Abstract

PURPOSE: To obtain an enzyme capable of reducing an aromatic 2,4-hexadienoic acid, by cultivating a microorganism belonging to the genus Pseudomonas, and separating a 2-hydroxy-6-oxo-6-substituted-2,4-hexadienoic acid from the resultant produced microbial cell.
CONSTITUTION: A microorganism, e.g. Pseudomonas cruciviae S93B1 (FERM-P No. 5426) is cultivated in a broth culture medium containing glucose, etc. at pH 4W9 for 10W30hr aerovically, and then a 2-hydroxy-6-oxo-6-substituted-2,4-hexadienoic acid reductase is separated and purified from the microbial cell by the ultrasonic treatment, fractionation with ammonium sulfate, ion exchange chromatography, etc. The resultant enzyme has a wide substrate specificity to reduce a compound expressed by formula I into a compound expressed by formula II and is capable of reducing even 2,4-hexadienoic acid regardless of natural or unnatural origin. The formed compound is used as an intermediate for medicines and agricultural chemicals.
