Abstract
PURPOSE: The titled reagent eliminating or correcting defects, e.g., inacurate quantitative determination, complex handling, etc. resulting from conventional electrophoresis, method by carrying out at enzyme activity ratio of FDP/FIP, etc., comprising muscular aldolase antibody of enzyme marker and its determination.
CONSTITUTION: An antibody combined with a carrier to be in solid phase is brought into contact with a specimen, e.g., serum, etc. so that the antigen in the specimen (human muscular aldolase) is reacted with the antibody in solid phase, the reaction solution is removed and the solid phase is washed with a buffer solution, etc. [process (a)]. The solid phase obtained in the process (a) is brought into contact with an antibody of enzyme marker so that the antibody of enzyme marker is the first bonded to the antigen which is linked to the antibody in solid phase, the reaction solution is removed and the solid phase is washed [process (b)]. The solid phase obtained in the process (b) is brought into contact and reacted enzymically with a base of enzyme [process (c)]. The absorbency of the decomposed product of the base in the reaction solution of the process (c) is measured by a spectrophotometer using wavelength suitable for the qualitative determination [process (d)]. In the above-mentioned measuring system, the absorbencies of standard specimens whose amounts have been known are previously measured so that a calibration curve between amounts of the antigen and the absorbencies is determined. An amount of the antigen in the specimen is found from the absorbency of the specimen using the calibration curve.
