Abstract

PURPOSE: To obtain the titled substance useful as an antibiotic readily, by reacting 3-formylrifamycin S with a condensation product prepared from a primary amine and a specific amount of formaldehyde, etc., and then with a reducing agent.
CONSTITUTION: 3-Formylrifamycin S of formula I is reacted with a condensation product prepared from a primary amine of the formula R-NH2 (R is alkyl, aryl, aralkyl or cycloalkyl group) and formaldehyde in an amount of at least equimolar with the primary amine. The reaction product is then reduced with a mild reducing agent, e.g. ascorbic acid, to give a compound of formula II. Alternatively, the compound of formula I is reacted with a condensation product obtained from a primary amine of the formula R-NH2 and formaldehyde in a molar, amount of twice that of the the primary amine and an alcohol of the formula R'-OH (R' is lower alkyl group) in an amount of twice that of the primary amine, and then reduced with the mild reducing agent.
