Abstract
PURPOSE: To prepare pentoxifylline having an ameliorant effect on the blood circulation advantageously, by using a 1-(3-halogenated propyl)-theobromine as a starting material, and hydrolysis or alcoholysis of novel 1-(4-acetyl-5-oxohexyl)- theobromine.
CONSTITUTION: A compound of formula I (X is halogen) is reacted with acetylacetone in the presence of a base at a temperature of preferably 50W120°C to give a novel compound of formula II, e.g. 1-(4-acetyl-5-oxohexyl)-3,7-dimethylxanthine, which is then isolated and subjected to hydrolysis or alcoholysis in the presence of a base at a temperature of preferably 20W80°C to afford a compound of formula III, common name: pentoxifylline, having an ameliorant action on the blood circulation. Thus, the aimed compound can be obtained from inexpensive raw materials through safe stages industrially and very advantageously. The bases used in the two stages are preferably sodium alcoholates in a nonaqueous solvent in the first stage and in an aqueous solvent in the second stage.
