Abstract

PURPOSE: To prepare an optical active compound industrially advantageously, by subjecting a specific ester to asymmetric hydrolysis with a lipase capable of hydrolyzing the ester asymmetrically.
CONSTITUTION: A lipase, e.g., steapsin, or a bacterium, e.g., one belonging to the genus Geotrichum, capable of hydrolyzing an ester asymmetrically, and the ester shown by the formula (X is acyloxy; Y is hologen, acyloxy, or oxygen which forms oxirane ring in combination with Z; Z is holomethyl or acyloxymethyl when Y is halogen, Z is halomethyl or aryl when Y is acyloxy, and Z is methylene which forms oxirane ring in combination with Y when Y is oxygen) are added to a buffer solution sufficiently enough to neutralize an acid to be formed in asymmetric hydrolysis, and the ester is subjected to asymmetric hydrolysis. The ester and alcohol are extracted with an organic solvent, and the ester is separated from alcohol by washing with water, chromatography, etc. to give an optical active compound.
