Abstract

PURPOSE: A composition that is composed of o-nitrophenyl-β-galactopyroside and a specific alcohol, thus permitting very high-sensitivity determination of the activity of β-galactosidase.
CONSTITUTION: o-Nitrophenyl-β-D-galactopyranoside is dissolved in 5W100, preferably 20W80vol% of at least one alcohol selected from ethylene glycol, methanol, propylene glycol, n-propanol, glycerol, and triethylene glycol. The solution is added to a sample to be determined so that the alcohol concentration becomes 0.5W20% and the yellow color of the nitrophenol liberated by the action of β-D-galactosidase is determined by colorimetry.
