Abstract

PURPOSE: To prepare the title enzymes useful for the production of maltose, economically, in an industrial scale, by culturing bacteria belonging to Bacillus genus, and capable of simultaneously producing β-amylase and α-1,6-glucosidase, in a medium containing a manganese compound and a calcium salt.
CONSTITUTION: Bacteria belonging to Bacillus genus and capable of producing β- amylase and α-1,6-glucosidase simultaneously, e.g. Bacillus cereus var. mycoides FERM-P No.2391, are cultured in a nutrient medium containing (usually 1×10-6 W 1×10-3 M of) an organic or inorganic manganese compoound such as manganess sulfate, manganese acetate, etc., and (usually 0.01W0.5M of) a calcium salt such as calcium chloride, etc. After culturing, the insoluble matters are separated by centrifugal separation, to obtain a solution containing the title enzymes.
