Abstract

PURPOSE: To prepare cephapirin useful as an anti-bacterial agent, in high yield, by acylating 7-aminocephalosporanic acid (7-ACA) with 4-pyridine thioacetic acid in the presence of culturing product of bacteria belonging to Arthrobacter genus.
CONSTITUTION: Bacteria belonging to Arthrobacter genus and capable of producing an acylating enzyme, e.g. Arthrobacter viscosous ATCC 15924 are cutured in a medium. Cephapirin is prepared by the acylation reaction of 7-ACA and 4-pyridyl thioacetic acid or its reactive erivative (pref. methyl ester) in the presence of the above cultured product or the cultured medium, at 30W45°C, pref. 35W40°C and pH6.5W8, pref. 6.8W7.2 for 1W5 hours. The concentration of 7-ACA in the reaction mixture is 5W50 mg/ml, pref. 15W30 mg/ml, and the molar ratio of pyridyl thioacetic acid to 7-AVA is 1W20, pref. 2W5.
