Abstract
PURPOSE: To produce the title substance effectively, by collecting creatine amidohydrolase from a culture of AK-2 strains which belongs to the genus Alcaligenes.
CONSTITUTION: Strains of microorganisms AK-2 which belong to the genus Alcaligenes are cultured in a medium comprising creatine and/or creatinine as a main nitrogen and carbon sources, and an inorganic base, e.g. ammonium chloride or sulfate and an organic nutriment in a trace amount, e.g. yeast or malt extract, at about a neutral pH and 25W35°C for 1W2days aerobically with stirring to propagate the bacterium bodies wherein the enzyme is accumulated. The enzyme is then extracted from the bodies, separated, and purified.
EFFECT: Creatinine amidohydrolase and creatine amidinohydrolase can be produced at an enzyme activity ratio of 6:1W1:2.
