Abstract

PURPOSE: To prepare an aminoplycoside having excellent antibacterial activity, in high yield, by reacting an antibiotic substance, fortimicin, with an acid, thereby breaking the methyl ether group.
CONSTITUTION: A compound selected from an antibiotic substance, fortimicin of formula I (R1,2 are H or methyl; R3 is H or acyl; R4 is H or OH; the amino group may be protected) and an antibiotic substance KA-7038, is reacted with an acid such as an organic strong acid, a Lewis acid, etc., pref. at room temp. to 200°C for 1W30hr. The products are separated and purified, e.g. by column chromatography, etc. to obtain the objective compound of formula II. If necessary, the 4- position methylamino group is acylated, and the protecting group of the other amino group is optionally eliminated. In the compounds of the formula I, those of formula III (Z1,2 are H or methyl; Z3 is H or acyl; when all of Z1WZ3 are H, the methylamino group at the 4-position is not trans configuration regarding to the 3- and the 5-position OH groups) are novel.
