Abstract

PURPOSE: To prepare the title enzyme useful as a bioactive substance, from a dispersion of disintegrated cells obtained by the culturing of bacteria belonging to Thermus genus.
CONSTITUTION: Bacteria belonging to Thermus genus, e.g. highly thermophilic bacteria, Thermus thermophilus HB8, Thermus flavus BS1, AT61, AT62, etc., are cultured in a nutrient medium for about 6 hours, and collected at the latter period of logarithmic phase. The collected cells are disintegrated by, e.g. ultrasonic treatment. The objective phosphofructokinase is separated and purified from the dispersion of the disintegrated cells by centrifugal separation, ammonium sulfate fractionation, column chromatography, etc. The phosphofructokinase thus obtained is an allosteric enzyme, the activity of which is suppressed allosterically with phosphoenol pyruvic acid, and recovered to the original value with adenosine-diphosphoric acid.
