Abstract
PURPOSE: To determine the GOT activity of a specimen in high specificity, by adding the specimen to a reaction liquid containing a substrate consisting of cysteine sulfinate and 2-oxoglutarate or cysteine sulfinate and oxaloacetate, an intermediate electron carrier, and a tetrazolium salt compound.
CONSTITUTION: The specimen is added to a reaction liquid containing (A) a substrate consisting of cysteine sulfinate and 2-oxoglutarate or cysteine sulfinate and oxaloacetate, (B) an intermediate electron carrier such as phenazine, metho.sulfate, etc., and (C) a tetrazolium salt compound such as 3-(4,5-dimethyl-2-thiazolyl)-2,5- diphenyl-2H-tetrazolium bromide, and the absorbance of the mixture is measured. As an alternative method, the specimen is subjected to electrophoretic treatment and immersed in the above reaction liquid, and its color density is measured by densitmetry to determine the GOT activity.
