロンに特異的に結合することができることから高血圧症及び対AS
4S2O筋肉けいれんの処置に有用である。不発明のその他の混成タンパク質は
、キネマの選択されたニュープペプチドが待合するそれらの細胞をそこなったり
或は破壊する必要のある治療に有用である。
従って、不発明は線状ニューロペプチドをジスルフィドを通してCRM 45に
共有待合させて成り、線状二二一ロペプチドレセプターをゼする細胞に対し毒性
の混成タンパク質から成る。特に好適な生成物は、TRHをジスルフィド−合を
通してCRM 45に共有請合させて成01TRHレセプターを有する細胞に対
し毒性の混成タンパク質から成る。好適な実f!態様では、共有結合はTRfl
のヒスチジルイミダゾール基とCRM 45の任意の反応性基との間にあり1尚
一層好ましくは、結合はTREIのヒスチジルイミダゾール基とCRM 45の
アミノ又はアミド基との間にある。加えて、本発明は混成タンパク質を生理学的
に容認し得る毒性の無いキャリヤー、例えば塩類と一部にして成る、線状二ニー
四ペプチドレセプター、特にTRI’lし七ブタ−を達ぶ細胞に対してのみ毒性
の治療組成物、並びに該レセプターを運ぶ細2宛におけるタンパク質合成の抑制
方法であって、線状二二一ロペプチドから反る混成タンパク質、例えばCRM
45に共有待合させたTRT(を、例えば治療組成物を非経口注入によって細胞
に接触させることから成る前記方法とから成る。
線状二二一ロペプチドのC″Ryi45への共有結合は、各分子の選択された官
能基を共有反応によってジスルフィド結合を含む従来の橋かけ剤を通して互いに
カップリングさせる従来の任意の手順によって行うことができる。
一方では(CRM 45のンアミ7基と反応し、他方では(TRHの)ヒスチジ
ルイミダゾール基と何段碧かで反応する二官能性橋かけ剤を通しての間接カップ
リングが特に重要である。
次の実施例は本発明による混成タンパク質又はキネマの調製について例示する役
割を果すもので、本発明の蛇囲を制約するつもりのものではない。
実施例1
TRHとCRM 45とを二段階反応によって共有待合させた。第1段階で、T
RHとCRMとを各々独立に二官能性儒かけ剤に反応させて反応性の誘導体を形
成し、第2段階で、反応性誘導体を互いに反応させて共有結合した混成タンパク
質を形成した。
米国Proc 、 Natl 、 Aead 、 Set 、、 75巻、55
19−23(197B)、ギリランド等によって説明されている方法に類似の方
法で1−エチル−3(3−ジメチルアミツブ冒ピル)−カルボジイミドMCI
(EDAC) を触媒としてヨード酢酸とシスタミンとを棒金反応させて次式に
従って二官能性橋かけ剤を作った:1

30労後にpH25,6に上げ、市販の合成TRH2:、ηを亦工た。TRHと
のカルボキシメチル化反応をqttがりで、迩やかにふりまぜながら6時間次式
に従って運行させた:RH

完全に誘4化させた( derivatized )アセチロシスタミニル−T
RHをリンm 堰繰衝4で平衡にしたセファデックスG−10カラムでm!した
。ピーク留分を240 nmにおける光学1度によってめた。
CRM 45は据置(d@ferrated ) CY培地で生育させた溶原甜
Cy (βtox −45)の培養上澄み液から硫酸アンモニウム沈、投及びJ
、Bacteriol −i 56巻、1135−1142(197B)、バ
ハ等記載のDBAE イオン交遺クロマトグラフィーによって精製した。CRM
45 fi度は、J 、 Cl1n 、 Microbiol 、、 7巻、
91−96(197B)、マーフィー等に記載されるようにマケット免疫に%泳
動によってめた。
精製したCRM 45の第−及び第ニアミ7基を市販の腹素二官能性橋かけ剤で
あるN−スフシミジル−3(2−ビリジルジチオ)−ブマビオネー) (5PD
P ) で誘導化させた:
簡メには、αI M NaC1、α1Mリン酸ナトリウム援衝4(pH1sン中
のCRM45 (1m9/m)を、5PDPの2倍過剰量を少量の環水アルコー
ルに溶解した溶液に室温で30分間混合した。次に、混合物を(L 15 M
NaCl 。
[101Mリン酸ナトリウム(p H7,5) CPBS ) ニ対して透析さ
せたo fll変度、Biochem 、 J 、、 151巻、417−43
2(1975)、スタフベリー等によって先に説明されたように、2−ピリジル
ジスルフィド(FDP )結合を還元した後に345 nm における吸光度の
増大によってめた。平均では、CRM 45分子当りpDP fi分が1〜2で
あった。該調剤は生体外で測定してADPR−)ランスフェラーゼ活性を保持し
ていた。
TR)1とCRM 45との 結合
上述したように調部したアセチルシスタミニルーTRHをジチオトレイット5×
10”Mで室温において30tf間還元し、次にPBSで平衡させたセファデッ
クスG−10カラムで迅速な脱壜を行った。氏5′−ジチオビス−(2−二)口
安息香酸)で滴定してめるような遊浦のスルフヒドリル4で変性した次の構造を
有するTRH分子:

を含有する留分をCRM 45− PDPにおよそ10:1のモル比で直ちに混
合した。ジスルフィド反応を室温で進行させて343 nm における吸光度の
増大を分光光度計によって追った。反応を完結させた後に(通常20〜30労)
、混合物をリン眼堰a衝塩に対して大量に透析させて未反応TRH’ff子を取
り除いた。反応により次の構造の混成タンパク質が得られた:

μノ甲のNaN0t t 8 m9に10分間反応させて新しいジアゾ化(di
凰zot’1=ed ) p−アミノ安息香酸を−4した。アリ:ff ) 1

500μ!甲のTRH3O0μIに服えた。4 ’Cで24時間経過倹、シスタ
ミン6ダ、1−エチル−3(3−ジメチルアミツブ冒ピル)−カルボジイミドH
Cl5η、蒸留H,0500μlを反応配合物に皿えた。次に、pHを74Mし
て4.7に30分間保った。
完全に誘導化させたTRHをリン酸″14衝塩において平衡にした七ファデック
スG−10で楕裂し、次に実施例1で説明したア七チルシスタミニルーTRHと
同じ方法でジチオトレイットで還元し、脱壜し、CRM 45− PDPに反応
させた。債果は次の構造の混成タンパク質であった。
CH,ラット下垂体1瘍細胞(タシュジアン(Taahjian )等、196
B)を、馬血清15%、牛の冶児血清2.5k、グルタミン2X10 Mを補充
したノ)ンのF10培池r:L5d中のウェル当り5X10 細胞1度で24の
ウェルプレート(リンブマ)に接種した。72時間培養した〜10 M)を含有
する新しい培地で更に24時間再培養した。次に培地をl125μct c−o
イシン(280mci/ff1d 、ニューイングランドニュークリア)に直き
換えた。1時間後に培地を取り去り、付着した細胞を5%トリクロル酢ぼでよく
洗浄した。次に細胞をウルトラフルーオア(Ultrafluor ) (ナシ
ョナルダイアグノスチツクス)α5tILtGC町溶化した0中毒した培1によ
って加入したワイシンの量を未処理の対照培橿によって万口大したものの%とし
て地紋する投与量一応答カーブを作った。
特異的毒性は混成タンパク質のパッチからバッチへと変ったが、典型的には、実
施例1の混成タンパク質約5X10=Mの濃度がこの系におけるタンパク質合成
の50%抑制、即ち起源(orlgina )の毒性のおよそ200〜500倍
の毒性を引き起こした。実施例2の混成タンパク質は同じ系でlX10 Mの濃
度においてタンパク合成を50%抑制させた。
対照実験では、TRH−CRM 45 gE会合体増大した毒性が成分の生化学
的斐具によるよりもむしろ配合体自体に固有であることを示した、というのはア
セチルシスタミニルTRHはこの系で毒性が無く、かつCRM 45− PDP
がCRM 45単独の場合に予想されるのと1同じ程度にのみタンパク亘合成を
抑制したからである。1えて、配合体をジスルフィド橋かけを裟壊するだめのジ
チオトレイット(DTT ) 5 X 10 Mで或はジフテリア抗毒素で前処
理してその毒性を4滅させた。また、TRT(−CRM 45によるGH5細胞
の中毒を、細胞を過剰のTRHで1時間前培養して防いだ。同悌に、TRHレセ
プターを含有しない3T3ムース) (moust )線維芽細胞系統で配合体
の試験を行った場合には、’ CRM 45単独の毒性にすぎなかった。
配合体がaH3,Jlla上のレセプターについての” H−TRHに対抗する
能力を、J 、 Biol 、 Ch@m、、249 春、3086−3090
(1974Lヒンクル等の技法を直接用いて測定した。実施11の混成タンパク
質(KA ==3X10 )は・親和力が一層小さいが、TRHし七ブタ一部位
についての遊離のTRH(KA = I X 10 )に対抗することが明らか
であった、一方実施例2の混成タンパク質は親和力定数KA :10Mを示した
。
CRM 45或は混成タンパク質のどちらかをゆるやかに放射線ヨウ素で処理し
、クロマトグラフィー精製し、ラットの尾静脈中に注射し、15分後に−f!1
頭して行った局在研究では、前方下垂体剤、視床下部・畢丸においてCRM 4
5以上の混成物の増大した吸収を示したが、悩、肝、腎I、肺又は犀3において
は示さなかった。過剰の非放射性CRM 45を襟識混成吻として同時に注射す
ることにより、下垂体、視床下部、畢丸ii磁における吸収を低下した。
未処置対照に比べ、混成タンパク質で処置することによって培養中の正常ラット
下垂体細胞によるTRH及びプロラクチンの基礎分泌を低下させた。加えて、T
RH又は高カリウムパルスによって放出されるこれら2つのホルモン量もまた処
置細胞において顕著な程に一層少なかった。対照的に、TRHによって規制され
ないLH分泌はこれらの培養において影iを受けなかった。
また、乳を分泌する母体ラットを混成タンパク質で処置してもプロラクチン生成
を抑制し、ラットの子を死なせるか或は育たなかった。
