Abstract
PURPOSE: To simply eliminate interference due to constituents except for an objective constituent, which are present together in a sample, by using a physical value, which results from the addition of a reaction assisting reagent to the sample, to compensate another physical value which results from the following addition of a trigger reagent.
CONSTITUTION: A reaction assisting reagent, which does not contaon one of constituents necessary for reaction, is added to a sample. A first physical value resulting from the addition is measured. A trigger reagent, which contains the necessary constituent that is not contained in the former reagent, is added to the sample. A second physical value resulting from the latter addition is measured. To analyze glucose in blood serum by a hexonase method, a first reagent containing ATP, NADP and G6P-DH is added to the serum. The absorbance (b) of the serum mixed with the first reagnet is measured. A hexonase solution is then added as a trigger reagent to the serum. After that, the absorbance (c) of the serum is measured. The absorbance difference (c-b) is multiplied by a concentration converting coefficient to determine the quantity of the glucose in the serum.
