Claims (1)
- 【特許請求の範囲】 1. DNA由来の予め決定されている遺伝子エキソン類を分析する方法であっ て、遺伝子エキソンの連続するグループにプライマー対を添加し、続いて第1工 程である比較的長い多重ポリメラーゼ連鎖反応として1本の共通のチューブ内で それらのポリメラーゼ連鎖反応増幅を行う工程、この遺伝子エキソン類のそれぞ れに対するプライマー対をさらに添加しそして第2の工程である短鎖多重ポリメ ラーゼ連鎖反応としてその共通のチューブ内でそれらのポリメラーゼ連鎖反応増 幅を行う工程、及びその遺伝子断片を電気泳動的に分離する工程を含む方法。 2. 該方法が遺伝子断片を1次元に沿ってサイズに基づき電気泳動的に分離す るものである、請求項1記載の方法。 3. 該方法が、直交する次元に沿って配列特異的な位置に遺伝子断片を分布さ せるため、直交する次元に沿いそして温度勾配又は化学的変性勾配に沿って遺伝 子断片を塩基対配列に基づく更なる電気泳動的分離に付し、そしてこれらの位置 を正常な遺伝子断片の位置と比較して遺伝子の突然変異を検出するものである、 請求項2記載の方法。 4. プライマーが蛍光色素で標識されたオリゴヌクレオチド・プライマーであ る、請求項3記載の方法。 5. ポリメラーゼ連鎖反応に用いるヌクレオチド建築ブロックが蛍光的に標識 されているものである、請求項3記載の方法。 6. 電気泳動による分離が変性勾配ゲル中で行われるものである、請求項3記 載の方法。 7. 該方法が、断片を分離するため、ゲルに尿素とホルムアミドの勾配を適用 するものである、請求項6記載の方法。 8. 該方法が、断片を分離するため、ゲルに温度勾配を適用するものである、 請求項6記載の方法。 9. 長鎖−PCR遺伝子エキソングループが網膜芽腫遺伝子のエキソン1〜2 、3〜6、7〜11、12〜17、18〜23及び24〜27である、請求項3 記載の方法。 10. DNA由来の予め決定されている遺伝子エキソン類を分析する方法であ って、遺伝子エキソンの連続するグループにプライマー対を添加し、続いて第1 工程である比較的長い多重ポリメラーゼ連鎖反応として1本の共通のチューブ内 でそれらのポリメラーゼ連鎖反応増幅を行う工程、及びこの遺伝子エキソン類の それぞれに対するプライマー対をさらに添加しそして第2の工程である短鎖多重 ポリメラーゼ連鎖反応としてその共通のチューブ内でそれらのポリメラーゼ連鎖 反応増幅を行う工程を含む方法。
