Claims (1)
- 【特許請求の範囲】 1.新規抗生物質、抗菌剤又は抗微生物剤の同定に使用するための改善されたア ッセイ方法であって、 a.基質を固体担体に固定化する工程、 b.固体担体に固定化した基質とセンサータンパク質及び試験サンプルとを混 合する工程、 c.混合物をあらかじめ決定した時間インキュベートする工程、 d.固体担体から前記混合物の残りを分離する工程、及び e.前記固体担体を調査し、反応生成物が存在するかどうかを測定し、前記試 験サンプルの治療学的可能性を決定する工程、 からなることを特徴とする方法。 2.前記センサータンパク質が酵素又はその触媒作用的に活性な部分である、請 求の範囲第1項記載の方法。 3.前記酵素がキナーゼ酵素又はその酵素的に活性なポリペプチド部分である、 請求の範囲第2項記載の方法。 4.前記キナーゼがヒスチジンプロテインキナーゼである、請求の範囲第3項記 載の方法。 5.前記混合物が更に高エネルギーリン酸源を含む、請求の範囲第3項記載の方 法。 6.前記高エネルギーリン酸源がATP又はGTPである、請求の範囲第5項記 載の方法。 7.前記高エネルギーリン酸源が標識化されている、請求の範囲第5項記載の方 法。 8.前記基質が細菌のタンパク質又はポリペプチド分子を含む、請求の範囲第1 項記載の方法。 9.前記基質が細菌のSPoOFタンパク質又はそのポリペプチド部分を含む、 請求の範囲第1項記載の方法。 10.前記基質が更にリンカーを含む、請求の範囲第1項記載の方法。 11.前記リンカーが1以上のヒスチジン残基を含む、請求の範囲第10項記載の 方法。 12.前記固体担体が樹脂を含む、請求の範囲第1項記載の方法。 13.前記樹脂がニッケル結合交換樹脂(Ni樹脂)である、請求の範囲第12項 記載の方法。 14.新規抗生物質、抗菌剤又は抗微生物剤の同定方法であって、 a.基質と、キナーゼ酵素、高エネルギーリン酸源及び試験サンプルとを混合 する工程、 b.前記混合物をあらかじめ決定した時間インキュベートする工程、 c.工程bにおいて生成した反応生成物を、前記混合物の残りから分離する工 程、 d.形成した反応生成物量を確認し、前記試験サンプルの治療学的可能性を決 定する工程、 からなることを特徴とする方法。 15.前記基質又は前記酵素が標識化されている、請求の範囲第14項記載の方法 。 16.前記決定工程が、基質又は酵素と標識化された免疫複合体を形成する、標識 化された抗体の使用及び形成した標識化された免疫複合体量の定量化を含む、請 求の範囲第14項記載の方法。 17.前記標識が蛍光性標識、放射性標識、ビオチン等を含む、請求の範囲第15 項又は16項記載の方法。 18.前記基質が固体担体に固定化されている、請求の範囲第14項記載の方法。 19.前記固体担体が樹脂からなる、請求の範囲第18項記載の方法。 20.前記樹脂がニッケル結合交換樹脂(Ni樹脂)である、請求の範囲第19項 記載の方法。 21.前記高エネルギーリン酸源がATP又はGTPである、請求の範囲第14項 記載の方法。 22.前記酵素が、キナーゼ酵素又は前記酵素のアミノ酸残基配列の配列サブセッ トのアミノ酸残基配列に対応するアミノ酸残基配列を有するポリペプチドである 、請求の範囲第14項記載の方法。 23.前記キナーゼがヒスチジンプロテインキナーゼである、請求の範囲第22項 記載の方法。 24.前記基質が細菌のタンパク質又はポリペプチド分子を含む、請求の範囲第1 4項記載の方法。 25.高処理能アッセイ系又はキットであって、以下に示す試薬: a.固体担体に固定化した基質、 b.センサータンパク質及び、 c.高エネルギーリン酸源、 を、少なくとも1回のアッセイを行うのに十分な量で含むことを特徴とする系 又はキット。 26.前記の1種以上の試薬が標識化されている、請求の範囲第25項記載のアッ セイ系。 27.前記試薬のそれぞれが別々の容器に存在する、請求の範囲第25項記載のア ッセイ系。 28.前記容器がパッケージ内に封入されており、該パッケージが更に前記試薬の 使用の指示を含む、請求の範囲第27項記載のアッセイ系。 29.前記センサータンパク質が酵素又はその触媒作用的に活性な部分を含む、請 求の範囲第25項記載のアッセイ系。 30.前記高エネルギーリン酸源がATP又はGTPである、請求の範囲第25項 記載のアッセイ系。 31.前記酵素がキナーゼ酵素を含む、請求の範囲第25項記載のアッセイ系。 32.前記キナーゼがヒスチジンプロテインキナーゼである、請求の範囲第31項 記載のアッセイ系。 33.前記基質が細菌のタンパク質又はポリペプチド分子を含む、請求の範囲第2 5項記載のアッセイ系。 34.前記基質がSpoOFタンパク質又はそのポリペプチド部分を含む、請求の 範囲第32項記載のアッセイ系。 35.前記基質が更にリンカーを含む、請求の範囲第32項記載のアッセイ系。 36.前記リンカーが前記基質に結合する1以上のヒスチジン残基を含む、請求の 範囲第34項記載のアッセイ系。 37.前記固体担体が樹脂を含む、請求の範囲第25項記載のアッセイ系。 38.前記樹脂がNi樹脂である、請求の範囲第36項記載のアッセイ系。
