Description
Claims (1)
- 【特許請求の範囲】 1.骨膜の骨細胞刺激に有効な量のFGF-1、及び医薬として許容し得る担体を含 む骨組織を含む病理学的状態の治療用医薬組成物であって、前記FGF-1が、1以 上のシステイン残基が分子間又は分子内ジスルフィド結合を形成することができ ないアミノ酸で置き換えられたものである組成物。 2.さらにヘパリン又はヘパリンフラグメントを含む請求項1記載の医薬組成物 。 3.前記FGF-1が、少なくとも約0.1 mg/mlであり、前記ヘパリン又はヘパリンフ ラグメントが、少なくとも約50単位/mlである、請求項2記載の医薬組成物。 4.骨膜の骨の成長を刺激するのに有効な量のヒト繊維芽細胞成長因子を投与す ることを含む、骨組織を含む病理学的な状態により苦しめられているヒトの該状 態を予防又は治療する方法であって、前記成長因子が、1以上のシステイン残基 が分子間又は分子内ジスルフィド結合を形成することができないアミノ酸で置き 換えられたものである方法。 5.前記成長因子がFGF-1である請求項4記載の方法。 6.前記FGF-1が、ECGFα、aFGF 及び ECGFβである、請求項5記載の方法。 7.前記FGF-1が、ECGFαである請求項6記載の方法。 8.前記FGF-1が、ECGFβである請求項6記載の方法。 9.前記FGF-1が、全長FGF-1である請求項6 記載の方法。 10.前記FGF-1がdes 1-21 FGF-1である請求項6記載の方法。 11.治療を必要とするヒトに骨膜の骨成長を刺激するのに有効な量のFGF-1を投 与することを含む、ヒトの骨粗鬆症の治療方法であって、前記FGF-1が、1以上 のシステイン残基が分子間又は分子内ジスルフィド結合を形成することができな いアミノ酸で置き換えられたものである方法。 12.治療を必要とするヒトに、歯の病的状態の部位に局所的に、膜の骨成長を刺 激するのに有効な量のFGF-1を投与することを含む、ヒトの歯の病理学的状態の 治療方法であって、前記FGF-1が、1以上のシステイン残基が分子間又は分子内 ジスルフィド結合を形成することができないアミノ酸で置き換えられたものであ る方法。 13.前記骨組織を含む病理学的状態が、骨粗鬆症、変形性骨炎、歯周囲病、骨折 及び骨の欠陥からなる群から選ばれるものである請求項4記載の方法。 14.前記骨組織を含む病理学的状態が、骨折である請求項13記載の方法。 15.さらにヘパリン又はヘパリンフラグメントの投与を含む、請求項4記載の方 法。 16.さらにヘパリン又はヘパリンフラグメントの投与を含む、請求項11記載の方 法。 17.さらにヘパリン又はヘパリンフラグメントの投与を含む、請求項12記載の方 法。 18.前記繊維芽細胞成長因子がFGF-1である請求項13記載の方法。 19.前記繊維芽細胞成長因子がFGF-1である請求項14記載の方法。 20.前記繊維芽細胞成長因子がFGF-1である請求項15記載の方法。 21.さらにヘパリン又はヘパリンフラグメントの投与を含む、請求項19記載の方 法。 22.2個のシステイン残基が、分子間又は分子内ジスルフィド結合を形成するこ とができないアミノ酸で置き換えられている請求項4記載の方法。 23.全3個のシステイン残基が、分子間又は分子内ジスルフィド結合を形成する ことができないアミノ酸で置き換えられている請求項4記載の方法。 24.分子間又は分子内ジスルフィド結合を形成することができないアミノ酸が、 セリンである請求項4記載の方法。 25.前記の量が、一注入当たり約0.5〜約 500 ngである請求項19記載の方法。 26.前記の量が、一注入当たり約 50 ngである請求項25記載の方法。 27.前記の量が、一注入当たり約2 約 4 μg/kgである請求項19記載の方法。 28.前記の量が、28日間で、少なくとも約0.2 mg/kg/日である請求項11記載の方 法。 29.前記投与が、フィブリンシーラントの投与である請求項19記載の方法。 30.前記担体が、失活させた骨粉末、ヒドロキシアパタイト、及びフィブリンシ ーラントからなる群から選ばれる1種以上のものである、請求項1記載の医薬組 成物。 31.前記担体がフィブリンシーラントである、請求項30記載の医薬組成物。 32.ヒトにおける骨組織を含む病理学的状態の治療に用いる薬剤の製造へのヒト 繊維芽細胞成長因子の使用であって、前記成長因子が、骨膜の骨の成長を刺激す るのに有効な量であり、1以上のシステイン残基が、分子間又は分子内ジスルフ ィド結合を形成することができないアミノ酸で置き換えられている前記使用。
