Abstract

PURPOSE: To prepare apramycin, economically in an industrial scale, by culturing Sacchropolyspora hirsuta (ATCC No.27875), and separating from the cultured liquid.
CONSTITUTION: Saccharopolyspora hirsuta (ATCC No.27875) is inoculated in an aqueous nutrient medium containing a carbon source and a nitrogen source, and cultured aerobically, at 15W40°C for 2W6 days. The cultured liquid is filtered, adsorbed to an ion exchange resin, activated charcoal, etc., and eluted with a 0.1W3N acid or alkali. The active component thus collected is freeze-dried to obtain crude powders of apramycin. The crude apramycin is treated with a weakly acidic cation exchange resin, CM-Sephadex, etc., and then eluted with an ammonia water or an aqueous solution of ammonium formate to obtain apramycin as free base. It is treated with a strong basic ion exchange resin, and eluted with water. The active fractions thus collected are freeze-dried.
