NEW MATERIAL:The novel title enzyme produced by the culturing of bacterial belonging to Pseudomonas genus, and capable of irreversibly producing formic acid by the oxidation of formaldehyde through nicotinamide-adenine dinucletide. Molecular weight, 15,000±1,000; isoelectric point, 5.2±0.1; optimum pH, 7W8.
USE: Reagent for the determination of uric acid in blood.
PROCESS: Bacteria belonging to Pseudomonas genus and capable of producing the above enzyme, e.g. Pseudomonas aeruginosa IFO3445, are inoculated in a medium containing 0.05W1wt% of metabolic precursors such as sarcosine, betaine, methylamine, creatinine, formaldehyde, etc., and cultured at 15W40°C and pH6W8 for 1W2 days. The title enzyme is extracted from the medium and purified by salting- out with ammonium sulfate, ion exchange chromatography, etc.
