Abstract

PURPOSE: To prepare 1,4-dihydro-2,6-dimethyl-4-(2'-nitrophenyl)-3,5-pyridinecarboxylic dimethyl ester, by reacting 2-nitrofenzal halogenide with methyl acetoacetate and ammonia.
CONSTITUTION: A 2-nitrofenzal halogenide shown by the formula (X is halogen) is reacted with methyl acetoacetate and ammonia in an inert selvent to prepare the desired compound. One mole of the compound shown by the formula I is used with 2.0W5.0 moles of methyl acetoacetate and 1.0W8.5 moles of ammonia. Methanol, pyridine, etc. may preferably be cited as the solvent. The reaction is preferably carried out in the presence of an organic base e.g., a tertiary amine, as a catalyst. An amount of the catalyst is not less than 0.1 equivalent based on the compound shown by the formula. The reaction is carried out in a temperature range of room temperature to 150°C.
EFFECT: The desired compound can be obtained readily, safely, efficiently in high yield.
USE: Medicines for angina pectoris.
