Abstract

NEW MATERIAL:Formimidoylistamycin B of formula I and an acid addition salt thereof. Apperance: white powder. Decomposition point: 202W210°C [α]D23=+47° (C=1, water).
USE: A semisynthetic aminoglycoside antibiotic having a more powerful antimicrobial activity than fortimicin A and almost the same antimicrobial activity as istamycin B and with low toxicity.
PROCESS: 1,2',6'-Tri-N-protected istamycin B expressed by formula II (Rs are monovalent protecting groups of the amino groups) is reacted with an iminoether to convert the amino group in the glycine part thereof into an amidine group and give a compound of formula III, which is then hydrolyzed to eliminate the protecting groups of the amino groups at the 1-, 2'-, and 6'-positions and give the compound of formula I.
