Abstract

PURPOSE: To obtain the title compound useful as an antitumor agent in high yield industrially advantageously, by reacting 5-fluorouracil with an aromatic acid halide in the presence of an excess organic base under specific temperature conditions through simple procedures.
CONSTITUTION: 5-Fluorouracil (F) is reacted with an aromatic acid halide, e.g. o-toluyl or benzoyl chloride, in the presence of an organic base in excess, preferably in a molar amount of twice that of F at 50°C or below, preferably room temperature or below, to give a 5-fluorouracil derivative, e.g. o-toluyl-5-fluorouracil, of the formula (R1 is acyl; R2 is H or acyl). Pyridine and triethylamine may be used as the base for preparing a 3-substituted 5-fluorouracil derivative and a 1,3-disubstituted-5-fluorouracil derivative, respectively.
