Abstract

PURPOSE: To prepare the title compound useful as a commerciallized remedy for stenocardia, safely and in high yield, by reacting an easily available 2-nitrobenzal halogenide with 3-aminocrotonic acid methyl ester.
CONSTITUTION: 1, 4-Dihydro-2, 6-dimethyl-4-( 2'-nitrophenyl )-3, 5-pyridinecarboxylic acid methyl ester or its salt of formula II is prepared in high yield (e.g. 85.2%) by reacting a 2-nitrobenzal halogenide of formula I (X is halogen) with 3-aminocrotonic acid methyl ester in an inert solvent, pref. in the presence of an organic base such as pyridine or ammonia. The starting materials are prepared economically, in high yield by known processes, e.g. the compound I is prepared from 2-nitrotoluene in two steps.
